Purpose of this review: Manipulating or re-engineering the damaged human spinal cord to achieve neuro-recovery is one of the foremost challenges of modern science. Addressing the restricted permission of neural cells and topographically organised neural tissue for self-renewal and spontaneous regeneration, respectively, is not straightforward, as exemplified by rare instances of translational success. This review assembles an understanding of advances in nanomedicine for spinal cord injury (SCI) and related clinical indications of relevance to attempts to design, engineer, and target nanotechnologies to multiple molecular networks. Recent findings: Recent research provides a new understanding of the health benefits and regulatory landscape of nanomedicines based on a background of advances in mRNA-based nanocarrier vaccines and quantum dot-based optical imaging. In relation to spinal cord pathology, the extant literature details promising advances in nanoneuropharmacology and regenerative medicine that inform the present understanding of the nanoparticle (NP) biocompatibility–neurotoxicity relationship. In this review, the conceptual bases of nanotechnology and nanomaterial chemistry covering organic and inorganic particles of sizes generally less than 100 nm in diameter will be addressed. Regarding the centrally active nanotechnologies selected for this review, attention is paid to NP physico-chemistry, functionalisation, delivery, biocompatibility, biodistribution, toxicology, and key molecular targets and biological effects intrinsic to and beyond the spinal cord parenchyma. Summary: The advance of nanotechnologies for the treatment of refractory spinal cord pathologies requires an in-depth understanding of neurobiological and topographical principles and a consideration of additional complexities involving the research’s translational and regulatory landscapes.

Paclitaxel (PTX) and 5-fluorouracil (5-FU) are clinically relevant chemotherapeutics, but both suffer a range of biopharmaceutical challenges (e.g., either low solubility or permeability and limited controlled release from nanocarriers), which reduces their effectiveness in new medicines. Anticancer drugs have several major limitations, which include non-specificity, wide biological distribution, a short half-life, and systemic toxicity. Here, we investigate the potential of liposome-micelle-hybrid (LMH) carriers (i.e., drug-loaded micelles encapsulated within drug-loaded liposomes) to enhance the co-formulation and delivery of PTX and 5-FU, facilitating new delivery opportunities with enhanced chemotherapeutic performance. We focus on the combination of liposomes and micelles for co-delivery of PTX and 5_FU to investigate increased drug loading, improved solubility, and transport/permeability to enhance chemotherapeutic potential. Furthermore, combination chemotherapy (i.e., containing two or more drugs in a single formulation) may offer improved pharmacological performance. Compared with individual liposome and micelle formulations, the optimized PTX-5FU-LMH carriers demonstrated increased drug loading and solubility, temperature-sensitive release, enhanced permeability in a Caco-2 cell monolayer model, and cancer cell eradication. LMH has significant potential for cancer drug delivery and as a next-generation chemotherapeutic.

Quantum dot-based NO2 sensors often suffer from low sensitivity or long recovery times. Here, the authors report that bimetallic PbCdSe quantum dot gels containing atomically dispersed Pb ionic sites enable ultra-sensitive and fast NO2 sensing.

This paper describes the synthesis of a new type of nanowires, potassium tetracyanoplatinate sesquihydrate Krogmann salt or K(def)TCP. Seed-mediated electrocrystallization nucleates and confines the K(def)TCP crystals into nanowires. Our data show that the width of K(def)TCP scales proportionally with the diameter of the seed, which is consistent with the nanoconfinement effect of the seed. K(def)TCP nanowires are also electrodeposited on lithographic Au patterns as interconnects and tested for impedance-based gas sensing. The results demonstrate the use of electrocrystallization to construct nanowire sensing units directly on patterned substrates to enable scaling up of nanowire sensor technology. IMPACT STATEMENT We show for the first time nanowires directly formed as interconnects on electronic substrates by electrocrystallization are used as sensor materials to detect gases.

