Dyslipidemia, a major risk factor for cardiovascular diseases (CVDs), is modifiable by diet and lifestyle changes. A large population with mild to moderate dyslipidemia is at risk of developing CVDs, and early initiation of preventive measures can avert advancing into severe medical conditions. Studies suggest increasing slowly digestible starch (SDS) in diets can help lower blood lipids. We processed dehulled adlay, a cereal rich in bioactive compounds, such as polyphenols and phytosterols, into an instant meal by extrusion and milling and then assessed its starch composition and in vitro digestibility. The dehulled adlay was found to consist of 32% SDS and resistant starch combined. Then, eligible subjects with dyslipidemia were recruited to explore the adlay’s hypolipidemic potential, safety, and acceptability. Subjects consumed the dehulled adlay as the sole carbohydrate source in their breakfast, without changing other components in the diet or lifestyle, for 12 weeks. After intervention, serum total cholesterol (TC) decreased significantly in subjects with hypercholesterolemia. In addition, both TC and triglyceride levels decreased significantly in those above 50 years old. In conclusion, the extruded dehulled adlay displays potential for favorably modulating blood lipids, and the effect is more pronounced in the middle-aged population.

Abstract The relationship between Helicobacter pylori infection and rheumatoid arthritis has been investigated, but the results remain controversial. This study aims to determine the association between the two diseases via a 17-year retrospective cohort study. Using the National Health Insurance Research Database, a nationwide population based in Taiwan, we identified 97,533 individuals with H. pylori infection and matched controls between 2000 and 2017 using propensity score matching at a 1:1 ratio. The adjusted hazard ratio of rheumatoid arthritis was determined by multiple Cox regression. Abstract The most widely used method for intracellular RNA fluorescence labeling is MS2 labeling, which generally relies on the use of multiple protein labels targeted to multiple RNA (MS2) hairpin structures installed on the RNA of interest (ROI). While effective and conveniently applied in cell biology labs, the protein labels add significant mass to the bound RNA, which potentially impacts steric accessibility and native RNA biology. We have previously demonstrated that internal, genetically encoded, uridine-rich internal loops (URILs) comprised of four contiguous UU pairs (8 nt) in RNA may be targeted with minimal structural perturbation by triplex hybridization with 1 kD bifacial peptide nucleic acids (bPNAs). A URIL-targeting strategy for RNA and DNA tracking would avoid the use of cumbersome protein fusion labels and minimize structural alterations to the RNA of interest. Here we show that URIL-targeting fluorogenic bPNA probes in cell media can penetrate cell membranes and effectively label RNAs and RNPs in fixed and live cells. This method, which we call fluorogenic U-rich internal loop (FLURIL) tagging, was internally validated through the use of RNAs bearing both URIL and MS2 labeling sites. Notably, a direct comparison of CRISPR-dCas labeled genomic loci in live U2OS cells revealed that FLURIL-tagged gRNA yielded loci with signal to background up to 7X greater than loci targeted by guide RNA modified with an array of eight MS2 hairpins. Together, these data show that FLURIL tagging provides a versatile scope of intracellular RNA and DNA tracking while maintaining a light molecular footprint and compatibility with existing methods.

Nuclear chromosome compaction is non-random and dynamic. The spatial distance among genomic elements instantly modulates transcription. Visualization of the genome organization in the cell nucleus is essential to understand nuclear function. In addition to cell type-dependent organization, high-resolution 3D imaging shows heterogeneous compaction of chromatin organization among the same cell type. Questions remain to be answered if these structural variations were the snapshots of dynamic organization at different time points and if they are functionally different. Live-cell imaging has provided unique insights into dynamic genome organization at short (milliseconds) and long (hours) time scales. The recent development of CRISPR-based imaging opened windows for studying dynamic chromatin organization in single cells in real time. Here we highlight these CRISPR-based imaging techniques and discuss their advances and challenges as a powerful live-cell imaging method that poses high potential to generate paradigm-shifting discoveries and reveal functional implications of dynamic chromatin organization.

