Abstract Genetic studies elucidate a link between testis-specific serine/threonine kinases (TSSKs) and male infertility in mammals, but the underlying mechanisms are unclear. Here, we identify a TSSK homolog in Drosophila, CG14305 (termed dTSSK), whose mutation impairs the histone-to-protamine transition during spermiogenesis and causes multiple phenotypic defects in nuclear shaping, DNA condensation, and flagellar organization in spermatids. Genetic analysis demonstrates that kinase catalytic activity of dTSSK, which is functionally conserved with human TSSKs, is essential for male fertility. Phosphoproteomics identify 828 phosphopeptides/449 proteins as potential substrates of dTSSK enriched primarily in microtubule-based processes, flagellar organization and mobility, and spermatid differentiation and development, suggesting that dTSSK phosphorylates various proteins to orchestrate postmeiotic spermiogenesis. Among them, the two substrates, protamine-like protein Mst77F/Ser9 and transition protein Mst33A/Ser237, are biochemically validated to be phosphorylated by dTSSK in vitro, and are genetically demonstrated to be involved in spermiogenesis in vivo. Collectively, our findings demonstrate that broad phosphorylation mediated by TSSKs plays an indispensable role in spermiogenesis.

The histone H4 basic patch is critical for chromatin structure and regulation of the chromatin machinery. However, the biological roles of these positively charged residues and the mechanisms by which they regulate gene expression remain unclear. In this study, we used histone mutagenesis to investigate the physiological function and downstream regulatory genes of H4 residues R17 and R19 in Drosophila. We found all histone mutations including R17A/E/H and R19A/E/H (R17 and R19 of H4 are substituted by A, E and H respectively) result in a range of growth defects and abnormalities in chromosomal high-order structures, whereas R17E mutation is embryonic lethal. RNA-seq demonstrates that downregulated genes in both R17A and R19A show significant overlap and are enriched in development-related pathways. In addition, Western and cytological analyses showed that the R17A mutation resulted in a significant reduction in H4K16 acetylation and male offspring, implying that the R17 may be involved in male dosage compensation mechanisms. R19 mutation on the other hand strongly affect Gpp (Dot1 homologue in flies)-mediated H3K79 methylation, possibly through histone crosstalk. Together these results provide insights into the differential impacts of positive charges of H4 basic patch R17/R19 on regulation of gene transcription during developmental processes.

As a hot research topic in urban geography, spatiotemporal interaction analysis has been used to detect the hotspot mobility patterns of crowds and urban structures based on the origin-destination (OD) flow data, which provide useful information for urban planning and traffic management applications. However, existing methods mainly focus on the detection of explicit spatial interaction patterns (such as spatial flow clusters) in OD flow data, with less attention to the discovery of underlying crowd travel demands. Therefore, this paper proposes a framework to discover the crowd travel demands by associating the dynamic spatiotemporal interaction patterns and the contextual semantic features of the geographical environment. With urban functional zones (UFZs) as the basic units of human mobility in urban spaces, this paper gives a case study in Wuhan, China, to detect and interpret the human mobility patterns based on the characteristics of spatiotemporal interaction between UFZs. Firstly, we build the spatiotemporal interaction matrix based on the OD flows of different UFZs and analyze the characteristics of the interaction matrix. Then, hotspot poles, defined as the local areas where people gather significantly, are extracted using the Gi-statistic-based spatial hotspot detection algorithm. Next, we develop a frequent interaction pattern mining method to detect the frequent interaction patterns of the hotspot poles. Finally, based on the detected frequent interaction patterns, we discover the travel demands of crowds with semantic features of corresponding urban functional zones. The characteristics of crowd travel distance and travel time are further discussed. Experiments with floating car data, road networks, and POIs in Wuhan were conducted, and results show that the underlying travel demands can be better discovered and interpreted by the proposed framework and methods in this paper. This study helps to understand the characteristics of human movement and can provide support for applications such as urban planning and facility optimization.