Active droplets play important roles in microfluidics, robotics, and micro-electromechanical systems. As a special class of active droplets that are conductive, reactive, and of high surface tension, liquid metal droplets (LMDs) can be driven by electric-field-induced surface (Marangoni) flows to function as reconfigurable components in actuators, sensors, catalytic reactors, and antennas. Stimulating LMDs using an electric field induces concurrent electro-hydrodynamic flows and electrochemical surface oxidation (passivation). It is however difficult to decouple these two effects which brings complexity in controlling LMD motions. To address this challenge, pulse time modulation (PTM) signals are used. PTM enables controlled LMD displacement by propelling the droplets forward during the voltage-on phases and facilitating surface recovery from oxidation during the voltage-off phases. Counterintuitively, by taking such intermittent "rests", the LMDs effectively inhibit the unfavorable impact of oxidation, granting high motion controllability. Combining high-speed imaging, motion tracking, machine learning, and electrochemical analysis, the study reveals how electro-hydrodynamic flows and surface oxide formation/dissolution interplay to generate well-defined motion regimes. The study further develops a quasi-analytical model to describe droplet motions and designs a rotary LMD motor to showcase the versatility of the approach. Effects of the Adenosine A1 blockade using 8-cyclopentyl-1,3-diprophyxanthine (DPCPX) nanoconjugate on inducing recovery of the hemidiaphragm paralyzed by hemisection have been thoroughly examined previously; however, the toxicology of DPCPX nanoconjugate remains unknown. This research study investigates the therapeutic efficacy and toxicology of the nanoconjugate DPCPX in the cervical spinal cord injury (SCI) rat model. We hypothesized that a single injection of nanoconjugate DPCPX in the paralyzed left hemidiaphragm (LDH) of hemisected rats at the 2nd cervical segment (C2Hx) would lead to the long-term recovery of LDH while showing minimal toxicity. Adult male rats underwent left C2Hx surgery and the diaphragms' baseline electromyography (EMG). Subsequently, rats were randomized into a control group and four treated subgroups. Three subgroups received a single intradiaphragmatic dose of either 0.09, 0.15, or 0.27 μg/kg, and one subgroup received 0.1 μg/kg of native DPCPX two times per day intravenously (i.v.) for 3 days (total 0.6 μg/kg). Rats were monitored for a total of 56 days. Compared with control, the treatment with nanoconjugate DPCPX at 0.09 μg/kg, 0.15 μg/kg, and 0.27 μg/kg doses elicited significant recovery of paralyzed LDH (i.e., 67% recovery at 8 wk) (P < 0.05). DPCPX nanoconjugate-treated rats had significant weight loss for first 2 wk but recovered significantly by day 56 (P < 0.05). The levels of gold in the blood and body tissues were below the recommended levels. No sign of weakness, histology of tissue damage, or organ abnormality was observed. A dose of DPCPX nanoconjugate can induce long-term diaphragm recovery after SCI without observed toxicity. [ABSTRACT FROM AUTHOR]

Sleep-disordered breathing (SDB) is very common after spinal cord injury (SCI). The present study was designed to evaluate the therapeutic efficacy of adenosine A1 receptor blockade (8-cyclopentyl-1,3-dipropylxanthine, DPCPX) on SDB in a rodent model of SCI. We hypothesized that SCI induced via left hemisection of the second cervical segment (C2Hx) results in SDB. We further hypothesized that blockade of adenosine A1 receptors following C2Hx would reduce the severity of SDB. In the first experiment, adult male rats underwent left C2Hx or sham (laminectomy) surgery. Unrestrained whole body plethysmography (WBP) and implanted wireless electroencephalogram (EEG) were used for assessment of breathing during spontaneous sleep and for the scoring of respiratory events at the acute (~1 wk), and chronic (~6 wk) time points following C2Hx. During the second experiment, the effect of oral administration of adenosine A1 receptor antagonist (DPCPX, 3 times a day for 4 days) on SCI induced SDB was assessed. C2Hx animals exhibited a higher apnea-hypopnea index (AHI) compared with the sham group, respectively (35.5 ± 12.6 vs. 19.1 ± 2.1 events/h, P < 0.001). AHI was elevated 6 wk following C2Hx (week 6, 32.0 ± 5.0 vs. week 1, 42.6 ± 11.8 events/h, respectively, P = 0.12). In contrast to placebo, oral administration of DPCPX significantly decreased AHI 4 days after the treatment (159.8 ± 26.7 vs. 69.5 ± 8.9%, P < 0.05). Cervical SCI is associated with the development of SDB in spontaneously breathing rats. Adenosine A1 blockade can serve as a therapeutic target for SDB induced by SCI. [ABSTRACT FROM AUTHOR]

