However, research focusing on this population remains limited.Methods: This study analyzed 17,434 psychiatric inpatients at Huzhou Third Municipal Hospital, incorporating data on demographics, psychiatric diagnoses, physical illnesses, laboratory results, and medication use. Predictive models for DVT were developed using logistic regression, random forest, support vector machine (SVM), and XGBoost (Extreme Gradient Boosting). Feature importance was assessed using the random forest model.Results: The DVT incidence among psychiatric inpatients was 1.6%. Predictive model performance, measured by the area under the curve (AUC), showed logistic regression (0.900), random forest (0.885), SVM (0.890), and XGBoost (0.889) performed well. Logistic regression and random forest models exhibited optimal overall performance, while XGBoost excelled in recall. Significant predictors of DVT included elevated D-dimer levels, age, Alzheimer’s disease, and Madopar use.Conclusion: Psychiatric inpatients require vigilance for DVT risk, with factors like D-dimer levels and age serving as critical indicators. Questionnaires were administered at two-time points (T1: March 1– 31, 2022; T2: March 1– 31, 2023), which included an Insomnia Severity Index (ISI) and questions related to sleep risk factors, including the COVID-19 pandemic, familial influences, work and study conditions, social activities, physical health, use of electronic devices before sleep, sleep environment, food intake and exercise before sleep, etc. Insomnia characteristics were compared at two points, with logistic regression testing associations with sociodemographic covariates and risk factors. Six machine learning models were employed to develop a predictive model for insomnia, namely logistic regression, random forest, neural network, support vector machine, CatBoost, and gradient boosting decision tree.Results: The study obtained 2769 and 1161 valid responses in T1 and T2, respectively. The prevalence of insomnia increased from 23.4% in T1 to 34.83% in T2. Univariate analyses indicated the factors of the COVID-19 pandemic, familial influences, social activity, physical health, food intake, and exercise before sleep significantly differed in T1 (p< 0.05) between insomnia and non-insomnia groups. In T2, significant differences (p< 0.05) were observed between the two groups, including the factors of the COVID-19 pandemic, family structure, work and study conditions, social activity, and physical health status. The random forest model had the highest prediction accuracy (90.92% correct and 86.59% correct in T1 and T2, respectively), while the pandemic was the most critical variable at both time points.Conclusion: The prevalence and severity of insomnia have worsened in the post-pandemic period, highlighting an urgent need for effective interventions. Notably, the COVID-19 pandemic and physical health status were identified as significant risk factors for insomnia.Keywords: COVID-19, public, insomnia, risk factors, machine learning

The application of photodynamic therapy (PDT) has aimed at destroying tumor cells and microbiological control in infections. Experimentation in vitro with individual elements of a biological system enables the study of mechanism of action and provides directions to in vivo applications where cancer coexists with normal cells and microorganisms, whether opportunistic or not. The action of the combined set of elements leads to a different behavior from each of the components isolated. In this study, we created a simple model to investigate the effects of PDT (1) on the coculture of macrophages (2) and melanoma cells (3) infected with E. coli (4) under different combinations. Comparing the 4 individual elements of the system interacting with each other to all of them interacting together, we found that macrophage phagocytosis depends on the order in which the elements are exposed to PDT. That indicates the behavior of macrophages depends on their relationship with the microenvironment. Applying the photodynamic effect to each component separately or to all of them together results in different outcomes. These in vitro experiments provide pathways to understand or to design new and more efficient applications of PDT.

