In this study, based on three tumor samples obtained from patients with sporadic vestibular schwannoma, 32,011 cells were obtained by single-cell transcriptome sequencing, and 22,309 high-quality cells were obtained after quality control and double cells removal. Then, 18 cell clusters were obtained after cluster analysis, and each cluster was annotated as six types of cells. Afterward, an in-depth analysis was conducted based on the defined six cell clusters, including characterizing the functional characteristics of each cell subtype, describing the cell development and differentiation pathway, exploring the interaction between cells, and analyzing the transcriptional regulatory network within the clusters. Based on these four dimensions, various types of cells in sporadic vestibular schwannoma tumor tissues were described in detail. For the first time, we expanded on the functional state of cell clusters that have been reported and described Schwann cells in the peripheral nervous system, which have not been reported in previous studies. Combined with the data of sporadic vestibular schwannoma and normal tissues in the gene expression omnibus (GEO) database, the candidate biomarkers of sporadic vestibular schwannoma were explored. Overall, this study described the single-cell map of sporadic vestibular schwannoma for the first time, revealing the functional state and development trajectory of different cell types. Combined with the analysis of data in the GEO database and immunohistochemical verification, it was concluded that HLA-DPB1 and VSIG4 may be candidate biomarkers and potential therapeutic targets for patients with sporadic vestibular schwannoma.

Whether to use face priors in the face super-resolution (FSR) methods is a symmetry problem.Various face priors are used to describe the overall and local face features, making the generation of super-resolution face images expensive and laborious. FSR methods that do not require any prior information tend to focus too much on the local features of the face, ignoring the modeling of global information. To solve this problem, we propose a dual-path facial image super-resolution network (SwinDPSR) fused with Swin Transformer. The network does not require additional face priors, and it learns global face shape and local face components through two independent branches. In addition, the channel attention ECA module is used to aggregate the global and local face information in the above dual-path sub-networks, which can generate corresponding high-quality face images. The results of face super-resolution reconstruction experiments on public face datasets and a real-scene face dataset show that SwinDPSR is superior to previous advanced methods both in terms of visual effects and objective indicators. The reconstruction results are evaluated with four evaluation metrics: peak signal-to-noise ratio (PSNR), structural similarity (SSIM), learned perceptual image patch similarity (LPIPS), and mean perceptual score (MPS). Head and neck squamous carcinoma (HNSCC) is the seventh most common cancer worldwide. Targeted therapeutic drugs for HNSCC are still being explored. Among them, (S)-10-hydroxycamptothecin (10-HCPT), a specific inhibitor of TOP1, functions by DNA double-strand breaks that can inhibit DNA replication and trigger apoptotic cell death subsequently. Previous studies have reported that MLN4924 exerts potent anti-tumor effects by inhibiting cullin–RING ligases and causing substrate accumulation in a variety of cancers. Here, we show that MLN4924 effectively causes dose-dependent accumulation of topoisomerase I (TOP1) and blocks TOP1 ubiquitination. Importantly, neddylation inhibition with MLN4924 acts synergistically with 10-HCPT to suppress cell growth, migration and apoptosis in HNSCC cells. Mechanistically, transcriptome sequencing shows that the cytotoxic effects of the combination of MLN4924 and 10-HCPT may involve activation of the NFKB1 pathway. Taken together, our results suggest that combined treatment with MLN4924 and 10-HCPT may be an effective strategy in HNSCC. [ABSTRACT FROM AUTHOR]

Time‐restricted eating (TRE) is a new therapeutic strategy for the management of weight loss and dysmetabolic diseases. At present, TRE (8/16, 8 h eating:16 h fasting) is the most common form of TRE. Therefore, this meta‐analysis included randomized controlled trials (RCTs) on TRE (8/16) in overweight and obese adults to determine its impact on body weight and metabolism. Articles reviewed from PubMed, Ovid MEDLINE, Embase, and Cochrane Central Register for the relevant RCTs that compared TRE (8/16) to non‐TRE in overweight and obese adults. Eight RCTs were included in this meta‐analysis. Participants following TRE (8/16) showed significant body weight reduction (mean difference [MD]: −1.48 kg, 95% confidence interval [CI]: −2.53 to −0.44) and fat mass reduction (MD: −1.09 kg, 95% CI: −1.55 to −0.63). There was no significant difference in lean mass change with TRE intervention (MD: −0.48 kg, 95% CI: −1.02 to 0.05, p =.08, I2 = 41%). The energy restriction and early TRE (eTRE) subgroups resulted in greater weight loss. TRE (8/16) showed beneficial effects on the homeostatic model assessment of insulin resistance (HOMA‐IR, MD: −0.32, 95% CI: −0.59 to −0.06), but had no significant effect on other parameters of glucose metabolism and lipid profiles. In conclusion, TRE (8/16), especially eTRE, or in combination with caloric intake restriction, is a potential therapeutic strategy for weight control in overweight and obese adults. TRE (8/16) also reduced HOMA‐IR; therefore, it may have a positive effect on glucose metabolism. [ABSTRACT FROM AUTHOR]
