The soil microbiota is a key component of agroecosystems, and understanding its traits is crucial for effective agronomic management. Among beneficial microorganisms, arbuscular mycorrhizal fungi (AMFs) are mutually associated with grapevine (Vitis vinifera L.), enhancing the ability of this cropping system to adapt to soil conditions and bolstering its resistance and resilience against abiotic stresses, particularly drought, by promoting root growth and enhancing the roots’ absorption surface. The objective of this on-field study was to determine AMF species richness and diversity along with their relation to soil chemical, physical, and biological characteristics in two adjacent organic vineyards in Central Italy. The two tested vineyards of the autochthonous cv. Aleatico differed by the presence of grafted (Vitis berlandieri × V. riparia rootstock; AL-420) or own-rooted (ungrafted V. vinifera L.; AL-ORV) vines. To this aim, soil and root samples were collected and geo-referenced. Analysis of the AMF species colonizing roots of both AL-ORV and AL-420 revealed the presence of four species: Scutellospora alterata, Paraglomus laccatum, Acaulospora laevis, and A. baetica, with S. alterata being the most frequent. Mycorrhization parameters were higher in the roots of grafted plants compared to ungrafted ones. A high beta-glucosidase (BG):N-acetylglusosaminidase (NAG) ratio in two tested vineyards indicated that microbes utilized more cellulose than chitin and peptidoglycan as dominant C resources. A negative correlation between mycorrhization rate (MyCP) and BG was observed, likely because AMFs form mutualistic relationships with plants, depending on the host plant for carbon. Results revealed a positive correlation between the degree of mycorrhizal association and the species involved, with the presence of copper and nickel among metals. Negative correlations were found concerning soil clay content along with beta-glucosidase. In conclusion, the grapevine root system was characterized by a differential symbiotic relationship with AMF species, whose development is influenced by the root genotype, soil texture, and biochemistry. Specifically, the increased frequency of AMFs in relation to copper content strengthens the evidence of their role in maintaining a vine’s production capacity in the event of soil contamination by this element.

Abstract Background Biodynamic agriculture is a management approach that aims to reduce the reliance on agrochemicals for production by emphasizing the use of specific natural preparations. A 2-year field trial spanning 2019–2020 was conducted in an established vineyard (Vitis vinifera L., cv. Cesanese d’Affile) to elucidate the impact of the cow horn silica biodynamic preparation (BD-501) on leaf vine physiology, potential resistance via chitinase activity, and analysis of secondary metabolites. The vineyard under biodynamic management was divided into two plots: one treated with BD-501 (BD-501) and the other untreated (BD). Throughout the vine growth season, measurements of carotenoid and chlorophyll levels, polyphenols, and chitinase activity were taken around key phenological phases (BBCH scale). During the ripening phase, a fluorometer was employed to assess chlorophyll fluorescence in the leaves. Results Leaves treated with BD-501 exhibited elevated concentrations of polyphenols and increased chitinase activity during the later phenological phases. In contrast, the untreated BD samples demonstrated high values primarily in the central phase of the observation period but not consistently throughout. At the time of harvest, chlorophyll concentration and quantum yield exhibited no statistically significant differences. BD-501 triggered a distinct response in terms of potential defense mechanisms (elevated polyphenols and chitinase activity) during the veraison phase. However, conversely, lower levels of chlorophylls and carotenoids were observed. Conclusions Nevertheless, a further round of experimental work is required to thoroughly comprehend the regulatory mechanisms behind this adaptive response and to ascertain the efficacy of BD-501. Agrobiodiversity represents a system of biological organisms that contribute to agri-food production. In a context marked by a significant loss of food-relevant species and a reduction in their genetic diversity, the adoption of strategies to preserve and enhance the diversity of genetic resources that support and complement agricultural production has become a global challenge. Many sustainable development strategies outlined in recent years directly and indirectly attribute a crucial role to agrobiodiversity in meeting food needs, ensuring food system security, promoting food justice, and enhancing well-being in modern living environments. This contribution aims to analyze the process of knowledge and awareness that has led many cities to plan their urban development by investing in the agricultural matrix and to address the design of open spaces with agricultural biodiversity.

