Water availability is the dominant driver of microbial community structure and function in desert soils. However, these habitats typically only receive very infrequent large-scale water inputs (e.g., from precipitation and/or run-off). In light of recent studies, the paradigm that desert soil microorganisms are largely dormant under xeric conditions is questionable. Gene expression profiling of microbial communities in desert soils suggests that many microbial taxa retain some metabolic functionality, even under severely xeric conditions. It, therefore, follows that other, less obvious sources of water may sustain the microbial cellular and community functionality in desert soil niches. Such sources include a range of precipitation and condensation processes, including rainfall, snow, dew, fog, and nocturnal distillation, all of which may vary quantitatively depending on the location and geomorphological characteristics of the desert ecosystem. Other more obscure sources of bioavailable water may include groundwater-derived water vapour, hydrated minerals, and metabolic hydro-genesis. Here, we explore the possible sources of bioavailable water in the context of microbial survival and function in xeric desert soils. With global climate change projected to have profound effects on both hot and cold deserts, we also explore the potential impacts of climate-induced changes in water availability on soil microbiomes in these extreme environments. Soils store about four times as much carbon as plant biomass (1), and soil microbial respiration releases about 60 petagrams of carbon per year to the atmosphere as carbon dioxide (2). Short-term experiments have shown that soil microbial respiration increases exponentially with temperature (3). This information has been incorporated into soil carbon and Earth-system models, which suggest that warming-induced increases in carbon dioxide release from soils represent an important positive feedback loop that could influence twenty-first-century climate change (4). The magnitude of this feedback remains uncertain, however, not least because the response of soil microbial communities to changing temperatures has the potential to either decrease (5-7) or increase (8,9) warming-induced carbon losses substantially. Here we collect soils from different ecosystems along a climate gradient from the Arctic to the Amazon and investigate how microbial community-level responses control the temperature sensitivity of soil respiration. We find that the microbial community-level response more often enhances than reduces the mid- to long-term (90 days) temperature sensitivity of respiration. Furthermore, the strongest enhancing responses were observed in soils with high carbon-to-nitrogen ratios and in soils from cold climatic regions. After 90 days, microbial community responses increased the temperature sensitivity of respiration in high-latitude soils by a factor of 1.4 compared to the instantaneous temperature response. The climate of maritime Antarctica has altered since the 1950s. However, the effects of increased temperature, precipitation and organic carbon and nitrogen availability on the fungal communities inhabiting the barren and oligotrophic fellfield soils that are widespread across the region are poorly understood. Here, we test how warming with open top chambers (OTCs), irrigation and the organic substrates glucose, glycine and tryptone soy broth (TSB) influence a fungal community inhabiting an oligotrophic maritime Antarctic fellfield soil. In contrast with studies in vegetated soils at lower latitudes, OTCs increased fungal community alpha diversity (Simpson's index and evenness) by 102-142% in unamended soil after 5 years. Conversely, OTCs had few effects on diversity in substrate-amended soils, with their only main effects, in glycine-amended soils, being attributable to an abundance of Pseudogymnoascus. The substrates reduced alpha and beta diversity metrics by 18-63%, altered community composition and elevated soil fungal DNA concentrations by 1-2 orders of magnitude after 5 years. In glycine-amended soil, OTCs decreased DNA concentrations by 57% and increased the relative abundance of the yeast Vishniacozyma by 45-fold. The relative abundance of the yeast Gelidatrema declined by 78% in chambered soil and increased by 1.9-fold in irrigated soil. Fungal DNA concentrations were also halved by irrigation in TSB-amended soils. In support of regional- and continental-scale studies across climatic gradients, the observations indicate that soil fungal alpha diversity in maritime Antarctica will increase as the region warms, but suggest that the accumulation of organic carbon and nitrogen compounds in fellfield soils arising from expanding plant populations are likely, in time, to attenuate the positive effects of warming on diversity. Abstract Mass populations of toxin-producing cyanobacteria commonly develop in fresh-, brackish- and marine waters and effective strategies for monitoring and managing cyanobacterial health risks are required to safeguard animal and human health. A multi-interdisciplinary study, including two UK freshwaters with a history of toxic cyanobacterial blooms, was undertaken to explore different approaches for the identification, monitoring and management of potentially-toxic cyanobacteria and their associated risks. The results demonstrate that (i) cyanobacterial bloom occurrence can be predicted at a local- and national-scale using process-based and statistical models; (ii) cyanobacterial concentration and distribution in waterbodies can be monitored using remote sensing, but minimum detection limits need to be evaluated; (iii) cyanotoxins may be transferred to spray-irrigated root crops; and (iv) attitudes and perceptions towards risks influence the public's preferences and willingness-to-pay for cyanobacterial health risk reductions in recreational waters.