A spin-coating method is described to nucleate organic molecular rods of uniform size from an inorganic nanoparticle at a solid surface. The result suggests that nanoparticles can be regarded as nanoseeds for the nucleation of guest crystals.

Nanowire sensors are promising to deliver greater performance than conventional sensors due to their size confinement as well as low cost and power consumption. However, scalable assembly of nanowire sensors remains a big challenge, which requires the synthesis of uniform nanowires and organization of them. Here, we report a simple method to control the growth and organization of a promising nanowire material for gas sensing, tetrathiafulvalene bromide ((TTF)Br), by mass transfer in electrodeposition.We found that the use of Pt microdisk electrodes for electrodeposition of (TTF)Br wires resulted in wires with higher uniformity and larger aspect ratio (length to width) than their counterparts deposited on Pt films under the same deposition conditions. More interestingly, we found that the number of wires grown from a Pt microdisk electrode exhibited a limiting number of 3 or 4 per electrode when the electrode diameters are smaller than ~1.4 μm. Both experimental and simulation results suggest that the morphological differences and the constant minimum wire density are caused by the different TTF flux distributions at a microdisk versus a film electrode. Purpose: Bombarding high-Z material with x-ray radiation releases Auger electrons and Coster-Kronig electrons, along with deeper penetrating fluorescent x-rays and photoelectrons. The Auger and Coster-Kronig electron penetration distance is on the order of nanometers to micrometers in water or tissue, creating a large dose enhancement accompanied by a RBE greater than 1 at the cellular level. The authors' aim is to measure the gold nanofilm dose enhancement factor (DEF) at the cellular level with unlaminated radiochromic film via primary 50 kVp tungsten x-ray spectrum interaction, similar to an electronic brachytherapy spectrum. Methods: Unlaminated Gafchromic® EBT2 film and Monte Carlo modeling were combined to derive DEF models. Gold film of thickness 23.1± 4.3 nm and surface roughness of 1.2±0.2 nm was placed in contact with unlaminated radiochromic film in a downstream orientation and exposed to a 50 kVp tungsten bremsstrahlung, mean energy 19.2 keV. Film response correction factors were derived by Monte Carlo modeling of electron energy deposition in the film's active layer, and by measuring film energy dependence from 4.5 keV to 50 kVp. Results: The measured DEF within a 13.6 µm thick water layer was 0.29 with a mean dose of 94±9.4 cGy from Au emissions and 324±32.4 cGy from the 50 kVp primary beam. Monte Carlo derived correction factors allowed determination of Au contributed dose in shallower depths at 0.25 µm intervals. Maximum DEF of 18.31 was found in the first 0.25 µm water depth. Conclusions: Dose enhancement from Au nanofilm can be measured at the cellular level using unlaminated radiochromic film. Complementing the measured dose value with Monte Carlo calculations allows estimation of dose enhancement at depth increments within the cellular range. [ABSTRACT FROM AUTHOR]