We aimed to compare the baseline characteristics and clinical outcomes after EVT for anterior circulation LVO with LAA and CE in the Chinese population.Methods: Patients were selected from the ANGEL registry and divided into LAA and CE groups. The primary outcome was the 90-day modified Rankin Scale (mRS) 0– 2. The secondary outcomes were 90-day mRS distribution, 90-day mRS 0– 1, 90-day mRS 0– 3, and early neurological improvement. The safety outcomes included death, symptomatic intracranial hemorrhage, and any intracranial hemorrhage. We conducted logistic regression models with adjustments to compare the outcomes.Results: A total of 632 patients were included, of whom, 488 were in the LAA group and 144 were in the CE group. No significant difference in 90-day mRS 0– 2 was observed between LAA and CE groups (55.7%vs.43.1%, odds ratio[OR] 1.19, 95% confidence interval(CI), 0.92– 1.53, P=0.190). The LAA group exhibited a higher frequency of mRS 0– 3 compared to the CE group (69.1% vs 32.6%, OR1.32, 95% CI 1.02– 1.72, P=0.038). However, the incidence of death within 90 days did not significantly differ between the LAA and CE groups (10.9%vs.24.3%, OR0.91, 95% CI0.66– 1.25, P=0.545), nor did the occurrences of symptomatic intracranial hemorrhage(SICH) (4.5%vs.9.7%,OR1.08, 95% CI 0.65– 1.78, P=0.779) or intracranial hemorrhage(ICH) (21.9%vs.30.6%, OR 0.94, 95% CI0.71– 1.25, P=0.680). Moreover, no significant disparities were detected in other outcomes between the two groups (All P> 0.05).Conclusion: In the ANGEL registry, a higher prevalence of patients undergoing EVT for acute anterior circulation LVO with LAA was found than those with CE. Changes in the vaginal microecological environment lead to multiple gynecological diseases, including cervical cancer. Recent research has shown that Lactobacillus may play an important role in the occurrence and development of cervical cancer. This review explores the role of Lactobacillus in cervical cancer. A total of 29 articles were included after identification and screening. The pertinent literature on Lactobacillus in cervical cancer from two perspectives, including clinical studies and experimental studies, was analyzed. An association network for the mechanism by which Lactobacillus induces cervical cancer was constructed. The structural and dielectric properties of BaMoO4 - CaTiO3 advanced ceramics in the Radio Frequency (RF) region (< 300 MHz) were studied in the present work. X-ray diffraction and Rietveld refinement showed no evidence of secondary phases, indicating no reaction between the BaMoO4 (BMO) and CaTiO3 (CTO) phases. Complex Impedance Spectroscopy (CIS) analysis demonstrated that BMO presents greater permittivity than composites can be explained by the low densification of the composites. Nyquist diagrams were fitted employing an equivalent circuit with two associations of R-CPE (constant phase element) related to the grain and the grain boundary effects. The composites showed a decrease in the values of Temperature Capacitance Coefficient (TCC) with CTO (CaTiO3) content increase, whereas composites BMO20 and BMO40 showed TCC values equal to zero at 1 MHz. Moreover, as obtained by CIS analysis, the activation energy of thermo-activated processes decreases with CTO concentration increase. [ABSTRACT FROM AUTHOR]

Serological tests are critical tools in the fight against infectious disease. They detect antibodies produced during an adaptive immune response against a pathogen with an immunological reagent, whose antibody binding characteristics define the specificity and sensitivity of the assay. While pathogen proteins have conveniently served as reagents, their performance is limited by the natural grouping of specific and non-specific antibody binding sites, epitopes. An attractive solution is to build synthetic proteins that only contains pathogen-specific epitopes, which could theoretically reach 100% specificity. However, the genesis of de novo proteins remains a challenge. To address the uncertainty of producing a synthetic protein, we have repurposed the beta barrel of fluorescent proteins into a receptacle that can receive several epitope sequences without compromising its ability to be expressed. Here, two versions of a multiepitope protein were built using the receptacle that differ by their grouping of epitopes specific to the parasite Trypanosoma cruzi, the causative agent for Chagas disease. An evaluation of their performance as the capture reagent in ELISAs showed near-complete agreement with recommended diagnostic protocols. The results suggest that a single assay could be developed for the diagnosis of Chagas disease and that this approach could be applied to other diseases. [ABSTRACT FROM AUTHOR]

The present work shows the dielectric properties of (100-X)Ba2TiSi2O8–(X)TiO2 composites (X in wt%) at radio frequency (RF) region (< 300 MHz). X-ray diffraction and Rietveld's refinement showed the presence of Ba2(Ti7/9Si2/9)9O20 (BTSO) phase in the studied composites. To analyze the relationship between the electrical properties and the microstructure of the matrix and composites, Impedance spectroscopy (IS) was used. The applied model that best describes the behavior found in the samples is the Havriliak–Negami model. From the IS it was observed that the dielectric relative permittivity presented high values in BTS10 (X = 10%) and BTS20 (X = 20%) which allows classifying them as Colossal permittivity materials. This is an important result due to the potential for applications in new storage systems, capacitive devices and microelectronics. Nyquist diagrams were employed to evaluate the contribution of the grain and grain boundary in the electrical response of the samples and fitted employing an equivalent circuit with two associations of R-CPE (constant phase element). Temperature coefficient of capacitance (TCC) was also obtained and it was possible to observe that there was an increase in the TCC value for the compounds in all frequencies. In addition, IS demonstrated that the activation energy decreasing with TiO2 addition indicating a decrease in the resistive character of the materials. [ABSTRACT FROM AUTHOR]

