Light intensity and spectrum have a direct effect on the growth and development of plants and on the qualitative characteristics of their productions. LED technology seems to offer interesting prospects for its possible use in confined environments (growth chambers, bioreactors, greenhouses, etc). On the basis of these considerations, we tested the effects on the in vitro growth and development of micropropagated pineapple (Ananas comosus (L.) Merr.) plantlets using four different light spectra: (1) fluorescence light (FL—control); (2) white LED (WL—blue 20%, green 36%, red 38%, far red 6%, and UV 1%); (3) predominantly red LED (RL—blue 12%, green 19%, red 61%, and far red 8%); (4) Red/Blue LED (RL/BL—67% red and 33% blue) and two light intensities: 80 and 100 μmol m−2s−1 (16/8 h light/dark photoperiod). A. comosus showed a different morphogenetic response to the light spectra and their intensity and to their interaction. Among the LED lights to date tested, at 80 μmol m−2s−1 the best results on shoot multiplication were obtained under the RL light and to a lesser extent under the WL. This last treatment resulted also in a significant improvement in shoot quality in terms of secondary shoot dry weight and total chlorophyll concentration when compared to all the other light spectra tested. Plant tissue cultures depend entirely upon artificial light sources for illumination. The illumination should provide light in the appropriate regions of the electromagnetic spectrum for photomorphogenic responses and photosynthetic metabolism. Controlling light quality, irradiances and photoperiod enables the production of plants with desired characteristics. Moreover, significant money savings may be achieved using both more appropriate and less consuming energy lamps. In this review, the attention will be focused on the effects of light characteristics and plant growth regulators on shoot proliferation, the main process in in vitro propagation. The effects of the light spectrum on the balance of endogenous growth regulators will also be presented. For each light spectrum, the effects on proliferation but also on plantlet quality, i.e., shoot length, fresh and dry weight and photosynthesis, have been also analyzed. Even if a huge amount of literature is available on the effects of light on in vitro proliferation, the results are often conflicting. In fact, a lot of exogenous and endogenous factors, but also the lack of a common protocol, make it difficult to choose the most effective light spectrum for each of the large number of species. Nowadays, Light-Emitting Diodes (LEDs) have been demonstrated to offer interesting prospects for use in plant lighting designs in controlled environment agriculture (greenhouses) and growth chambers for in vitro cultures. Light is an electromagnetic radiation that occurs in a narrow range of over an extremely wide range of wavelengths, from gamma rays with wavelengths to radio waves measured in meters. Light detected by human eyes and plant photoreceptors is a group of wavelengths from about 700 nanometers (nm) for red light down to about 400 nm for violet light: the visible light. However, anyone knows that light is necessary to the aesthetic appreciation of the visual world, and through the sense of sight, light is a primary tool for perceiving the world and communicating within it. Alzheimer's dementia is a neurodegenerative disease that affects the elderly population and causes memory impairment and cognitive deficit. Manifestation of this disease is associated to acetylcholine decrease; thus, Cholinesterase inhibition is the main therapeutic strategy for the treatment of Alzheimer's disease. In the present study, a series of aporphinoid alkaloids were tested as potential acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitors in vitro. Alkaloids liriodenine (3) and cassythicine (10) were the best inhibitors of both cholinesterases with IC50 values lower than 10 Î?M. In addition, these alkaloids demonstrated better inhibition of BChE than reference drug galantamine. In addition, some alkaloids showed selective inhibition. Laurotetatine clorhydrate (13) selectively inhibit AChE over BChE. On the contrary, pachyconfine (7) interacted more efficiently with BChE active site. Molecular modelling studies were performed in order to illustrate key interactions between most active compounds and the enzymes and to explain their selectivity. These studies reveal that the benzodioxole moiety exhibits strong interactions due to hydrogen bonds that form with the Glu201 (AChE) and Tyr440 (BChE) residues, which is reflected in the IC50 values. The term biorefinery is related to the sustainable production of value-added bioproducts and bioenergy from biomass. Esters from fatty acids are important compounds synthesized from by-products of the oleochemical industry. In agreement with the biorefinery concept, it is important to search for catalysts that reduce the consumption of energy and water, using moderate