American lotus (Nelumbo lutea) is one of the two species in Nelumbo and has only yellow flower. Identification of total flavonoids showed wild American lotus contained almost only flavonols with quercetin 3-O-glucuronide to be the dominant pigment. The variation tendency of the total flavonol content was coincident with yellow color variation of petals during flower development. To understand the mechanism of accumulation and constituent of pigments in petals, three pivotal genes, NlFLS1, NlFLS2 and NlFLS3, which were predicted to encode flavonol synthases were isolated and characterized by analyses of basic bioinformatics, temporal and spatial expression patterns and enzymatic activity. Their temporal expression levels showed the same variation tendency, which was also consistent with the development-dependent variation of total flavonol content. Spatial expression patterns indicated the three genes should function in petals. All the three proteins were demonstrated to be bifunctional dioxygenase, possessing both flavonol synthase activity and flavanone 3-hydroxylase activity. Besides, other flavonol biosynthesis related genes were also investigated on their expression levels to give more clues on the mechanism. Substrate preferences of the three FLSs, substrate competitions between the FLSs and other flavonol biosynthesis related enzymes, and the greatly differential expression levels between F3’H (flavonoid 3′-hydroxylase) and F3’5’H (flavonoid 3′,5′-hydroxylase) contributed to the flavonol constituent in the petals of America lotus, namely abundant quercetin-derivatives while very few kaempferol-derivatives and myricetin-derivatives.

Asian lotus (Nelumbo nucifera Gaertn.) is an aquatic plant with ornamental, cultural, economic, and ecological values. China has abundant germplasm resources of Asian lotus. However, in many areas, the wild Asian lotuses have been destroyed and the germplasms is now facing extinction. In addition, the knowledge of the genetic diversity of the wild Asian lotus in China is poor. To identify and protect the germplasms of Chinese wild Asian lotus, eleven genomic-SSR primers, three EST-SSR primers, and three chloroplast DNA primers were used to investigate the genetic diversity among 69 samples of wild Asian lotus from 25 locations in northern China. The genetic diversity of 27 samples of wild Asian lotus from southern China and other countries, the ancient Asian lotus, Asian lotus cultivars from China, and Asian-American hybrids was also compared. The genetic diversity of the wild Asian lotus from northern China was characterized as medium, and the mean values of observed heterozygosity (Ho) and expected heterozygosity (He) were 0.087 and 0.552, respectively. Based on a UPGMA dendrogram and STRUCTURE analysis, the wild Asian lotus samples in northern China were divided into three groups. The wild Asian lotus samples from northern China contained 16 haplotypes. The Nei's genetic distance between the wild Asian lotus samples from the Songhua River basin and the Liao River basin in northeastern China was relatively small, and these germplasms might be relatively primitive compared to those from other regions. This study provides essential information regarding the genetic diversity of the wild Asian lotus resources in northern China, and provides a basis for further analysis of population-level genetic evolution through high-throughput sequencing.

Lotus (Nelumbo nucifera) is a large aquatic plant that accumulates pharmacologically significant benzylisoquinoline alkaloids (BIAs). However, little is known about their biosynthesis and regulation. Here, we show that the two group III WRKY transcription factors (TFs), NnWRKY70a and NnWRKY70b, positively regulate the BIA biosynthesis in lotus. Both NnWRKY70s are jasmonic acid (JA) responsive, with their expression profiles highly correlated to the BIA concentration and BIA pathway gene expression. A dual-luciferase assay showed that NnWRKY70a could transactivate the NnTYDC promoter, whereas NnWRKY70b could activate promoters of the three BIA structural genes, including NnTYDC, NnCYP80G, and Nn7OMT. In addition, the transient overexpression of NnWRKY70a and NnWRKY70b in lotus petals significantly elevated the BIA alkaloid concentrations. Notably, NnWRKY70b seems to be a stronger BIA biosynthesis regulator, because it dramatically induced more BIA structural gene expressions and BIA accumulation than NnWRKY70a. A yeast two-hybrid assay further revealed that NnWRKY70b physically interacted with NnJAZ1 and two other group III WRKY TFs (NnWRKY53b and NnWRKY70a), suggesting that it may cooperate with the other group III WRKYs to adjust the lotus BIA biosynthesis via the JA-signaling pathway. To illustrate the mechanism underlying NnWRKY70b-mediated BIA regulation in the lotus, a simplified model is proposed. Our study provides useful insights into the regulatory roles of WRKY TFs in the biosynthesis of secondary metabolites.