Objective: This retrospective case series aimed to assess the stability of the papilla around four single crowns supported by narrow‐diameter implants replacing all maxillary incisors. Secondary objectives included assessment of marginal bone level stability, incidence of technical and biological complications, and patient satisfaction. Materials and Methods: Individuals with four adjacent implants in maxillary incisor sites, placed with a 3 mm inter‐implant distance and rehabilitated with single crowns were included. Retrospective data were obtained from photographs and radiographs taken at the delivery of the prosthesis (baseline—T0). Patients were then recalled (≥2 years after T0) for clinical and radiographic examination (follow‐up—T1). Photographs were obtained and patient satisfaction was assessed using a visual analogue scale. Papilla height and marginal bone level were compared over time. Results: Data from 10 patients with medium‐low smile lines and rehabilitated with 40 implants, in function for 5.4 ± 1.9 years, were analyzed. The papilla height between implants (T0: 2.3 ± 0.9 mm; T1: 2.6 ± 0.7 mm; p =.011) and between tooth and implant (T0: 3.4 ± 0.9 mm; T1: 3.8 ± 0.8 mm; p =.025) increased significantly over the years. The marginal bone level remained stable over time (T0: 0.88 ± 0.57 mm; T1: 0.71 ± 0.67 mm; p =.007). Patients were highly satisfied (97.7 ± 0.3%) with the treatment outcome. Conclusion: Within its limitations, this study demonstrated that four single implant‐supported crowns placed at maxillary incisor sites may exhibit soft tissue and marginal bone stability over a long period of time. This treatment approach, however, should be restricted to few patients as it requires a proper case selection and skillful execution of all surgical and prosthetic steps. [ABSTRACT FROM AUTHOR]

This work studies the dielectric properties in the microwave region (MW) of the Ba2TiSi2O8 (BTS) ceramic with TiO2 additions and its applications as a dielectric resonator antenna (DRA). In this study, structural characterization through x-ray diffraction (XRD) is performed and the Rietveld refinement is used to confirm the phases formed. Analysis of the morphology of the materials is performed using scanning electron microscopy (SEM). The resonant frequency temperature coefficient (τf) reveals a variation from − 47.0 ppm°C−1 to + 16.5 ppm°C−1. The dielectric properties in the MW region reveal an increase in the dielectric permittivity (εr) and a decrease in the loss tangent (tanδ) of the samples. Numerical simulation shows good fits of the experimental data, with gain and directivity standing out, ranging from 4 dBi to 6 dBi and radiation efficiency below 80%. Abstract Clinically, the body mass index remains the most frequently used metric of overall obesity, although it is flawed by its inability to account for different adipose (i.e., visceral, subcutaneous, and inter/intramuscular) compartments, as well as muscle mass. Numerous prior studies have demonstrated linkages between specific adipose or muscle compartments to outcomes of multiple diseases. Although there are no universally accepted standards for body composition measurement, many studies use a single slice at the L3 vertebral level. In this study, we use computed tomography (CT) studies from patients in The Cancer Genome Atlas (TCGA) to compare current L3-based techniques with volumetric techniques, demonstrating potential limitations with level-based approaches for assessing outcomes. In addition, we identify gene expression signatures in normal kidney that correlate with fat and muscle body composition traits that can be used to predict sex-specific outcomes in renal cell carcinoma.

