21. Effects of temperature on mortality of quagga mussels (Dreissena bugensis) exposed to potassium chloride and copper-based molluscicides in high conductivity waters.

Abstract

Dreissenid mussels (quagga and zebra mussels, Dreissena bugensis and polymorpha) continue to spread throughout inland waterbodies of North America. Although there are considerable management and mitigation efforts underway to prevent infestation of new waterbodies, there have been relatively few successful efforts to eradicate mussels once detected due to a lack of cost-effective treatments. Two potentially cost-effective molluscicidal agents, potassium chloride (KCl, also called muriate of potash) and copper (formulated as Earthtec QZ®), have successfully treated infested waters, but have varying efficacy under different water quality and temperature conditions, and to date have only been applied in moderate conductivity waters (defined here as < 400 µS/cm) common throughout North America. Natural resource managers need information on the efficacy of these chemicals in high conductivity waters more typical of riverine systems and arid climates, in addition to the range of water temperatures typically encountered in the field. We evaluated the doseresponse of quagga mussels to KCl and Earthtec QZ® in high conductivity water (≈ 700-1,400 µS/cm) across a range of temperatures (10, 18, 22 °C) to determine appropriate doses and treatment duration resulting in 100% mortality. Our data indicate that treatments in cool temperatures (i.e., 10 °C) may be challenging for eradication efforts in high conductivity waters because none of the KCl treatments and only one copper treatment resulted in 100% mortality within the experimental period. However, both KCl (> 200 ppm) and copper (120 and 180 ppb) were consistently able to induce 100% mortality in quagga mussels in warmer temperatures (i.e., = 18 °C) and thus may be good candidates for field scale application in high conductivity waters. The results of this study indicate that field-scale application should strongly consider environmental conditions during the period of application when selecting dosages and treatment duration. Abstract

Human glyoxylate reductase/hydroxypyruvate reductase (GRHPR) is a D-2-hydroxyacid dehydrogenase found predominantly within the liver, and is essential for the removal of the reactive metabolic product glyoxylate. Primary hyperoxaluria type 2 (PH2) disease, which is characterised by increased urinary oxalate and L-glycerate levels leading to calcium oxalate deposition and decreased renal function, is caused by mutations in the gene for GRHPR. In this study the first crystal structures of the human GRHPR enzyme are reported. Structures of apo, binary and ternary forms of GRHPR are reported, the latter being the first structure of a true ternary complex of an enzyme from the D-2-hydroxyacid dehydrogenase family.

Abstract

Fish movement in freshwater streams is often tied to patterns of flow, especially when the stream is intermittent, flashy, or some combination thereof. Relationships between flow and fish movement are exacerbated in small urban streams in impervious surface‐dominated watersheds that lead to extreme flow conditions. Here we examined the response of Creek Chub (Semotilus atromaculatus), a common pioneer fish species in eastern North America, to flow events of various sizes in a small intermittent urban headwater stream. Our primary goals were to determine (1) the level of Creek Chub movement occurring in an intermittent headwater stream, (2) the influence of flow on movement direction and magnitude, (3) the degree of variation in individual movement behaviour, and (4) mortality rates in the system. The study used PIT tagging and telemetry surveys to monitor Creek Chub movement over a six‐month period. We found that individual Creek Chub in our study system were generally mobile (71%), but usually moved short distances only; however, a few individuals moved much farther, with a maximum single movement of 521 m. Moderate or major flow events increased the probability and magnitude of Creek Chub movement, but there was no detectable asymmetry towards upstream or downstream directionality. Movement was variable among individual fish and fish were able to respond to large and small flow events to move among habitats. Finally, we observed relatively high, but episodic, mortality in our study site, potentially related to pollution events. Evidence of elevated mortality points to the potential importance of movement in highly disturbed streams, as it may enable recolonization after local extirpation. [ABSTRACT FROM AUTHOR]

33. River connectivity increases the diversity of fish communities in gravel pit lakes.

Abstract

Objective: Off‐channel floodplain features are important components of a river system that provide habitat and boost regional species pools, but river–floodplain connectivity is disrupted by anthropogenic activities. The Campbell Lakes in Harrison, Ohio, are formerly isolated gravel pits along the Whitewater River and are connected to the river through flooding‐related erosion. The lakes were first stocked with sport fish by Great Parks of Hamilton County to promote recreational fishing, but stocking ended in 2019 due to frequent connection with the Whitewater River during high flow. We explored how the fish community composition of the Campbell Lakes reflects increasing connectivity with the Whitewater River and among the lakes. Methods: Using historical aerial photographs and river discharge estimates, we established an approximate timeline of permanent connectivity and potential breaches between the lakes and the Whitewater River from 2000 to 2020. Fish community data collected via electrofishing surveys of the Campbell Lakes in 2004, 2007, 2012, and 2020 were compared to data from surveys of the Whitewater and Great Miami rivers in 1995 and 2013. Result: The lakes were dominated by centrarchid species in early surveys but showed increased species and family richness with time. Based on an analysis of similarity and nonmetric multidimensional scaling, the lakes' fish communities differed significantly among years and gradually increased in resemblance to the river communities. Conclusion: Our results suggest that the Campbell Lakes fish communities are broadening to include taxa from the Whitewater and Great Miami River communities and are shifting from lentic to lotic species. The change in fish community assemblages in the Campbell Lakes demonstrates the potential for gravel pit lakes to serve as artificial floodplain features that can enhance river–floodplain connectivity. Impact StatementAs four gravel pit lakes became increasingly connected with an adjacent river through temporary flooding and permanent channels, fish community assemblages in the lakes diversified and transitioned from stocked lentic species to riverine backwater species. Abstract