The purpose of this study was to explore college students’ purchase behaviors using a multivariate statistical model. Because many college students today have part-time jobs, they have increased opportunities to consume higher-quality brands. In Taiwan, Starbucks is regarded as a high-quality coffee brand, costing more than most local coffee brands. To clarify why such students in Taiwan are willing to pay more for Starbucks, data were collected from a sample of 402 college students. The results showed differences in purchase decisions between students majoring in different disciplines and lifestyles. The findings had theoretical and practical implications especially for the marketing strategy of the Taiwanese coffee industry.

This study proposes an assessment method to quantify the risks of the smoke environment for road tunnel fire safety based on previous studies. The assessment method integrates visibility and toxic gases to address the hazards of smoke distribution more comprehensively. Considering that the hazards of visibility reduction and toxic gases for tunnel users vary with exposure time and location in a fire event, the smoke environment (SE) levels are defined as a function of longitudinal location and time. The SE levels simplify smoke distribution as calculated from 3D computational fluid dynamics (CFDs). For easily identifying SE risks, SE levels are illustrated on a 2D map to analyze the potential hazard by quantifying specific areas and times of smoke exposure. To demonstrate the applicability of the assessment method of this study, cases are carried out using CFD simulation to investigate the risks associated with tunnel fires with various tunnel cross-section types, longitudinal velocities, and gradients. In the analysis of the SE level in different cross-section types and longitudinal velocities under the condition of no vehicle, a velocity of 0.9–1.1 m/s can maintain a less serious SE level both upstream and downstream in a horizontal rectangular tunnel, and 0.3–0.5 m/s in a horizontal horseshoe-shaped tunnel. Both rectangular and horseshoe-shaped tunnels reveal an obvious rise within 10–15 min. In the case of inclined tunnels, for both rectangular and horseshoe-shaped tunnels, the SE level near the fire source obviously deteriorates. Thus, the longitudinal velocity range for the purpose of maintaining a relatively less serious SE level should be slightly reduced for inclined tunnels compared with horizontal tunnels.

Japan and Europe have adopted significantly different ventilation modes with regard to traffic conditions (with or without congestion). This study focuses on the smoke behavior of different ventilation modes by conducting a 3D CFD analysis of smoke distribution and CO concentration variation and then comparing these modes based on two groups (relatively low target velocity: 0 and 1 m/s; relatively high target velocity: 2 and 3 m/s). The considered fire size is 30 MW for four longitudinal gradients (0, 2, 4, and 6%). In the simulation results, velocities of both 0 and 1 m/s reveal good performance in maintaining the stratification of smoke and ensuring the safety of the environment in 10 min in the occurrence of traffic jams. However, in 15 min, the smoke conditions change. Thus, it is difficult to distinguish which ventilation mode (0 and 1 m/s) shows relatively better performance during traffic congestion. When traffic is uncongested, the comparison of Um = 2 and 3 m/s reveals that a target velocity of Um = 2 m/s (lower than critical velocity) can also prevent the risk of smoke on the upstream side because no descending phenomenon is observed. Moreover, Um = 2 m/s causes the relatively slow propagation of descending smoke, increasing the possibility of evacuation once a second traffic incident occurs on the downstream side of the fire source.

In countries with a high motorbike utilization rate, road tunnels can feature motorbike lanes, bringing an additional risk to evacuation from tunnels during a fire or emergency. To better understand the walking speed in motorbike lanes to enhance risk assessment in tunnels, in the present study, we conducted evacuation experiments to investigate the influence of motorbike and evacuee density on the walking speed of motorbike users. According to the experimental results, the walking speed was slightly reduced even when the evacuee density was relatively lower (around 0.1 person/m2). To further analyze the influence of motorbikes in the lane, the walking speed decreased significantly with the increase in motorbike density. The decrease in walking speed presented an exponential relationship with evacuee and motorbike density. Considering this exponential relationship, nonlinear regression was applied to estimate the parameters of the walking speed model. The proposed model consisting of the evacuee density, motorbike density, and free walking speed as variables can serve as an approach to describe the walking speed of motorbike lane evacuation in tunnels.