Melanophoroma is a neoplasm of reptilian pigment cells, considered uncommon and part of a group of neoplasms called chromatophoromas. The objective of this work was to describe a case of melanophoroma in a free‐living Lichtenstein's green racer snake (Philodryas olfersii), presenting with an ulcerative nodular neoformation in the integument of the head region. In the neurologic evaluation, a proprioceptive deficit was observed. Ultrasound, X‐ray, and mass cytology examinations were performed. Radiographic and ultrasound findings of the tumor indicated infiltrative behavior, and cytology indicated a presumptive diagnosis of a melanocytic neoplasm. Based on the results of the examinations and the patient's clinical condition, euthanasia was chosen. At necropsy, there was a nodule measuring 4.5 × 2.5 × 2.0 cm, with a blackish‐colored cut surface, mottled by gray areas, which infiltrated soft tissue and bone, and extended dorsally from the mouth to the cervical musculature. On microscopy, a non‐delimited, non‐encapsulated, and invasive neoplastic proliferation was observed, with moderate cellularity, which was predominantly composed of fusiform cells with distinct borders and foci of epithelioid cells. The cells had ample cytoplasm, which had a brown to black (melanocytic) granular pigment compatible with a melanophoroma. Summary: Background: MET exon14 skipping mutations (METex14) is an established actionable driver oncogene of non-small-cell lung cancer (NSCLC). While ensartinib is a known second-generation tyrosine kinase inhibitor with primary activity against ALK translocation, it is also classified as a type Ia MET inhibitor. We have previously shown anti-tumor activity against METex14 positive NSCLC both in vivo and in vitro. The EMBRACE trial aims to evaluate the clinical efficacy and safety of ensartinib for treatment of METex14 positive NSCLC. Methods: This is a multicenter single arm phase II investigator-initiated study that enrolled METex14 positive lung cancer after failing first line chemotherapy and/or immunotherapy. Eligible patients received ensartinib 225 mg orally once daily in a continuous 28-day treatment cycle until disease progression, unacceptable side effect, or death. Primary endpoint was investigator-assessed objective response rate (ORR), and the secondary end point included disease control rate (DCR), progression-free survival (PFS), duration of response (DoR) and safety profiles. The study was registered with the Chinese Clinical Trial Registry (ChiCTR2100048767). Findings: From July 2021 to February 2024, a total of 31 patients were enrolled and received ensartinib. Median follow-up time of the 30 evaluable patients was 9.2 months (95% Confidence Interval [CI], 6.3-not estimable). The ORR was 53.3% (16/30; 95% CI, 35.5–71.2) and DCR was 86.7% (26/30; 95% CI, 74.5–98.8). Median PFS was 6.0 months (95% CI, 3.0–8.8) and median DoR was 7.9 months (95% CI, 4.8–8.7). Adverse events (AEs) were reported in 24 patients (80%), with 7 (23.3%) of grade 3. The most common AEs were rash (14/30, 46.7%), followed by anemia (7/30, 23.3%), increased ALT (7/30, 23.3%), increased AST (7/30, 23.3%), and pruritus (6/30, 20%). No serious adverse events or treatment-related deaths occurred. Importantly, the exploratory ctDNA analysis indicates that clearance of circulating tumor DNA (ctDNA) at four weeks treatment was associated with more favorable treatment outcomes comparing with patients having positive ctDNA. Interpretation: Ensartinib has a promising anti-tumor activity and manageable safety in previously treated patients with METex14 positive lung cancer. This article evaluates a diagnostic algorithm for mastocytosis in skin biopsies and CD25 expression in skin mast cells of 49 patients at a Tertiary Hospital in Sao Paulo, Brazil. Mastocytosis is characterized by the accumulation of neoplastic mast cells in the skin or other organs. The study found that the algorithm was able to correctly diagnose all mastocytosis cases, but the significance of CD25 expression in skin mast cells is still unclear. Further research is needed to identify skin mast cell markers that can correlate with disease chronicity and severity. [Extracted from the article]

This research aimed to analyze the perception of tourism stakeholders regarding the potential of gastronomy to revitalize tourism in the Mirandela region. The sample consisted of 95 stakeholders (e.g., managers, team leaders, directors, entrepreneurs) operating in the tourism sector. The study was operationalized using a qualitative methodology, and the data were analyzed using MAXQDA software. The results demonstrated that food quality, identity gastronomy, and food heritage are considered differentiating factors of the local cuisine. Additionally, it was found that diversification of tourism offerings, variety of traditional dishes, and multiplier effect are considered essential elements to enhance local gastronomy and boost tourism in the region. It was possible to ascertain that the promotion of local gastronomy as a tourist product depends on the organization of gastronomic events, preservation of culture, investment in digital marketing, and immersive experiences. Finally, it was observed that among the main measures to position the region as a gastronomic tourist destination are support from the private sector, synergies among local stakeholders, investment in infrastructure, and integration of local gastronomy with other tourism activities.