The difluoroamino derivatives of prismane were designed and calculated at the B3LYP/6-311G** level. The detonation performances, as well as the band gap, were investigated to look for high energy density compounds. Our calculations showed that difluoroamino group is an effective substitute group for increasing density and heats of formation. All compounds have large heats of formation, and there is a linear relationship between the heats of formation (HOFs) and the substituent numbers. Bond dissociation energies of trigger bonds are all over 200kJ/mol, which indicated that these prismane derivatives have good stability. More than three substituted groups made the detonation performances meet the requirement of energetic materials. Our calculations provided basic information for the further syntheses of title molecules.

Plant virus diseases, also known as “plant cancers”, cause serious harm to the agriculture of the world and huge economic losses every year. Antiviral agents are one of the most effective ways to control plant virus diseases. Ningnanmycin is currently the most successful anti-plant virus agent, but its field control effect is not ideal due to its instability. In recent years, great progress has been made in the research and development of antiviral agents, the mainstream research direction is to obtain antiviral agents or lead compounds based on structural modification of natural products. However, no antiviral agent has been able to completely inhibit plant viruses. Therefore, the development of highly effective antiviral agents still faces enormous challenges. Therefore, we reviewed the recent research progress of anti-plant virus agents based on natural products in the past decade, and discussed their structure-activity relationship (SAR) and mechanism of action. It is hoped that this review can provide new inspiration for the discovery and mechanism of action of novel antiviral agents.

Piezoelectric print-heads (PPHs) are used with a variety of fluid materials with specific functions. Thus, the volume flow rate of the fluid at the nozzle determines the formation process of droplets, which is used to design the drive waveform of the PPH, control the volume flow rate at the nozzle, and effectively improve droplet deposition quality. In this study, based on the iterative learning and the equivalent circuit model of the PPHs, we proposed a waveform design method to control the volume flow rate at the nozzle. Experimental results show that the proposed method can accurately control the volume flow of the fluid at the nozzle. To verify the practical application value of the proposed method, we designed two drive waveforms to suppress residual vibration and produce smaller droplets. The results are exceptional, indicating that the proposed method has good practical application value.

This study focused on cadmium (Cd) uptake by two rice varieties, Yuzhenxiang (YZX) and Xiangwanxian 12 (XWX), which differ in their capacity to accumulate Cd, i.e., XWX > YZX. Treatments with three different gradients of soil Cd concentrations showed that with the increase in soil Cd concentration gradient, the Cd content in each rice plant organ also increased, i.e., Cd-3 > Cd-2 > Cd-1. The trend in the Cd content of each organ was such that the farther the organ from the root, the lower its Cd content, i.e., root > stem and sheath > leaf > grain. We observed that for all four growth stages, the booting stage is the key stage in terms of Cd absorption, where the highest levels of accumulation are observed, that is, booting stage > full heading stage > tillering stage > maturity stage. Of the two cultivars, XWX had higher SOD, POD, and CAT activities but lower MDA content. In contaminated soils, SOD, POD, and CAT activities increased gradually with the increase in Cd concentration, while MDA content decreased, which indicated that the low Cd variety XWX had an advantage over the high Cd variety YZX. Through the comparative analysis of photosynthetic physiology, it was found that the low-Cd-accumulating rice variety XWX appeared more tolerant to Cd, while the high-Cd-accumulating rice YZX was more sensitive. Therefore, the low Cd rice variety XWX was more suitable for planting safe rice in Cd-polluted paddy fields.

In some applications of piezoelectric three-dimensional inkjet printing, the materials used are power-law fluids as they are shear thinning. Their time-varying viscosities affect the droplet formation, which is determined by the volume flow rate at the nozzle outlet. To obtain a fine printing effect, it is necessary to present a driving waveform design method that considers the shear-thinning viscosities of materials to control the volume flow rate at the nozzle outlet, which lays the foundation for the single and stable droplet generation during the printing process. In this research, we established the relationship between the driving waveform and the volume flow rate at the nozzle outlet by modifying a model that describes the inkjet mechanism of power-law fluid. The modified model was used to present a driving waveform design method based on iterative learning control. The iterative learning law of the method was designed based on the gradient descent algorithm and demonstrated its convergence. The driving waveform design method was verified to be practical and feasible by implementing drop generation experiments.