Soil CO2 efflux is a pivotal component of agro-ecosystem C budgets. It is considered a proxy indicator of biological activity and a descriptor of soil quality that is strongly linked to agricultural soil management. We investigated the effects of soil fertilization practices (organo-mineral (OMN) versus chemical (C)) on soil under-vine CO2 efflux (TSR) in an Italian rainfed vineyard (cv Chardonnay). The TSR was measured using the chamber technique as follows: a close multi-chamber system (prototype) was placed under a vine. Data (CO2, temperature, and moisture) were acquired hourly during two consecutive years (2021 and 2022) from flowering to berry ripening. Physical–hydrological soil parameters were determined, and the seasonal trends of the TSR, soil temperature, and soil moisture were assessed. The TSR measurements fluctuated for the 2021 season, ranging from 1.03 to 1.97 µmol CO2∙m−2∙s−1 for the C treatment, while for the OMN treatment, the TSR measurements ranged from 1.24 to 1.71 µmol CO2∙m−2∙s−1. Extreme weather conditions (2022) highlighted the differences between the two agronomical practices, and a decoupling was found between the TSR and the soil water content, with the TSR being controlled primarily by the soil temperature. At the daily scale, the findings showed that the TSR reached its minimum in the early morning hours (5:00–8:00). The results promote organic–mineral nutrition as an improved practice for soil carbon storage (restoration of the organic fraction) by reducing the TSR, permitting the preservation of soil quality and stabilizing the hydrological traits by preserving the biotic activities.

Plant–soil biota represent a unique living system crucial for improving crops’ adaptation to climate change. In vineyards, plant–soil relations are mediated by rootstock–scion interaction, with grafted vines being the main plant material employed in vineyard planting. The interaction between two deeply different biological systems such as Vitis vinifera sativa and the American Vitis species may modify vines’ adaptation to abiotic stress. The aim of this study was to investigate the physiological response (chlorophyll content—CHL; stomatal conductance—gs) of grafted and ungrafted vines and assess the involvement of Arbuscular Mycorrhizal Fungi (AMF) in this response. In two vineyards located in Central Italy, the presence of AMF was assessed in the roots of grafted and ungrafted cv ‘Aleatico’ vines. The morphological traits of AMF and root mycorrhizal colonization differed in the grafted and ungrafted vines. Under limited climate conditions, ungrafted vines showed better leaf resilience traits (high CHL and gs values) and larger AMF storage organs (vesicles). On the other hand, the grafted ones—which are more sensitive to climate conditions (lower gs and CHL)—involved AMF colonization strategies (greater abundance of arbuscoles and mycorrhizal colonization potential) linked to the improved uptake and transport of water from the bulk soil to the vine. Taken together, these findings highlight different mycorrhizal colonization strategies and storage behaviors in grafted and ungrafted vineyards and with respect to different physical and chemical soil traits.

The European Community has recently imposed considerable restrictions on the use of pesticides, with the establishment of a regulatory framework for the sustainable use of agro-chemicals. However, in the viticulture sector, the intensive use of chemical pesticides, as well as sulfur and copper, is often required. Recently, ozone has been proposed as a possible environmentally friendly tool for controlling the development of pests on vines. However, little is known about the parameters linked to the practical application of ozone for controlling grapevine pests and how it triggers plant defence mechanisms. The main aim of this preliminary study was to determine the concentration of ozone and exposure duration in a treatment for stimulating the expression of systemic acquired resistance (SAR)-related genes, without inducing toxic effects and affecting vine health. In the first trial, three different combinations of ozone concentration and duration of treatment were tested on potted grapevines: i) gaseous ozone at 300 ppb for 12 hours, ii) gaseous ozone at 100 ppb for 6 hours, and iii) gaseous ozone at 100 ppb for 3 hours. Based on the results of the first trial, the potted vines were treated with just 100 ppb for 3 hours in a second trial. Leaves at different developmental stages were sampled. The expression level of systemic acquired resistance-related genes was analysed 12 hours and 7 days after each treatment. Furthermore, physiological parameters and NIR spectra were analysed. Ozone induced a transient up-regulation (limited to 12 hours after the treatments) of chitinases, β-1,3-glucanase and glutathione-S-transferase. On the other hand, pathogen-related (PR) genes showed a more persistent over-expression. The ozone treatment selectively affected the stomatal conductance depending on the different ozone concentrations. Detected NIR spectra revealed significant structural changes in ozone-treated plants, especially in leaves exposed to higher concentrations of ozone. These results suggest that ozone is able to transiently stimulate the expression of some resistance-related genes even at low and non-toxic doses for the vine leaves.