Abstract Background Low anterior resection syndrome (LARS) is a distressing condition that affects approximately 25–80% of patients following surgery for rectal cancer. LARS is characterized by debilitating bowel dysfunction symptoms, including fecal incontinence, urgent bowel movements, and increased frequency of bowel movements. Although biofeedback therapy has demonstrated effectiveness in improving postoperative rectal control, the research results have not fulfilled expectations. Recent research has highlighted that stimulating the pudendal perineal nerves has a superior impact on enhancing pelvic floor muscle function than biofeedback alone. Hence, this study aims to evaluate the efficacy of a combined approach integrating biofeedback with percutaneous electrical pudendal nerve stimulation (B-PEPNS) in patients with LARS through a randomized controlled trial (RCT). Methods and analysis In this two-armed multicenter RCT, 242 participants with LARS after rectal surgery will be randomly assigned to undergo B-PEPNS (intervention group) or biofeedback (control group). Over 4 weeks, each participant will undergo 20 treatment sessions. The primary outcome will be the LARS score. The secondary outcomes will be anorectal manometry and pelvic floor muscle electromyography findings and the European Organization for the Research and Treatment of Cancer Quality of Life Questionnaire-Colorectal 29 (EORTC QLQ-CR29) scores. Data will be collected at baseline, post-intervention (1 month), and follow-up (6 months). Discussion We anticipate that this study will contribute further evidence regarding the efficacy of B-PEPNS in alleviating LARS symptoms and enhancing the quality of life for patients following rectal cancer surgery. This paper reviews the dielectric properties of Sr2TiSi2O8 (STS) ceramic in the radio frequency (RF) and microwave (MW) regions. X-ray diffraction (XRD) analysis is used to demonstrate that a small secondary phase of SrTiO3 is present in a synthesised STS ceramic. Complex impedance spectroscopy is performed, and the typical permittivity values of ceramic materials are observed. Nyquist diagrams are fitted based on an equivalent circuit using two associations of R-CPE related to the grain and the grain boundary effects. A dependence study of the changes in AC conductivity with frequency at different temperatures demonstrates that the conduction process is thermally activated, and a value of 1.0 eV is obtained for the activation energy. In the MW range, we observe values of εr = 12.7, tan δ = 1.3 × 10–2, and τf = − 6.1 ppm/°C, indicating interesting properties for applications in devices that operate in the MW region. A numerical simulation is employed to evaluate STS ceramic as a dielectric resonator antenna, and we observe a reflection coefficient of below − 10 dB, a realised gain of 4.6 dBi, a bandwidth of 475 MHz, and a radiation efficiency of around 75%. The properties of the STS matrix presented here indicate that this ceramic would be a suitable candidate for the operation of devices in the C-band, as well as in devices operating in the RF range. Abstract INTRODUCTION Neighborhood disadvantage may be an important determinant of cardiometabolic health and cognitive aging. However, less is known about relationships among individuals with mild cognitive impairment (MCI). METHODS The objective of this study is to investigate the relationship between neighborhood disadvantage measured by national Area Deprivation Index (ADI) rank with measures of cardiometabolic health and cognition among Wake Forest (WF) Alzheimer's Disease Research Center (ADRC) participants, with and without MCI. RESULTS ADI was positively associated with blood pressure and cardiometabolic index (CMI), and negatively associated with global and Preclinical Alzheimer's Cognitive Composite (PACC5) scores, in cognitively unimpaired (CU) individuals. ADI was only positively associated with hemoglobin A1c (HbA1c) in MCI. DISCUSSION Neighborhood disadvantage is associated more strongly with measures of cardiometabolic health and cognition among CU individuals rather than MCI. These findings demonstrate a need for structural solutions to address social determinants of health in an attempt to reduce cardiometabolic and cognitive risks.