Temperatures approaching or exceeding 20°C have been measured during summer in polar regions at the surfaces of barren fellfield soils under cloudless skies around solar noon. However, despite the upper temperature limit for the growth of cold‐adapted microbes—which are abundant in polar soils and have pivotal roles in nutrient cycling—typically being close to this temperature, previous studies have not addressed the consequences of climate change for the metabolism of these organisms in the natural environment. Here in a 5‐year field experiment on Alexander Island in the southern maritime Antarctic, we show that the abundance of Pseudogymnoascus roseus, the most widespread decomposer fungus in maritime Antarctic fellfield soils, is reduced by 1–2 orders of magnitude when irrigated and nutrient‐amended soils are warmed to >20°C during summer. Laboratory experiments under conditions mimicking those during midsummer in the natural environment indicated that the hyphal extension rates of P. roseus isolates and the activities of five extracellular enzymes are reduced by 54%–96% at high water availability after exposure to temperatures cycling daily from 2 to 21°C and 2 to 24°C, relative to temperatures cycling from 2 to 18°C. Given that the temperatures of surface soils at the study site already reach 19°C during midsummer, the observations reported here suggest that, at predicted rates of warming arising from moderate greenhouse gas emissions, inhibitory effects of climate change on the metabolism of P. roseus could manifest themselves within the next few decades. Furthermore, with peak temperatures at the surfaces of fellfield soils at other maritime Antarctic locations and in High Arctic and alpine regions already exceeding 20°C during summer, the observations suggest that climate warming has the potential to inhibit the growth of other cold‐adapted microbes, with negative effects on soils as the Earth's climate continues to warm. We report a metabarcoding study documenting the fungal taxa in 29 barren fellfield soils sampled from along a 1,650 km transect encompassing almost the entire maritime Antarctic (60–72°S) and the environmental factors structuring the richness, relative abundance, and taxonomic composition of three guilds and growth forms. The richness of the lichenised fungal guild, which accounted for 19% of the total fungal community, was positively associated with mean annual surface air temperature (MASAT), with an increase of 1.7 operational taxonomic units (OTUs) of lichenised fungi per degree Celsius rise in air temperature. Soil Mn concentration, MASAT, C:N ratio, and pH value determined the taxonomic composition of the lichenised guild, and the relative abundance of the guild was best predicted by soil Mn concentration. There was a 3% decrease in the relative abundance of the saprotrophic fungal guild in the total community for each degree Celsius rise in air temperature, and the OTU richness of the guild, which accounted for 39% of the community, was negatively associated with Mn concentration. The taxonomic composition of the saprotrophic guild varied with MASAT, pH value, and Mn, NH4+-N, and SO42− concentrations. The richness of the yeast community, which comprised 3% of the total fungal community, was positively associated with soil K concentration, with its composition being determined by C:N ratio. In contrast with a similar study in the Arctic, the relative abundance and richness of lichenised fungi declined between 60°S and 69°S, with those of saprotrophic Agaricales also declining sharply in soils beyond 63°S. Basidiomycota, which accounted for 4% of reads, were much less frequent than in vegetated soils at lower latitudes, with the Ascomycota (70% of reads) being the dominant phylum. We conclude that the richness, relative abundance, and taxonomic composition of guilds and growth forms of maritime Antarctic soil fungi are influenced by air temperature and edaphic factors, with implications for the soils of the region as its climate changes during the 21st century. The soils of the Maritime and sub‐Antarctic experience extreme environmental conditions but nonetheless host biological communities that can survive low temperatures, limited water availability and short day lengths or even the complete absence of solar radiation during the winter. We determined the soil organic carbon (SOC) and total nitrogen (N), soil microbial biomass (SMB), labile carbon (LC) and respiration rate (RR) in soil samples from the longest latitudinal transect (approximately 2,000 km) ever sampled exclusively in the Maritime and sub‐Antarctic, comprising 69 sites located between South Georgia (54°S) and south‐eastern Alexander Island (72°S). With the exception of the most northerly location (South Georgia), the greatest SMB values occurred at 67–68°S, despite the harsh environmental conditions. This is consistent with the relative nutrient richness of the soils at these latitudes, as indicated by large SOC and total soil N concentrations, which are in turn probably linked to more abundant guano and excreta deposition by sea birds and seals, which have fewer and smaller areas of ice‐free terrain to land or haul out on further south. South of 68°S, SOC values declined with increasing latitude, which is probably due to increasingly extreme environmental conditions. We also found that the SOC and SMB values, the proportion of SOC considered labile (LC/SOC) and the carbon mineralization rates expressed as either RR/SOC or RR/SMB were all small compared with values from less extreme temperate and tropical regions. However, the proportion of the SOC in the microbial biomass (SMB/SOC) was substantially greater than that reported for non‐polar soils. We conclude that although the soils of the Maritime and sub‐Antarctic have small and relatively inactive microbial communities, they are characterized by efficient conversion of organic resources into microbial biomass and large affinities for added substrates. There are few reliable data sets to inspire confidence in policymakers that soil organic carbon (SOC) can be measured on farms. We worked with farmers in the Tamar Valley region of southwest England to select sampling sites under similar conditions (soil type, aspect and slope) and management types. Topsoils (2–15 cm) were sampled in autumn 2015, and percentage soil organic matter (%SOM) was determined by loss on ignition and used to calculate %SOC. We also used the stability of macroaggregates in cold water (WSA) ('soil slaking') as a measure of 'soil health' and investigated its relationship with SOC in the clay‐rich soils. %SOM was significantly different between management types in the order woodland (11.1%) = permanent pasture (9.5%) > ley‐arable rotation (7.7%) = arable (7.3%). This related directly to SOC stocks that were larger in fields under permanent pasture and woodland compared with those under arable or ley‐arable rotation whether corrected for clay content (F = 8.500, p <.0001) or not (F = 8.516, p <.0001). WSA scores were strongly correlated with SOC content whether corrected for clay content (SOCadjR2 =.571, p <.0001) or not (SOCunadjR2 = 0.490, p =.002). Time since tillage controlled SOC stocks and WSA scores, accounting for 75.5% and 51.3% of the total variation, respectively. We conclude that (1) SOC can be reliably measured in farmed soils using accepted protocols and related to land management and (2) WSA scores can be rapidly measured in clay soils and related to SOC stocks and soil management. The legacy nitrate–nitrogen (NO3−−N) in the soils is a long‐term threat to groundwater in regions with thick unsaturated zones, and it is thus important to investigate the amounts and sources of NO3−−N for effective environmental management. Given the substantial conversions of arable farmlands to non‐fertilizer land uses for the restoration of degraded land, the legacy effects of NO3−−N in thick loessial unsaturated zones in China remain uncertain. We collected soil samples from loess profiles >13‐m deep under arable farmland and grassland, apricot, pine, peashrub, willow, and poplar converted from arable farmland over 15–35 years. We determined their nitrate content and isotope compositions to quantify the accumulation and sources of the former and its potential threat to groundwater. The seven NO3−−N profiles exhibited a parabolic shape with peak depths at 2.6–9.2 m. Greater peak depths corresponded to greater NO3−−N values, and the peak nitrate level varied from 0 to 10 m. The total NO3−−N accumulation ranged 3,181–9,018 kg N ha−1, and the non‐fertilizer profiles accounted for 35–73% of the arable farmland. The nitrate sources varied with depths. According to the isotope mass balance, the nitrate above, near, and below the peak depths mainly originated from atmospheric NO3− (25–34%), synthetic N fertilizer (49–68%), and soil organic N and manure (50–73%). The estimated residence time of nitrate in the soil ranged from 270 to 620 years. In contrast to the situation in plants inhabiting most of the world's ecosystems, mycorrhizal fungi are usually absent from roots of the only two native vascular plant species of maritime Antarctica, Deschampsia antarctica and Colobanthus quitensis. Instead, a range of ascomycete fungi, termed dark septate endophytes (DSEs), frequently colonise the roots of these plant species. We demonstrate that colonisation of Antarctic vascular plants by DSEs facilitates not only the acquisition of organic nitrogen as early protein breakdown products, but also as non‐proteinaceous d‐amino acids and their short peptides, accumulated in slowly‐decomposing organic matter, such as moss peat. Our findings suggest that, in a warming maritime Antarctic, this symbiosis has a key role in accelerating the replacement of formerly dominant moss communities by vascular plants, and in increasing the rate at which ancient carbon stores laid down as moss peat over centuries or millennia are returned to the atmosphere as CO2. Temporal changes