34. Novel Temperature-Dependent Development Rate Models for Postdiapause Egg Eclosion of Three Important Arthropod Pests Found in Commercial Christmas Tree Plantations of Southern Québec, Canada.

Abstract

The balsam twig aphid Mindarus abietinus Koch (Hemiptera: Aphididae), the spruce spider mite Oligonychus ununguis (Jacobi) (Acari: Tetranychidae), and the pine needle scale Chionaspis pinifoliae (Fitch) (Hemiptera: Diaspididae) are three important pests for the Christmas tree industry of Québec, Canada. Temperature-dependent development rates for postdiapause overwintering eggs of these species have never been studied for populations of southern Québec, where the vast majority of Christmas trees are grown. The accumulation of physiological time in growing degree days (GDD) is a standard tool to predict egg eclosion dates for pests, when first generation immatures are most vulnerable to insecticidal control. Development rates for postdiapause eggs were tested at four or five constant temperatures in the 11–23°C range under controlled conditions, based on time before hatching for eggs collected in a balsam fir plantation in late winter. The standard linear model and three published nonlinear models were fitted to the data and compared for their ability to estimate key biological temperature parameters and to predict development rates of postdiapause eggs. Validation of model predictions of egg eclosion time on balsam fir in the field was reasonably accurate using the classical linear model where field-accumulated GDD were calculated with a particular interpretation of the average method based on daily maximum and minimum temperatures. The linear model could predict mean egg eclosion within around 5 d of eclosion dates observed in the field. Le développement des œufs hivernants postdiapausants, en fonction de la température, n'a jamais été étudié pour les populations du sud du Québec, où cette culture est concentrée. L'accumulation du temps physiologique en degrés-jours de croissance (DJ) est un outil essentiel pour prédire la date d'éclosion des œufs de ravageurs, alors que les immatures de première génération sont vulnérables aux traitements insecticides. Le taux de développement des œufs hivernants a été mesuré en conditions contrôlées pour quatre ou cinq températures constantes dans la gamme 11–23°C, calculé selon le temps de développement jusqu'à l'éclosion d'œufs collectés sur le sapin baumier. Le modèle linéaire standard et trois modèles non linéaires publiés ont été évalués pour leur capacité à prédire les paramètres biologiques de réponse à la température et le taux de développement. La prédiction de l'éclosion des œufs sur le terrain a été validée avec le calcul du temps physiologique accumulé en DJ avec une interprétation particulière de la méthode des moyennes basée sur les températures quotidiennes maximales et minimales, en association avec le modèle linéaire. Abstract

