20. Interaction and immune regulations of innate lymphoid cells and plasmacytoid dendritic cells in skin inflammatory diseases

Abstract

The skin is equipped with an immune network providing surveillance and effector function. Plasmacytoid dendritic cells (pDCs) are found to accumulate in skin wounds, yet the mechanisms of their role in skin wound healing are hitherto unstudied. Here, BDCA-2+ DCs were shown to infiltrate in vivo human wounds, showing heterogeneity based on differential expression of CD123, with a CD123int DC subset expressing CD1a at high levels. The CD1a+-bearing subsets could be derived from blood pDCs and presented self-lipid antigens to CD1a-reactive T cells, inducing interleukin (IL)-22 production. Using single-cell RNA sequencing, human skin wound CD123intBDCA 2+ CD1a+ DCs were shown to acquire features compatible with a role in lymph node homing and antigen presentation. Plasmacytoid DCs and group 3 innate lymphoid cells (ILC3) have emerged as important mediators of skin inflammation, but, the mechanisms of their interaction have remained elusive. Here, it was identified that type I interferons (IFN-I), produced by pDCs, negatively regulated IL-22 producing capability of ILC22, an ILC3 subset, and positively mediated their co-stimulatory marker expression. ILC22 also presented peptide antigens to CD4+ T cells. While exogenous IL 1beta and IL 23 dampened the antigen presenting capacity of ILC22, addition of IFN I rescued their ability, and the study of psoriatic ILC22 revealed a hyper responsiveness to IFN-I. These data demonstrate early infiltration of a previously unrecognised subset of CD123intBDCA-2+CD1a+ DCs in human skin wounds which can present lipid antigens to autoreactive T cells, contributing to the wound healing process. Furthermore, the blood CD123hiBDCA 2+ pDCs produce IFN-I and regulate ILC22 to promote T cell functions. Overall, this study reveals a new route for potential therapeutic manipulations in maintaining skin homeostasis: the pDC-IFN-I-ILC22-T cell axis.

21. From Laissez Faire to a Market Mechanism: the Formation of Housing Finance in Taiwan.

Abstract

The literature on East Asian developmental states has mostly focused on economic policies; few studies have investigated how developmentalism affects urbanisation. This study addresses this gap by analysing the formation of a housing system by the developmental state and the role of the housing sector in the transition from an economic miracle in the 1960s to stagnation after the 1990s. Wang, Lee, and Chen (2017) argued that Taiwan’s state bureaucrats played a gradually diminishing role in directing the market. This article takes a different approach by arguing that the government’s interventions on housing intensified after the 1990s; housing finance sectors were formalised, squatters were removed, and interventions based on market mechanisms were employed to stimulate the housing market. However, the lack of an effective state intervention on housing made speculation difficult to control and created an obstacle to economic development. Abstract

The CD1 and MR1 protein families present lipid antigens and small molecules to T cells, complementing well-studied major histocompatibility complex–peptide mechanisms. The CD1a subtype is highly and continuously expressed within the skin, most notably on Langerhans cells, and has been demonstrated to present self and foreign lipids to T cells, highlighting its cutaneous sentinel role. Alteration of CD1a-dependent T-cell responses has recently been discovered to contribute to the pathogenesis of several inflammatory skin diseases. In this review, we overview the structure and role of CD1a and outline the current evidence implicating CD1a in the development of psoriasis, atopic dermatitis and allergic contact dermatitis. Abstract

Multiple forms of interference and noise that impact the receiver's capacity to receive and interpret satellite signals, and consequently the preciseness of positioning and navigation, may be present during the processing of Global Positioning System (GPS) navigation. The non-Gaussian noise predominates in the signal owing to the fluctuating character of both natural and artificial electromagnetic interference, and the algorithm based on the minimum mean-square error (MMSE) criterion performs well when assuming Gaussian noise, but drops when assuming non-Gaussian noise. The maximum correntropy criteria (MCC) adaptive filtering technique efficiently reduces pulse noise and has adequate performance in heavy-tailed noise, which addresses the issue of filter performance caused by the presence of non-Gaussian or heavy-tailed unusual noise values in the localizing measurement noise. The adaptive kernel bandwidth (AKB) technique employed in this paper applies the calculated adaptive variables to generate the kernel function matrix, in which the adaptive factor can modify the size of the kernel width across a reasonably appropriate spectrum, substituting the fixed kernel width for the conventional MCC to enhance the performance. The conventional maximum correntropy criterion-based extended Kalman filter (MCCEKF) algorithm's performance is significantly impacted by the value of the kernel width, and there are certain predetermined conditions in the selection based on experience. The MCCEKF with a fixed adaptive kernel bandwidth (MCCEKF-AKB) has several advantages due to its novel concept and computational simplicity, and gives a qualitative solution for the study of random structures for generalized noise. Abstract