operation conditions and low reaction times. In this work, response surface methodology (RSM) was used to optimize the enzymatic synthesis of pentyl oleate using Candida antarctica lipase B (CALB) immobilized on a polyethylene-aluminum structured support. A factorial design was employed to evaluate the effects of several parameters on the ester yield. To obtain a model with a good fit, an approach to reaction mechanism and enzyme kinetics was taken into consideration. Experimental findings were correlated and explained using equations of a ping-pong bi-bi kinetic model and considering the inhibitory effects of both substrates. The developed model was consistent with the experimental data predicting an increase in pentyl oleate production with increasing temperature and a decrease with higher oleic acid amounts and alcohol to acid molar ratios. Abstract Giant reed (Arundo donax L.) is a warm-season, fast growing lignocellulosic crop suitable to Mediterranean and semi-arid environments. However, its sterility limits a cost-effective propagation along with environmental impacts associated with rhizome processing. In the present study, the effects of intrinsic (node size and node chemical composition) and extrinsic factors (node pretreatment with growth regulators) on root and shoot development were investigated under different excision time of node-cuttings in a controlled and open field conditions. Results indicated that nutrients availability of the propagation material plays a key role in rooting development. To enhance the success of giant reed propagation by single node stem-cuttings, the best period for node excision is July in open field trial, followed by the late spring (May). Nodes whose diameter exceed 2 cm resulted in higher rooting and plantlets development even in the less favorable excision dates. However, when big nodes are adopted in controlled environments even winter periods are eligible, being particularly useful for spring open field transplants of rooted plantlets. Among the growth regulators used in the present study, indole-3-butyric acid - IBA (in controlled environment) resulted effective in promoting rooting particularly in the worse excision date (April); in open field, IBA significantly improved rooting in the excision dates of February and May in the big nodes, and only in February in the small ones. NAA (1-naphthaleneacetic acid) was almost ineffective in enhancing rooting as compared to the control. The study aimed to evaluate, in a marginal semi-arid Mediterranean agro-ecosystem (Sicily-Italy), the effects of arbuscular mycorrhizal fungi (AMF) inoculation and raw olive mill wastewater (OMW) (40 and 80 m3 ha−1) on forage (durum wheat-snail medick intercropping) yield, and grain production of broad bean and chickpea. AMF inoculation significantly increased (+13.6%) forage dry biomass and durum wheat nitrogen (+22.8%) and phosphorus (+32.5%) uptake. AMF inoculation, significantly promoted broad bean phosphorus uptake (+11.5%) and root nodule number (+13.9%) in the absence of OMW. OMW spreading reduced weeds in the forage (−31.3%), root nodule number (−29.7%) and dry weight (−22.7%) in broad bean. OMW also significantly increased snail medick dry biomass (+19.3%) as compared to control treatments (0, 40 and 80 m3 H2O ha−1, average production 361 g m−2), and broad bean grain yield with a production of 2.46 ± 0.12 and 1.94 ± 0.09 Mg ha−1 with and without OMW, respectively. During the experiment AMF colonization was not affected by OMW volumes. The results obtained showed that in a marginal Mediterranean agro-ecosystem: 1) OMW, notwithstanding spreading volumes, is a valuable amendment to maximize legume yield while 2) AMF inoculation is a valuable practice to improve biomass production and N and P uptake in wheat. A series of thirty-three alkynyl and β-ketophosphonates were evaluated for their in vitro acetyl- and butyryl-cholinesterase (AChE and BChE) inhibitory activities using Ellman’s spectrophotometric method. None of the examined compounds inhibited AChE activity at tested concentrations while twenty-nine of them showed significant and selective inhibition of BChE with IC 50 values between 38.60 µM and 0.04 µM. In addition, structure-activity relationships were discussed. The most effective inhibitors were the dibutyl o -methoxyphenyl alkynylphosphonate 3dc and dibutyl o -methoxyphenyl β-ketophosphonate 4dc . Activities of most potent compounds were also compared with a commercial organophosphorus compound. These results could inspire the design of new inhibitors with stronger activity against BChE. Candida antarctica lipase B was immobilized for the first time (at the author's knowledge) onto linear low density polyethylene (LLDPE) films. Polymer films were previously bonded to a commercial aluminum sheet using a simple support preparation method. Biocatalyst performance was evaluated in penthyl oleate synthesis at room temperature. Two different catalyst geometries were tested and compared: one aluminum‐polyethylene 50 mm × 50 mm foil (50CAT) or near 5 mm × 5 mm aluminum‐polyethylene foils (5CAT). The obtained results demonstrate that the biocatalyst obtained with 50 mm × 50 mm aluminum‐polyethylene foil or 50CAT is reusable in up to 7 cycles, easy to separate from reaction products, and economical in comparison with commercial Novozym 435. Novel and economical CALB/LLDPE/Al biocatalyst is an attractive alternative for possible applications in a continuous monolithic reactor and future industrial scaling up. [ABSTRACT FROM AUTHOR]

