In recent years, the temperature-adjusted asphalt pavement has been an extensive concern by scholars in various countries, and this pavement can reduce temperature-related diseases. In this study, the shaped composite phase change materials (CPCMs) were successfully synthesized by two processes, which are vacuum impregnation and epoxy curing. Firstly, the applicability of CPCMs in asphalt mixtures was evaluated by microscopic characterization, chemical compatibility, thermal properties, durability, and leakage stability. Secondly, CPCMs were applied to the asphalt mixture to evaluate its temperature-adjusted characteristics and pavement performance. Finally, the performance of the temperature-adjusted asphalt mixture was analyzed by integrating all factors. The research shows that the prepared CPCMs have excellent thermal properties and durability, the phase transition temperature is 48.93 °C, and the phase transition enthalpy is 106.5 J/g, which fully meets the requirements for use in pavement. The temperature-adjusted asphalt mixture could alleviate the occurrence of extreme temperature, which was 4.9 °C lower than the conventional mixture. The pavement performance of the temperature-adjusted asphalt mixture can meet the specified standards for humid areas. Considering the factors, the recommended amount of CPCMs is 1.5%. The research results provide a basis for the promotion of temperature-adjusted asphalt pavement and effectively support the development of pavement engineering technology.

ObjectiveTo study the current status of hemophilia B (HB) patients in the central and western regions of China.MethodsThis cross-sectional, multicenter study was conducted in seven provinces in the central and western regions of China from April 2019 to June 2023. Samples were collected for the factor IX activity, inhibitor screen, and gene mutation. Furthermore, the status of six index joints and quality of life (QoL) were assessed.ResultsA total of 185 HB patients (mild 15, moderate 75, and severe 95) with a median age of 12.17 years were enrolled. 30.3% (56/185) of patients had a family history of HB. 34.6% (64/185) of HB patients had diagnostic delay and 38.5% (69/179) experienced treatment delay. The incidence of inhibitors was 6.1% (11/179). We identified 123 genetic variants in this study, with missense mutations being the most common. 84.0% (89/106) of HB mothers were genetically identified as carriers, with 27.7% (13/47) of carriers having clotting factor levels less than 0.40 IU/ml. 71.4% (132/185) of HB patients had a history of joint hemorrhage, with a rate of target joint in these patients was 64.4% (85/132). Lower extremity joints were most often affected in patients. ABSTRACTObjective The prognosis of acute myeloid leukemia (AML) remains poor although the basic and translational research has been highly productive in understanding the genetics and pathopoiesis of AML and a plethora of targeted therapies have been developed. Consequently, it is crucial to deepen the knowledge of molecular pathogenesis underlying AML for the advancement of new treatment options.Method A RSK gene family-related signature was constructed to investigate whether RSK gene family members were useful in predicting the prognosis of AML patients. The relationship between the RSK gene family-related signature and the infiltration of immune cells was further assessed using the CIBERSORT algorithm. The ‘oncoPredict’ package was used to analyze relationships between the RSK gene family-related signature and the sensitivity to drugs or small molecules.Results Patients were classified into two groups using the RSK gene family-related signature following the median risk score. Overall survival (OS) was significantly longer in patients with low-risk scores than that in patients with high-risk scores as showed by both training and validation datasets. Moreover, the signature was helpful in predicting 1-year, 3-year, and 5-year OS in training and validation datasets. In addition, it was identified that low-risk patients exhibited greater sensitivity to 20 drugs or small molecules and that high-risk patients had higher sensitivity to 38 drugs or small molecules.Conclusion RSK gene family members, particularly RPS6KA1 and RPS6KA4, may help to predict prognosis for AML patients. Furthermore, RPS6KA1 may serve as a novel drug target for AML.

In order to significantly improve the viscoelasticity and durability of asphalt materials, Spray polyurea (SPUA) resin modified materials with different particle sizes were obtained by low-temperature fine grinding process in this study, and the corresponding SPUA modified asphalt was prepared. The basic properties of resin modified asphalt were compared and analyzed. The influence of SPUA resin modification materials on the apparent viscosity of asphalt was analyzed. Through dynamic shear rheometer (DSR) and bending beam rheometer (BBR) test, the effect of SPUA resin material on rheological properties of asphalt was systematically investigated. Furthermore, the modifier and asphalt binder were characterized by scanning electron microscopy (SEM) and X-ray energy dispersive spectroscopy (EDS). Results show that decreasing the particle size or increasing the dosage of the SPUA modifier could improve the softening point and apparent viscosity and reduce the ductility, which indicate SPUA modifier could improve the high temperature shear resistance and elastic recovery performance of asphalt binder. The SPUA modifier with smaller particle size has better high temperature rheological modification effect and less negative effect on low temperature performance deterioration. Although part of the functional spherical structure was damaged, the 0.075 mm SPUA modifier could closely interact with the base asphalt perfectly. This study would provide the scientific references for the application of Spray polyurea resin materials in the field of asphalt pavement materials.

