Abstract

Turbulent jet ignition is a pre-chamber ignition system for an otherwise standard gasoline spark ignition engine. Turbulent jet ignition works by injecting chemical active turbulent jets to initiate combustion in a premixed fuel/air mixture. The main advantage of turbulent jet ignition is its ability to ignite and burn completely very lean fuel/air mixtures in the main chamber charge. This occurs with a very fast burn rate due to the widely distributed ignition sites that consume the main charge rapidly. Rapid combustion of lean mixtures leads to lower exhaust emissions due to more complete combustion at lower combustion temperature. The purpose of the paper is to study the combustion characteristics of gasoline, ethanol, and wet ethanol when operated with the pre-chamber combustion system and the ability of the pre-chamber ignition to extend the lean-burn limits of such fuels. The combustion and heat release process was analyzed and exhaust emissions measured. Results show that the effect of turbulent jet ignition system on the lean-burn limit and exhaust emissions varied with fuels. The lean limit was extended by using fueled pre-chamber furthest, to λ = 1.71 with gasoline, followed by λ = 1.77 with wet ethanol and λ = 1.9 with ethanol. NOx emissions were significantly reduced with increased lambda for each fuel under stable combustion conditions. For ethanol, at maximum lean limit lambda 1.9, the NOx emissions were almost negligible due to lower combustion temperature. [ABSTRACT FROM AUTHOR]

Abstract

The use of molecular typing methods for defining human leukocyte antigen (HLA) Class I and Class II alleles is now commonplace. Molecular methods offer a greater accuracy than traditional serological methods (1-5) which is reflected in many disease and transplantation studies (6-9). This chapter describes a polymerase chain reaction sequence-specific primer (PCR-SSP) typing system that is applicable to the identification of all HLA class I and class II alleles, as well as alleles from non-HLA loci (10,11) . 31. Cw * 1701 defines a divergent African HLA-C allelic lineage.

Abstract

The complete sequence of a new HLA-C allele, Cw * 1701, was determined from a South African Zulu individual. Unique features that distinguish Cw * 1701 from other HLA-C alleles include multiple point substitutions and an 18 nucleotide insertion in exon 5, which encodes the transmembrane domain. In a phylogenetic analysis of HLA-C sequences, Cw * 1701 forms a third, distinct allelic lineage. A comparison of the transmembrane domain of Cw * 1701 with other HLA-B and -C alleles reveals that duplications and deletions have been common in the evolution of these loci. A polymerase chain reaction based typing method was used to determine the distribution of this unusual allele in human populations. In contrast to the other two lineages of HLA-C alleles, the Cw * 17 lineage is found at high frequencies only in populations of African descent. Abstract

Recurrent respiratory papillomatosis (RRP) is characterised by multiple laryngeal papillomas. Left untreated, the lesions enlarge, spread, and endanger the airway. Medical treatments are unsatisfactory, and repeated surgery remains the mainstay of therapy. RRP is caused by human papillomavirus (HPV) infection. However, since oral HPV infection is common and RRP is rare, other host and/or viral factors may contribute to pathogenesis. In an attempt to identify such factors, we have investigated 60 patients. The patients were HLA class I, II, and tumor necrosis factor TNF typed by sequence-specific primer PCR, and the results compared to those for 554 healthy controls by using Fisher's exact test. Peripheral blood mononuclear cell proliferative responses of 25 controls and 10 patients to HPV-11 L1 virus-like particles (VLP) were compared. Short-term VLP-specific T-cell lines were established, and recognition of L1 was analyzed. Finally, the L1 open reading frames of HPV isolates from four patients were sequenced. Susceptibility to RRP was associated with HLA DRB1[sup *]0301 (33 of 60 patients versus 136 of 554 controls, P < 0.0001). The three most severely affected patients were homozygous for this allele. A range of T-cell proliferative responses to HPV-11 VLP were observed in DRBl[sup *]0301-positive healthy donors which were comparable to those in DRBl[sup *]0301-negative controls. Individuals with juvenile-onset RRP also mounted a range of VLP responses, and their magnitude was negatively correlated with the clinical staging score (P = 0.012 by the Spearman rank correlation). DRBl[sup *]0301-positive patients who responded to L1 recognized the same epitope as did matched controls and produced similar cytokines. Sequencing of clinical isolates excluded the possibility that nonresponsiveness was the result of mutation(s) in L1. [ABSTRACT FROM AUTHOR]

Abstract

HLA typing by sequence-specific primers (PCR-SSP) is a commonly used technique in HLA typing in which multiple pairs of cis-located allele-specific primers are used to determine the alleles present in a given DNA sample. Although the technique is around two decades old, it still offers a relatively straight forward way of typing and has the benefit of using commonly available laboratory equipment. Here we describe the background of the PCR-SSP, how to design and validate reactions, and common problems that arise.