Carob is a valuable leguminous species for its productions but also for forestation of arid and degraded areas threatened by soil erosion and desertification processes. However, large-scale cultivation of this species is hampered by the difficult propagation by cuttings or seeds. Researches to evaluate genotype tolerance to saline and drought stress during germination and first plant establishment, are also inhibited by the scarce seed germination. In this study, carob seeds, from wild and domesticated carob genotypes collected in a representative area of the Mediterranean basin (Sicily, Italy), were subjected to germination tests under isotonic solutions of polyethylene glycol (PEG) and sodium chloride (NaCl) at −0.5, −1.0, −1.5 MPa. Before germination tests, a 20 min pre-treatment with 96% sulphuric acid was necessary to overcome seed coat dormancy which does not permit germination. Differences in drought and salinity tolerance were determined in the different genotypes during germination. All genotypes exhibited a higher sensitivity to drought stress as compared to salinity stress. The observed differences in some genetically inherited germination characteristics (i.e. base Ψb(50)s and hydrotimes) suggest genetic differences even in seeds of individual old trees situated in close areas, as previously found in other countries of the Mediterranean basin. A greater sensitivity to stress was determined during early radicle growth, an essential feature for early plant establishment in marginal dry areas. In this paper, a protocol to test germination under simulated stress conditions (drought and salinity stress) was established. Including in the analysis other parameters representative of early embryo growth (i.e. radicle elongation), this protocol may be useful to select genotypes for breeding programs towards the selection of tolerant cultivars or rootstocks. The agronomic use of olive mill wastewaters (OMWs) is limited by some constraints, such as high salinity, acidity, and phenolic compounds that exert phytotoxic and antimicrobial effects. Many researches have been focused on OMW polyphenol effects on germination, but only few on the combined effects of salinity and acidity. In this paper, on Lolium multiflorum Lam., the effects on germination of pH and osmotic potentials similar or higher to those found in OMWs have been investigated. The studied salinity and pH levels did not affect final germination of L. multiflorum , but in some cultivars they significantly enhanced T 50 and consequently reduced initial root growth. Thus, in this species moderately tolerant to salinity, the reduction of germination determined by OMWs cannot be attributed to the osmotic stress and/or to the low pH, but mainly to polyphenols. This research emphasizes the possibility of obtaining good crop establishments after spreading OMWs on soil, choosing herbaceous species tolerant to salinity once polyphenols have been inactivated. Giant reed (Arundo donax L.), a promising energy crop, is vegetatively-propagated from fragments of stems and rhizomes. This may limit large-scale cultivation, since it is time-consuming and involves considerable cost and effort. Tissue culture is an alternative to conventional methods of vegetative propagation and may represent a useful tool for large-scale propagation of plants for biomass production programmes. This report describes a protocol for the large-scale in vitro propagation of giant reed by adventitious bud formation. Stem nodes with dormant buds proved to be the most effective to initiate in vitro cultures giving the highest percentage of differentiated shoots (77%) compared to the other plant fractions tested. A sterilisation procedure using 5 g 1-1 HgCl2 enabled the production of sterile explants. Moreover, early results indicated that late Autumn excision dates not only gave a higher percentage of well-developed shoots ( > 80%), but also a lower level of bacterial contamination (15 - 20%). 