Piezoelectric three-dimensional inkjet printing has been used to manufacture heterogeneous objects due to its high level of flexibility. The materials used are non-Newtonian inks with complex rheological properties, and their behavior in the context of inkjet printing has not been fully understood: for example, the fact that the shear-thinning viscosity affects the droplet generation. Therefore, a control strategy coping with shear-thinning behaviors is needed to ensure printing consistency. In this paper, a novel model-based approach is presented to describe the shear-thinning ink dynamics inside the piezoelectric inkjet printhead, which provides the basis to design the excitation parameters in a systematic way. The dynamic equation is simplified into a quasi-one-dimensional equation through the combination of the boundary layer theory and the constitutive equation of the power-law fluid, of which the viscosity is shear-thinning. Based on this, a nonlinear time-varying equivalent circuit model is presented to simulate the power-law fluid flow rate inside the tube. The feasibility and effectiveness of this model can be evaluated by comparing the results of computational fluid dynamics and the experimental results.

Abstract During neuronal pruning, phagocytes engulf shed cellular debris to avoid inflammation and maintain tissue homeostasis. How phagocytic receptors recognize degenerating neurites had been unclear. Here, we identify two glucosyltransferases Alg8 and Alg10 of the N-glycosylation pathway required for dendrite fragmentation and clearance through genetic screen. The scavenger receptor Draper (Drpr) is N-glycosylated with complex- or hybrid-type N-glycans that interact specifically with galectins. We also identify the galectins Crouching tiger (Ctg) and Hidden dragon (Hdg) that interact with N-glycosylated Drpr and function in dendrite pruning via the Drpr pathway. Ctg and Hdg are required in hemocytes for expression and function, and are induced during dendrite injury to localize to injured dendrites through specific interaction with exposed phosphatidylserine (PS) on the surface membrane of injured dendrites. Thus, the galectins Ctg and Hdg bridge the interaction between PS and N-glycosylated Drpr, leading to the activation of phagocytosis.

IntroductionPrevious studies have shown that stroke patients exhibit greater neuroimaging-derived biological “brain age” than control subjects. This difference, known as the brain age gap (BAG), is calculated by comparing the chronological age with predicted brain age and is used as an indicator of brain health and aging. However, whether stroke accelerates the process of brain aging in patients with small-volume infarcts has not been established. By utilizing longitudinal data, we aimed to investigate whether small-volume infarctions can significantly increase the BAG, indicating accelerated brain aging.MethodsA total of 123 stroke patients presenting with small-volume infarcts were included in this retrospective study. The brain age model was trained via established protocols within the field of machine learning and the structural features of the brain from our previous study. We used t-tests and regression analyses to assess longitudinal brain age changes after stroke and the associations between brain age, acute stroke severity, and poststroke outcome factors.ResultsSignificant brain aging occurred between the initial and 6-month follow-ups, with a mean increase in brain age of 1.04 years (t = 3.066, p 3 at admission presented more pronounced adverse effects on brain aging, even after adjusting for confounders such as chronological age, sex, and total intracranial volume (F1,117 = 7.339, p = 0.008, η2 = 0.059). There were significant differences in the proportional brain age difference at 6 months among the different functional outcome groups defined by the Barthel Index (F2,118 = 4.637, p = 0.012, η2 = 0.073).ConclusionStroke accelerates the brain aging process, even in patients with relatively small-volume infarcts. This phenomenon is particularly accentuated in elderly patients, and both stroke severity and poststroke functional outcomes are closely associated with accelerated brain aging. Further studies are needed to explore the mechanisms underlying the accelerated brain aging observed in stroke patients, with a particular focus on the structural alterations and plasticity of the brain following minor strokes.