Trace elements and minerals play a significant role in human health and diseases. In recent years, ionomics has been rapidly and widely applied to explore the distribution, regulation, and crosstalk of different elements in various physiological and pathological processes. On the basis of multi-elemental analytical techniques and bioinformatics methods, it is possible to elucidate the relationship between the metabolism and homeostasis of diverse elements and common diseases. The current review aims to provide an overview of recent advances in the application of ionomics in metabolic disease research. We mainly focuses on the studies about ionomic or multi-elemental profiling of different biological samples for several major types of metabolic diseases, such as diabetes mellitus, obesity, and metabolic syndrome, which reveal distinct and dynamic patterns of ion contents and their potential benefits in the detection and prognosis of these illnesses. Accumulation of copper, selenium, and environmental toxic metals as well as deficiency of zinc and magnesium appear to be the most significant risk factors for the majority of metabolic diseases, suggesting that imbalance of these elements may be involved in the pathogenesis of these diseases. Moreover, each type of metabolic diseases has shown a relatively unique distribution of ions in biofluids and hair/nails from patients, which might serve as potential indicators for the respective disease. Overall, ionomics not only improves our understanding of the association between elemental dyshomeostasis and the development of metabolic disease but also assists in the identification of new potential diagnostic and prognostic markers in translational medicine.

The incorporation of crumb rubber (CR) into asphalt pavement materials can improve the performance of asphalt pavement and generate environmental benefits. However, the storage stability of the crumb rubber asphalt (CRA) remains an issue that needs to be resolved. This study explores the interaction laws among various modified materials based on the response surface methodology. Optimal preparation dosages of each material are determined, and performance predictions and validations are conducted. The storage stability of the CRA compounded with epoxidized soybean oil (ESO) and polyester fiber (PF) is investigated by combining traditional compatibility testing methods with refined characterization methods. The results indicate that the modification of CRA exhibits better rheological properties when the percentages of CR, PF, and ESO are 22%, 0.34%, and 3.21%, respectively. The addition of ESO effectively complements the light components of CRA to improve asphalt compatibility, and the addition of PF alleviates the adverse effects of ESO’s softening effect on rheological properties through stabilization and three-dimensional strengthening. The scientifically compounded additions of ESO and PF can effectively enhance the storage stability and rheological properties of CRA, promoting the development of sustainable and durable roads.

Zinc and cadmium are environmental contaminants that can cause disease by affecting the activity of DNA-repair proteins. In this study, we investigated the effect of Zn2+ and Cd2+ on the Candida albicans Pif1, a DNA-repair helicase that plays a critical role in ensuring genomic stability. We show that Zn2+ and Cd2+ strongly inhibit both the ATPase and the unwinding activities of CaPif1, but have no effect on its DNA binding activity. High concentrations of Cd2+ may bind to the cysteine residues of CaPif1, and its inhibition appears to be difficult to be restored by ethylene diamine tetraacetic acid, while inhibition due to Zn2+ can. When the two ions are at low concentrations, increasing the concentration of ATP in the reaction can appropriately weaken the inhibitory effect of Zn2+, while cysteine can reduce the inhibitory effect of Cd2+. In addition, we found that for both Cd2+ and Zn2+ the inhibition effects were nearly 100 times greater in reduced environments than in non-reducing environments. When heavy metals stimulate the body’s response, the environment of the body becomes less reducing, and thus the tolerance of CaPif1 to heavy metals will be stronger. We propose that CaPif1 may resist the toxicity of heavy metals through this mechanism. Altogether, our results provide new insights into the mechanisms by which heavy metals are toxic to DNA-repair proteins.