6-Benzylaminopurine (BAP) at 3.0 mg 1-1 was most effective in promoting shoot multiplication when added to a basal medium containing Murashige and Skoog (MS) macro- and micro-nutrients, Morel's vitamins, 30 g 1 sucrose, and 7.0 g l-1 bacteriological agar supplemented with 1.0 mg l-1 indole-3-acetic acid (IAA) and 0.05 mg l l-1 gibberellic acid (GA3). Rooting was successfully induced on the same basal medium used for proliferation, modified by halving the MS macro-nutrients and replacing BAP and the other growth regulators with 2.0 mg l l-1 l-naphthaleneacetic acid (NAA). Abstract: The effects of reduced water potential (ψ) in NaCl and pre-osmopriming in PEG, on seed germination and early radicle growth at different temperatures were assessed in the laboratory for sweet sorghum [Sorghum bicolor (L.) Moench] cv. Roce. Five salt solutions (with a ψ of 0-control, −0.22, −0.42, −0.82 or −1.23MPa) and four temperatures of 10, 15, 25 or 35°C were used for germination tests. Daily and final germination, seed moisture at 4 and 24h of imbibition and radicle length 2 days after initial germination were recorded. Sorghum seeds seem to be more tolerant to reduced ψ at optimum temperature. Indeed, at 25°C, down to −0.82MPa seed germination was not affected. The increase in incubation temperature to 35°C determined a greater seed water content and a faster germination than at 25°C at all ψ s, despite the lower final germination percentage. With the lowering of temperature to 15°C, germination percentage significantly declined with the decrease of ψ and at −0.82MPa it reduced to less than 50%. At 10°C seeds failed to germinate at ψ <−0.22MPa due to combined depressive effects of reduced ψ and low temperature. Seed priming enhanced germination and shortened the delay in germination time due to the increase in saline stress, at suboptimal temperatures only. This effect could be explained by the faster water absorption occurring in primed seeds as compared to those unprimed, at these temperatures, regardless of salt concentration of the solution. Root growth was more sensitive than germination to salt stress. Indeed, the increase in NaCl concentration adversely affected root elongation at all temperatures. The beneficial effects of PEG-osmopriming were evident on root growth at all temperatures except the optimal one. From a practical point of view, when early sowings of sweet sorghum are requested, the use of primed seeds is suggested, as PEG-osmopriming is helpful in overcoming the negative effect imposed by reduced water potential upon seed germination under suboptimal thermal conditions. When saline water is available for irrigation at sowing, it may be useful to sow when optimal or nearly optimal temperature may be encountered in the soil in order to avoid the combined stress of temperature and salinity. Summary: Light and temperature-response curves and their resulting coefficients, obtained within ecophysiological characterization of gas exchanges at the leaf level, may represent useful criteria for breeding and cultivar selection and required tools for simulation models aimed at the prediction of potential plant behaviour in response to environmental conditions. Leaf-scale gas exchanges, by means of an IRGA open-flow system, were measured in response to light intensity (8 levels from 0 up to 2000μmolm−2 s−1), CO2 concentrations (ambient—350μmolmol−1 and short-term enriched—700μmolmol−1) and air temperature (from 7 up to 35°C) on three Vicia faba L. genotypes, each representing one of the three cultivated groups: major, equina and minor. The net assimilation rate response to light intensity was well described by an exponential rise to max function. The short-term CO2 enrichment markedly increased the values of light response curve parameters such as maximum photosynthetic rate (+80%), light saturation point (+40%) and quantum yield (+30%), while less homogenous behaviour was reported for dark respiration and light compensation point. For each light intensity level, the major and minor genotypes studied showed assimilation rates at least a 30% higher than equina. The positive effects of short-term CO2 enrichment on photosynthetic water use efficiency (WUE) indicate a relevant advantage in doubling CO2 concentration. In the major and minor genotypes studied, similar assimilation rates, but different WUE were observed. The optimum leaf temperature for assimilation process, calculated through a polynomial function, was 26–27°C and no relevant limitations were observed in the range between 21 and 32°C. Analysis at the single leaf level provided both rapid information on the variations in gas exchange in response to environmental factors and selection criteria for the screening of genotypes. To improve microgreen yield and nutritional quality, suitable light spectra can be used. Two species—amaranth (Amaranthus tricolor L.) and turnip greens (Brassica rapa L. subsp. oleifera (DC.) Metzg)—were studied. The experiment was performed in a controlled LED environment growth chamber (day/night temperatures of 24 ± 2 °C, 16 h photoperiod, and 50/60% relative humidity). Three emission wavelengths of a light-emitting diode (LED) were adopted for microgreen lighting: (1) white LED (W); (2) blue LED (B), and (3) red LED (R); the photosynthetic photon flux densities were 200 ± 5 µmol for all light spectra. The response to light spectra was often species-specific, and the interaction effects were significant. Morphobiometric