in the concentrations of plant-available phosphorus (P) in soil (Olsen-P), total soil-P and P activation coefficient (the ratio of Olsen-P to residual-P (i.e. an approximation to total-P)) were measured in plots that received consistent inorganic nitrogen, phosphorus and potassium plus organic fertilizers annually. Maize and winter wheat crops were grown in rotation for 24 years. Olsen-P and P activation coefficient declined significantly in the earlier years (< 12 years) for treatments that did not include any P fertilizer, and increased over the same period for the P-fertilized treatments. The rates of change in the Olsen-P and P activation coefficient values were positively related to P balance. Global patterns in diversity were estimated for cyanobacteria-dominated hypolithic communities that colonize ventral surfaces of quartz stones and are common in desert environments. A total of 64 hypolithic communities were recovered from deserts on every continent plus a tropical moisture sufficient location. Community diversity was estimated using a combined t-RFLP fingerprinting and high throughput sequencing approach. The t-RFLP analysis revealed desert communities were different from the single non-desert location. A striking pattern also emerged where Antarctic desert communities were clearly distinct from all other deserts. Some overlap in community similarity occurred for hot, cold and tundra deserts. A further observation was that the producer-consumer ratio displayed a significant negative correlation with growing season, such that shorter growing seasons supported communities with greater abundance of producers, and this pattern was independent of macroclimate. Highthroughput sequencing of 16S rRNA and nifH genes from four representative samples validated the t-RFLP study and revealed patterns of taxonomic and putative diazotrophic diversity for desert communities from the Taklimakan Desert, Tibetan Plateau, Canadian Arctic and Antarctic. All communities were dominated by cyanobacteria and among these 21 taxa were potentially endemic to any given desert location. Some others occurred in all but the most extreme hot and polar deserts suggesting they were relatively less well adapted to environmental stress. The t-RFLP and sequencing data revealed the two most abundant cyanobacterial taxa were Phormidium in Antarctic and Tibetan deserts and Chroococcidiopsis in hot and cold deserts. The Arctic tundra displayed a more heterogenous cyanobacterial assemblage and this was attributed to the maritime-influenced sampling location. The most abundant heterotrophic taxa were ubiquitous among samples and belonged to the Acidobacteria, Actinobacteria, Bacteroidetes, and Proteobacteria. Sequencing using nitrogenase gene-specific primers revealed all putative diazotrophs were Proteobacteria of the orders Burkholderiales, Rhizobiales, and Rhodospirillales. We envisage cyanobacterial carbon input to the system is accompanied by nitrogen fixation largely from non-cyanobacterial taxa. Overall the results indicate desert hypoliths worldwide are dominated by cyanobacteria and that growing season is a useful predictor of their abundance. Gelifluction, thixotropy and yield stress were measured from < 5 g soil samples taken from Signy, Alectoria, Greenwich, Wiencke and Livingston islands in the maritime Antarctic using a temperature-controlled rotational rheometer. The small sample size that this method permitted is compatible with sampling from sensitive sample locations. An oscillating 10 Pa shear stress was applied to samples at -0.5 kPa water potential. Two freeze-thaw cycles had temperature ramps from 5°C to -10°C over 2 h, followed by -10°C to 5°C over 2 h and finally at 5°C for 1 h. At freezing onset, the shear modulus, G, dropped to 4-50 per cent of thawed G, with no differences between locations. At thawing onset, G dropped to 8-32 per cent of thawed G, with significant differences between locations ( P < 0.001). Thixotropy was then measured by applying a 2 kPa oscillating shear stress for 10 min, followed by relaxation at 10 Pa for 2 h. The increased shear stress caused G to drop to less than 8 per cent of the pre-stressed value, with no difference between locations. After 0.1 and 2 h, G was 18-65 per cent and 31-82 per cent of the pre-stressed value, respectively. A shear ramp determined yield stresses ranging from 494-2217 Pa. These findings demonstrate the potential risk of more frequent freeze-thaw cycles or the occurrence of thawed soil to the stability of polar soils. Gelifluction through more frequent freeze-thaw cycles could result in increased slope movement, whereas thixotropy caused by trampling of thawed soils could exacerbate mechanical damage of surface soils. Rising global temperatures may increase the rates of soil organic matter decomposition by heterotrophic microorganisms, potentially accelerating climate change further by releasing additional carbon dioxide (CO2) to the atmosphere. However, the possibility that microbial community responses to prolonged warming may modify the temperature sensitivity of soil respiration creates large uncertainty in the strength of this positive feedback. Both compensatory responses (decreasing temperature sensitivity of soil respiration in the long-term) and enhancing responses (increasing