Most waterlily flowers open at dawn and close after noon usually for three to four days, and thereafter wilt. The short lifespan of flowers restricts the development of the flower postharvest industry. The termination of flower movements is a key event during flower aging process. However, it is still unclear when the senescence process initiates and how it terminates the movement rhythm. In this study, we observed that the opening diameter of flowers was the smallest on the fourth (last) flowering day. Subsequent transcriptome profiles generated from petals at different flowering stages showed that the multiple signaling pathways were activated at the last closure stage (Time 3, T3) of the flowers, including Ca2+, reactive oxygen species and far red light signaling pathways, as well as auxin, ethylene and jasmonic acid signaling pathways. Moreover, In terms of cell metabolism regulation, the genes related to hydrolase (protease, phospholipase, nuclease) were upregulated at T3 stage, indicating that petals entered the senescence stage at that time; and the genes related to water transport and cell wall modification were also differentially regulated at T3 stage, which would affect the ability of cell expand and contract, and eventually lead to petal not open after the fourth day. Collectively, our data provided a new insight into the termination of flower opening in the waterlilies, and a global understanding of the senescence process of those opening-closure rhythm flowers.

Begonia, one of the most diverse plant taxa and the fifth or sixth largest angiosperm genus, consists of over 1800 accepted species. The number of species recognized within this genus has greatly increased over the past 20 years, rising from 80 to 200 species in China alone. Based on recent field surveys, the number of begonia species in China is predicted to be between 250 and 300. Given the large number of begonia species that still remain to be described, further taxonomical work is urgently required. This is especially true for Chinese Begonia, in which there is a huge diversity of habitat, habit, plant size, leaf type, flower and fruit morphology, and most species are narrowly distributed in isolated habitats that are subject to negative disturbances from climate change, as well as agricultural and industrial activities. Although the conservation status for the majority of species has been evaluated using the standards of the International Union for Conservation of Nature, the results don't represent the truth in many species, and also about 11.5% of which are data-absent. In addition, illegal collection and over-harvesting of wild begonias for ornamental or medicinal use has increased due to the rapid development of internet commerce. Far more often than predicted, these species should be categorized as rare and endangered and require immediate protection. Ex situ conservation of Chinese begonias started in 1995 and over 60% of the total species have been so far introduced into cultivation by several major botanical gardens in China. However, only few research institutions, limited funds and human resources have been involved in Begonia conservation; moreover, no project has conducted reintroduction. Therefore, more conservation-based work remains to be done. Improved conservation of Chinese begonias in the future depends on further field survey, an improved understanding of population diversity, and integrative approaches, including in situ and ex situ conservation, seed banking, and plant reintroduction. Species-targeted conservation zones should be established for endangered species excluded from the existing nature reserves. Additionally, laws pertaining to plant protection should be extended to prevent the illegal collection and transaction of wild plants, particularly for those species with unique habitats and small populations. Compared with rose, chrysanthemum, and water lily, the absence of short-wide and long-narrow tepals of ornamental lotus (Nelumbo Adans.) limits the commercial value of flowers. In this study, the genomes of two groups of lotus mutants with wide-short and narrow-long tepals were resequenced to uncover the genomic variation and candidate genes associated with tepal shape. In group NL (short for N. lutea, containing two mutants and one control of N. lutea), 716,656 single nucleotide polymorphisms (SNPs) and 221,688 insertion-deletion mutations (Indels) were obtained, while 639,953 SNPs and 134,6118 Indels were obtained in group WSH (short for ‘Weishan Hong’, containing one mutant and two controls of N. nucifera ‘Weishan Hong’). Only a small proportion of these SNPs and Indels was mapped to exonic regions of genome: 1.92% and 0.47%, respectively, in the NL group, and 1.66% and 0.48%, respectively, in the WSH group. Gene Ontology (GO) analysis showed that out of 4890 (NL group) and 1272 (WSH group) annotated variant genes, 125 and 62 genes were enriched (Q < 0.05), respectively. Additionally, in the Kyoto Encyclopedia of Genes and Genomes (KEGG) database, 104 genes (NL group) and 35 genes (WSH group) were selected (p < 0.05). Finally, there were 306 candidate genes that were sieved to determine the development of tepal shape in lotus plants. It will be an essential reference for future identification of tepal-shaped control genes in lotus plants. This is the first comprehensive report of genomic variation controlling tepal shape in lotus, and the mutants in this study are promising materials for breeding novel lotus cultivars with special tepals.