parameters were influenced by species, light, and their interaction; at harvest, in both species, the fresh weight was significantly greater under B. In amaranth, Chl a was maximized in B, whereas it did not change with light in turnip greens. Sugar content varied with the species but not with the light spectra. Nitrate content of shoots greatly varied with the species; in amaranth, more nitrates were measured in R, while no difference in turnip greens was registered for the light spectrum effect. A light-emitting diode (LED) system covering plant-receptive wavebands from ultraviolet to far-red radiation (360 to 760 nm, "white" light spectrum) was investigated for greenhouse productions of Thymus vulgaris L. Biomass yields and amounts of terpenoids were examined, and the lights' productivity and electrical efficiency were determined. All results were compared to two conventionally used light fixture types (high-pressure sodium lamps (HPS) and fluorescent lights (FL)) under naturally low irradiation conditions during fall and winter in Berlin, Germany. Under LED, development of Thymus vulgaris L. was highly accelerated resulting in distinct fresh yield increases per square meter by 43% and 82.4% compared to HPS and FL, respectively. Dry yields per square meter also increased by 43.1% and 88.6% under LED compared to the HPS and FL lighting systems. While composition of terpenoids remained unaffected, their quantity per gram of leaf dry matter significantly increased under LED and HPS as compared to FL. Further, the power consumption calculations revealed energy savings of 31.3% and 20.1% for LED and FL, respectively, compared to HPS. In conclusion, the implementation of a broad-spectrum LED system has tremendous potential for increasing quantity and quality of Thymus vulgaris L. during naturally insufficient light conditions while significantly reducing energy consumption. Light emitting diodes (LEDs) have recently been considered an efficient artificial light source in plant factories for enhancing plant growth and nutritional quality. Accordingly, this study aimed to review blue, red, and white LED light sources for efficiency and length of the growing period to produce seedlings of Scutellaria baicalensis with high nutritional value. The roots, stems, and leaves of S. baicalensis seedlings were grown under different LED lights and harvested after two and four weeks, and analyzed using high-performance liquid chromatography and gas chromatography time-of-flight mass spectrometry to identify and quantify primary and secondary metabolites. Roots, particularly in the seedlings treated with white LEDs were determined to contain the greatest concentrations of the representative compounds present in S. baicalensis: baicalin, baicalein, and wogonin, which show highly strong biological properties compared to the other plant organs. A total of 50 metabolites (amino acids, sugars, sugar alcohols, organic acids, phenolic acids, and amines) were detected in the roots, stems, and leaves of S. baicalensis seedlings, and the concentrations of primary and secondary metabolites were generally decreased with the increasing duration of LED illumination. Therefore, this study suggests that white LED light and a 2-week growing period are the most efficient conditions for the production of baicalin, baicalein, and wogonin. [ABSTRACT FROM AUTHOR]

It has been shown that monochromatic red and blue light influence photosynthesis and morphology in cucumber. It is less clear how green light impacts photosynthetic performance or morphology, either alone or in concert with other wavelengths. In this study, cucumber (Cucumis sativus) was grown under monochromatic blue, green, and red light, dichromatic blue–green, red–blue, and red–green light, as well as light containing red, green, and blue wavelengths, with or without supplemental far-red light. Photosynthetic data collected under treatment spectra at light-limiting conditions showed that both red and green light enhance photosynthesis. However, photosynthetic data collected with a 90% red, 10% blue, 1000 µmol photons m−2 s−1, saturating light show significantly lower photosynthesis in the green, red, and red–green treatments, indicating a blue light enhancement due to photosystem stoichiometric differences. The red–green and green light treatments show improved photosynthetic capacity relative to red light, indicating partial remediation by green light. Despite a lower quantum efficiency and the lowest ambient photosynthesis levels, the monochromatic blue treatment produced among the tallest, most massive plants with the greatest leaf area and thickest stems. Within the project "BIOSEA" funded by the Italian Ministry of Agriculture and Forestry, a preliminary laboratory test was conducted to assess the variability for cold tolerance during germination in 30 cultivars of biomass sorghum, among fiber and sweet types. Seed germination (%) and mean germination time (MGT) were examined at seven