temperature sensitivity) have been reported, but the mechanisms underlying these responses are poorly understood. In this study, microbial biomass, community structure and the activities of dehydrogenase and ß-glucosidase enzymes were determined for 18 soils that had previously demonstrated either no response or varying magnitude of enhancing or compensatory responses of temperature sensitivity of heterotrophic microbial respiration to prolonged cooling. The soil cooling approach, in contrast to warming experiments, discriminates between microbial community responses and the consequences of substrate depletion, by minimising changes in substrate availability. The initial microbial community composition, determined by molecular analysis of soils showing contrasting respiration responses to cooling, provided evidence that the magnitude of enhancing responses was partly related to microbial community composition. There was also evidence that higher relative abundance of saprophytic Basidiomycota may explain the compensatory response observed in one soil, but neither microbial biomass nor enzymatic capacity were significantly affected by cooling. Our findings emphasise the key importance of soil microbial community responses for feedbacks to global change, but also highlight important areas where our understanding remains limited. The McMurdo Dry Valleys of Antarctica are an extreme polar desert. Mineral soils support subsurface microbial communities and translucent rocks support development of hypolithic communities on ventral surfaces in soil contact. Despite significant research attention, relatively little is known about taxonomic and functional diversity or their interrelationships. Here we report a combined diversity and functional interrogation for soil and hypoliths of the Miers Valley in the McMurdo Dry Valleys of Antarctica. The study employed 16S rRNA fingerprinting and high throughput sequencing combined with the GeoChip functional microarray. The soil community was revealed as a highly diverse reservoir of bacterial diversity dominated by actinobacteria. Hypolithic communities were less diverse and dominated by cyanobacteria. Major differences in putative functionality were that soil communities displayed greater diversity in stress tolerance and recalcitrant substrate utilization pathways, whilst hypolithic communities supported greater diversity of nutrient limitation adaptation pathways. A relatively high level of functional redundancy in both soil and hypoliths may indicate adaptation of these communities to fluctuating environmental conditions. [ABSTRACT FROM AUTHOR]

Abstract: We investigated the relationships between fungal community composition, latitude and a range of physicochemical parameters in 58 soils sampled from a 2370 km latitudinal gradient between South Georgia (54°S, 38°W) in the sub-Antarctic and Mars Oasis (72°S, 68°W) on Alexander Island in the southern maritime Antarctic. Our study, which is based on approximately ten times the number of samples used in previous similar studies, indicates that latitude and its associated environmental parameters are not related to fungal community composition. Significant changes in the composition of soil fungal communities were observed in relation to gradients of the ratio of total organic carbon to nitrogen, and, to a lesser extent, soil pH. A novel bacteriophage infecting Staphylococus pasteuri was isolated during a screen for phages in Antarctic soils. The phage named SpaA1 is morphologically similar to phages of the family Siphoviridae. The 42,784 bp genome of SpaA1 is a linear, double-stranded DNA molecule with 3' protruding cohesive ends. The SpaA1 genome encompasses 63 predicted proteincoding genes which cluster within three regions of the genome, each of apparently different origin, in a mosaic pattern. In two of these regions, the gene sets resemble those in prophages of Bacillus thuringiensis kurstaki str. T03a001 (genes involved in DNA replication/transcription, cell entry and exit) and B. cereus AH676 (additional regulatory and recombination genes), respectively. The third region represents an almost complete genome (except for the short terminal segments) of a distinct bacteriophage, MZTP02. Nearly the same gene module was identified in prophages of B. thuringiensis serovar monterrey BGSC 4AJ1 and B. cereus Rock4-2. These findings suggest that MZTP02 can be shuttled between genomes of other bacteriophages and prophages, leading to the formation of chimeric genomes. The presence of a complete phage genome in the genome of other phages apparently has not been described previously and might represent a 'fast track' route of virus evolution and horizontal gene transfer. Another phage (BceA1) nearly identical in sequence to SpaA1, and also including the almost complete MZTP02 genome within its own genome, was isolated from a bacterium of the B. cereus/B. thuringiensis group. Remarkably, both SpaA1 and BceA1 phages can infect B. cereus and B. thuringiensis, but only one of them, SpaA1, can infect S. pasteuri. This finding is best compatible with a scenario in which MZTP02 was originally contained in BceA1 infecting Bacillus spp, the common hosts for these two phages, followed by emergence of SpaA1 infecting S. pasteuri. Mechanisms to mitigate global