The lotus (Nelumbo Adans.) is an important aquatic plant with ornamental, medicinal and edible values and cultural connotations. It has single-, semi-double-, double- and thousand-petalled types of flower shape and is an ideal material for developmental research of flower doubling. The lotus is a basal eudicot species without a morphological difference between the sepals and petals and occupies a critical phylogenetic position in flowering plants. In order to investigate the genetic relationship between the sepals and petals in the lotus, the class E genes which affect sepal formation were focused on and analyzed. Here, SEPALLATA 1(NnSEP1) and its homologous genes AGAMOUS-LIKE MADS-BOXAGL9 (NnAGL9) and MADS-BOX TRANSCRIPTION FACTOR 6-like (NnMADS6-like) of the class E gene family were isolated from the flower buds of the Asian lotus (Nelumbo nucifera Gaertn.). The protein structure, subcellular localization and expression patterns of these three genes were investigated. All three genes were verified to locate in the nucleus and had typical MADS-box characteristics. NnSEP1 and NnMADS6-like were specifically expressed in the sepals, while NnAGL9 was highly expressed in the petals, suggesting that different developmental mechanisms exist in the formation of the sepals and petals in the lotus. The significant functional differences between NnSEP1, NnMADS6-like and NnAGL9 were also confirmed by a yeast two-hybrid assay. These results expand our knowledge on the class E gene family in sepal formation and will benefit fundamental research on the development of floral organs in Nelumbo.

Asian lotus (Nelumbo nucifera Gaertn.) is the national flower of India, Vietnam, and one of the top ten traditional Chinese flowers. Although lotus is highly valued for its ornamental, economic and cultural uses, genomic information, particularly the expressed sequence based (genic) markers is limited. High-throughput transcriptome sequencing provides large amounts of transcriptome data for promoting gene discovery and development of molecular markers.In this study, 68,593 unigenes were assembled from 1.34 million 454 GS-FLX sequence reads of a mixed flower-bud cDNA pool derived from three accessions of N. nucifera. A total of 5,226 SSR loci were identified, and 3,059 primer pairs were designed for marker development. Di-nucleotide repeat motifs were the most abundant type identified with a frequency of 65.2%, followed by tri- (31.7%), tetra- (2.1%), penta- (0.5%) and hexa-nucleotide repeats (0.5%). A total of 575 primer pairs were synthesized, of which 514 (89.4%) yielded PCR amplification products. In eight Nelumbo accessions, 109 markers were polymorphic. They were used to genotype a sample of 44 accessions representing diverse wild and cultivated genotypes of Nelumbo. The number of alleles per locus varied from 2 to 9 alleles and the polymorphism information content values ranged from 0.6 to 0.9. We performed genetic diversity analysis using 109 polymorphic markers. A UPGMA dendrogram was constructed based on Jaccard's similarity coefficients revealing distinct clusters among the 44 accessions.Deep transcriptome sequencing of lotus flower buds developed 3,059 genic SSRs, making a significant addition to the existing SSR markers in lotus. Among them, 109 polymorphic markers were successfully validated in 44 accessions of Nelumbo. This comprehensive set of genic SSR markers developed in our study will facilitate analyses of genetic diversity, construction of linkage maps, gene mapping, and marker-assisted selection breeding for lotus.