constant temperatures (from 8 °C to 35 °C) and base temperature (Tb) and thermal time (θT) for 50% germination were calculated. A wide genetic diversity in the germination response of sorghum was ascertained at 8 °C (CV 45%) and 10 °C (CV 25.4%). At 8 °C, in cultivars of 'Padana 4', 'PR811F', 'PSE24213', 'PR849' and 'Zerberus', seed germination exceeded 80%. Seeds of 'Zerberus' were also the fastest, requiring less than 13 days for final germination at this low temperature. Great differences were found in Tb and θT among cultivars. Tb varied between 7.44 °C ('PR811F') and 13.48 °C ('Nectar'). Thermal time (θT) was, on average, 24.09 °Cd−1, and ranged between 16.62 ('Nectar') and 33.42 °Cd−1 ('PSE24213'). The best combination of the two germination parameters (i.e., low Tb and θT) corresponded to 'Zerberus', 'Sucrosorgo 506', 'Jumbo' and 'PR811F'. Accordingly, these cultivars are more tolerant to cold stress during germination and, thus, more adapt to early spring sowings in Mediterranean areas (March-April). Cultivars 'PR811F' (fiber type) and 'Sucrosorgo 506' (sweet type) also combine high cold tolerance with good productivity in terms of final dry biomass, as assessed in open-field conditions (late spring sowing). The genetic variation in the germination response to a low temperature is useful for the identification of genotypes of sorghum suitable to early sowings in semi-arid areas. Selection within existing cultivars for cold tolerance during germination may also contribute to the expansion of biomass sorghum into cooler cultivation areas, such as those of Northern Europe, which are less suitable to this warm season crop. Plant specialized metabolites (SMs) play an important role in the interaction with the environment and are part of the plant defense response. These natural products are volatile, semi-volatile and non-volatile compounds produced from common building blocks deriving from primary metabolic pathways and rapidly evolved to allow a better adaptation of plants to environmental cues. Specialized metabolites include terpenes, flavonoids, alkaloids, glucosinolates, tannins, resins, etc. that can be used as phytochemicals, food additives, flavoring agents and pharmaceutical compounds. This review will be focused on Mediterranean crop plants as a source of SMs, with a special attention on the strategies that can be used to modulate their production, including abiotic stresses, interaction with beneficial soil microorganisms and novel genetic approaches. The aim of the presented studies was to evaluate the magnetic field (MF) effect on changes in some enzymes and phytohormones that takes place in the process of seed germination and growth of seedlings. Studies were led in the climatic chambers HERAEUS, on Petri dishes during six consecutive days. Pea seeds were divided into three groups from which one was the control (without stimulation) and two were treated with different doses of magnetic field (30 and 85 mT, respectively). Contents of amylolytic enzymes (AE) and phytohormones were determined at seven terms (0; 24; 48; 72; 96; 120 and 144 h) after placing them on the dishes. A favorable effect of seed stimulation with MF was found on the biochemical processes in the germinating seeds and pea seedlings. The size of changes in enzyme concentration was dependent mainly on the advancement of germination process (i.e., on length of time in which the seeds were on the dishes) and the MF treatment. The use of MF also had an effect on the increase in hormone content in the seeds and organs of seedlings, but values from objects treated with different doses did not differ significantly. Manipulation of the LED illumination spectrum can enhance plant growth rate and development in grow tents. We report on the identification of the illumination spectrum required to significantly enhance the growth rate of sweet basil (Ocimum basilicum L.) plants in grow tent environments by controlling the LED wavebands illuminating the plants. Since the optimal illumination spectrum depends on the plant type, this work focuses on identifying the illumination spectrum that achieves significant basil biomass improvement compared to improvements reported in prior studies. To be able to optimize the illumination spectrum, several steps must be achieved, namely, understanding plant biology, conducting several trial-and-error experiments, iteratively refining experimental conditions, and undertaking accurate statistical analyses. In this study, basil plants are grown in three grow tents with three LED illumination treatments, namely, only white LED illumination (denoted W*), the combination of red (R) and blue (B) LED illumination (denoted BR*) (relative red (R) and blue (B) intensities are 84% and 16%, respectively) and a combination of red (R), blue (B) and far-red (F) LED illumination (denoted BRF*) (relative red (R), blue (B) and far-red (F) intensities are 79%, 11%, and 10%, respectively). The photosynthetic photon flux density (PPFD) was