1. DFU (5,5-dimethyl-3-(3-fluorophenyl)-4-(4-methylsulphonyl)phenyl-2(5H)-furan one) was identified as a novel orally active and highly selective cyclo-oxygenase-2 (COX-2) inhibitor. 2. In CHO cells stably transfected with human COX isozymes, DFU inhibited the arachidonic acid-dependent production of prostaglandin E2 (PGE2) with at least a 1,000 fold selectivity for COX-2 (IC50 = 41 +/- 14 nM) over COX-1 (IC5050 microM). Indomethacin was a potent inhibitor of both COX-1 (IC50 = 18 +/- 3 nM) and COX-2 (IC50 = 26 +/- 6 nM) under the same assay conditions. The large increase in selectivity of DFU over indomethacin was also observed in COX-1 mediated production of thromboxane B2 (TXB2) by Ca2+ ionophore-challenged human platelets (IC5050 microM and 4.1 +/- 1.7 nM, respectively). 3. DFU caused a time-dependent inhibition of purified recombinant human COX-2 with a Ki, value of 140 +/- 68 microM for the initial reversible binding to enzyme and a kappa 2 value of 0.11 +/- 0.06 s-1 for the first order rate constant for formation of a tightly bound enzyme-inhibitor complex. Comparable values of 62 +/- 26 microM and 0.06 +/- 0.01 s-1, respectively, were obtained for indomethacin. The enzyme-inhibitor complex was found to have a 1:1 stoichiometry and to dissociate only very slowly (t1/2 = 1-3 h) with recovery of intact inhibitor and active enzyme. The time-dependent inhibition by DFU was decreased by co-incubation with arachidonic acid under non-turnover conditions, consistent with reversible competitive inhibition at the COX active site. 4. Inhibition of purified recombinant human COX-1 by DFU was very weak and observed only at low concentrations of substrate (IC50 = 63 +/- 5 microM at 0.1 microM arachidonic acid). In contrast to COX-2, inhibition was time-independent and rapidly reversible. These data are consistent with a reversible competitive inhibition of COX-1. 5. DFU inhibited lipopolysaccharide (LPS)-induced PGE2 production (COX-2) in a human whole blood assay with a potency (IC50 = 0.28 +/- 0.04 microM) similar to indomethacin (IC50 = 0.68 +/- 0.17 microM). In contrast, DFU was at least 500 times less potent (IC5097 microM) than indomethacin at inhibiting coagulation-induced TXB2 production (COX-1) (IC50 = 0.19 +/- 0.02 microM). 6. In a sensitive assay with U937 cell microsomes at a low arachidonic acid concentration (0.1 microM), DFU inhibited COX-1 with an IC50 value of 13 +/- 2 microM as compared to 20 +/- 1 nM for indomethacin. CGP 28238, etodolac and SC-58125 were about 10 times more potent inhibitors of COX-1 than DFU. The order of potency of various inhibitors was diclofenacindomethacin approximately naproxennimesulide approximately meloxicam approximately piroxicamNS-398 approximately SC-57666SC-58125CGP 28238 approximately etodolacL-745,337DFU. 7. DFU inhibited dose-dependently both the carrageenan-induced rat paw oedema (ED50 of 1.1 mg kg-1 vs 2.0 mg kg-1 for indomethacin) and hyperalgesia (ED50 of 0.95 mg kg-1 vs 1.5 mg kg-1 for indomethacin). The compound was also effective at reversing LPS-induced pyrexia in rats (ED50 = 0.76 mg kg-1 vs 1.1 mg kg-1 for indomethacin). 8. In a sensitive model in which 51Cr faecal excretion was used to assess the integrity of the gastrointestinal tract in rats, no significant effect was detected after oral administration of DFU (100 mg kg-1, b.i.d.) for 5 days, whereas chromium leakage was observed with lower doses of diclofenac (3 mg kg-1), meloxicam (3 mg kg-1) or etodolac (10-30 mg kg-1). A 5 day administration of DFU in squirrel monkeys (100 mg kg-1) did not affect chromium leakage in contrast to diclofenac (1 mg kg-1) or naproxen (5 mg kg-1). 9. The results indicate that COX-1 inhibitory effects can be detected for all selective COX-2 inhibitors tested by use of a sensitive assay at low substrate concentration. The novel inhibitor DFU shows the lowest inhibitory potency against COX-1, a consistent high selectivity of inhibition of COX-2 over COX-1 (300 fold) with enzyme, whole cell and whole blood assays, with no detectable loss of integrity of the gastrointestinal tract at doses200 fold higher than efficacious doses in models of inflammation, pyresis and hyperalgesia. These results provide further evidence that prostanoids derived from COX-1 activity are not important in acute inflammatory responses and that a high therapeutic index of anti-inflammatory effect to gastropathy can be achieved with a selective COX-2 inhibitor.

Abstract

The Rupert River is one of the largest tributaries on the east coast of James Bay. Lake sturgeon ( Acipenser fulvescens) is present all along the main stem where several spawning grounds have been located, four of which are major spawning grounds that have been studied at km 216, 281, 290 and 362. The total number of drifting larvae was estimated with drift nets set along transverse transects at km 212, 276, and km 287 from 2007 to 2009, and at km 361 in 2008 and 2009, using a new technique, namely, a Doppler current meter to measure water velocity within transect sub-sections corresponding to Voronoï polygons. There was a substantial, persistent difference in the number of larvae produced by the four main spawning areas. On average, the most productive site (km 276) produced over five times more larvae than the least productive site (km 361). Average estimated numbers were 41,194 at km 212, 176,840 at km 276, 106,212 at km 287, and 30,642 at km 361. Temporal variations were of much less amplitude than spatial differences. Between 2007 and 2009, interannual variations were not significant, except at km 212, despite differences in river flow during incubation and larval drift. The number of gravid females and the quality of spawning grounds would likely be the main factors influencing the total number of larvae. Vertical distribution of larvae is variable between sites and years, and shows a slight tendency for larvae to be more surface oriented. Higher flow near the surface would partly explain larger surface drifting of larvae. Transverse distribution is uneven and often associated with the location of the spawning grounds and the river flow. Given the uneven vertical and transverse distribution of larvae, an effective sampling strategy should cover the complete water column and full river width. Where depth exceeds 3 m, at least two stacked nets are recommended. In large rivers, filtering close to 1% of total river flow should result in acceptable confidence intervals, allowing a good comparison of the number of larvae in space and time. Abstract

