28. Hemodynamic and Clinical Profiles of Pulmonary Arterial Hypertension Patients with GDF2 and BMPR2 Variants.

Abstract

Asians have a higher carrier rate of pulmonary arterial hypertension (PAH)-related genetic variants than Caucasians do. This study aimed to identify PAH-related genetic variants using whole exome sequencing (WES) in Asian idiopathic and heritable PAH cohorts. A WES library was constructed, and candidate variants were further validated by polymerase chain reaction and Sanger sequencing in the PAH cohort. In a total of 69 patients, the highest incidence of variants was found in the BMPR2, ATP13A3, and GDF2 genes. Regarding the BMPR2 gene variants, there were two nonsense variants (c.994C>T, p. Arg332*; c.1750C>T, p. Arg584*), one missense variant (c.1478C>T, p. Thr493Ile), and one novel in-frame deletion variant (c.877_888del, p. Leu293_Ser296del). Regarding the GDF2 variants, there was one likely pathogenic nonsense variant (c.259C>T, p. Gln87*) and two missense variants (c.1207G>A, p. Val403Ile; c.38T>C, p. Leu13Pro). The BMPR2 and GDF2 variant subgroups had worse hemodynamics. Moreover, the GDF2 variant patients were younger and had a significantly lower GDF2 value (135.6 ± 36.2 pg/mL, p = 0.002) in comparison to the value in the non-BMPR2/non-GDF2 mutant group (267.8 ± 185.8 pg/mL). The BMPR2 variant carriers had worse hemodynamics compared to the patients with the non-BMPR2/non-GDF2 mutant group. Moreover, there was a significantly lower GDF2 value in the GDF2 variant carriers compared to the control group. GDF2 may be a protective or corrected modifier in certain genetic backgrounds. 33. The expression of matrix metalloproteinases in macrophage phenotypes

Abstract

Macrophages, matrix metalloproteinases (MMPs) and tissue inhibitor of MMPs (TIMPs) are key factors in atherosclerotic plaque formation and plaque rupture, both of which are driven by macrophage accumulation and activation. The thesis investigated whether distinct phenotypes of macrophages may play different roles in MMP or TIMP expression during inflammation or plaque formation. The results showed that classically activated Ml macrophages significantly up-regulated MMP-l, MMP-3, MMP-7, MMP-IO, MMP-12, MMP-14 and MMP-25 and down-regulated TIMP-3, which suggests a strong increase in matrix-degrading potential. Patterns of response to classical activation in vitro are dominated by LPS and pro-inflammatory cytokines, only MMP-12, MMP-14 and MMP-25 expressions being partly affected by interferon-y. On the other hand, alternatively activated M2 macrophages strongly up-regulated MMP-II, MMP-12 and MMP-25, and this was accompanied by lower expression of many other MMPs and greater expression of TIMP-3. Hence alternative activation may rather selectively increase the potential for elastolysis by MMP-12, without promoting degradation of other matrix components. The MMP expression stimulated by classical activation depended on activation of mitogen activated protein kinases, phosphinositide-3 kinase, and inhibitor of K.8 kinase-2. In addition, MMP-l, and MMP-I0 co-localised with nuclear localised NF-KB in human atherosclerotic plaques. Furthermore, unstable plaques have higher COX-2, MMP-I, MMP-3, MMP-IO and MMP-14 expression in foam cell macro phages when compared to stable human coronary atherosclerotic plaques. By contrast, stable plaques had higher expression of the M2 marker, CD206, and TIMP-3 than human unstable plaques. In conclusion, Ml macrophages selectively increase the balance of several MMPs over TIMPs in macrophages in vitro and in unstable plaques, an example of inflammation in vivo. However, only limited groups of MMPs and TIMP-3 were significantly over-expressed in M2 macrophages.

Abstract

Aims Pulmonary arterial hypertension (PAH) is characterized by extensive pulmonary arterial remodelling. Although mesenchymal stem cell (MSC)-derived exosomes provide protective effects in PAH, MSCs exhibit limited senescence during in vitro expansion compared with the induced pluripotent stem cells (iPSCs). Moreover, the exact mechanism is not known. Methods and results In this study, we used murine iPSCs generated from mouse embryonic fibroblasts with triple factor (Oct4, Klf4, and Sox2) transduction to determine the efficacy and action mechanism of iPSC-derived exosomes (iPSC-Exo) in attenuating PAH in rats with monocrotaline (MCT)-induced pulmonary hypertension. Both early and late iPSC-Exo treatment effectively prevented the wall thickening and muscularization of pulmonary arterioles, improved the right ventricular systolic pressure, and alleviated the right ventricular hypertrophy in MCT-induced PAH rats. Pulmonary artery smooth muscle cells (PASMC) derived from MCT-treated rats (MCT-PASMC) developed more proliferative and pro-migratory phenotypes, which were attenuated by the iPSC-Exo treatment. Moreover, the proliferation and migration of MCT-PASMC were reduced by iPSC-Exo with suppression of PCNA, cyclin D1, MMP-1, and MMP-10, which are mediated via the HIF-1α and P21-activated kinase 1/AKT/Runx2 pathways. Conclusion IPSC-Exo are effective at reversing pulmonary hypertension by reducing pulmonary vascular remodelling and may provide an iPSC-free therapy for the treatment of PAH. [ABSTRACT FROM AUTHOR]

41. Pyrene-based covalent organic polymers with nano carbonaceous composites for efficient supercapacitive energy storage.

