Abstract Coronaviruses being capable of spreading through droplet contamination have raised significant concerns regarding high-capacity public rail transport, such as the metro. Within a rapidly moving railcar cabin, the internal airflow lags behind the bulkhead, generating internally induced airflow that accelerates droplet dispersion within a non-inertial reference system. This study investigates the impact of acceleration on the diffusion of cough droplets of varying sizes using computational fluid dynamics. The modified k–ε equation in ANSYS® Fluent was utilized to simulate droplet diffusion under different body orientations by adjusting the inertial force correction source term. Results indicate that droplets in the middle size range (50–175 μm) are primarily influenced by inertial forces, whereas smaller droplets (3.5–20 μm) are predominantly controlled by air drag forces. Regardless of facial orientation, the outlet of high-capacity public rail transport poses the highest risk of infection.

The conventional image matting algorithms needed priori manual Trimap information to produce excellent matting results which made real time matting impossible. To tackle the problem, a Trimap-free image matting network, TFMNet, is proposed in this paper. The proposed network consists of four modules, ConvNeXt backbone module for image features extraction, Trimap prediction module for normalized Trimap generation, glance matting module for rough matting results prediction, and post-processing module for exact matting results production. To further optimize the training process of the proposed model, an improved Loss function based on frequency domain information is proposed. In experiment, Sets of Experiments designed by variable controlling approach prove that the proposed TFMNet do well in real time image matting. The TFMNet model achieves 8.99, 0.011, 12.31, 11.15 in the accuracy metrics of SAD, MSE, GRAD, CONN, respectively, costs 51ms for one image averagely which meet the real-time requirements, and model size is 671M. Besides, further experiments conducted by comparing with five state-of-the-art models based on three typical matting databases demonstrate the superiority of the proposed algorithm.

FAIR Principles are a set of high-level guidelines for sharing digital resources. The growing global adoption of the FAIR Principles by policymakers, funders, and organizations compels data professionals, projects, and repositories to demonstrate the level of FAIR-compliance (referred to as FAIRness) of their digital data, metadata, and infrastructures. Because the FAIR Principles offer general objectives rather than specific implementation instructions, discrepancies exist due to different interpretations, domain-specific requirements, and intended applications. These discrepancies hinder direct comparisons and integration of assessment outcomes. To address this issue, we propose a novel framework, including a consolidated FAIR vocabulary. This framework establishes quality measures upfront in FAIRness assessment workflows to surpass the intricacies arising from the aforementioned dependencies. The established quality measures encapsulate the distinctive core concepts inherent in individual FAIR principles and can serve as common, fundamental pillars of holistic FAIRness assessment workflows. Building upon this fundamental set of the quality measures, we introduce a FAIRness quality maturity matrix (FAIR-QMM) as a structured, tiered, and progressive approach for evaluating and reporting the degree of FAIR-compliance. The FAIR-QMM can be used as a FAIRness assessment tool independently and/or as a translator between other FAIRness assessment tools or models.

Abstract Background Chronic primary pain (CPP) is an intractable pain of unknown cause with significant emotional distress and/or dysfunction that is a leading factor of disability globally. The lack of a suitable animal model that mimic CPP in humans has frustrated efforts to curb disease progression. 2R, 6R-hydroxynorketamine (2R, 6R-HNK) is the major antidepressant metabolite of ketamine and also exerts antinociceptive action. However, the analgesic mechanism and whether it is effective for CPP are still unknown. Methods Based on nociplastic pain is evoked by long-term potentiation (LTP)-inducible high- or low-frequency electrical stimulation (HFS/LFS), we wanted to develop a novel CPP mouse model with mood and cognitive comorbidities by noninvasive low-frequency percutaneous electrical nerve stimulation (LF-PENS). Single/repeated 2R, 6R-HNK or other drug was intraperitoneally (i.p.) or intrathecally (i.t.) injected into naïve or CPP mice to investigate their analgesic effect in CPP model. A variety of behavioral tests were used to detect the changes in pain, mood and memory. Immunofluorescent staining, western blot, reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR) and calcium imaging of in cultured dorsal root ganglia (DRG) neurons by Fluo-8-AM were used to elucidate the role and mechanisms of 2R, 6R-HNK in vivo or in vitro. Results Intrathecal 2R, 6R-HNK, rather than intraperitoneal 2R, 6R-HNK or intrathecal S-Ketamine, successfully mitigated HFS-induced pain. Importantly, intrathecal 2R, 6R-HNK displayed effective relief of bilateral pain hypersensitivity and depressive and cognitive comorbidities in a dose-dependent manner in LF-PENS-induced CPP model. Mechanically, 2R, 6R-HNK markedly attenuated neuronal hyperexcitability and the upregulation of calcitonin gene-related peptide (CGRP), transient receptor potential ankyrin 1 (TRPA1) or vanilloid-1 (TRPV1), and vesicular glutamate transporter-2 (VGLUT2) in peripheral nociceptive pathway. In addition, 2R, 6R-HNK suppressed calcium responses and CGRP overexpression in cultured DRG neurons elicited by the agonists of TRPA1 or/and TRPV1. Strikingly, the inhibitory effects of 2R, 6R-HNK on these pain-related molecules and mechanical allodynia were substantially occluded by TRPA1 antagonist menthol. Conclusions In the newly designed CPP model, our findings highlighted the potential utility of intrathecal 2R, 6R-HNK for preventing and therapeutic modality of CPP. TRPA1-mediated uprgulation of CGRP and neuronal hyperexcitability in nociceptive pathways may undertake both unique characteristics and solving process of CPP.

