Submesoscale processes significantly influence the air‐sea interaction, heat budget and the distribution of physical and biogeochemical tracers in the upper‐ocean. We study the spatiotemporal characteristics and generation mechanisms of submesoscale vertical heat transport (SVHT) at the Kuroshio Extension using a submesoscale‐permitting simulation. Compared with the commonly used Boxcar and Hanning filters, the clean‐cut feature of the Butterworth filter in the wavenumber domain makes it a proper filter to diagnose SVHT. SVHT is systematically upward, peaking in winter. Through causality analysis, we find that, among the basic factors (mixed layer depth, strain rate, surface wind stress, and horizontal buoyancy gradient) from the conventional submesoscale generation mechanism scalings, mixed layer depth plays the leading role in modulating the intraseasonal variation of SVHT. Using the budget of submesoscale temperature variance and causality analysis, we find that advection also plays an important role in regulating SVHT. This work suggests that the choice of appropriate spatial filter is important for SVHT diagnosis and including advection may help improve submesoscale parameterization schemes. Plain Language Summary: Ocean currents with horizontal spatial scales of O(0.1–10) km and temporal scales of hours‐days are defined as submesoscale processes. Submesoscale processes are characterized by large vertical velocity, driving significant vertical heat transport. In this study, we investigate the spatiotemporal features of submesoscale vertical heat transport (SVHT) and the corresponding mechanisms in the Kuroshio Extension region using a high‐resolution numerical simulation. We find that the diagnostics of SVHT is sensitive to the extraction method of submesoscale processes based on spatial scales. SVHT is systematically upward with pronounced seasonal variability, reaching its peak in winter. The intraseasonal variation of SVHT is controlled by multiple factors simultaneously, among which the mixed layer depth plays the most important role. Advection also has influence on SVHT variability. This study offers a better understanding of the dynamic mechanisms driving SVHT, while also highlights the importance of proper filter selection in SVHT diagnostics. Bay scallops (Argopecten irradians; A. irradians) are shellfish with high nutritional and economic value. However, nutritional studies on A. irradians with different shell colors are limited. This study examines the hazardous, nutritional, and taste-contributing compounds during the growth of A. irradians with different shell colors. During the growth of A. irradians, the hazardous contents were below the standard limit. Changes in the nutritional and taste-contributing compounds between months were more significant than shell color. Bay scallops had more fats, total fatty acids, and taste-contributing compounds in August and more proteins, essential fatty acids, vitamin D, vitamin B12, Cu, and Zn in September and October. In October, the golden shell color strain had more proteins, essential fatty acids, vitamin D, vitamin B12, Cu, and Zn, while the purple shell color strain had more taste-contributing compounds. A. irradians had better taste in August and higher nutritional value in September and October. In October, the golden shell color strain has higher nutritional value, and the purple shell color strain has better commercial value and taste. Protoplasts are widely used in gene function verification, subcellular localization, and single-cell sequencing because of their complete physiological activities. The traditional methods based on tissues and organs cannot satisfy the requirement. Therefore, the isolation and capture of a single protoplast are most important to these studies. In this study, a dual-channel microfluidic chip based on PDMS with multi-capture cavities was designed. The design theory of the dual-channel microfluidic chip's geometry was discussed. The capture mechanism of the single cell in a dual-channel microfluidic chip was studied by simulation analysis. Our results showed that a single polystyrene microsphere or tobacco protoplast was successfully isolated and trapped in this chip. The capture efficiency of the chip was 83.33% for the single tobacco protoplast when the inlet flow rate was 0.75 μL/min. In addition, the dynamic capture of the polystyrene microsphere and tobacco protoplasts was also presented. Overall, our study not only provided a new strategy for the subsequent high throughput single protoplast research, but also laid a theoretical foundation for the capture mechanism of the single cell. Although the distribution of 8:2 polyfluoroalkyl phosphate diester (8:2 diPAP) in aquatic environments has been reported, details on its uptake, tissue