climate change by sequestering carbon ( C) in different 'sinks' have been proposed as at least temporary measures. Of the major global C pools, terrestrial ecosystems hold the potential to capture and store substantially increased volumes of C in soil organic matter ( SOM) through changes in management that are also of benefit to the multitude of ecosystem services that soils provide. This potential can only be realized by determining the amount of SOM stored in soils now, with subsequent quantification of how this is affected by management strategies intended to increase SOM concentrations, and used in soil C models for the prediction of the roles of soils in future climate change. An apparently obvious method to increase C stocks in soils is to augment the soil C pools with the longest mean residence times ( MRT). Computer simulation models of soil C dynamics, e.g. RothC and Century, partition these refractory constituents into slow and passive pools with MRTs of centuries to millennia. This partitioning is assumed to reflect: (i) the average biomolecular properties of SOM in the pools with reference to their source in plant litter, (ii) the accessibility of the SOM to decomposer organisms or catalytic enzymes, or (iii) constraints imposed on decomposition by environmental conditions, including soil moisture and temperature. However, contemporary analytical approaches suggest that the chemical composition of these pools is not necessarily predictable because, despite considerable progress with understanding decomposition processes and the role of decomposer organisms, along with refinements in simulation models, little progress has been made in reconciling biochemical properties with the kinetically defined pools. In this review, we will explore how advances in quantitative analytical techniques have redefined the new understanding of SOM dynamics and how this is affecting the development and application of new modelling approaches to soil C. Nitrogen is a key regulator of primary productivity in many terrestrial ecosystems. Historically, only inorganic N (NH4 + and NO3 -) and L-amino acids have been considered to be important to the N nutrition of terrestrial plants. However, amino acids are also present in soil as small peptides and in D-enantiomeric form. We compared the uptake and assimilation of N as free amino acid and short homopeptide in both L- and D-enantiomeric forms. Sterile roots of wheat (Triticum aestivum L.) plants were exposed to solutions containing either 14C-labelled L-alanine, D-alanine, L-trialanine or D-trialanine at a concentration likely to be found in soil solution (10 μM). Over 5 h, plants took up L-alanine, D-alanine and L-trialanine at rates of 0.9±0.3, 0.3±0.06 and 0.3±0.04 mmol g-1 root DW h-1, respectively. The rate of N uptake as L-trialanine was the same as that as Lalanine. Plants lost ca.60% of amino acid C taken up in respiration, regardless of the enantiomeric form, but more (ca.80%) of the L-trialanine C than amino acid C was respired. When supplied in solutions of mixed N form, N uptake as D-alanine was ca.5-fold faster than as NO3 -, but slower than as L-alanine, L-trialanine and NH4 +. Plants showed a limited capacity to take up D-trialanine (0.04±0.03 mmol g-1 root DW h-1), but did not appear to be able to metabolise it. We conclude that wheat is able to utilise L-peptide and D-amino acid N at rates comparable to those of N forms of acknowledged importance, namely L-amino acids and inorganic N. This is true even when solutes are supplied at realistic soil concentrations and when other forms of N are available. We have measured total soil organic carbon (SOC), dissolved organic carbon (DOC), and microbial lipid contents (as indices of microbial biomass and community structure), and their distributions to 60 cm depth in soils from replicated medium-term (2003-2008) experimental arable plots subject to different tillage regimes in Scotland. The treatments were zero tillage (ZT), minimum tillage (MT; cultivation to 7 cm), the conventional tillage (CT) practice of ploughing to 20 cm, and deep ploughing (DP) to 40 cm depth. In the 0-30 cm depth range, SOC content (corrected for bulk density differences between tillage treatments) was greatest under ZT and MT, but over 0-60 cm depth the SOC contents of these treatments were similar to the CT and DP treatments. DOC concentrations declined with increasing depth in ZT and MT above 20 cm, but there were no significant differences with depth in the CT and DP treatments. Beneath 20 cm, there was little change in DOC concentration with depth for all treatments, although for the MT treatment, there was less DOC beneath the depth of cultivation. The total microbial biomass decreased with increasing depth over the 0-60 cm range in the ZT and MT treatments, whereas it decreased with depth only below 30-40 cm in the CT and DP treatments. The microbial biomass was significantly different only between 0-5 cm in the ZT, CT and DP treatments, but not for other depths between all treatments. The bacterial biomass was greater in the ZT treatment than in MT, CT and DP near the soil surface, but not significantly different over the whole profile (0-60 cm). The fungal biomass decreased with depth in the ZT and MT treatments over the whole 0-60 cm depth