AbstractThis study focuses on the issue of uncontrollable debris generated by laser-matter interactions in high-power laser facilities, which significantly impairs operational efficiency. The maximum motion distance of debris, referred to as the safety distance, is a crucial parameter that this work investigates. Three prediction methods that utilize aerodynamic coefficients and trajectories are proposed, with input parameters derived from velocity curves extracted from high-resolution time-resolved images. The results obtained from these methods show good agreement with experimental data collected under laser fluence conditions ranging from 18 to 79 J/cm2 that damage the rear surface of fused silica. The analytically predicted safety distance, with an input velocity fitted at 0 ns and an upper diameter limit of 10 µm, was closest to the experimental result at 155 mm. Analytically predicted values with input velocities fitted at 13,000 ns and all diameter ranges did not exceed 192.3 mm. Numerically predicted values, with input velocity fitted at 13,000 ns and a median diameter of small debris (5 µm), were 140.4 and 295.2 mm for aspect ratios of 2 and 4, respectively. The numerical method is recommended for predicting the safety distance in high-power laser facilities, but obtaining accurate estimates of transient velocity is crucial for the method’s effectiveness.

Abstract Background Polycystic ovarian syndrome (PCOS) is one of the most common causes of infertility in reproductive-age women. However, the efficacy and optimal therapeutic strategy for reproductive outcomes are still under debate. We conducted a systematic review and network meta-analysis to compare the efficacy of different first-line pharmacological therapies in terms of reproductive outcomes for women with PCOS and infertility. Methods A systematic retrieval of databases was conducted, and randomized clinical trials (RCTs) of pharmacological interventions for infertile PCOS women were included. The primary outcomes were clinical pregnancy and live birth, and the secondary outcomes were miscarriage, ectopic pregnancy and multiple pregnancy. A network meta-analysis based on a Bayesian model was performed to compare the effects of the pharmacological strategies. Results A total of 27 RCTs with 12 interventions were included, and all therapies tended to increase clinical pregnancy, especially pioglitazone (PIO) (log OR 3.14, 95% CI 1.56 ~ 4.70, moderate confidence), clomiphene citrate (CC) + exenatide (EXE) (2.96, 1.07 ~ 4.82, moderate confidence) and CC + metformin (MET) + PIO (2.82, 0.99 ~ 4.60, moderate confidence). Moreover, CC + MET + PIO (2.8, -0.25 ~ 6.06, very low confidence) could increase live birth most when compared to placebo, even without a significant difference. For secondary outcomes, PIO showed a tendency to increase miscarriage (1.44, -1.69 ~ 5.28, very low confidence). MET (-11.25, -33.7 ~ 0.57, low confidence) and LZ + MET (-10.44, -59.56 ~ 42.11, very low confidence) were beneficial for decreasing ectopic pregnancy. MET (0.07, -4.26 ~ 4.34, low confidence) showed a neutral effect in multiple pregnancy. Subgroup analysis demonstrated no significant difference between these medications and placebo in obese participants. Conclusions Most first-line pharmacological treatments were effective in improving clinical pregnancy. CC + MET + PIO should be recommended as the optimal therapeutic strategy to improve pregnancy outcomes. However, none of the above treatments had a beneficial effect on clinical pregnancy in obese PCOS. Trial registration CRD42020183541; 05 July 2020
