ABSTRACT: Milk coagulation is an important step in the production of fermented dairy products such as yogurt and cheese. Jujube is gaining popularity and acceptance as a food ingredient. In China, jujube yogurt is popular among consumers. However, there is limited information on the effect of jujube on acid- and rennet-induced coagulation properties of milk. The objective of this study was to evaluate the effects of jujube pulp at different concentrations on acid- and rennet-induced coagulation kinetics of milk and the microstructure of acid- and rennet-induced gels. During acid-induced coagulation, with increasing jujube pulp concentration, the initial pH value decreased; however, the final pH value increased. The initial elasticity index (EI) value increased, and the time point at which the mean square displacement curves lost the linear trend advanced. The sample with 10% jujube pulp had the densest structure and highest EI value. During rennet-induced coagulation, with increasing jujube pulp concentration, the production rate and amount of caseinomacropeptide decreased, and the final EI value increased. Protein aggregates in rennet-induced gels became rough, and the sample with 20% jujube pulp had the highest EI value. This study provides a new perspective and understanding of the application of jujube in fermented dairy products.

Abstract This study explored the relationship between parenting stress, parenting style, parenting quality, and young children's executive function. In total, 243 firstborns aged 2–9 years old (SD = 3.82) and their parents from two-child families in Beijing participated in the study, which used executive function tasks and parenting questionnaires. The results found that (1) parenting stress negatively predicted parenting quality; (2) parenting style partially mediated the relationship between parenting stress and parenting quality; (3) children's executive function partially mediated the relationship between parenting stress and parenting quality; and (4) the spoiled, democratic, permissive, and authoritarian parenting styles each play a chain mediating role with young children's executive function between parenting stress and parenting quality. Taken together, these findings provide implications for scientific parenting of children with different psychological characteristics (such as executive function) in multiple-child families under Parenting stress.

Abstract Chinese first-born children need to learn how to get along with their siblings after the implementation of the universal two-child policy in 2016. We investigated the relationships between temperament, parenting style, and psychological adjustment among firstborns. The current study employed a questionnaire survey conducted in four regions in China. A total of 524 Chinese two-child families participated in the study; the firstborns were between 3 and 8 years old and their younger siblings were between 1 month and 5 years old. The results indicated that (1) children’s temperament subscales were significantly related to parenting style subscales and psychological adaptation. Moreover, the parenting style subscales were significantly related to psychological adaptation, and (2) authoritarian parenting partially mediated the relationship between approach or withdrawal and psychological adjustment.

As a representative kind of soft-shelled turtles, Pelodiscus sinensis has soft apron termed calipash around its pliable carapace as the most distinctive trait, which is functionally important in turtle defense. It is of great significance to explore the physiological function of stress resistance and the possible regulation of anti-stress nutrition for the survival of aquaculture. To address this issue, a fibroblast cell line from the embryonic calipash tissue of P. sinensis (PSCF) was established from the perspective of ethics, repeatability, and convenience. Effect of glycine, the main amino acid component in calipash extracellular matrix, on the proliferation of PSCF treated with 160 μg/mL lipopolysaccharide (LPS) and the underlying mechanisms were also studied. The results showed that adding 100 μg/mL glycine significantly improved the fibroblast survival, increased the mRNA levels of collagen biosynthesis genes, prolyl hydroxylase and collagen degradation genes, and inhibited reactive oxygen species (ROS) production. It suggested that glycine shows positive effects on the cell proliferation of PSCF, and could regulate the oxidative stress and collagen metabolism. To our knowledge, this report is the first to describe cell cultures established at specific calipash tissue-forming stages during the embryonic development in P. sinensis and to reveal the anti-stress effect of glycine in turtles. The results will contribute to the application of functional calipash tissue, or its manipulation by nutrition additives, such as glycine, to prevent or ameliorate stress in turtle industry.

This study aimed to investigate relationships among parental psychological control, adolescent emotion regulation, and social problems in China. In total, 1,145 adolescents aged 12–15 years participated in the study, which used the Parental Psychological Control Scale, Adolescent Problem Behavior Scale, and Emotion Regulation Scale. The results indicated the following: (1) Compared with only-child teens, adolescents in multi-child families had significant social problems; (2) parental psychological control significantly predicted adolescents’ social problems; (3) there was a partially mediating effect of adolescents’ emotion regulation between parental psychological control and adolescents’ social problems.