Urban agriculture is an emerging challenge. Identifying suitable agrosystems that allow for the multiple functions of urban agriculture represents a key issue for the reinforcement of the agricultural matrix in cities, with the aims of counteracting and adapting to climate change and providing economic and social benefits. This study aims to produce a preliminary assessment of the adaptability of Italian native and non-native Vitis vinifera L. cultivars to the stressors of an urban environment. The investigation was carried out on the grapevine collection of the Botanical Garden of Rome (“Vigneto Italia”). A total of 15 grapevine varieties were selected for the evaluation of leaf chlorophyll content, stomatal conductance, and chlorophyll fluorescence under abiotic conditions during the growing season of 2021. Spectral signatures were collected from mature leaves, and several vegetation indices (LWI, MCARI, and WBI) were calculated. Our preliminary results highlighted differences in the behavior of the grapevine cultivars. The native ones showed a medium-high level for leaf chlorophyll content (greater than 350 mol m−2), good photosynthetic efficiency (QY > 0.75), and optimal stomatal behavior under drought stress (200 > gs > 50 mmol H2O m−2 s−1). The data allowed for the classification of the tested genotypes based on their site-specific resistance and resilience to urban environmental conditions. The grapevine proved to be a biological system that is highly sensitive to climate variables, yet highly adaptable to limiting growing factors.

Fluorescent lights are typically used as light source in indoor horticultural production, including micropropagation. However, light emitting diodes (LEDs) have been recently used for plant growth under controlled environment. Major advantage of LEDs is wavelength specificity, that allows to adequately adjust the spectra according to plant needs. The possibility of using LED as primary light source for the micropropagation of Pyrus communis L. has been investigated in this work. It was proceeded to the optimization of a protocol of micropropagation using a LED lamp as the primary light source inside of the growth chamber with specific wavelengths, to reduce energy consumption and increase the quality of the micro propagated plants. Explants were maintained in a growth chamber and exposed to three different continuous spectrum LED lamps (AP67, NS1, G2) as a primary light source and fluorescent lamps (control) for 4 weeks. At the end of four weeks period, it was proceeded to the morphometric and biochemical analysis. Shoot and leaf growths were more influenced by LED lamps as compared to fluorescent lamps (control) in both cultivars. The results showed that the shoots of William and San Giovanni cultivars showed significant differences in morphological and physiological traits, as well as in chlorophyll, carotenoid, and MDA contents. Highest number of neo-formed shoots and nodes were observed in the plantlets of cv William under AP67 LED followed by NS1 and G2 LED lights respectively as compared to the white light (control), whereas same trend was observed in cv San Giovanni under AP67 LED, but it showed higher shoots and node numbers under control LED lamps as compared to both NS1 and G2. The photosynthetic pigments were significantly decreased in leaves of both cultivars when grown under LEDs as compared to the control fluorescent lamps. Moreover, the AP67 LED light had also significant effects on the protein and MDA contents in the leaves of both cultivars in comparison to all other treatments. This work underlines the importance of the modulation of light sources in relation with different species and varieties, allowing optimizing the proliferation phase and the efficiency of Pyrus communis L. micropropagation protocol. Moreover, this protocol can be improved with further studies to examine the response of the plantlets to the ex-vitro acclimatization because the possibility of using LED light for the micropropagation of pear can be considered as a valuable alternative for its ecologically sustainable production.