range, whereas it decreased with depth only below 20 cm in the CT and DP treatments. Abstract: Global warming in the Arctic may alter decomposition rates in Arctic soils and therefore nutrient availability. In addition, changes in the length of the growing season may increase plant productivity and the rate of labile C input below ground. We carried out an experiment in which inorganic nutrients (NH4NO3 and NaPO4) and organic substrates (glucose and glycine) were added to soils sampled from across the mountain birch forest-tundra heath ecotone in northern Sweden (organic and mineral soils from the forest, and organic soil only from the heath). Carbon dioxide production was then monitored continuously over the following 19 days. Neither inorganic N nor P additions substantially affected soil respiration rates when added separately. However, combined N and P additions stimulated microbial activity, with the response being greatest in the birch forest mineral soil (57% increase in CO2 production compared with 26% in the heath soil and 8% in the birch forest organic soil). Therefore, mineralisation rates in these soils may be stimulated if the overall nutrient availability to microbes increases in response to global change, but N deposition alone is unlikely to enhance decomposition. Adding either, or both, glucose and glycine increased microbial respiration. Isotopic separation indicated that the mineralisation of native soil organic matter (SOM) was stimulated by glucose addition in the heath soil and the forest mineral soil, but not in the forest organic soil. These positive ‘priming’ effects were lost following N addition in forest mineral soil, and following both N and P additions in the heath soil. In order to meet enhanced microbial nutrient demand, increased inputs of labile C from plants could stimulate the mineralisation of SOM, with the soil C stocks in the tundra-heath potentially most vulnerable. Global environmental change, related to climate change and the deposition of airborne N-containing contaminants, has already resulted in shifts in plant community composition among plant functional types in Arctic and temperate alpine regions. In this paper, we review how key ecosystem processes will be altered by these transformations, the complex biological cascades and feedbacks that might result, and some of the potential broader consequences for the earth system. Firstly, we consider how patterns of growth and allocation, and nutrient uptake, will be altered by the shifts in plant dominance. The ways in which these changes may disproportionately affect the consumer communities, and rates of decomposition, are then discussed. We show that the occurrence of a broad spectrum of plant growth forms in these regions (from cryptogams to deciduous and evergreen dwarf shrubs, graminoids and forbs), together with hypothesized low functional redundancy, will mean that shifts in plant dominance result in a complex series of biotic cascades, couplings and feedbacks which are supplemental to the direct responses of ecosystem components to the primary global change drivers. The nature of these complex interactions is highlighted using the example of the climate-driven increase in shrub cover in low-Arctic tundra, and the contrasting transformations in plant functional composition in mid-latitude alpine systems. Finally, the potential effects of the transformations on ecosystem properties and processes that link with the earth system are reviewed. We conclude that the effects of global change on these ecosystems, and potential climate-change feedbacks, cannot be predicted from simple empirical relationships between processes and driving variables. Rather, the effects of changes in species distributions and dominances on key ecosystem processes and properties must also be considered, based upon best estimates of the trajectories of key transformations, their magnitude and rates of change. Mass populations of toxin-producing cyanobacteria commonly develop in fresh-, brackish- and marine waters and effective strategies for monitoring and managing cyanobacterial health risks are required to safeguard animal and human health. A multi-interdisciplinary study, including two UK freshwaters with a history of toxic cyanobacterial blooms, was undertaken to explore different approaches for the identification, monitoring and management of potentially-toxic cyanobacteria and their associated risks. The results demonstrate that (i) cyanobacterial bloom occurrence can be predicted at a local- and national-scale using process-based and statistical models; (ii) cyanobacterial concentration and distribution in waterbodies can be monitored using remote sensing, but minimum detection limits need to be evaluated; (iii) cyanotoxins may be transferred to spray-irrigated root crops; and (iv) attitudes and perceptions towards risks influence the public's preferences and willingness-to-pay for cyanobacterial health risk reductions in recreational waters. Warming-induced release of CO2 from the large carbon (C) stores in arctic soils could accelerate climate change. However, declines in the response of soil respiration to warming in long-term experiments suggest that microbial activity acclimates to temperature, greatly reducing the potential for enhanced C losses. As