Abstract

mRNA vaccines were highly effective in response to the COVID-19 pandemic, making them an attractive platform to address cancers and other infectious diseases. Many new mRNA vaccines in development are multivalent, which represents a difficulty for the standard assays commonly used to characterize the critical quality attributes of monovalent formulations. Here, we present a multiplexed analytical tool with nucleic acid microarray technology using the VaxArray platform that measures the identity and quantity of mono- and multivalent mixtures of naked mRNA and mRNA encapsulated in lipid nanoparticle formulations in under 2 h without any additional preparation steps, such as extraction or RT-PCR. Using a quadrivalent mixture of encapsulated mRNA constructs that encode for four unique proteins in a vaccine formulation, the VaxArray mRNA assay was demonstrated to be highly specific for each mRNA with sensitivity < 1 µg/mL. The quantification of individual mRNAs within the lipid nanoparticle mixture resulted in a precision of ≤10% RSD and an accuracy of 100 ± 9%. [ABSTRACT FROM AUTHOR]

13. Industrialization potential analysis of citrus waste in Colombia.

Abstract

Citrus production in Colombia is divided into six productive centers, with a total production of 1,450 thousand tons in 2021. The greater entrepreneurial activity stands out in the West and a lower level in the South, which generates 230 thousand tons per year of citrus waste, posing challenges in terms of pollution and wet/semi-solid waste management. This study evaluates the industrial potential of these waste products for strengthening the national citrus industry, considering three aspects: raw materials availability, technological opportunities, and market opportunities. The results indicate that Colombia has a robust citrus production, with an emphasis on oranges, mandarins, and lemons, and significant potential in the industrialization of the peel, which constitutes between 40% and 60% of the total fruit. To industrialization were identified six key research areas of citrus waste in terms of technological opportunities. Regarding market opportunities, there is observed growth both nationally and internationally in products derived from citrus waste, including essential oils, terpenes, dietary fiber, pectin, citric acid, and biofuels. Colombia is considered a country with great potential for the industrialization of citrus waste, supported by its abundant raw materials, promising technological opportunities, and a growing market. However, improvements in infrastructure, investment in research and development, and the promotion of entrepreneurial culture in the citrus sector are needed to fully capitalize on this opportunity. Abstract

The adoption of more sedentary residential practices is a long process in which groups shift in size, permanence, makeup, and even location in response to the social, demographic, ecological, and subsistence pressures associated with settling down. Coastal peoples living in the Southeast U.S. are among the first to adopt more sedentary lifeways in North America as evidenced by their creation of dozens of large circular or arcing shell middens during the end of the Middle Holocene and start of the Late Holocene. Relying on material culture studies and AMS radiocarbon data refined using Bayesian statistical modeling, we investigate the sequential establishment and abandonment of three village sites built over more than 300 years and located only a few kilometers apart. These studies reveal important insights into the process of adopting more sedentary practices in the region and how local communities adapted to associated pressures. [ABSTRACT FROM AUTHOR]

Abstract

BACKGROUND: Young Indigenous people, particularly those involved in the child welfare system, those entrenched in substance use and those living with HIV or hepatitis C, are dying prematurely. We report mortality rates among young Indigenous people who use drugs in British Columbia and explore predictors of mortality over time. METHODS: We analyzed data collected every 6 months between 2003 and 2014 by the Cedar Project, a prospective cohort study involving young Indigenous people who use illicit drugs in Vancouver and Prince George, BC. We calculated age-standardized mortality ratios using Indigenous and Canadian reference populations. We identified predictors of mortality using time-dependent Cox proportional hazard regression. RESULTS: Among 610 participants, 40 died between 2003 and 2014, yielding a mortality rate of 670 per 100 000 person-years. Young Indigenous people who used drugs were 12.9 (95% confidence interval [CI] 9.2-17.5) times more likely to die than all Canadians the same age and were 7.8 (95% CI 5.6-10.6) times more likely to die than Indigenous people with Status in BC. Young women and those using drugs by injection were most affected. The leading causes of death were overdose (n = 15 [38%]), illness (n = 11 [28%]) and suicide (n = 5 [12%]). Predictors of mortality included having hepatitis C at baseline (adjusted hazard ratio [HR] 2.76, 95% CI 1.47-5.16), previous attempted suicide (adjusted HR 1.88, 95% CI 1.01-3.50) and recent overdose (adjusted HR 2.85, 95% CI 1.00-8.09). INTERPRETATION: Young Indigenous people using drugs in BC are dying at an alarming rate, particularly young women and those using injection drugs. In this article, two in-service English teachers and two preservice English teachers, who participated in a shared mentoring relationship during the student-teaching semester, share their experiences, conflicts, and how they were resolved. The authors provide first-person narratives of all four teachers and suggestions for improving mentoring experiences for both mentor teachers and student teachers. Important issues addressed include territoriality of classroom spaces, students, decision making, and time. The authors provide suggestions for improving communication, use of shared spaces, and planning.