Background: The Identification and Management of Feeding Difficulties (IMFeD™) tool was introduced in 2011 for management of children with feeding difficulties and has been used in various countries around the world. This study aimed to assess its acceptance and effectiveness in clinical practice in Taiwan. Methods: This was a prospective, observational, multicenter study where subjects were followed over 3 months. Outpatients identified as picky eaters, having poor appetite, poor weight gain, or weight loss were enrolled. An optical fiber sensing system integrating a fiber Bragg grating (FBG) sensor, a long-period fiber grating (LPFG) optical filter and a photodetector is presented to monitor the dynamic response of a structure subjected to base excitation and impact loading. The FBG sensor is attached to a test specimen and connected to an LPFG filter. As the light reflected from the FBG sensor is transmitted through the long-period fiber grating filter, the intensity of the light is modulated by the wavelength, which is affected by the strain of the FBG. By measuring the intensity of the light using a photodetector, the wavelength reflected from the FBG sensor can be demodulated, thus leading to the determination of the strain in the structure. To demonstrate its effectiveness, the proposed sensing system was employed to measure the dynamic strain of a beam subjected to mechanical testing. The mechanical tests comprised three load scenarios: base excitation by a shaker at resonant frequency, impact loading by a hammer and shock test on a drop table. To monitor the dynamic strain during the test and validate the accuracy of the measurement of the FBG sensor, strain gauge was used as reference. Experimental results show good correlation between the measurements of FBG sensor and strain gauge. The present work provides a fast response and easy-to-implement optical fiber sensing system for structural health monitoring based on real-time dynamic strain measurements.

Lifelong overexpression of heat shock protein (HSP) 72 in skeletal muscle is known to protect against age-related oxidative stress and muscle damage. This study aimed to ascertain whether exhaustive exercise (EE)-induced muscle fatigue and damage can be prevented by lifelong overexpression of HSP72 in skeletal muscle. Methods: Transgenic mice heterozygous for the porcine HSP70.2 gene ([+]HSP72) and transgene-negative littermate controls ([–]HSP72) were subjected to an EE protocol. Mice were randomly divided into four groups: sedentary [–]HSP72, sedentary [+]HSP72, EE [–]HSP72, and EE [+]HSP72. Animals were killed 82 minutes after the start of EE to determine muscular levels of HSP72, serum levels of superoxide dismutase (SOD, an antioxidant enzyme) and lactate (an indicator of muscle fatigue), muscular levels of matrix metalloproteinase (an indicator of inflammatory myopathies), and muscular damage. Results: During the test, the latency value for the occurrence of EE was 79–85 minutes and 100–110 minutes for [–]HSP72 and [+]HSP72 mice, respectively. After EE, [+]HSP72 mice had significantly higher serum SOD and significantly lower serum lactate, muscular matrix metalloproteinase or myeloperoxidase activity, and muscle damage compared to [–]HSP72 mice. Conclusion: The results suggest that HSP72 overexpression in skeletal muscle may improve muscle fatigue and damage in EE by reducing oxidative damage and phagocytic infiltration, at least in mice.

Dynamic chromatin organization instantly influences DNA accessibility through modulating local macromolecular density and interactions, driving changes in transcription activities. Chromatin dynamics have been reported to be locally confined but contribute to coherent chromatin motion across the entire nucleus. However, the regulation of dynamics, nuclear orientation and compaction of subregions along a single chromosome are not well-understood. We used CRISPR-based real-time single-particle tracking and polymer models to characterize the dynamics of specific genomic loci and determine compaction levels of large human chromosomal domains. Our studies showed that chromosome compaction changed during interphase and that compactions of two arms on chromosome 19 were different. The dynamics of genomic loci were subdiffusive and dependent on chromosome regions and transcription states. Surprisingly, the correlation between locus-dependent nuclear localization and mobility was negligible. Strong tethering interactions detected at the pericentromeric region implies local condensation or associations with organelles within local nuclear microenvironments, such as chromatin--nuclear body association. Pumpkin (Cucurbita moschata) is a popular and nutritious vegetable consumed worldwide. The overall purpose of this study was to evaluate the effects of C. moschata fruit extract (CME) on anti-fatigue and ergogenic functions following physiological challenges. Male ICR mice from four groups designated vehicle, CME-50, CME-100 and CME-250, respectively (n = 8 per group in each test) were orally administered CME for 14 days at 0, 50, 100 and 250 mg/kg/day. The anti-fatigue activity and exercise performance were evaluated using exhaustive swimming time, forelimb grip strength, as well as levels of plasma lactate, ammonia, glucose, and creatine kinase after an acute swimming exercise. The resting muscular and hepatic glycogen was also analyzed after 14-day supplementation with CME. Trend analysis revealed that CME treatments increased grip strength. CME dose-dependently increased 5% body weight loaded swimming time, blood glucose, and muscular and hepatic glycogen levels. CME dose-dependently decreased plasma lactate and ammonia levels and creatine kinase activity after a 15-min swimming test. The mechanism was relevant to the increase in energy storage (as glycogen) and release (as blood glucose), and the decrease of plasma levels of lactate, ammonia, and creatine kinase. Therefore, CME may be potential for the pharmacological effect of anti-fatigue.