BackgroundPatients with locally advanced non-small cell lung cancer (LA-NSCLC) usually bear high tumor burden and are not tolerated well to concurrent chemoradiation therapy (CRT) followed by consolidation immunotherapy. We investigated the feasibility of chemoimmunotherapy as induction therapy before CRT for LA-NSCLC.MethodsWe retrospectively analyzed data from 91 patients with unresectable stage III NSCLC treated with either induction chemoimmunotherapy or chemotherapy before CRT. Tumor responses, survival statistics, and toxic effects were compared. The dosimetric parameters of the RT protocol were evaluated. The primary endpoint was progression-free survival (PFS). The overall response (ORR), the depth of response (DpR) were accessed at the end of CRT (ORRinduc+CRT, DpRinduc+CRT) and induction therapy (ORRinduc, DpRinduc).ResultsThe median PFS (mPFS) were significantly longer in the chemoimmunotherapy induction group (13.5 months vs. 11.2 months; HR, 0.56; 95% CI, 0.32–0.97; p=0.036). The ORRinduc+CRT, median DpRinduc+CRT (mDpRinduc+CRT) and mDpRinduc were significantly higher in the chemoimmunotherapy induction group (ORRinduc+CRT, 84.0% vs. 65.9%, p=0.044; mDpRinduc+CRT, 49.5% vs. 39.0%, p = 0.012; mDpRinduc, 38.5% vs. 28.0%, p=0.044). Incidence of treatment-related adverse events (AE) was similar between groups, with myelosuppression being the most common grade ≥ 3 AE. Regarding radiotherapy, adopting a mapping strategy with a 5–8 mm margin for clinical tumor volume resulted in decreased radiation doses to critical organs in the chemoimmunotherapy induction group.ConclusionsChemoimmunotherapy induction therapy before CRT improves efficacy with comparable incidence of AEs compared to chemotherapy induction in LA-NSCLC patients. Further studies are warranted to validate these findings.

Objetivo: Descrever um caso de satelitismo plaquetário como causa de pseudotrombocitopenia detectado na rotina de um grande laboratório. Introdução: O satelitismo plaquetário é um fenômeno raro que ocorre somente in vitro e pode ser visto à hematoscopia de indivíduos saudáveis ou com outras patologias, inclusive hematológicas. Caracteriza-se por trombocitopenia espúria ao hemograma completo e pela disposição de grande número de plaquetas em torno de neutrófilos na lâmina de esfregaço de sangue periférico preparados a partir da amostra colhida em tubo com Ácido EtilenoDiamonoTetra-Acético de sódio (EDTA). A formação dessas rosetas plaquetárias pode ser ainda observada ocasionalmente em torno de outras células como eosinófilos, basófilos, linfócitos ou monócitos. Embora o mecanismo que leva a essa alteração não seja plenamente compreendido, acredita-se que o EDTA possa agir diretamente modificando proteínas de membrana das plaquetas e dos leucócitos, resultando em pontes de interação entre ambos. Há, ainda, a possibilidade de ação indireta promovida pelo EDTA expondo criptoantígenos plaquetários que poderiam então interagir com autoanticorpos contra epítopos de plaquetas e neutrófilos simultaneamente. Relato de caso: Em novembro de 2023, deu entrada em nossa área técnica do Rio de Janeiro amostra colhida em tubo EDTA para a realização de hemograma completo de paciente do sexo masculino, 25 anos, sem comorbidades. O analisador XN-Sysmex liberouo resultado com contagem de plaquetas de 98.000/mm3, com sinalização de presença de grumos. Seguindo a rotina de nosso laboratório, amostra foi encaminhada para distensão automatizada e análise no equipamento CellaVision. À análise da hematoscopia, observamos numerosos neutrófilos com grande quantidade de plaquetas ao redor de seu citoplasma com aspecto de rosetas, configurando o satelitismo plaquetário como causa provável da trombocitopenia espúria. Como não dispúnhamos de amostra coletada em anticoagulante diferente (citrato ou CPDA) e não tínhamos acesso direto ao cliente para realizarmos distensão direta a partir de digito-punção, optamos por submeter a amostra ao agitador de alta frequência (Digital Vortex – ThermoScientific), com a correção da contagem plaquetária para a faixa da normalidade de 223.000/mm3. Discussão: A trombocitopenia é alteração frequentemente encontrada nos ambulatórios de Hematologia. As etiologias vão desde autoimunes, passando por doenças metabólicas / carenciais até falências medulares. No entanto, é fundamental a exclusão das trombocitopenias espúrias, principalmente em reduções leves a moderadas das contagens. Isso evita investigações desnecessárias, procedimentos invasivos, custos em saúde e estresse psicológico nos pacientes. Apesar do satelitismo ser evento raro, a pseudotrombocitopenia é relativamente frequente e deve estar sempre no leque de diagnósticos diferenciais. Conclusão: Seja em indivíduos saudáveis ou em pacientes portadores de doenças hematológicas ou autoimunes, a trombocitopenia espúria pode levar à investigação desnecessária ou, em casos mais graves, a procedimentos mal indicados. Por essa razão, a análise da hematoscopia por técnicos experientes e hematologistas deve sempre ser realizada como o primeiro passo na investigação de trombocitopenias detectadas ao hemograma completo.