set at 155 µmol m−2 s−1 for all illumination treatments, and the photoperiod was 20 h per day. Experimental results show that a combination of blue (B), red (R), and far-red (F) LED illumination leads to a one-fold increase in the yield of a sweet basil plant in comparison with only white LED illumination (W*). On the other hand, the use of blue (B) and red (R) LED illumination results in a half-fold increase in plant yield. Understanding the effects of LED illumination spectrum on the growth of plant sweet basil plants through basic horticulture research enables farmers to significantly improve their production yield, thus food security and profitability. Nasturtium is a popular herbal plant, widely cultivated as culinary and medicinal plants all over the world. However, the seed propagation of nasturtium is inefficient, and in-vitro propagation is sophisticated and high-cost. In this study, the cutting propagation method was employed to produce nasturtium seedlings. We aimed to determine the optimal conditions for cutting propagation of nasturtium seedlings by investigating the effects of node position and electric conductivity (EC) of nutrient solution on the root formation of the cuttings. Cuttings from five node positions (apical bud, 2nd node, 3rd node, 4th node, and 5th node) were subjected to water and five EC (1.0, 2.0, 3.0, 4.0, and 5.0 dS m−1) treatments with a hydroponic cultivation system in a plant factory. Results showed that all cuttings rooted successfully within two weeks. The cuttings from the apical bud position rooted earliest and produced the most roots regardless of EC level. Cuttings from other node positions produced longer roots and heavier root fresh and dry weights than those from the apical bud position. The cuttings under EC of 1.0 dS m−1 had the greatest root number, the longest root length, and the heaviest root fresh and dry weights regardless of node positions. The EC of 1.0 dS m−1 is considered the best condition for nasturtium cuttings for the range of EC tested in this study, and the cuttings from all the five node positions can be used as seedling materials. Abstract: Constructed wetlands (CWs) could play a strategic role in wastewater reclamation and reuse in arid and semiarid regions, such as the Mediterranean area. The aim of this study was to analyse the phytoremoval effectiveness of Phragmites australis and spontaneous plant species (SPS) in two CW systems: a horizontal (H-SSF) and a vertical (V-SSF) subsurface flow. P. australis showed a higher biomass growth than SPS and always covered 100% of the bed''s surface. The H-SSF system was more suitable for enhancing biomass growth of P. australis (4701 gm −2 as compared to 3088 gm −2 detected in the V-SSF CWs) whereas V-SSF of SPS (1700 gm −2 and 100% total cover as compared to 240 gm −2 and 40% total cover observed in H-SSF). SPS included species belonging to different botanical families in relation to the system adopted. Some of these species (i.e. Phalaris spp. and Chrysantemum segetum) were dominant in aboveground floral composition. Beds with SPS showed the same efficiency of the Phragmites ones in TSS, COD and pathogen removal. Phragmites was more effective than SPS in removing BOD5 (in horizontal system), total phosphorus and nitrates. V-SSF systemwas more effective than H-SSF in removing total nitrogen. SPS presence did not decrease the efficiency of unplanted beds. Irrespective of the different plant species covering the beds, CWs were overall efficient in removing pollutants confirming their potential as an economic alternative to traditional urban wastewater treatments in small and medium-sized communities. Since the development of the "cholinergic hypothesis" as an important therapeutic approach in the treatment of Alzheimer's disease (AD), the scientific community has made a remarkable effort to discover new and effective molecules with the ability to inhibit the enzyme acetylcholinesterase (AChE). The natural function of this enzyme is to catalyze the hydrolysis of the neurotransmitter acetylcholine in the brain. Thus, its inhibition increases the levels of this neurochemical and improves the cholinergic functions in patients with AD alleviating the symptoms of this neurological disorder. In recent years, attention has also been focused on the role of another enzyme, butyrylcholinesterase (BChE), mainly in the advanced stages of AD, transforming this enzyme into another target of interest in the search for new anticholinesterase agents. Over the past decades, Nature has proven to be a rich source of bioactive compounds relevant to the discovery of new molecules with potential applications in AD therapy. Bioprospecting of new cholinesterase inhibitors among natural products has led to the discovery of an important number of new AChE and BChE inhibitors that became potential lead compounds for the development of anti-AD drugs. Plants of the Amaryllidaceae family are well-known (not only) for their ornamental value but