DNA release dynamics from layer-by-layer (LbL) films is an important aspect to consider with regards to localized gene delivery systems. The rate of DNA release and the condensation state of DNA during release are of particular interest in the field of gene delivery. A hyperbranched poly(amido amine) (RHB) containing bioreducible disulfide bonds is used to form interpolyelectrolyte complexes with DNA during LbL film assembly. During film disassembly, DNA is released in physiologic conditions due to the reducing nature of the RHB. Uncondensed DNA deposited on the surface was compared to DNA condensed by RHB in polyplex form by using two types of LbL films, RHB/DNA/RHB and polyplex terminated films, RHB/DNA/polyplex. LbL films with up to three layers are used in order to facilitate high-resolution atomic force microscopy (AFM) imaging. X-ray reflectivity, ellipsometry, and Fourier transform infrared spectroscopy are also used. The film disassembly, rearrangement, and release of molecules from the surface due to thiol−disulfide exchange is conducted in reducing dithiothreitol (DTT) solutions. Salt is found to accelerate the overall rate of film disassembly. Additionally, it was found that the polyplex layer disassembles faster than the DNA layer. The predominant intermediate structure is the toroid structure for the polyplex layer and the fiber bundle structure for the DNA layer during film disassembly. This study offers a simple means to modulate DNA release from LbL films by utilizing both condensed and uncondensed DNA in different layers. This paper reports the synthesis and characterization of polystyrene nanorods in hemicylindrical hemimicelles of a nonionic polyoxyethylene surfactant, C12E5, on graphite. The surface structure is characterized by atomic force microscopy (AFM), Fourier transform infrared spectroscopy, and contact angle goniometry. Uniformly aligned polystyrene nanorods are captured by AFM. The nanorod dimensions are studied as a function of the reaction time and styrene monomer concentration. The template synthesis using self-assembled surfactant surface aggregates promises to create functional and stable nanostructures for optoelectronics and surface engineering. [ABSTRACT FROM AUTHOR]

Abstract Electrocrystallization is a promising method for controlled charge‐transfer complex (CTC) deposition on microfabricated electrodes for gas sensing applications. However, there remains a gap in our understanding of CTC electrodeposition. In this study, we focus on investigating the electrocrystallization of cobalt tetracyanoquinodimethane (Co‐TCNQ) on a microdisk electrode to elucidate and control the process. Leveraging the microelectrode technique, we conduct steady‐state measurements to observe nucleation and crystal growth dynamics, particularly in the early stages of electrocrystallization. We use cyclic voltammetry and chronoamperometry to examine Co‐TCNQ electrocrystallization under various electrolytic conditions. We identify electrocrystallization kinetics, ranging from electrokinetic to diffusion‐limited growth, governing the nucleation and growth of Co‐TCNQ crystals. Notably, we pinpoint the applied overpotential and precursor concentration range necessary for a single nucleation site on the microelectrode. Moreover, we demonstrate control over crystal orientation and morphology. Our findings reveal a nonclassical growth pathway for Co‐TCNQ crystals characterized by oriented attachment of small crystallites along the conductive long axis. Importantly, electrodeposited Co‐TCNQ on patterned microelectrodes exhibits selective sensing capabilities for nitrogen dioxide gas. Overall, this study sheds light on CTC electrodeposition through a proof‐of‐concept demonstration involving Co‐TCNQ electrodeposition on microelectrodes, presenting potential applications across diverse materials.

Poor drug encapsulation efficiency and rapid release of the encapsulated drug limit the use of nanoparticles in biomedical applications involving water-soluble drugs. We have developed a novel polymer-surfactant nanoparticle formulation, using the anionic surfactant Aerosol OT™ (AOT) and polysaccharide polymer alginate, for sustained release of water-soluble drugs. Particle size of nanoparticles, as determined by atomic force microscopy and transmission electron microscopy, was in the range of 40–70 nm. Weakly basic molecules like methylene blue, doxorubicin, rhodamine, verapamil, and clonidine could be encapsulated efficiently in AOT-alginate nanoparticles. In vitro release studies with basic drug molecules indicate that nanoparticles released 60–70% of the encapsulated drug over 4 weeks, with near zero-order release during the first 15 days. Studies with anionic drug molecules demonstrate poorer drug encapsulation efficiency and more rapid drug release than those observed with basic drugs. Further studies investigating the effect of sodium concentration in the release medium and the charge of the drug suggest that calcium-sodium exchange between nanoparticle matrix and release medium and electrostatic interaction between drug and nanoparticle matrix are important determinants of drug release.
