2. Yijing Decoction regulates MMPs/TIMPs-related molecules to antagonize high glucose-induced basement membrane damage in iBRB model

Abstract

AIM: To investigate the mechanism by which Yijing Decoction antagonist high glucose-induced damage to the basement membrane(BM)in an in vitro inner blood-retinal barrier(iBRB)model.METHODS:Rat retinal microvascular pericytes(RMPs)and endothelial cells(ECs)were isolated and cultured to establish an in vitro iBRB model. The cells were randomly divided into four groups: low glucose group(LG), high glucose group(HG), minocycline group(MG)and Yijing Decoction group(YG). The LG group received 25 mmol/L glucose, the HG group received 60 mmol/L glucose, the MG group received 60 mmol/L glucose + 10 μg/mL minocycline, and the YG group received 60 mmol/L glucose + 10% Yijing Decoction-containing serum. Incubation for each group were terminated after intervention for 12 h. Next, the Western blot analysis was performed to assess the protein expression of BM-related proteins, including collagen Ⅳ(CⅣ)and laminin(LN), as well as matrix metalloproteinase(MMPs)/tissue inhibitor of matrix metalloproteinases(TIMPs)such as MMP-2, MMP-3, MMP-9, TIMP-1, TIMP-2.RESULTS:Compared to the LG group, the protein expressions of CⅣ increased in the HG, MG, and YG groups, as did LN in the HG and MG groups(all P0.05).CONCLUSION:Yijing Decoction can potentially intervene in DR by modulating the protein expression of MMP-2, MMP-3, MMP-9, CⅣ, and LN, suppressing high glucose-induced BM remodeling, and mitigating damage to iBRB.

Abstract

One of the key factors determining the degree of erosion in the outlet parts of refractory interfaces is diffusion. By studying the representative diffusion behavior of lanthanum, cerium, and manganese in molten rare‐earth high‐carbon heavy rail steel using different refractory interfaces, the diffusion kinetics mechanism of each alloying element is illustrated. The results show that the concentration of diffused lanthanum, cerium, and manganese in rare‐earth high‐carbon heavy rail steel imparts minor changes on the two aluminum–magnesium refractories used in our experiments, whereas the other four refractories show unstable behavior. Furthermore, it is revealed that the smaller the porosity of a refractory material, the greater the decrease in the diffusion channel of each element in molten steel, which in turn increases the diffusion activation barrier and results in slower diffusion. Overall, the suitability degree of the refractory materials tested in experiments for processing molten rare‐earth high‐carbon heavy rail steel with regard to cost, adhesion strength, and erosion degree is as follows: Al2O3–MgO > Al2O3–MgO–SiO2 coating > Al2O3–SiO2–SiO2 coating > Al2O3–SiO2 > Al2O3 > ZrO2. Abstract

This article aims to investigate the agglomeration behavior of cerium oxide particles at the interface of high‐temperature melts. The high‐temperature experiment with 0.15% cerium addition is carried out at 1600 °C and the type of cerium oxide is detected to be Ce–O–S particles according to thermodynamic calculation and energy‐dispersive analysis. The attractive force between particles is analyzed by high‐temperature confocal laser scanning microscopy and the Kralchevsky–Paunov model. The in situ observation indicates that the largest acting distance between Ce–O–S particle pairs is measured to be 80 μm. The attractive force becomes weak as the distance between Ce–O–S particle pairs (L) decreases, and a stronger attractive force exists between Ce–O–S particle pairs with a smaller radius ratio (R1/R2). The capillary force for Ce–O–S particles is weaker than that for Ce2O3 particles. The first‐principles results indicate that the wettability at the interface of the Fe/Ce2O3–S system is weaker than the Fe–S/Ce2O3 system, which is in agreement with the in situ observation and Kralchevsky–Paunov model results.