also for the alkaloids that they produce. In this report, the first phytochemical study of Clinanthus genus was carried out. The chemical composition of alkaloid fractions from Clinanthus microstephium was analyzed by GC/MS and NMR. Seven known compounds belonging to three structural types of Amaryllidaceae alkaloids were identified. An epimeric mixture of a haemanthamine-type compound (6-hydroxymaritidine) was tested as an inhibitor against acetyl- and butyrylcholinesterase enzymes (AChE and BChE, respectively), two enzymes relevant in the treatment of Alzheimer's disease, with good results. A collection of sixteen semisynthetic 17-hydroxycativic acid esters with alcohols containing a tertiary amine group was evaluated for their in vitro cytotoxicity against two human cancer cell lines, THP-1 and U937, and for their effects on the cell cycle and cell death. While 17-hydroxycativic acid itself is not cytotoxic, all the esters displayed cytotoxic activity, with 50% growth inhibition (GI 50 ) values ranging between 3.2 and 23.1 µM. In general, the most potent compounds in both cell lines were esters with four carbon long alcohol residues. There was no clear relationship between the identity of the terminal secondary amine and the activity of the compound. Experiments using the 6-(pyrrolidin-1-yl)pentyl ester, 2c, revealed that this compound activates caspases-3/7 and causes poly(ADP-ribose)polymerase 1 (PARP-1) fragmentation in THP-1 and U937 cells, indicating the induction of apoptotic cell death. These results suggest that further investigation into the anticancer activity of diterpene derivatives and other labdane diterpenes may be fruitful.

Alzheimer's disease (AD) is a neurodegenerative disorder associated with memory impairment and cognitive deficit. Most of the drugs currently available for the treatment of AD are acetylcholinesterase (AChE) inhibitors. Plants of the Amaryllidaceae family are known to synthesize alkaloids, which have shown AChE inhibitory activity. Habranthus tubispathus and H. jamesonii are two Amaryllidaceae that can be found growing wild to the southwest of Buenos Aires in Argentina. Acetyl- and butyrylcholinesterase inhibition was observed for the extracts obtained from bulbs of H. tubispathus and bulbs and aerial parts of H. jamesonii. The strongest cholinesterase inhibition was observed for the alkaloid extract obtained from the aerial parts for H. jamesonii (AChE IC50 = 0.7 microg/mL; BChE IC50 = 6.7 microg/mL). The AChE inhibition observed for H. jamesonii could be explained by the presence of galanthamine and sanguinine, two potent AChE inhibitors. The levels of lycorine and hippeastidine, moderate AChE inhibitors, observed in the bulbs of H. tubispathus could be responsible for the significant AChE inhibition observed. The alkaloids present in these Amaryllidaceae were identified by means of GC-MS analysis. In the case of H. tubispathus, hippeastidine and 3-O-demethylhippeastidine, were isolated and completely characterized by 1H and 13C NMR spectroscopy.

Light and temperature-response curves and their resulting coefficients, obtained within ecophysiological characterization of gas exchanges at the leaf level, may represent useful criteria for breeding and cultivar selection and required tools for simulation models aimed at the prediction of potential plant behaviour in response to environmental conditions. Leaf-scale gas exchanges, by means of an IRGA open-flow system, were measured in response to light intensity (8 levels from 0 up to 2000 micromol m(-2) s(-1)), CO(2) concentrations (ambient-350 micromol mol(-1) and short-term enriched-700 micromol mol(-1)) and air temperature (from 7 up to 35 degrees C) on three Vicia faba L. genotypes, each representing one of the three cultivated groups: major, equina and minor. The net assimilation rate response to light intensity was well described by an exponential rise to max function. The short-term CO(2) enrichment markedly increased the values of light response curve parameters such as maximum photosynthetic rate (+80%), light saturation point (+40%) and quantum yield (+30%), while less homogenous behaviour was reported for dark respiration and light compensation point. For each light intensity level, the major and minor genotypes studied showed assimilation rates at least a 30% higher than equina. The positive effects of short-term CO(2) enrichment on photosynthetic water use efficiency (WUE) indicate a relevant advantage in doubling CO(2) concentration. In the major and minor genotypes studied, similar assimilation rates, but different WUE were observed. The optimum leaf temperature for assimilation process, calculated through a polynomial function, was 26-27 degrees C and no relevant limitations were observed in the range between 21 and 32 degrees C. Analysis at the single leaf level provided both rapid information on the variations in gas exchange in response to environmental factors and selection criteria for the screening of genotypes.
