Background and Purpose: IL‐1β produced by macrophages via the NOD‐, LRR‐ and pyrin domain‐containing 3 (NLRP3) inflammasome, mediates the inflammatory crosstalk between macrophages and adipocytes. In our previous study, (16S,20S,24R)‐12β‐acetoxy‐16,23‐epoxy‐24,25‐dihydroxy‐3β‐(β‐D‐xylopyranosyloxy)‐9,19‐cyclolanost‐22(23)‐ene (AEDC), a cycloartane triterpenoid isolated from Actaea vaginata (Ranunculaceae), was found to possess anti‐inflammatory effect on LPS‐treated RAW264.7 macrophages. This study was designed to investigate whether AEDC modulates macrophage–adipocyte crosstalk to alleviate adipose tissue inflammation. Experimental Approach The anti‐inflammatory effect of AEDC was evaluated on LPS plus ATP‐induced THP‐1 macrophages and C57BL/6J mice. The expression of autophagy‐related and NLRP3 inflammasome complex proteins was analysed by western blots, immunofluorescence staining and co‐immunoprecipitation. The pro‐inflammatory cytokines levels were determined by ELISA kits. The adipose tissue inflammation was evaluated by histological analysis and immunohistochemical staining. Key Results: AEDC (5 and 10 μM) activated autophagy, which in turn suppressed the NLRP3 inflammasome activation and IL‐1β secretion in THP‐1 macrophages. AEDC increased the expression of SIRT3 deacetylase and enhanced its deacetylating activity to reverse mitochondrial dysfunction and activate AMP‐activated protein kinase, which together induced autophagy. Moreover, AEDC (10 μM) attenuated macrophage conditioned medium‐induced inflammatory responses in adipocytes and blocked THP‐1 macrophages migration towards 3T3‐L1 adipocytes. In inflammation mice, AEDC (5 and 20 mg·kg−1) treatment reduced the levels of pro‐inflammatory cytokines in serum and epididymal adipose tissue and reduced macrophage infiltration to alleviate adipose tissue inflammation. Conclusion and Implications: AEDC attenuated the inflammatory crosstalk between macrophages and adipocytes through SIRT3‐autophagy‐mediated NLRP3 inflammasome inhibition, which might used for the treatment of adipose tissue inflammation‐related metabolic disorders. To reduce the manufacturing difficulty and improve the robustness of traditional boron-lined detectors that may replace the 3He counter, the honeycomb neutron converter-based gaseous neutron detector has been proposed. A drift electric field is applied to drive electrons ionized by $\alpha$ or 7Li after the 10B(n, $\alpha$ )7Li reaction from their origination positions to the incident surface of the gas electron multiplier (GEM), which multiplies electrons and forms the neutron signal. As the working gas affects the energy deposition of $\alpha$ or 7Li, the transverse diffusion of electrons in the migration process, as well as the multiplication of electrons in the GEM detector, the working gas selection of the honeycomb converter-based detector would be very important. Fourteen different working gases are investigated in detail through simulation research. Four working gases, Ar:iC4H10:CF4 = 90:7:3, Ar:CO2 = 95:5, Ar:CH4 = 90:10, and Ar:DME = 95:5, are experimentally tested. Both the simulation and experimental results demonstrate that working gases of Ar:iC4H10:CF4 = 90:7:3, Ar:CO2 = 95:5, and Ar:DME = 95:5 show good performances benefitting from both the large stopping powers of $\alpha$ or 7Li and the small transverse diffusion coefficients of electrons. The simulation results indicate that the detection efficiency with one of the three gases is 1.33 to 1.48 times the Ar:CH4 = 90:10, while the experimental results demonstrate that there is 1.34–1.49 times of the detection efficiency. A new design of the boron-lined gaseous neutron detector composed of a boron-lined honeycomb neutron converter and an electron multiplier is proposed in this paper. The motivation for this research was to decrease the manufacturing difficulty and improve the robustness of the boron-lined gaseous neutron detector. The numerous anode wires in the traditional designs were removed, and the gas electron multiplier (GEM) was used as the electron multiplier. To drive the ionized electrons produced inside the honeycomb structure out to the incident surface of the GEM, a drift electric field was applied inside the holes of the honeycomb structure. The design principles of this detector were discussed. Geant4, Maxwell11, and Garfield9 were used to estimate the neutron absorption efficiency and the electron migration process. A prototype detector was constructed and experimentally evaluated. Both the simulation and experimental results indicate that this detector has the potential to be used in the applications of small angle neutron scattering for scientific research, and to replace the currently used 3He detectors, which have the trouble of very limited supply of 3He gas. To identify the chemical components responsible for the anti-hyperglycemic effect of Cyclocarya paliurus (Batal.) Iljinsk (Juglandaceae) leaves, an ethanol extract (CPE) and a water extract (CPW) of C. paliurus leaves, as well as their total flavonoids (CPF), triterpenoids (CPT) and crude polysaccharides (CPP), were prepared and assessed on streptozotocin (STZ)-induced diabetic mice. After being orally administrated once a day for 24 days, CPF (300 mg/kg), CPP (180 mg/kg), or CPF+CPP (300 mg/kg CPF + 180 mg/kg CPP) treatment reversed STZ-induced body weight and muscle mass losses. The glucose tolerance tests and insulin tolerance tests suggested that CPF, CPP, and CPF+CPP showed anti-hyperglycemic effect in STZ-induced diabetic mice. Furthermore, CPF enhances glucose-stimulated insulin secretion in MIN6 cells and insulin-stimulated glucose uptake in C2C12 myotubes. CPF and CPP suppressed inflammatory cytokine levels in STZ-induced diabetic mice. Additionally, CPF and CPP improved STZ-induced diabetic nephropathy assessed by H&E staining, blood urea nitrogen content, and urine creatinine level. The molecular networking and Emperor analysis results indicated that CPF showed potential anti-hyperglycemic effects, and HPLC–MS/MS analysis indicated that CPF contains 3 phenolic acids and 9 flavonoids. In contrast, CPT (650 mg/kg) and CPC (300 mg/kg CPF + 180 mg/kg CPP + 650 mg/kg CPT) did not show anti-hyperglycemic effect. Taken together, polysaccharides and flavonoids are responsible for the anti-hyperglycemic effect of C. paliurus leaves, and the clinical application of C. paliurus need to be refined. Aims: The purpose of this study was to investigate the prognostic significance of pigment epithelium-derived factor (PEDF) in human clear cell renal cell carcinoma (CCRCC) tissues., Patients and Methods: We investigated the expression of PEDF and its correlation with clinicopathological features in 203 patients who underwent radical nephrectomy for CCRCC. Expression of PEDF and CD34-positive vessels (microvessel density, MVD) in tumor samples was examined by immunohistochemistry. The relationship between PEDF expression and clinicopathologic features including tumor diameter, MVD, and survival was evaluated by univariate and multivariate analyses., Results: Of 203 patients, 116 cases (57.1%) were positive for PEDF. A significant association was found between PEDF expression and high MVD (p = 0.012). PEDF expression was inversely correlated with grade (p = 0.004) and pT stage (p = 0.001). Multivariate analysis using the Cox regression model indicated that PEDF expression was an independent favorable prognostic factor (risk ratio, 4.105; p = 0.001). However, no significant correlation was found between PEDF expression and age, gender, tumor diameter or lymph node invasion., Conclusion: Our findings suggest that PEDF may be a favorable prognostic indicator in patients with CCRCC., (Copyright 2010 S.