Global warming has two dangerous global consequences for agriculture: drought, due to water scarcity, and salinization, due to the prolonged use of water containing high concentrations of salts. Since the global climate is projected to continue to change over this century and beyond, choosing salt-tolerant plants could represent a potential paramount last resort for exploiting the secondary saline soils. Olive is considered moderately resistant to soil salinity as compared to other fruit trees, and in the present study, we investigated the influence of NaCl solutions (ranging from 0 to 200 mM) in a salt-tolerant (cv Canino) and two of its transgenic lines (Canino AT17-1 and Canino AT17-2), overexpressing tobacco osmotin gene, and in a salt-sensitive (Sirole) olive cultivar. After four weeks, most of the shoots of both Canino and Sirole plants showed stunted growth and ultimate leaf drop by exposure to salt-enriched media, contrary to transgenic lines, that did not show injuries and exhibited a normal growth rate. Malondialdehyde (MDA) content was also measured as an indicator of the lipid peroxidation level. To evaluate the role of the S assimilatory pathway in alleviating the adverse effects of salt stress, thiols levels as well as extractable activities of ATP sulfurylase (ATPS) and O-acetyl serine(thiol)lyase (OASTL), the first and the last enzyme of the S assimilation pathway, respectively, have been estimated. The results have clearly depicted that both transgenic lines overexpressing osmotin gene coped with increasing levels of NaCl by the induction of S metabolism, and particularly increase in OASTL activity closely paralleled changes of NaCl concentration. Linear correlation between salt stress and OASTL activity provides evidence that the S assimilation pathway plays a key role in adaptive response of olive plants under salt stress conditions.

Anorexia nervosa (AN) is associated with a wide range of disturbances of the autonomic nervous system. The aim of the present study was to monitor the heart rate (HR) and the heart rate variability (HRV) during light physical activity in a group of adolescent girls with AN and in age-matched controls using a wearable, minimally obtrusive device. For the study, we enrolled a sample of 23 adolescents with AN and 17 controls. After performing a 12-lead electrocardiogram and echocardiography, we used a wearable device to record a one-lead electrocardiogram for 5 min at baseline for 5 min during light physical exercise (Task) and for 5 min during recovery. From the recording, we extracted HR and HRV indices. Among subjects with AN, the HR increased at task and decreased at recovery, whereas among controls it did not change between the test phases. HRV features showed a different trend between the two groups, with an increased low-to-high frequency ratio (LF/HF) in the AN group due to increased LF and decreased HF, differently from controls that, otherwise, slightly increased their standard deviation of NN intervals (SDNN) and the root mean square of successive differences (RMSSD). The response in the AN group during the task as compared to that of healthy adolescents suggests a possible sympathetic activation or parasympathetic withdrawal, differently from controls. This result could be related to the low energy availability associated to the excessive loss of fat and lean mass in subjects with AN, that could drive to autonomic imbalance even during light physical activity.

Developing adaptation strategies in Vitis vinifera, a crop sensitive to climate change, is crucial for resilience of traditional viticultural systems, especially in climate-vulnerable areas like the Mediterranean basin. A progressive warming is demonstrated to alter the geographical distribution of grapevine, reducing land capability for typical grapes and vine productions in most Southern European districts traditionally specialized in tree crops. Grapevine growth and reproduction under climate change require a continuous monitoring to adapt agronomic practices and strategies to global change. The present study illustrates an empirical approach grounded on a set of bio-physical indicators assessing the genotype-related response to climate variation. This approach was tested in Umbria, central Italy, to verify the response of some major international and local grapevine varieties to climate variation during a relatively long time interval (1995−2015). Long-term data for ripening time and berry quality collected in the study area were correlated to representative bioclimatic indices including Winkler, Huglin, and Cool night indicators. Results of this study highlighted the increase of air temperature (reflecting the inherent growth in thermal availability for maturation) and the alteration of precipitation patterns toward more intense precipitation. Climate variability exerted distinctive impacts on grapevine phenology depending on the related genotype. Empirical findings underline the usefulness of a permanent field monitoring of the relationship between selected climate variables and grape ripening with the aim to develop adaptive viticultural practices at farm’s scale.