Background: Resistance to standard chemotherapy is a critical problem for breast cancer patients. The ATP-binding cassette (ABC) superfamily transporters actively pump out drugs and play an important role in chemoresistance. ABCB1 (ABC subfamily B, member 1, also named as multidrug resistance protein 1, MDR1) and suppressive myeloid-derived suppressor cells (MDSCs) potentially involve in chemoresistance of breast cancer. The relationship between ABCB1 and immune genes in breast cancer has not been widely studied.Methods: Microarray and RNA sequencing data were obtained from The Cancer Genome Atlas Breast Invasive Carcinoma in Genomic Data Commons Data Portal and Gene Expression Omnibus database. A patient-derived xenograft (PDX) model of HER2+ breast cancer was established to investigate the association between ABCB1 and immune genes in breast cancer.Results: Expression of ABCB1 increased in doxorubicin-selected MCF-7/ADR cells. High expression of ABCB1 mRNA is correlated with lymph-node metastasis and worse overall survival in patients with breast cancer. ABCB1 is positively correlated with IL6, CSF1, CSF3, and PTGS2. In the HER2+ stage IIA breast cancer PDX model, both doxorubicin and paclitaxel suppressed growth of P2 tumors. IL6, CSF1, CSF3, and PTGS2 expression were suppressed by paclitaxel but not doxorubicin. Intrasplenic MDSCs, including CD11b+Ly6G+ and CD11b+Ly6C+ cells, were more abundant than intratumor MDSCs in PDX-carrying nude mice. Clinically, the patient developed cancer recurrence after adjuvant chemotherapy with doxorubicin-based regimen and was well controlled after paclitaxel-trastuzumab combined therapy.Conclusion: ABCB1 was a poor predictor of HER2+ LN– breast cancer. Regulation of immune genes by ABCB1 contributed to cancer recurrence and treatment effect. The PDX model was suitable for investigation the expression of target genes and expansion of immune cells. 40. The formation of parental language ideology in a multilingual context : a case study in Taiwan

Abstract

In the past ten years, the number of English cram schools has tripled in Taiwan and more than 70% of fifth and sixth graders claim they attended cram schools before receiving formal English education in primary schools. In response to pressures of both globalisation and localisation, the government introduced a school policy of learning a Taiwanese minority language, in addition to Mandarin, in 2005. The majority of parents, however, are not keen to encourage their children to learn a Taiwanese second language compared to the ‘trend’ of learning English (as a foreign language). This study explores family language policies using a multiple-case-study strategy with twelve families whose children attend a language school in Taichung, Taiwan, with two additional cases from different geolinguistic areas. The main focus is on how parents form their ideologies about language and language learning. The data were gathered using semi-structured interviews. The macro- and micro-factors which underpin the parents’ language ideologies are the central focus of analysis. Amongst these families the process of ideology formation involves more than three languages, English, Mandarin, Minnan and Hakfa (which, in this study, is the only representative of other minority languages spoken in Taiwan). The three Chinese languages appear as ‘mother tongue’ in various combinations amongst the parents in the study. The findings indicate that the influence of macro- and micro-factors on parents’ language ideologies is complex and interactive, rather than linear. Significant macro-factors identified include the local, national and global sociolinguistic environments, government policies and economic factors, notably the labour market. Macro-factors, as well as micro-factors, do not influence parents’ ideologies in isolation from each other. Similar, shared macro-contexts are responded to in diverse ways by the parents in the study, with familial mother tongue, educational experiences and different perceptions of the social roles of language all playing a part. Parents’ language ideologies are, therefore, clearly not structurally determined, but neither do the parents act as ‘free agents’ in their ideological choices which, in turn, have an impact on family language management and language practices. The complexity and fluidity of the Taiwanese language situation and the rapid social, political and economic changes that are taking place in the community make this study particularly valuable in enhancing our understanding of how personal language ideologies evolve.

Abstract

The convergence of text, visual, and audio data is a key step towards human-like artificial intelligence, however the current Vision-Language-Speech landscape is dominated by encoder-only models which lack generative abilities. We propose closing this gap with i-Code V2, the first model capable of generating natural language from any combination of Vision, Language, and Speech data. i-Code V2 is an integrative system that leverages state-of-the-art single-modality encoders, combining their outputs with a new modality-fusing encoder in order to flexibly project combinations of modalities into a shared representational space. Next, language tokens are generated from these representations via an autoregressive decoder. The whole framework is pretrained end-to-end on a large collection of dual- and single-modality datasets using a novel text completion objective that can be generalized across arbitrary combinations of modalities. i-Code V2 matches or outperforms state-of-the-art single- and dual-modality baselines on 7 multimodal tasks, demonstrating the power of generative multimodal pretraining across a diversity of tasks and signals.

Abstract

Simple Summary: We aimed to investigate if VDT could be applied as a predictor of clinical outcome in segmentectomy and wedge resection. We retrospectively studied 96 NSCLC patients post sublobar resection from 2012 to 2018, collecting two chest CT scans preoperatively of each case and calculating VDT. The receiver operating characteristic curve was constructed to identify the optimal cut-off point of VDTs as 133 days. We divided patients into two groups: VDT < 133 days (n = 22) and VDT ≥ 133 days (n = 74). Univariable and multivariable analyses were performed for comparative purposes. Our study demonstrated that the five year OS rates of patients with VDTs ≧ 133 days and VDTs < 133 days, respectively, were 89.9% and 71.9% (p = 0.003), and the five year DFS rates were 95.9% and 61.5% (p = 0.002). Thus, we concluded that VDT can be a powerful prognostic predictor and provides an essential role in planning surgical procedures. Univariable and multivariable analyses were performed for comparative purposes. Results: Univariable and multivariable analyses revealed that the consolidation and tumor diameter ratio was the factor of overall survival (OS), and VDT was the only factor of disease-free survival (DFS). The five year OS rates of patients with VDTs ≥ 133 days and VDTs < 133 days, respectively, were 89.9% and 71.9%, and the five year DFS rates were 95.9% and 61.5%. Conclusion: As VDT serves as a powerful prognostic predictor and provides an essential role in planning surgical procedures, the evaluation of VDT preoperatively is highly suggested.