Studies have shown that Sargassum fusiforme and its extracts are effective herbal treatments for leukemia. We previously found that a polysaccharide from Sargassum fusiforme, SFP 2205, stimulated apoptosis in human erythroleukemia (HEL) cells. However, the structural characterization and antitumoral mechanisms of SFP 2205 remain uncertain. Here, we studied the structural characteristics and anticancer mechanisms of SFP 2205 in HEL cells and a xenograft mouse model. The results demonstrated that SFP 2205, with a molecular weight of 41.85 kDa, consists of mannose, rhamnose, galactose, xylose, glucose, and fucose with monosaccharides composition of 14.2%, 9.4%, 11.8%, 13.7%, 11.0%, and 38.3%, respectively. On animal assays, SFP 2205 significantly inhibited growth of HEL tumor xenografts with no discernible toxicity to normal tissues. Western blotting showed that SFP 2205 therapy improved Bad, Caspase-9, and Caspase-3 protein expression, and ultimately induced HEL tumor apoptosis, indicating mitochondrial pathway involvement. Furthermore, SFP 2205 blocked the PI3K/AKT signaling pathway and 740 Y-P, an activator of the PI3K/AKT pathway, rescued the effects of SFP 2205 on HEL cell proliferation and apoptosis. Overall, SFP 2205 may be a potential functional food additive or adjuvant for preventing or treating leukemia.

Pelodiscus sinensis (P. sinensis) is a commonly cultivated turtle species with a habit of hibernation. To study the changes in histone expression and methylation of P. sinensis during hibernation induction, a model was established by artificial induction. Physiological and metabolic indices were measured, and the expression and localization of histone (H1, H2A, H2B, H3, and H4) and methylation-related genes (ASH2L, KMT2A, KMT2E, KDM1A, KDM1B, and KDM5A) were measured by quantitative PCR, immunohistochemistry, and Western blot analysis. The results indicated that the metabolism, antioxidation index, and relative expression of histone methyltransferase were significantly decreased (p < 0.05), whereas the activity and expression of histone demethyltransferase were significantly increased (p < 0.05). Although our results showed significant changes in physiological and gene expression after hibernation induction, we could not confirm that P. sinensis entered deep hibernation. Therefore, for the state after cooling-induced hibernation, cold torpor might be a more accurate description. The results indicate that the P. sinensis can enter cold torpor through artificial induction, and the expression of histones may promote gene transcription. Unlike histones expressed under normal conditions, histone methylation may activate gene transcription during hibernation induction. Western blot analysis revealed that the ASH2L and KDM5A proteins were differentially expressed in the testis at different months (p < 0.05), which may perform a role in regulating gene transcription. The immunohistochemical localization of ASH2L and KDM5A in spermatogonia and spermatozoa suggests that ASH2L and KDM5A may perform a role in mitosis and meiosis. In conclusion, this study is the first to report changes in histone-related genes in reptiles, which provides insight for further studies on the physiological metabolism and histone methylation regulation of P. sinensis during the hibernation induction and hibernation period.

Participating in mobile services by synthesizing trajectories with consistent lifestyle and meaningful mobility as actual traces are the most popular way to protect location privacy. However, recent trajectory synthesizing techniques are still threatened by the information that the attacker inevitably obtains, such as the locations of the accepted tasks in the crowdsourcing application. With this information and the spatiotemporal correlation hidden in the user's mobility, the attacker can infer the user's actual location and even future behaviors. It remains open to defend against such inferential attacks in the continual crowdsourcing scenarios. In this paper, we propose a mobility-aware differentially private solution, ConCrowdDP, for achieving the privacy-preserving continual crowdsourcing application. Specifically, before starting the application, we first construct a spatiotemporal mobile model, STMarkov, to model the spatiotemporal correlation in users' mobility. Then, a perturbed location is generated for the user to participate in the crowdsourcing application, according to STMarkov and K-norm DP. Finally, we eliminate the privacy threat brought by the accepted task based on K-norm DP and Bayesian posterior theorem. With ConCrowd-DP in place, a mobility-aware differentially private trace is generated for the user to participate in the application continually. Extensive experiments with real-world datasets demonstrate that ConCrowd-DP guarantees the usability of the synthesized trajectory effectively, while providing the DP protection for defending against the inferential attacks which stem from the multiple accepted tasks.