Abstract

Covalent organic polymers (COPs) show great potential as supercapacitor electrodes due to their tunable properties, stability, and high capacitance. In this work, we prepared a covalent organic polymer (Py-DSDA-COP) through a Schiff base reaction of the 1,3,6,8-tetrakis(4-formylphenyl)pyrene monomer (Py-Ph-CHO) and 4,4′-disulfanediyldianiline (DSDA), validated its chemical structure, and investigated its thermal stability, porosity, morphology, and electrochemical properties. The Py-DSDA-COP displayed high semi-crystallinity, good porosity, and a specific capacitance of 56.18 F g−1. To improve its electrochemical performance, we incorporated different nano-carbonaceous materials [fullerene (C60), MWCNTs, and SWCNTs] into Py-DSDA-COP and compared their characteristics. The Py-DSDA-COP/SWCNTs showed the highest specific capacitance (171 F g−1) at 1 A g−1and highest energy density (23.7 W h kg−1) with a capacity retention of 93% after 2000 cycles, which is superior to those of Py-DSDA-COP/MWCNTs and Py-DSDA-COP/C60 nanocomposites. These superior electrochemical characteristics of Py-DSDA-COP/SWCNTs are attributed to the excellent π–π stacking interactions, higher EDLC behavior, higher conductivity, lower diameter, and higher surface area of SWCNTs than those of MWCNTs. Abstract

BACKGROUND: Pulmonary arterial hypertension (PAH), defined as the presence of a mean pulmonary artery pressure > 20 mmHg, pulmonary artery wedge pressure ≤ 15 mmHg, and pulmonary vascular resistance (PVR) > 2 Wood units based on expert consensus, is characterized by a progressive and sustained increase in PVR, which may lead to right heart failure and death. PAH is a well-known complication of connective tissue diseases (CTDs), such as systemic sclerosis, systemic lupus erythematosus, Sjogren’s syndrome, and other autoimmune conditions. In the past few years, tremendous progress in the understanding of PAH pathogenesis has been made, with various novel diagnostic and screening methods for the early detection of PAH proposed worldwide. OBJECTIVES: This study aimed to obtain a comprehensive understanding and provide recommendations for the management of CTD-PAH in Taiwan, focusing on its clinical importance, prognosis, risk stratification, diagnostic and screening algorithm, and pharmacological treatment. METHODS: The members of the Taiwan Society of Cardiology (TSOC) and Taiwan College of Rheumatology (TCR) reviewed the related literature thoroughly and integrated clinical trial evidence and real-world clinical experience for the development of this consensus. CONCLUSIONS: Early detection by regularly screening at-risk patients with incorporations of relevant autoantibodies and biomarkers may lead to better outcomes of CTD-PAH. This consensus proposed specific screening flowcharts for different types of CTDs, the risk assessment tools applicable to the clinical scenario in Taiwan, and a recommendation of medications in the management of CTD-PAH.

CPs 4–6 are 2D layers, showing (64·8·10)(6)-2,4L3, (36·46·53)-hxl and (4·82)-fes topologies, respectively, whereas CP 7 displays a 3-fold interpenetrated 3D net with a (45·8)-dmp topology and 8 forms a 1D → 2D polycatenation with a (63·82·10)(6)2-2,2,4C5 topology. A structural comparison of 1–8 indicates that the flexibility of the bis-pyridyl-bis-amide plays an important role in the formation of entangled Co(II) CPs auxiliarily supported by the angular dicarboxylate ligand. Abstract

Protein oligomerization occurs frequently both in vitro and in vivo, with specific functionalities associated with different oligomeric states. The YqiC protein from Salmonella Typhimurium forms a homotrimer through its C‐terminal coiled‐coil domain, and the protein is closely linked to the colonization and invasion of the bacteria to the host cells. To elucidate the importance of the oligomeric state of YqiC in vivo and its relation with bacterial infection, we mutated crucial residues in YqiC's coiled‐coil region and confirmed the loss of trimer formation using chemical crosslinking and size exclusion chromatography coupled with multiple angle light scattering (SEC‐MALS) techniques. The yqiC‐knockout strain complemented with mutant YqiC showed significantly reduced colonization and invasion of Salmonella to host cells, demonstrating the critical role of YqiC oligomerization in bacterial pathogenesis. Furthermore, we conducted a protein–protein interaction study of YqiC using a pulled‐down assay coupled with mass spectrometry analysis to investigate the protein's role in bacterial virulence. The results reveal that YqiC interacts with subunits of Complex II of the electron transport chain (SdhA and SdhB) and the β‐subunit of F0F1‐ATP synthase. These interactions suggest that YqiC may modulate the energy production of Salmonella and subsequently affect the assembly of crucial virulence factors, such as flagella. Overall, our findings provide new insights into the molecular mechanisms of YqiC's role in S. Typhimurium pathogenesis and suggest potential therapeutic targets for bacterial infections.
