Abstract Coupling high-capacity cathode and Li-anode with solid-state electrolyte has been demonstrated as an effective strategy for increasing the energy densities and safety of rechargeable batteries. However, the limited ion conductivity, the large interfacial resistance, and unconstrained Li-dendrite growth hinder the application of solid-state Li-metal batteries. Here, a poly(ether-urethane)-based solid-state polymer electrolyte with self-healing capability is designed to reduce the interfacial resistance and provides a high-performance solid-state Li-metal battery. With its dynamic covalent disulfide bonds and hydrogen bonds, the proposed solid-state polymer electrolyte exhibits excellent interfacial self-healing ability and maintains good interfacial contact. Full cells are assembled with the two integrated electrodes/electrolytes. As a result, the Li||Li symmetric cells exhibit stable long-term cycling for more than 6000 h, and the solid-state Li-S battery shows a prolonged cycling life of 700 cycles at 0.3 C. The use of ultrasound imaging technology shows that the interfacial contact of the integrated structure is much better than those of traditional laminated structure. This work provides an interesting interfacial dual-integrated strategy for designing high-performance solid-state Li-metal batteries.

The sparse leaf patch of seashore paspalum (Paspalum vaginatum Sw.) caused by Microdochium paspali seriously impacts the landscape value of turf and poses a challenge to the maintenance and management of golf courses. Little is known about the genome of M. paspali or the potential genes underlying pathogenicity. In this study, we present a high-quality genome assembly of M. paspali with 14 contigs using the Nanopore and Illumina platform. The M. paspali genome is roughly 37.32 Mb in size and contains 10,365 putative protein-coding genes. These encompass a total of 3,830 pathogen-host interactions (PHI) genes, 481 carbohydrate-active enzymes (CAZymes) coding genes, 105 effectors, and 50 secondary metabolite biosynthetic gene clusters (SMGCs) predicted to be associated with pathogenicity. Comparative genomic analysis suggests M. paspali has 672 species-specific genes (SSGs) compared to two previously sequenced non-pathogenic Microdochium species, including 24 species-specific gene clusters (SSGCs). Comparative transcriptomic analyses reveal that 739 PHIs, 198 CAZymes, 40 effectors, 21 SMGCs, 213 SSGs, and 4 SSGCs were significantly up-regulated during the process of infection. In conclusion, the study enriches the genomic resources of Microdochium species and provides a valuable resource to characterize the pathogenic mechanisms of M. paspali.

Abstract Through agronomic traits and sequencing data, the cultivated and wild varieties of grapes and peaches were analyzed and compared in terms of fruit size, fruit flavor, fruit resistance, and fruit color. Cultivated grapes and peaches have advantages in fruit size, soluble sugar content, sugar and acid ratio, etc. Wild grapes and peaches have utility value in resistance. The results showed that there were 878 and 301 differentially expressed genes in cultivated and wild grapes and peaches in the three growth stages, respectively based on the next-generation sequencing study. Ten and twelve genes related to the differences between cultivated and wild grapes and peaches were found respectively. Among them, three genes, namely chalcone synthase (CHS), glutathione S-transferase (GST) and malate dehydrogenase (MDH1) were present in both cultivated and wild grapes and peaches.

Abstract Background Pears and apples are both perennial deciduous trees of the Rosaceae family, and both are important economic fruit trees worldwide. The emergence of many varieties in the market has been mostly domesticated from wild to cultivated and regulated by the differential expression of genes. However, the molecular process and pathways underlying this phenomenon remain unclear. Four typical wild and cultivar pear and apple trees at three developmental stages were used in our study to investigate the molecular process at the transcriptome level. Result Physiological observations indicated the obvious differences of size, weight, sugar acid content and peel color in wild and cultivar fruit among each developmental stage. Using next-generation sequencing based RNA-seq expression profiling technology, we produced a transcriptome in procession of a large fraction of annotated pear and apple genes, and provided a molecular basis underlying the phenomenon of wild and cultivar fruit tree differences. 5921 and 5744 differential expression genes were identified in pear and apple at three developmental stages respectively. We performed temporal and spatial differential gene expression profiling in developing fruits. Several key pathways such as signal transduction, photosynthesis, translation and many metabolisms were identified as involved in the differentiation of wild and cultivar fruits. Conclusion In this study, we reported on the next-generation sequencing study of the temporal and spatial mRNA expression profiling of pear and apple fruit trees. Also, we demonstrated that the integrated analysis of pear and apple transcriptome, which strongly revealed the consistent process of domestication in Rosaceae fruit trees. The results will be great influence to the improvement of cultivar species and the utilization of wild resources.