With the deepening of the medical and health system, high-quality nursing service with the reform of nursing service model and the provision of holistic nursing care for patients as the core connotation is being comprehensively carried out. With the continuous improvement of medical quality, people’s awareness of health has gradually increased. They have put forward higher requirements for medical quality, nursing service quality, and medical care safety. Under the influence of these conditions, the workload and work pressure of clinical nurses continue to increase, and mental health problems become increasingly prominent. According to relevant data, the detection rate of occupational stress among nurses was 100%, and the proportion of nurses who perceived considerable stress was 60.9%, which seriously affected their work efficiency and quality of life. However, the physical and mental health of the frontline nurses working in psychiatric hospitals is not optimistic. This study explored the effects of psychological education among nurses in the department of psychiatry. The results showed that psychological education intervention among nurses in the department of psychiatry could alleviate their professional tiredness, effectively improve their psychological elasticity and happiness index, and thus improve sleep quality and promote their physical and mental health development.

This study aimed to examine first-born children's sibling jealousy and explore the relationships among first-born children's sibling jealousy, temperament, and emotion regulation in China during COVID-19 pandemic. The research hypotheses of this study are empirically examined through online and offline surveys. A sample of 304 two-child families from China participated in the study; the first-born children were aged between 1.17 and 7 years. The results indicated the following: (1) the older the first-born children and the greater the age difference between siblings, the lower the sibling jealousy. (2) Difficult temperament of first-born children could predict sibling jealousy significantly and positively, and emotion regulation could predict sibling jealousy negatively. (3) There was a partially mediating effect of emotion regulation between temperament and sibling jealousy. Compared with intermediate temperament, first-born children with difficult temperament had weaker emotion regulation and higher sibling jealousy. Overall, findings have important implications for psychological interventions for families of first-born children with difficult temperament.

This study aimed to investigate mothers’ parenting stress and explore its relationship with associated demographic variables in two-child families involving preschool children. A sample of 621 two-child families and a comparison group of 319 one-child families from China participated in the study; the children were aged between 3 and 7. The results showed that (1) mothers of two-child families had higher parenting stress than those of one-child families; (2) within the two-child families, demographic variables, such as birth order, gender combination, and the age gap were found to have significant effects on maternal stress levels; and (3) in two-child families, families with an income of less than 3000 yuan had significantly higher maternal stress than families with an income of more than 6000 yuan.

Background: Conformational change induced by the binding of a substrate or coenzyme is a poorly understood stage in the process of enzyme catalysed reactions. For enzymes that exhibit a domain movement, the conformational change can be clearly characterized and therefore the opportunity exists to gain an understanding of the mechanisms involved. The development of the non-redundant database of protein domain movements contains examples of ligand-induced domain movements in enzymes, but this valuable data has remained unexploited. Description: The domain movements in the non-redundant database of protein domain movements are those found by applying the DynDom program to pairs of crystallographic structures contained in Protein Data Bank files. For each pair of structures cross-checking ligands in their Protein Data Bank files with the KEGG-LIGAND database and using methods that search for ligands that contact the enzyme in one conformation but not the other, the non-redundant database of protein domain movements was refined down to a set of 203 enzymes where a domain movement is apparently triggered by the binding of a functional ligand. For these cases, ligand binding information, including hydrogen bonds and salt-bridges between the ligand and specific residues on the enzyme is presented in the context of dynamical information such as the regions that form the dynamic domains, the hinge bending residues, and the hinge axes. Conclusion: The presentation at a single website of data on interactions between a ligand and specific residues on the enzyme alongside data on the movement that these interactions induce, should lead to new insights into the mechanisms of these enzymes in particular, and help in trying to understand the general process of ligand-induced domain closure in enzymes. The increasing popularity of smartphones and location-based service (LBS) has brought us a new experience of mobile crowdsourcing marked by the characteristics of network-interconnection and information-sharing. However, these mobile crowdsourcing applications suffer from various inferential attacks based on mobile behavioral factors, such as location semantic, spatiotemporal correlation, etc. Unfortunately, most of the existing techniques protect the participant’s location-privacy according to actual trajectories. Once the protection fails, data leakage will directly threaten the participant’s location-related private information. It open the issue of participating in mobile crowdsourcing service without actual locations. In this paper, we propose a mobility-aware trajectory-prediction solution, TMarkov, for achieving privacy-preserving mobile crowdsourcing. Specifically, we introduce a time-partitioning concept into the Markov model to overcome its traditional limitations. A new transfer model is constructed to record the mobile user’s time-varying behavioral patterns. Then, an unbiased estimation is conducted according to Gibbs Sampling method, because of the data incompleteness. Finally, we have the TMarkov model which characterizes the participant’s dynamic mobile behaviors. With TMarkov in place, a mobility-aware spatiotemporal trajectory is predicted for the mobile user to participate in the crowdsourcing application. Extensive experiments with real-world dataset demonstrate that TMarkov well balances the trade-off between privacy preservation and data usability.