Southern Xizang harbors the highest diversity of plants in Xizang of China, and recently there have been many new species discovered in this region. Based on field surveys and review of literature and herbarial specimens, one new species (Begonia renqinbengensis D.K.Tian & W.G. Wang) and one new record (B. leptoptera Hara) of Begonia are diagnosed and reported in this paper. Also B. leptoptera is redescribed with illustrations. The article provides an overview of the aquatic plant industry in China in 2023, highlighting various lotus festivals and events that took place in different cities. Despite travel restrictions due to the COVID-19 pandemic, Chinese colleagues made significant contributions to the field. These events showcased different cultivars of ornamental lotus and waterlilies, and included exhibitions, symposiums, and cultural activities. China is highly rich in diversity of Begonia, and in which Guangxi Zhuangzu Autonomous Region, following Yunnan province, has the second highest number of Begonia species. As a distribution center of Begonia sect. Coelocentrum, Guangxi already had 58 species recognized in this section. In this paper, Begonia ornithopedata, a new taxon of B. sect. Coelocentrum from southwestern Guangxi is described. With the bird-foot-shape lobed leaves, the new species is easily distinguished from any of other taxa, excepting few similarities only in leaf morphology of B. laminariae Irmsch. and B. circumlobata Hance. China has an exceptional diversity in Begonia, holding the second largest number of described species in this genus at the national level. Many new taxa need to be explored and described. This paper reports two new species, Begonia ruchengensis (B. sect. Reichenheimia) and B. auritalata (B. sect. Platycentrum), from Hunan province and Guizhou province, respectively. Besides the detailed morphology description, color illustrations, and line drawings, the diagnoses of these two species distinguished from their allied taxa are provided. Begonia floriprolifera is a new species of Begonia sect. Coelocentrum distributed in the karst area of southwestern Guangxi of China and northern Vietnam. Morphologically, it is mostly similar to B. bonii, but differs mainly by its nearly glabrous stipules, shorter inflorescence, bilaterally symmetric and upward curved androecium with fewer stamens, and concave apex of anthers. Due to the narrow distribution area and unfavorably environmental changes of its natural habitat, the new species is assigned to Endangered according to the IUCN Red List Categories and Criteria. Information about the topics discussed at the 26th National Lotus Show and International Symposium on Nelumbo that was held at Guyi Garden in Shanghai, China from July 6-7, 2012 is presented. Topics include the culture and history of lotus in the country, the color pigment formation in waterlilies, and the applications of water plants. The event also featured new cultivars of aquatic plants, particularly water lilies and lotuses.

Information on the First Symposium on Lotus Breeding and International Nelumbo Registration held in Shanghai, China on November 22-24, 2013 is presented. Topics include the principles of lotus breeding and registration, the methods of cultivating plants, and the challenges of international Nelumbo registration. The symposium featured several speakers including professors Qibai Xiang and Xiaobai Jin, and Yuchu Chen, vice-president of Zhejiang Cultural Garden Co. Ltd.

The article discusses the potential of the new on-farm American lotus or Nelumbo lutea at the Lake Eufaula National Wildlife Refuge in Alabama. The scientists have found that the American lotus has been treated as a sacred plant with mystical powers by the Native American Indian tribes who also used parts of the plant as a supplemental food. However, the American lotus is listed as a potentially invasive and noxious weed and has been banned in Connecticut.