We show that ablation features in poly-methyl methacrylate (PMMA) induced by a single femtosecond laser pulse are imposed by light polarization. The ablation craters are elongated along the major axis of the polarization vector and become increasingly prominent as the pulse energy is increased above the threshold energy. We demonstrate ~40% elongation for linearly and elliptically polarized light in the fluence range of 4-20 J cm-2, while circularly polarized light produced near circular ablation craters irrespective of pulse energies. We also show that irradiation with multiple pulses erases the polarization-dependent elongation of the ablation craters. However, for line ablation the orientation of the electric field vector is imprinted in the form of quasi-periodic structures inside the ablated region. Theoretically, we show that the polarization dependence of the ablation features arises from a local field enhancement during light-plasma interaction. Simulations also show that in materials with high nonlinearities such as doped PMMA, in addition to conventional explosive boiling, sub-surface multiple filamentation can also give rise to porosity. Abstract

Background and objectives: To evaluate the effects of cardiopulmonary bypass (CPB) on platelet function in children undergoing open-heart surgery. Methods: Data from 16 consecutive children undergoing cardiac surgery with CPB were prospectively collected. Blood samples of 10 mL were collected via the central venous line immediately before and after CPB for CD62 measurements by flow cytometry. Results: Ten children had acyanotic heart disease (median age 3yr, range 1.8-14) and six had a cyanotic defect (median age 4yr, range 2-14). The platelet count decreased significantly with CPB in both groups: from 163.5 (130-201) to 93.5 (57-186) × 10³ μL-1 in acyanotic children and from 139.5 (77-212) to 75 (43-99) × 10³ μL-1 in cyanotic children (P < 0.0001). The percentage of activated platelets was significantly lower in acyanotic children at baseline: 1% (0-23%) vs. 5% (3-8%) (P = 0.07). CPB increased the percentage of activated platelets significantly in both groups: post-bypass the values were 10% (range 1-17%) in acyanotic children and 7% (range 1-30%) in cyanotic children (P = 0.03). The increase in the percentage of activated platelets did not differ between the two study groups (P = 0.11). Abstract

Background and objective: This study was undertaken to investigate the role played by antidiuretic hormone (ADH), the renin-angiotensin system and atrial natriuretic factor (ANF) in the oliguria in patients undergoing spinal fusion. Methods: Sixteen patients undergoing posterior spinal fusion using isoflurane and fentanyl (n = 8) or sufentanil (n = 8) had renin, aldosterone, ADH and ANF measurements. Results: Compared to the non-oliguric patients, the oliguric patients had a higher number of fused vertebrae 10.5 ± 1.3 vs. 9.0 ± 0.5 (P = 0.01) and higher renin values at 12 h (3.3 ± 3.2 vs. 0.7 ± 0.6 ng L-1 s-1, P = 0.04). Hormonal values that had a significant correlation with intraoperative diuresis were: renin at 0.5 h (r2 = 0.26, P = 0.04), aldosterone at 0.5 h (r2 = 0.30, P = 0.03) and ANF at 0.5 h (r2 = 0.32, P = 0.02). Those that had a significant correlation with the mean postoperative diuresis were: renin at 6 h (r2 = 0.29, P = 0.03), 8 h (r2 = 0.26, P = 0.04) and 12 h (r2 = 0.31, P = 0.03), aldosterone at 6 h (r2 = 0.54, P = 0.001), 8 h ( r2 = 0.40, P = 0.01) and 12 h (r2 = 0.32, P = 0.03), ADH at 24 h (r2 = 0.38, P = 0.01) and ANF at 6 h (r2 = 0.26, P = 0.045). Using stepwise regression, excluding hormonal values, only two continuous variables had a significant correlation with the mean postoperative diuresis: the number of fused vertebrae (P = 0.02) and the length of surgery (P = 0.02). Conclusion: Activation of the renin-angiotensin system is the major cause of the early intraoperative oliguria. The victim-choice polymorphia of 178 sexual aggressors divided into six subtypes, incest offenders, pseudoincest offenders, sexual aggressors of familiar children, sexual aggressors of unfamiliar children, sexual aggressors of familiar women, and sexual aggressors of unfamiliar women, was compared. Results showed that sex offenders remained stable in their choice of victim from one offence to another in terms of victim age, victim gender, and aggressor-victim relationship. Subjects characterised by high levels of polymorphia were pseudoincest offenders and sexual aggressors of familiar women, whereas sexual aggressors of both unfamiliar women and unfamiliar children were characterised by low levels of polymorphia.