28. Cognitive Dissonance Among Chinese Gamblers: Cultural Beliefs Versus Gambling Behavior

Abstract

This study examined the extent to which cognitive dissonance exists among Chinese gamblers as a consequence of gambling while holding negative attitudes toward gambling, which are inherent in China’s traditional cultural values. Using the behavioral variable of actual gambling and an attitudinal variable of negative beliefs about gambling, a third, practical measure of cognitive dissonance was developed. By using questionnaires completed by 200 adult Chinese respondents, these measures were examined in relation to a set of relevant independent variables frequently tested in the gambling literature. Cognitive dissonance was expected to have significant negative correlations with traditional Chinese values and family support, and a significant positive correlation with neuroticism. Cognitive dissonance was also expected to be negatively correlated with two personal outcomes, i.e. self-actualization and life satisfaction. The results supported these hypotheses, which confirmed the validity of the new measures, and that cognitive dissonance does indeed exist among Chinese gamblers. The results also found that Chinese gamblers, even though they do gamble, also hold negative attitudes toward gambling, with more cognitive dissonance strongly associated with higher levels of gambling. This provides a new perspective on studying Chinese gambling, and offers a possible strategy to help pathological gamblers, for example, by advising them that their negative beliefs about gambling reflect the positive moral values of their society’s traditional culture, an approach that may be effective in reducing excessive gambling.

Abstract

Vaccines contain residual DNA derived from the cells used to produce them. As part of our investigation to assess the risk of this cellular DNA, we are developing a quantitative in vivo assay to assess the oncogenicity of DNA. In an earlier study, we had generated expression plasmids for two oncogenes - human activated T24-H-ras and murine c-myc - and had shown that these two plasmids, pMSV-T24-H-ras and pMSV-c-myc, could act in concert to induce tumors in mice, although the efficiency was low. In this study, we took two approaches to increase the oncogenic efficiency: 1) both oncogene-expression cassettes were placed on the same plasmid; 2) transfection facilitators, which increase DNA uptake and expression in vitro, were tested. The dual-expression plasmid, pMSV-T24-H-ras/MSV-c-myc, is about 20-fold more efficient at tumor induction in newborn NIH Swiss mice than the separate expression plasmids, with tumors being induced with 1 µg of the dual-expression plasmid DNA. However, none of the transfection facilitators tested increased the efficiency of tumor induction. Based on these data, the dual-expression plasmid pMSV-T24-H-ras/MSV-c-myc will be used as the positive control to develop a sensitive and quantitative animal assay that can be used to assess the oncogenic activity of DNA.

Abstract

The COVID-19 pandemic highlighted mRNA as a promising platform for vaccines and therapeutics. Many of the analytical tools used to characterize the critical quality attributes of mRNA are inherently singleplex and are not necessarily optimal from a labor and cost perspective. Here, we demonstrate the feasibility of a multiplexed platform (VaxArray) for efficient identity verification and concentration determination for both monovalent and multivalent mRNA formulations. A model system comprising mRNA constructs for influenza hemagglutinin and neuraminidase was used to characterize the analytical performance metrics for a VaxArray mRNA assay. The assay presented herein had a time to result of less than 2 h, required no PCR-based amplification nor extraction of mRNA from lipid nanoparticles, and exhibited high construct specificity that enabled application to the bivalent mixture. The sensitivity for influenza hemagglutinin and neuraminidase mRNA was sub-µg/mL, which is vaccine-relevant, and the average accuracy (%recovery of a check standard) and precision were 104 ± 2% and 9 ± 2%, respectively. Abstract

Archaeological investigations of Mississippian platform mounds have traditionally required invasive excavation or coring. Excavations are damaging to sites, and in many cases, invasive or destructive research methods on Native American mounds are forbidden or inappropriate. Non‐invasive geophysical investigations avoid these issues but have their own challenges in terms of resolving the interior of the mound, particularly if electrically conductive materials, such as clay, are present. Here, we present a multi‐method non‐invasive geophysical approach using ground penetrating radar, electrical resistivity tomography, time‐domain induced polarization, and electrical resistance mapping to study the Mississippian platform mound at Snow's Bend (1TU2/3), a late Moundville II/III (ca. AD 1300 to 1520) site located near Moundville, Alabama. From our data, we interpreted at least two construction stages and found indications of remnants of summit architecture on each. The final, as well as earlier, construction stage of the mound had a two‐tier summit with a lower platform in the northern half of the mound. Summit buildings were identified on the lower platforms of each mound stage. We acknowledge that there is inherent uncertainty with any non‐invasive approach, but demonstrate the capabilities of geophysics for new understandings of the life‐histories of Mississippian platform mounds.