Wilson's disease (WD) is an autosomal recessive defect of cellular copper export. Early diagnosis in children is difficult due to its obscure clinical presentations. The efficacy of zinc salts is well documented, although there are limited data concerning zinc use in pediatric patients with WD. Methods: We performed a retrospective analysis of clinical features, laboratory results and treatment responses in children with WD diagnosed at Taichung Veterans General Hospital between 1996 and 2008. Diagnosis was established by low serum ceruloplasmin, high 24-hour urinary copper excretion, presence of Kayser-Fleischer rings, and mutation analysis. Results: Eleven children were included in this study. The main initial presentations were impaired liver function tests (6/11) and hemolytic anemia (2/11). Gene studies in seven children showed six different mutations (G934D, R778Q, C490X, 304insC, IVS4-1 G > C, P992I) and one possible novel mutation (L1181P). All patients had improved liver function tests and hemoglobin levels after treatment with D-penicillamine, trientine and zinc supplement therapy. During a mean period of 3.4 ± 2.1 years with zinc therapy, six patients had serum zinc levels above the normal limit, and seven patients had serum copper levels below the normal range. Conclusion: Serum ceruloplasmin and 24-hour urinary copper examinations could be used to rule out WD in children with chronic hepatitis and hemolytic anemia. Gene analysis is helpful for prompt diagnosis of asymptomatic siblings and patients with atypical features. Zinc treatment is generally safe in pediatric patients with WD. However, its adverse effects should be monitored.

We assessed the characteristics and perception of telephone appointments among outpatients and medical staff during the COVID-19 pandemic in Taiwan. Our survey was performed by giving self-administered questionnaires to the enrollees. Basic socioeconomic status data were collected. We used a valid and reliable telehealth usability questionnaire (TUQ) to assess the telemedicine experience among outpatients and medical staff. Only outpatients with chronic illness and who had regular visits before the pandemic were enrolled. We delivered the questionnaire survey to participants who used telephone appointments from 20 May 2021 to 31 July 2021 in Taichung Veterans General Hospital. A total of 471 outpatients and 203 medical staff completed the survey. Most of the respondents were aged 30–69, college-educated, women, and married. Outpatients have higher scores in all dimensions of TUQ than medical staff, especially in the dimensions of ease of use and effectiveness. Age, gender, education, and marriage have no significant associations in the medical staff group. In the outpatient group, gender is the only significant factor in the six dimensions of TUQ. We found a significant disparity in the perception gap of telemedicine among outpatient and medical staff. Outpatients are satisfied with telephone appointments during the COVID-19 pandemic, but medical staff are concerned about the ease of use and effectiveness. [ABSTRACT FROM AUTHOR]

On the basis of the multivariate MGF (moment-generating function) of Weibull statistics and an alternative expression of the Q-function, an approximate BER (bit-error rate) expression is derived for an MC-DS-CDMA system combined with MRC (maximal ratio-combining) diversity. The Weibull fading model is applied to analyze the BER performance of an MC-DS-CDMA system by considering multi-users with both multi-carriers and the phenomena of PBI (partial-band interference) cases. Regardless of the channel model assumed or the user number considered, it is noteworthy that the PBI is definitely the most important factor dominating the performance of an MC-DS-CDMA system, according to the results obtained in this study. The reliability of the proposed schemes can be explicitly verified in accordance with the previously reported results. Thus, we assert that the aim of the performance analysis of an MC-DS-CDMA system operating under Weibull fading has been completely achieved. A study was conducted to show how rapidly evolving information technology has altered the knowledge dissemination process. Findings show that e-learning could be the paradigm shift of traditional education. Organizations are also provided with a methodology to deliberately evaluate their e-learning programs.