reduced respiration rates with time could be equally caused by substrate depletion, evidence for thermal acclimation remains controversial. To overcome this problem, we carried out a cooling experiment with soils from arctic Sweden. If acclimation causes the reduction in soil respiration observed after experimental warming, then it should subsequently lead to an increase in respiration rates after cooling. We demonstrate that thermal acclimation did not occur following cooling. Rather, during the 90 days after cooling, a further reduction in the soil respiration rate was observed, which was only reversed by extended re-exposure to warmer temperatures. We conclude that over the time scale of a few weeks to months, warming-induced changes in the microbial community in arctic soils will amplify the instantaneous increase in the rates of CO2 production and thus enhance C losses potentially accelerating the rate of 21st century climate change. Nostoc commune Vaucher (a cyanobacterium) is a very conspicuous terrestrial primary producer in Victoria Land, continental Antarctica. Because polar ecosystems are considered to be especially sensitive to environmental changes, understanding the environmental constraints on net carbon (C) fixation by N. commune is necessary to determine the effects of environmental changes on the ecological functioning of ice-free areas of the continent. A model describing net C fixation in terrestrial populations of N. commune in an Antarctic dry valley was constructed using field and laboratory measurements in which N. commune colonies were exposed to different combinations of incident irradiance (400–700 nm), temperature, and degree of desiccation. For desiccated N. commune mats with water content ≤ 30% saturation, net C fixation was highly variable between replicates and could not be modelled. However, for colonies at > 30% saturation, rates of net C fixation and dark respiration depended strongly on irradiance and temperature. Net C fixation reached a maximum rate of 21.6 μg C m− 2 s− 1 at irradiance of approximately 250 μmol m− 2 s− 1 and the optimum temperature of 20.5 °C. Agreement between predicted short-term net C fixation and field and laboratory measurements allowed estimation of total seasonal fixation, using previously published environmental data. Annual net C fixation was estimated in the range 14.5–21.0 g C fixed m− 2 Nostoc mat, depending on year/season. Estimates for different seasons correlated with thermal time (accumulated hours above 0 °C during the year) rather than irradiance, in contrast to communities in local lacustrine environments, where irradiance is the main driver of primary productivity. In the terrestrial habitat, N. commune appears to compromise between an ability to capitalize on short periods of higher temperature and efficient utilization of lower irradiance at low temperature. Lignin is a major determinant of the decomposition of plant materials in soils. Advances in transgenic technology have led to the possibility of modifying lignin to improve the pulping properties of plant materials for papermaking. Previous studies have shown that lignin modifications also affect the rate of plant material decay in soil. The aim of this work was to investigate short-term changes in soil microbial community structures when tobacco residues with reduced activity of enzymes in the monolignol pathway decompose. The residues from lignin-modified plants all decomposed faster than unmodified plant materials. The relative proportions of some of the structural groups of microbial phospholipid fatty acids were affected by genetic modifications, especially the proportion of double unsaturated chain fatty acids, indicative of fungi. The agronomic and pulping performance of transgenic trees with altered lignin has been evaluated in duplicated, long-term field trials. Poplars expressing cinnamyl alcohol dehydrogenase (CAD) or caffeate/5-hydroxyferulate O-methyltransferase (COMT) antisense transgenes were grown for four years at two sites, in France and England. The trees remained healthy throughout the trial. Growth indicators and interactions with insects were normal. No changes in soil microbial communities were detected beneath the transgenic trees. The expected modifications to lignin were maintained in the transgenics over four years, at both sites. Kraft pulping of tree trunks showed that the reduced-CAD lines had improved characteristics, allowing easier delignification, using smaller amounts of chemicals, while yielding more high-quality pulp. β-Glucosidase is involved in the degradation of cellulose in soils and has potential for monitoring biological soil quality. We assayed β-glucosidase activity in 29 permanent grassland soils from England and Wales with contrasting physico-chemical and biological properties (clay content 22–68%; total carbon 29–80 mg g−1; microbial carbon 412–3412 μg g−1). Substrate induced β-glucosidase activity ranged between 1.12 and 6.12 μmol para-nitrophenol g−1 soil h−1 and was positively correlated with concentrations of clay, total carbon and microbial carbon. This suggests that substrate-induced β-glucosidase activity is an integrative measure of physico-chemical and biological soil properties and may have applications in monitoring biological soil quality.