This study focuses on using spodumene flotation tailings (SFT) to prepare phosphoric acid-activated metakaolin geopolymer, in which the replacement of metakaolin (MK) by a high percentage (up to 75 wt.%) of tailings was achieved. The compressive strength of geopolymer mortar was significantly improved with SFT as aggregates. In addition, the mechanical properties could also be enhanced by an increased concentration of phosphoric acid (H3PO4) solution or a decreased aggregate particle size. The optimized geopolymer mortar composite was SFT:MK = 3:1, which was activated by H3PO4 solution with a concentration of 51 vol%, followed by curing at 55 °C for 24 h. On the other hand, properties of the geopolymer mortar could also be affected by the morphology of the aggregates. For example, SFT as aggregates could produce more interconnected pores compared to standard sand. The major chemical structural units of geopolymer mortar were -P-O-Al- and AlPO4, which could be spontaneously generated according to the thermodynamic calculation results. Finally, many aluminum ions and a small amount of silicon ions could be leached from the tailings under acidic conditions.

Road asphalt pavements cover a high percentage of urban size and contribute to heat islands. This study proposed a new method to cool asphalt pavement by incorporating a kind of hybrid mineral filler (HMF) with high emissivity into a reference asphalt mixture prepared with limestone mineral filler (LMF). The physical, emissive, solar reflective, and rheological properties of asphalt mastic and the thermal performances of asphalt mixture were covered to investigate the possibility of the proposed strategy. From Fourier transform infrared spectrum test, it can be found that HMF was physically blended with asphalt. The emissivity results show that HMF increased the emissivity of asphalt mastic from 0.9204 to 0.9820. The asphalt mastic containing HMF had similar solar reflectance with the control one. In addition, HMF could enhance the rutting resistance of asphalt mastic according to the results of multiple stress creep recovery tests. When HMF replaced LMF, the thermal conductivity of the asphalt mixture with HMF increased by 0.26 W/(m·K) (the reference value was 1.72 W/(m·K)). The combined effect of high emissivity and thermal conductivity led to a lower surface temperature (i.e., −5.4 °C) in the tests. The results of this study demonstrate that HMF is a potential material to cool asphalt pavements.

Selenium (Se) is an important trace element that mainly occurs in the form of selenocysteine in selected proteins. In prokaryotes, Se is also required for the synthesis of selenouridine and Se-containing cofactor. A large number of selenoprotein families have been identified in diverse prokaryotic organisms, most of which are thought to be involved in various redox reactions. In the last decade or two, computational prediction of selenoprotein genes and comparative genomics of Se metabolic pathways and selenoproteomes have arisen, providing new insights into the metabolism and function of Se and their evolutionary trends in bacteria and archaea. This review aims to offer an overview of recent advances in bioinformatics analysis of Se utilization in prokaryotes. We describe current computational strategies for the identification of selenoprotein genes and generate the most comprehensive list of prokaryotic selenoproteins reported to date. Furthermore, we highlight the latest research progress in comparative genomics and metagenomics of Se utilization in prokaryotes, which demonstrates the divergent and dynamic evolutionary patterns of different Se metabolic pathways, selenoprotein families, and selenoproteomes in sequenced organisms and environmental samples. Overall, bioinformatics analyses of Se utilization, function, and evolution may contribute to a systematic understanding of how this micronutrient is used in nature.

Background/Aims: Most of estrogen receptor positive breast cancer patients respond well initially to endocrine therapies, but often develop resistance during treatment with selective estrogen receptor modulators (SERMs) such as tamoxifen. Altered expression and functions of microRNAs (miRNAs) have been reportedly associated with tamoxifen resistance. Thus, it is necessary to further elucidate the function and mechanism of miRNAs in tamoxifen resistance. Methods: Tamoxifen sensitivity was validated by using Cell Counting Kit-8 in tamoxifen-sensitive breast cancer cells (MCF-7, T47D) and tamoxifen-resistant cells (MCF-7/TAM, T47D/ TAM). Quantitative real-time polymerase chain reaction (qRT-PCR) was performed to detect the expression level of miR-449a in tamoxifen-sensitive/-resistant cells and patient serums. Dual-luciferase assay was used to identify the binding of miR-449a and predicted gene ADAM22. The expression level of ADAM22 was determined by qRT-PCR and western blotting in miR-449a +/- breast cancer cells. Subsequently, rescue experiments were carried out to identify the function of ADAM22 in miR-449a-reduced tamoxifen resistance. Finally, Gene ontology (GO) and Protein-protein interaction analyses were performed to evaluate the potential mechanisms of ADAM22 in regulating tamoxifen resistance. Results: MiR-449a levels were downregulated significantly in tamoxifen-resistant breast cancer cells when compared with their parental cells, as well as in clinical breast cancer serum samples. Overexpression of miR-449a re-sensitized the tamoxifen-resistant breast cancer cells, while inhibition of miR-449a conferred tamoxifen resistance in parental cells. Luciferase assay identified ADAM22 as a direct target gene of miR-449a. Additionally, silencing of ADAM22 could reverse tamoxifen resistance induced by miR-449a inhibition in ER-positive breast cancer cells. GO analysis results showed ADAM22 was mainly enriched in the biological processes of cell adhesion, cell differentiation, gliogenesis and so on. Protein-protein interaction analyses appeared that ADAM22 might regulate tamoxifen resistance through PPARG, LGI1, KRAS and LYN. Conclusion: Decreased miR-449a causes the upregulation of ADAM22, which induces tamoxifen resistance of breast cancer cells. These results suggest that miR-449a, functioning by targeting ADAM22, contributes to the mechanisms underlying breast cancer endocrine resistance, which may provide a potential therapeutic strategy in ER-positive breast cancers.