Titanium dioxide (TiO2) paste was prepared by sol-gel and hydrothermal method with various precursors. Nanostructured mesoporous TiO2 thin-film back electrode was fabricated from the nanoparticle colloidal paste, and its performance was compared with that made of commercial P25 TiO2. The best performance was demonstrated by the DSSC having a 16 μm-thick TTIP-TiO2 back electrode, which gave a solar energy conversion efficiency of 6.03%. The ability of stong adhesion on ITO conducting glass substrate and the high surface area are considered important characteristics of TiO2 thin film. The results show that a thin film with good adhesion can be made from the prepared colloidal paste as a result of alleviating the possibility of electron transfer loss. One can control the colloidal particle size from sol-gel method. Therefore, by optimizing the preparation conditions, TiO2 paste with nanoparticle and narrow diameter distribution was obtained.

OBJECTIVE. We aimed to detect biliary atresia (BA) in early infancy to prevent additional liver damage because of the delay of referral and surgical treatment and to investigate the incidence rate of BA in Taiwan. METHODS. A pilot study to screen the stool color in infants for the early diagnosis of BA was undertaken from March 2002 to December 2003. We had designed an "infant stool color card" with 7 numbers of different color pictures and attached it to the child health booklet. Parents were then asked to observe their infant's stool color by using this card. The medical staff would check the number that the parents chose according to their infant's stool color at 1 month of age during the health checkup and then send the card back to the stool color card registry center. RESULTS. The average return rate was ~65.2% (78 184 infants). A total of 29 infants were diagnosed as having BA, and 26 were screened out by stool color card before 60 days of age. The sensitivity, specificity, and positive predictive value were 89.7%, 99.9%, and 28.6%, respectively. Seventeen (58.6%) infants with BA received a Kasai operation within 60-day age period. The estimated incidence of BA in screened newborns was 3.7 of 10 000. CONCLUSIONS. The stool color card was a simple, efficient, and applicable mass screening method for early diagnosis and management of BA. The program can also help in estimating the incidence and creating a registry of these patients. A nonstatistical behavior is observed in the photodissociation of two acyl cyanide compounds. The analysis reveals that the excited-state dissociation rate does not decrease with molecular size as much as statistical as statistical rate theory predicts. This suggests that a nonergodic system in which the available vibrational energy does not redistribute statistically among all modes before the bond breakage occurs.

Abstract Background Recent studies relating to the association between DNA repair-gene polymorphisms and colorectal cancer risk would, to the best of our knowledge, appear to be very limited. This study was designed to examine the polymorphisms associated with three DNA repair genes, namely: XRCC1 Arg399Gln, XRCC3 Thr241Met and XPD Lys751Gln, and investigate their role as susceptibility markers for colorectal cancer. Methods We conducted a case-control study including 727 cases of cancer and 736 hospital-based age- and sex-matched healthy controls to examine the role of genetic polymorphisms of three DNA-repair genes (XRCC1, XRCC3 and XPD) in the context of colorectal cancer risk for the Taiwanese population. Genomic DNA isolated from 10 ml whole blood was used to genotype XRCC1 Arg399Gln, XRCC3 Thr241Met and XPD Lys751Gln by means of polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) analysis. Results The risk for colorectal cancer did not appear to differ significantly amongst individuals featuring the XRCC1 399Arg/Arg genotype (OR = 1.18; 95% CI, 0.96–1.45), the XRCC3 241Thr/Thr genotype (OR = 1.25; 95% CI, 0.88–1.79) or the XPD 751Gln allele (OR = 1.20; 95% CI, 0.90–1.61), although individuals featuring a greater number of risk genotypes (genotype with OR greater than 1) did experience a higher risk for colorectal cancer when compared to those who didn't feature any risk genotypes (Trend test P = 0.03). Compared with those individuals who didn't express any putative risk genotypes, individuals featuring all of the putative risk genotypes did experience a significantly greater cancer risk (OR = 2.43, 95% CI = 1.21–4.90), particularly for individuals suffering tumors located in the rectum (OR = 3.18, 95% CI = 1.29–7.82) and diagnosed prior to the age of 60 years (OR = 4.90, 95% CI = 1.72–14.0). Conclusions Our results suggest that DNA-repair pathways may simultaneously modulate the risk of colorectal cancer for the Taiwanese population, and, particularly for rectal cancer and younger patients.