Abstract In vertebrates, the primary sex-determining signals that initiate sexual development are remarkably diverse, ranging from complete genetic to environmental cues. However, no sex determination-related genes have been functionally identified in reptiles. Here, we characterized a conserved DM domain gene, Dmrt1, in Chinese soft-shelled turtle Pelodiscus sinensis (P. sinensis), which exhibits ZZ/ZW sex chromosomes. Dmrt1 exhibited early male-specific embryonic expression, preceding the onset of gonadal sex differentiation. The expression of Dmrt1 was induced in ZW embryonic gonads that were masculinized by aromatase inhibitor treatment. Dmrt1 knockdown in ZZ embryos by RNA interference resulted in male to female sex reversal, characterized by obvious feminization of gonads, significant down-regulation of testicular markers Amh and Sox9, and remarkable up-regulation of ovarian regulators, Cyp19a1 and Foxl2. Conversely, ectopic expression of Dmrt1 led to largely masculinized genetic females, production of Amh and Sox9, and a decline in Cyp19a1 and Foxl2. These findings demonstrate that Dmrt1 is both necessary and sufficient to initiate testicular development, thereby acting as an upstream regulator of the male pathway in P. sinensis.

Immunoassays are among the frontline methods used for disease diagnosis and surveillance. Despite this, there are no immunoassays developed for the Chinese soft-shelled turtle (Pelodiscus sinensis), which has expanded into large scale commercial production in several Asian countries. One of the critical factors delaying the development of immunoassays is the lack of characterized soft-shelled turtle immunoglobulins. Herein, we used mass spectrometry together with the ProtQuest software to identify the soft-shelled turtle IgM heavy chain in serum, which again was used to produce a polyclonal anti-turtle-IgM in rabbits. Thereafter, the polyclonal anti-turtle-IgM was used as a secondary antibody in an indirect ELISA to evaluate antibody responses of soft-shelled turtles injected with the bovine serum albumin (BSA) model antigen. Our findings show that only turtle immunized with a water-in-oil BSA plus ISA 763A VG adjuvant (SEPPIC, France) emulsion had antibodies detected at 42 days post vaccination (dpv) while turtles injected with phosphate buffered saline (PBS) only as well as turtle injected with BSA dissolved in PBS had no significant antibody levels detected in serum throughout the study period. In summary, our findings show that rabbit polyclonal anti-turtle-IgM produced can be used in ELISA to measure serum antibody responses in immunized soft-shelled turtles. Future studies should explore its application in other immunoassays needed for the disease diagnosis and vaccine development for soft-shelled turtles.

Motivation: The biological function of proteins is associated with a variety of motions, ranging from global domain motion to local motion of side chain. We propose a method, dihedral transition analysis (DTA), to identify significant dihedral angle changes between two distinct protein conformations and for characterization of the effect of these transitions on both local and global conformation. Results: Applying DTA to a comprehensive and non-redundant dataset of 459 high-resolution pairs of protein structures, we found that a dihedral transition occurs in 82% of proteins. Multiple dihedral transitions are shown to occur cooperatively along the sequence, which allows us to separate a polypeptide chain into fragments with and without transitions, namely transition fragments (TFs) and stable fragments (SFs), respectively. By characterizing the magnitude of TF conformational change and the effect of the transition on the neighboring fragments, flap and hinge motions are identified as typical motions. DTA is also useful to detect protein motions, subtle in RMSD but significant in terms of dihedral angle changes, such as the peptide-plane flip, the side-chain flip and path-preserving motions. We conclude that DTA is a useful tool to extract potential functional motions, some of which might have been missed using conventional methods for protein motion analysis.