specificity, and elimination in bivalve mollusks remain to be clarified. The present study is the first report on the accumulation and elimination of 8:2 diPAP in mussels (Mytilus galloprovincialis). The tissue‐specific accumulation and depuration of 8:2 diPAP and its metabolites were investigated via semistatic seawater exposure (8:2 diPAP at a nominal concentration of 10 μg/L), through water‐borne exposure with static daily renewal over a 72‐h exposure period and a 360‐h depuration period. The digestive gland was found to be the target organ where accumulation and biotransformation primarily occur. The bioaccumulation factor values (mL/g dry wt) in different organs were in the following order: digestive gland (1249) > adductor muscle (315) > gills (289) > gonad (82.9) > mantle (33.0). Moreover, the distribution of 8:2 diPAP among tissues may be related to the total protein content. The 8:2 diPAP tended to be excreted in feces. The compounds 8:2 fluorotelomer carboxylic acid, 8:2 fluorotelomer unsaturated carboxylic acid, 7:3 fluorotelomer carboxylic acid, perfluorooctanoic acid, and perfluoroheptanoic acid were detected and quantified as phase I metabolites, and the concentration of all phase I metabolites relative to the 8:2 diPAP concentration (72 h) was 0.304 mol%. A phase II metabolite, 8:2 fluorotelomer alcohol conjugated with sulfate, was detected but not quantitated in the digestive gland. A biotransformation pathway of 8:2 diPAP in M. galloprovincialis was proposed on the basis of the results obtained in the present study and previous studies. Nitrogen (N) addition may have strong impacts on legume growth and their biological N fixation (BNF), but how legume N acquisition sources respond to N inputs have yet to be comprehensively assessed. We quantified the effects of N addition on the growth and BNF of Medicago sativa and to assess the response of legume N acquisition to N addition. We grew M. sativa in the greenhouse under gradients of added NH4NO3 and analysed the variables that were relative to growth and BNF, such as N concentration, biomass, δ15N values, nodule number, percentage of N derived from the atmosphere (Ndfa%). Nitrogen addition had marginal effects on plant biomass production and foliar N concentration. Foliar δ15N value increased with increasing added N, while Ndfa% decreased. The number of nodules formed also decreased with N addition while the nitrogenase (nifH)genecopies per unit nodule mass was not significantly different with N addition. These findings indicate that increasing mineral N availability decreases symbiotic investment into BNF, mainly by reducing nodule formation; this was found to have no significant impact on plant growth because the plant changes its N source from BNF-N to mineral N derived from the soil. The role of circular RNA in cancer is emerging. A newly reported circular RNA HIPK3 (circHIPK3) is critical in cell proliferation of various cancer types, although its role in non-small cell lung cancer (NSCLC), has yet to be elucidated. Our results provided evidence that silencing of circHIPK3 significantly impaired cell proliferation, migration, invasion and induced macroautophagy/autophagy. Mechanistically, we uncovered that autophagy was induced upon loss of circHIPK3 via the MIR124-3p-STAT3-PRKAA/AMPKa axis in STK11 mutant lung cancer cell lines (A549 and H838). STAT3 abrogation as well as transfection with a MIR124-3p mimic, recapitulated the induction of autophagy. We also demonstrated antagonistic regulation on autophagy between circHIPK3 and linear HIPK3 (linHIPK3). We therefore propose that the ratio between circHIPK3 and linHIPK3 (C:L ratio) may reflect autophagy levels in cancer cells. We observed that a high C:L ratio (>0.49) was an indicator of poor survival, especially in advanced-stage NSCLC patients. These results support that circHIPK3 is a key autophagy regulator in a subset of lung cancer and has potential clinical use as a prognostic factor. The circular RNA HIPK3 (circHIPK3) functions as an oncogene and autophagy regulator may potential use as a prognostic marker and therapeutic target in lung cancer. BACKGROUND: To enhance discovering efficiency of new herbicidal compounds, a strategy for rapidly screening out strongly herbicidal chemical components from natural resources is necessary. RESULTS: Seventeen essential oils selected from 14 Eucalyptus species and hybrids were evaluated for their herbicidal activities on annual ryegrass. A feasible strategy was established for rapidly pinpointing the chemicals contributing to strong herbicidal activities without the process of isolation and purification of individual compounds. This strategy was based on gas chromatography–mass spectrometry (GC–MS) coupled with principal component analysis (PCA) and verified