Lignin is one of the main components of plant cell walls, which provides mechanical support for plants and also contributes to resisting against plant pathogenic fungi. In the fruit industry, the lignin content can affect the quality of fruit. The biosynthesis of lignin involves a variety of enzymes, of which caffeoyl-CoA 3-O-methyltransferase (CCoAOMT) is a class of methyltransferases that plays an essential role in lignin biosynthesis. Studies have been conducted on the CCoAOMT gene family in several species, including arabidopsis (Arabidopsis thaliana), black poplar (Populus nigra), and cotton (Gossypium hirsutum). Still, there is relatively little research on this gene family in the Rosaceae. In this study, we used bioinformatics to identify and characterize the CCoAOMT gene family in apple (Malus domestica), chinese white pear (Pyrus bretschneideri), and peach (Prunus persica). In total, 35 CCoAOMT genes were identified in the three Rosaceae species: 8 from chinese white pear, 12 from apple, and 15 from peach. By using structure analysis and collinearity analysis, we found 12 conserved motifs and 12 pairs of CCoAOMT genes with collinearity. In the phylogenetic tree, the gene family was mainly divided into two groups. The genes had different expression patterns during the growth and development stage of fruit, a finding that is consistent with the pattern of lignin accumulation. This study will be beneficial for further study of CCoAOMT genes.

We sampled 127 turfgrass soil samples from 33 golf courses in NC, EC, and SC for plant-parasitic nematodes (PPNs). PPNs were extracted from soil samples using the shallow dish method and were identified at the genus or species levels with a combination of morphological and molecular methods. The results revealed 41 species of nematode belonging to 20 genera and 10 families. Nine genera are new records of PPNs associated with turfgrass in China. The PPNs show strong geographical distributions. Of the 20 genera, Helicotylenchus, Paratrichodorus, Hoplolaimus, Meloidogyne, Hemicriconemoides, and Mesocriconema showed higher infestation and frequency, and most of these genera had numbers in soil samples above established damage thresholds. Four golf courses had soil samples with PPNs > 30%, indicating the potential for nematode damage. The biodiversity indices H’, SR, J’, λ, and H2 showed significant differences among different regions and turfgrass species; H’, SR, J’, and H2 were significantly higher in EC than in NC and SC, while λ was lowest in EC. Creeping bentgrass had the highest H’, SR, J’, and H2 and the lowest λ in comparison with seashore paspalum and hybrid bermudagrass. These findings provide baseline information on the occurrence of turfgrass-associated PPNs in China, and have important implications for the effective management of PPNs causing damage on turfgrass.

Dollar spot is reported to be caused by multiple Clarireedia species and is a serious problem on many turfgrasses around the world. To our knowledge, the distribution of different Clarireedia species and their sensitivity profiles to fungicides remains unknown. In this study, a total of 275 isolates were characterized by ITS sequence. Amounts of 124, 59 and 75 isolates were identified as C. jacksonii, C. monteithiana and C. paspali, respectively, while each species of C. homoeocarpa and C. bennettii had only five isolates. Four and three isolates were identified as two potential new species, which remained to be further characterized. C. jacksonii and C. monteithiana were distributed worldwide, while C. paspali was restricted to China. Of the isolates with host information, 81% (93/115) and 19% (22/115) of C. jacksonii isolates were collected from C3 and C4 plants, respectively, 97% (56/58) of the C. monteithiana isolates were collected from C4 plants and all C. paspali isolates were collected from C4 plants. The coexistence of different Clarireedia species on the same C4 host type in the same locales was found in Shanghai (Paspalum vaginatum), Jiangsu (Paspalum vaginatum) and Florida (Cynodon dactylon). The study revealed that differential fungicide sensitivity patterns were observed in different species in Clarireedia for the first time. Similar differential sensitivity profiles were also found in the locales with coexistence of at least two species. The findings from this study suggest that the adjacent coexistence of different Clarireedia species and the differential fungicide sensitivity profiles of different species will complicate dollar spot disease control.