The nucleotide-binding oligomerization domain (NOD)-like receptor (NLR) protein 3 (NLRP3) inflammasome pathway is believed to mediate chronic inflammation in diabetic retinopathy (DR); however, its impact on the progression of DR remains to be elucidated. Therefore, the primary aim of this pilot study was to determine whether systemic inflammasome biomarkers interleukin (IL)-1β and IL-18 can be used to predict DR progression. DR screening results were analyzed against weight, level of glycated hemoglobin (HbA1c), and plasma levels of inflammasome biomarkers (IL-1β and IL-18), as well as general inflammation markers (C-reactive protein (CRP), IL-6, IL-8, tumor necrosis factor-alpha (TNF-α), and vascular endothelial growth factor (VEGF)) in patients with type 2 diabetes at baseline and 1 year post-bariatric surgery. Cross-sectional analysis demonstrated that weight, HbA1c, CRP, and IL-18 did not correlate with DR severity. The progressed group showed a higher relative change in IL-18 and CRP levels compared to the stable and regressed groups. Furthermore, relative changes in plasma CRP levels correlated with those of IL-18. Although further validation with larger cohorts is necessary, this pilot study supports the hypothesis that systemic inflammasome activation is associated with DR progression. [ABSTRACT FROM AUTHOR]

Abstract

This systematic review analyses research that introduces commercial design applications that could be adopted for suicide prevention in homes. Furthermore, this literature review captures social, spatial and biophilic design methods to improve wellness in homes using environmental design psychology. Safety and human wellness frame this spatial design research that examines means and access restriction to improve home safety and prevent suicides. Suicide is a growing phenomenon that deserves specific attention to how environments can impact or restrict events. There is a substantial evidence base to evaluate suicide prevention methods used in high-risk environments of health and healing environments, workplaces and incarceration facilities. This review outlines design methods using spatial arrangement and material choices to improve human wellness in homes. The effects of biochemical reactions, such as those studied in toxicology, and stress are considered in this research to suggest material choices and applications in design to improve mental health in homes. Spatial designs for suicide prevention can guide various prevention measures, such as adopting means and access restriction and environmental design methods for wellness and considering impacts during lockdown periods. Environmental design psychology research supplies evidence for improved spatial arrangements in homes, with evidence showing that design applications can restore and improve mental health. This systematic review shows evidence for planning methods to prevent suicides considering both means and access restriction with considerable biochemical impacts from design. Design methods discovered by this systematic review will be considered for future studies and used within economic modelling to demonstrate design guidelines that improve wellbeing and support existing suicide prevention methods for Australian homes. Abstract

Developing simple methods to silence antisense oligonucleotides (ASOs) using photocages opens up the possibility of precise regulation of biological systems. Here, we have developed a photocaging strategy based on 'handcuffing' two ASOs to a protein. Silencing was achieved by divalent binding of two terminally photocleavable biotin-modified ASOs to a single streptavidin. These 'handcuffed' oligonucleotides showed a drastic reduction in gene knockdown activity in cell-free protein synthesis and were unlocked through illumination, regaining full activity. 49. Relationships between Body Composition and Fundamental Movement Skills among Children and Adolescents

Abstract

The purpose of this study was to examine associations of fundamental movement skills (FMS) with measures of body composition. among children and adolescents. Secondary analyses of cross-sectional data collected from 4,363 children and adolescents in Grades 4, 6, 8, and 10 as part of the 1997 New South Wales Schools Fitness and Physical Activity Survey were conducted. Six FMS (run, vertical jump, throw, catch, kick, and strike) were assessed by observation. Height and weight (used to calculate body mass index; BMI) and waist circumference were directly measured. Results indicated that the children's and adolescents' ability to perform FMS was significantly related to BMI and waist circumference. Specifically, BMI and waist circumference were significant predictors for FMS in six of the eight demographic groups. Adjusted odds ratios revealed that overweight boys and girls in all grades were less likely to possess high levels of FMS and more likely to possess low levels of FMS than those who weren't overweight. When FMS were partitioned into locomotor and object-control skills, non-overweight buys and girls in each grade were two to there times more likely to possess more advanced locomotor skills than overweight boys and girls. However, for object-control skills, the only demographic groups in which non-overweight students possessed a greater number of advanced skills than overweight students were boys in Grades 6 and 10. There appear to be significant and important association between performance of locomotor skills and weight status among children and adolescents. This would suggest that intervention strategies to prevent unhealthy weight gain among children and youth might usefully include increasing proficiency of locomotor skills as a key component.