Objetivo: Analisar in silico as atividades biológicas de compostos químicos da própolis com potencial anticancerígenas. Metodologia: Através da plataformas PubChem foi realizado a busca dos compostos químicos específicos da própolis e direcionados ao Pass Online, para obter dados sobre atividades biológicas e farmacológicas. Os compostos selecionados e coletados pelo SMILES canônicos (estrutura de uma molécula) foram classificados de acordo com PA (Probabilidade de atividade) e PI (Probabilidade de inatividade), com PA > 0,7 sendo os compostos: 7-Hidroxiflavanona, Pinocembrina, Pinostrobina e Pinobanksin. Discussão: No estudo conduzido por Moghadari Masoud et al. (2022), foi explorado o potencial antitumoral da própolis de Kerman como tratamento das células NALM-6 com concentrações variáveis de própolis de Kerman e doxorrubicina durante 72 horas. Os resultados demonstraram que tanto a própolis de Kerman quanto a doxorrubicina induziram a morte celular por apoptose de forma dose-dependente, com uma regulação significativa da razão Bax/Bcl-2, sugerindo um efeito pró-apoptótico. O que endossa os estudo dirigido por Franchi Jr. et al. (2011), em que investigaram o efeito dos extratos etanólicos de própolis grupo 12 (G12) e própolis grupo 13 (G13) em células de leucemia humana, em que os valores de IC50 para os extratos de própolis vermelha (G13) e verde (G12) variaram significativamente entre diferentes linhagens de células de leucemia. A própolis vermelha demonstrou ser mais potente em termos citotóxicos, especialmente nas células K562 e HL60, e apresentou menor dispersão nos valores de IC50. Além disso, ao comparar o efeito da própolis vermelha com os fármacos clínicos Gleevec e citarabina, foi identificada uma diferença marcante nas células K562 e Nalm16, com a G12 mostrando um efeito citotóxico mais significativo. Conclusão: A composição química da própolis pode ser útil para linhagem de leucemia linfoblástica aguda (NALM-6), o que envolve principalmente compostos que possuem atividades anticancerígenas. Esses compostos podem interferir no ciclo celular, induzir apoptose e reduzir o estresse oxidativo nas células cancerígenas com potencial para afetar as células NALM-6.

Abstract Background The aggregation and spread of misfolded amyloid structured proteins, such as tau and α-synuclein, are key pathological features associated with neurodegenerative disorders, including Alzheimer’s and Parkinson’s disease. These proteins possess a prion-like property, enabling their transmission from cell to cell leading to propagation throughout the central and peripheral nervous systems. While the mechanisms underlying their intracellular spread are still being elucidated, targeting the extracellular space has emerged as a potential therapeutic approach. The glymphatic system, a brain-wide pathway responsible for clearing extracellular metabolic waste from the central nervous system, has gained attention as a promising target for removing these toxic proteins. Methods In this study, we investigated the impact of long-term modulation of glymphatic function on tau aggregation and spread by chronically treating a mouse model of tau propagation with a pharmacological inhibitor of AQP4, TGN-020. Thy1-hTau.P301S mice were intracerebrally inoculated with tau into the hippocampus and overlying cortex, and subsequently treated with TGN-020 (3 doses/week, 50 mg/kg TGN-020, i.p.) for 10-weeks. During this time, animal memory was studied using cognitive behavioural tasks, and structural MR images were acquired of the brain in vivo prior to brain extraction for immunohistochemical characterisation. Results Our findings demonstrate increased tau aggregation in the brain and transhemispheric propagation in the hippocampus following the inhibition of glymphatic clearance. Moreover, disruption of the glymphatic system aggravated recognition memory in tau inoculated mice and exacerbated regional changes in brain volume detected in the model. When initiation of drug treatment was delayed for several weeks post-inoculation, the alterations were attenuated. Conclusions These results indicate that by modulating AQP4 function and, consequently, glymphatic clearance, it is possible to modify the propagation and pathological impact of tau in the brain, particularly during the initial stages of the disease. These findings highlight the critical role of the glymphatic system in preserving healthy brain homeostasis and offer valuable insights into the therapeutic implications of targeting this system for managing neurodegenerative diseases characterized by protein aggregation and spread.