Abstract The Large Eddy Simulation (LES) / Conditional Moment Closure (CMC) model with detailed chemistry is used for modelling spark ignition and flame propagation in a turbulent methane jet in ambient air. Two centerline and one off-axis ignition locations are simulated. We focus on predicting the flame kernel formation, flame edge propagation and stabilization. The current LES/CMC computations capture the three stages reasonably well compared to available experimental data. Regarding the formation of flame kernel, it is found that the convection dominates the propagation of its downstream edge. The simulated initial downstream and radial flame propagation compare well with OH-PLIF images from the experiment. Additionally, when the spark is deposited at off-centerline locations, the flame first propagates downstream and then back upstream from the other side of the stoichiometric iso-surface. At the leading edge location, the chemical source term is larger than others in magnitude, indicating its role in the flame propagation. The time evolution of flame edge position and the final lift-off height are compared with measurements and generally good agreement is observed. The conditional quantities at the stabilization point reflect a balance between chemistry and micro-mixing. This investigation, which focused on model validation for various stages of spark ignition of a turbulent lifted jet flame through comparison with measurements, demonstrates that turbulent edge flame propagation in non-premixed systems can be reasonably well captured by LES/CMC. Large eddy simulation (LES) with three-dimensional conditional moment closure (CMC) subgrid model for combustion is applied to simulate a swirl-stabilized nonpremixed methane flame with localized extinction, with special focus on the effects of heat loss to the burner surface. The convective wall heat loss is modeled through introducing a source term in the conditionally filtered total enthalpy equation for the CMC cells adjacent to the wall. The mean heat flux is high on the middle surface of the bluff body, but relatively low near its edges. The turbulent heat flux based on the gradient of the resolved temperature is relatively low compared to the laminar counterpart, but increases with the turbulent intensity. The heat loss facilitates the occurrences of extinction and re-ignition for the CMC cells immediately adjacent to the wall, evidenced by comparing flame structures in the near-wall CMC cells. This can be directly linked to the increase of the mean conditional scalar dissipation near the wall in the heat loss case. Furthermore, the degree of local extinction near the bluff body measured by conditional reactedness at stoichiometry is intensified due to the wall heat loss. However, the results also show that there is negligible influence of wall heat loss on the probability density function (PDF) of the lift-off height, demonstrating the dominance of aerodynamic effects on flame stabilization. Emerging multidrug resistance (MDR) in Clarireedia spp. is a huge challenge to the management of dollar spot (DS) disease on turfgrass. Insight into the molecular basis of resistance mechanisms may help identify key molecular targets for developing novel effective chemicals. Previously, a MDR isolate (LT586) of C. jacksonii with significantly reduced sensitivities to propiconazole, boscalid, and iprodione, and a fungicide-sensitive isolate (LT15) of the same species were isolated from creeping bentgrass ( Agrostis stolonifera L.). The present study aimed to further explore the molecular mechanisms of resistance by using genome-wide transcriptional analyses of the two isolates. A total of 619 and 475 differentially expressed genes (DEGs) were significantly down and upregulated in the MDR isolate LT586, compared with the sensitive isolate LT15 without fungicide treatment. Three hundreds and six and 153 DEGs showed significantly lower and higher expression in the MDR isolate LT586 than those in the sensitive isolate LT15, which were commonly induced by the three fungicides. Most of the 153 upregulated DEGs were xenobiotic detoxification-related genes and genes with transcriptional functions. Fifty and 17 upregulated DEGs were also commonly observed in HRI11 (a MDR isolate of the C. jacksonii ) compared with the HRS10 (a fungicide-sensitive isolate of same species) from a previous study without and with the treatment of propiconazole, respectively. The reliability of RNA-seq data was further verified by qRT-PCR method using a few select potentially MDR-related genes.