by the bioassay results using several individual compounds. Two strong herbicidal compounds trans‐pinocarveol and α‐terpineol were screened out using the strategy. The strong herbicidal activity of trans‐pinocarveol was discovered for the first time. High rates of de novo lipid synthesis have been discovered in certain kinds of tumours, including gallbladder cancer (GBC). Unlike several other tumours, GBC is highly insensitive to standard adjuvant therapy, which makes its treatment even more challenging. Although several potential targets and signalling pathways underlying GBC chemoresistance have been revealed, the precise mechanisms are still elusive. In this study, we found that α‐Mangostin, as a dietary xanthone, repressed the proliferation and clone formation ability, induced cell cycle arrest and the apoptosis, and suppressed de novo lipogenesis of gallbladder cancer cells. The underlying mechanisms might involve the activation of AMPK and, therefore, the suppression of SREBP1 nuclear translocation to blunt de novo lipogenesis. Furthermore, SREBP1 silencing by siRNA or α‐mangostin enhanced the sensitivity of gemcitabine in gallbladder cancer cells. In vivo studies also displayed that MA or gemcitabine administration to nude mice harbouring NOZ tumours can reduce tumour growth, and moreover, MA administration can significantly potentiate gemcitabine‐induced inhibition of tumour growth. Corroborating in vitro findings, tumours from mice treated with MA or gemcitabine alone showed decreased levels of proliferation with reduced Ki‐67 expression and elevated apoptosis confirmed by TUNEL staining, furthermore, the proliferation inhibition and apoptosis up‐regulation were obviously observed in MA combined with gemcitabine treatment group. Therefore, inhibiting de novo lipogenesis via targeting the AMPK/SREBP1 signalling by MA might provide insights into a potential strategy for sensitizing GBC cells to chemotherapy. CDK2-associated cullin domain 1 (CAC1) is as a novel cell cycle regulator widely expressed in colorectal cancer (CRC). However, its expression and function in drug resistant CRC cells remains elusive. Therefore, the present study aimed to assess the biochemical function and relevance of CAC1 in drug resistant CRC cells, and detect the potential mechanism. For this purpose, a total of 83 CRC cases were collected for the immunohistochemical analysis of CAC1 expression. Functional studies (stable transfection, flow cytometry, colony formation, and invasion and migration assays) were performed in SW480, LoVo and their corresponding 5-FU resistant cells. In addition, a nude mice xenograft model was established for further observation in vivo. In the present study, CAC1 protein expression was higher in CRC tissues than that in normal tissues (P<0.05). Furthermore, CAC1 protein expression was higher in SW480/5-FU cells than in SW480 cells. CAC1 knockdown arrested 5-FU resistant cells at the G1/S phase and increased the sensitivity of 5-FU resistant cells to 5-FU by inducing apoptosis. In addition, CAC1 reduced the invasive and migration ability of SW480/5-FU and LoVo/5-FU cells in vitro, and reduced their tumorigenicity and metastatic ability in vivo. Finally, CAC1 knockdown resulted in decreased P-glycoprotein and MRP-1 protein expression. Based on these results, it can be concluded that CAC1 plays an important role in the occurrence and promotion of drug resistance in CRC. Circular RNAs are widely expressed in various cancers and play important roles in tumorigenesis and tumor progression. The function and mechanism of circSMARCA5 in lung adenocarcinoma however remains unclear. QRT-PCR analysis was applied for determining circSMARCA5 expression in lung adenocarcinoma patient tumor tissues and cells. Molecular biological assays were used for investigating the role of circSMARCA5 in lung adenocarcinoma progression. Luciferase reporter and bioinformatics assays were used for identifying the underlying mechanism. In this study, we observed that circSMARCA5 expression was decreased in lung adenocarcinoma tissues but silencing of circSMARCA5 in lung adenocarcinoma cells inhibited cell proliferation, colony formation, migration and invasion. Mechanistically, we found EGFR, c-MYC and p21 were down-regulated upon circSMARCA5 knockdown. MiR-17-3p efficiently down- regulated EGFR expression via directly binding to EGFR mRNA. These studies suggest that circSMARCA5 functions as an oncogene via targeting miR-17-3p-EGFR axis and may represent a promising therapeutic target for lung adenocarcinoma. [ABSTRACT FROM AUTHOR]

Chiral and racemic macromonomers with a polymerizable phenylacetylenyl end group (PA-PLLA and PA-PDLLA) were successfully synthesized through living ring-opening polymerization of chiral and racemic monomers ( L-lactide and DL-lactide), respectively. Then the homopolymerization of PA-PLLA and PA-PDLLA was successfully carried out to give poly(PA-PLLA) and poly(PA-PDLLA), respectively. Chiroptical properties, hydrodynamic radii and micromorphologies of poly(PA-PLLA) and poly(PA-PDLLA) were analyzed via circular dichroism, dynamic light scattering and atomic force microscopy (AFM), and the conformation of the polymers is discussed. The results indicate that poly(PA-PLLA) induced Cotton effects with different intensities in various solvents, and its hydrodynamic radii and dispersity were also different in various solvents, while poly(PA-PDLLA) induced no Cotton effects and its hydrodynamic radii and dispersity were approximately the same in various solvents. Furthermore, a helical conformation of poly(PA-PLLA) and the random main chains of poly(PA-PDLLA) were observed by AFM. Therefore, it is concluded that poly(PA-PLLA) possesses a dynamic one-handed helical conformation in various solvents due to chiral poly( L-lactide) pendants, while poly(PA-PDLLA) possesses racemic main chains because of the racemic poly( DL-lactide) pendants. Background: Schisandra chinensis (Turcz.) Baill bee pollen extract (SCBPE) is often used as a functional food in China due to its good antioxidant property. However, its chemical compositions and effects on H9c2 cardiomyocytes against H2O2-induced cell injury still lacks of reports thus far. This study aimed to characterize the main components of SCBPE and investigate its protective effects against H2O2-induced H9c2 cardiomyocyte injury. Methods: The main components of SCBPE were analyzed via ultraperformance liquid chromatography–quadrupole time-of-flight tandem mass spectrometry (UPLC–QTOF MS/MS). The three main nucleosides in SCBPE were quantitatively analyzed via ultraperformance liquid chromatography–diode array detection. Furthermore, the potential mechanism by which SCBPE exerts protective effects against H2O2-induced H9c2 cardiomyocyte injury was explored for the first time via cell survival rate measurements; cell morphological observation; myocardial superoxide dismutase (SOD) activity and malondialdehyde (MDA) and glutathione (GSH) level determination; flow cytometry; and quantitative polymerase chain reaction. Results: Two carbohydrates, three nucleosides, and nine quinic acid nitrogen-containing derivatives in SCBPE were identified or tentatively characterized via UPLC–QTOF MS/MS. The nine quinic acid nitrogen-containing derivatives were first reported in bee pollen. The contents of uridine, guanosine, and adenosine were 2.4945 ± 0.0185, 0.1896 ± 0.0049, and 1.8418 ± 0.0157 μg/mg, respectively. Results of in vitro experiments showed that cell survival rate, myocardial SOD activity, and GSH level significantly increased and myocardial MDA level significantly decreased in SCBPE groups compared with those in H2O2 group. Cell morphology in SCBPE groups also markedly improved compared with that in H2O2 group. Results indicated that SCBPE protected H9c2 cardiomyocytes from H2O2-induced apoptosis by downregulating the mRNA expressions of Bax, cytochrome C, and caspase-3 and upregulating the Bcl-2 mRNA expression. Conclusions: This study is the first to report that SCBPE could protect against oxidative stress injury and apoptosis in H2O2-injured H9c2 cells. Results indicated that the nucleosides and quinic acid nitrogen-containing derivatives could be the main substances that exert protective effects against H2O2-induced H9c2 cardiomyocyte injury. Oxidative stress plays an important role in the pathogenesis of myocardial infarction (MI). Schisandra chinensis bee pollen extract (SCBPE) possesses powerful antioxidant capacity. This study aimed to further explore the antioxidative and cardioprotective effects of SCBPE on acute MI induced by isoprenaline (ISO) in rats. The rats were intragastrically administrated with SCBPE (600, 1200, or 1800 mg/kg/day) and Compound Danshen dropping pills (270 mg/kg/day) for 30 days, then subcutaneously injected with ISO (65 mg/kg/day) on the 29th and 30th day. Compared with the model group, pretreatment with middle and high doses of SCBPE significantly reduced serum aspartate transaminase, lactate dehydrogenase, and creatine kinase activities and increased myocardial superoxide dismutase, glutathione peroxidase, and catalase activities. The histopathologic aspects showed that pathological heart change was found in the model group and reduced to varying degrees in the SCBPE groups. Moreover, the protein expression of nuclear factor-erythroid 2-related factor 2 (Nrf-2), heme oxygenase-1 (HO-1), and Bcl2 in the heart increased in the SCBPE groups, while that of Bax decreased compared to the model group. Besides this, uridine was isolated from S. chinensis bee pollen for the first time.