Abstract Natural halloysite nanotubes (HNTs) were hybridized with metal–organic frameworks (MOFs) to prepare novel composites. MOFs were transformed into carbon by carbonization calcination, and palladium (Pd) nanoparticles were introduced to build an emerging ternary compound system for hydrogen adsorption. The hydrogen adsorption capacities of HNT-MOF composites were 0.23 and 0.24 wt%, while those of carbonized products were 0.24 and 0.27 wt% at 25 °C and 2.65 MPa, respectively. Al-based samples showed higher hydrogen adsorption capacities than Zn-based samples on account of different selectivity between metal and hydrogen and approximate porous characteristics. More pore structures are generated by the carbonization reaction from metal–organic frameworks into carbon; high specific surface area, uniform pore size, and large pore volume benefited the hydrogen adsorption ability of composites. Moreover, it was also possible to promote hydrogen adsorption capacity by incorporating Pd. The hydrogen adsorption capacity of ternary compound, Pd-C-H3-MOFs(Al), reached 0.32 wt% at 25 °C and 2.65 MPa. Dissociation was assumed to take place on the Pd particles, then atomic and molecule hydrogen spilled over to the structure of carboxylated HNTs, MOFs, and the carbon products for enhancing the hydrogen adsorption capacity.

Photo-receptors are widely present in both prokaryotic and eukaryotic cells, which serves as the foundation of tuning cell behaviors with light. While practices in eukaryotic cells have been relatively established, trials in bacterial cells have only been emerging in the past few years. A number of light sensors have been engineered in bacteria cells and most of them fall into the categories of two-component and one-component systems. Such a sensor toolbox has enabled practices in controlling synthetic circuits at the level of transcription and protein activity which is a major topic in synthetic biology, according to the central dogma. Additionally, engineered light sensors and practices of tuning synthetic circuits have served as a foundation for achieving light based real-time feedback control. Here, we review programming bacteria cells with light, introducing engineered light sensors in bacteria and their applications, including tuning synthetic circuits and achieving feedback controls over microbial cell culture.

With the rapid development of economic construction, styrene-butadiene-styrene (SBS)-modified asphalt is being more and more widely used in highway engineering, but there are still many deficiencies in the process of its use. In order to further improve its performance for use, nano-organic palygorskite (A-Pal) and star-shaped SBS were compounded to obtain modified asphalt in this study. The high-temperature stability of SBS-modified asphalt was enhanced after incorporation with A-Pal for the high-temperature stability test by a dynamic shear rheometer. The A-Pal should improve the surface free energy and adhesion of SBS-modified asphalt by the water stability test analysis. The aging test shows that A-Pal can reduce the thermal oxygen decomposition of SBS and improve the anti-aging performance and the fatigue resistance of SBS-modified asphalt. A-Pal has a certain improvement effect on the low temperature performance of SBS-modified asphalt as shown by a low temperature crack resistance test. A-Pal-compounded SBS-modified asphalt features good storage stability in normal temperatures with the lowest critical compatibility temperature.

Patterning an oil-based ink on a solid surface based on a wettability difference is of significant importance for the application of offset printing. Herein, we describe a large-area patterning of oil-based ink on a self-organized TiO2 nanotubular layer based on a photocatalytic wettability conversion. The TiO2 nanotubular layer was fabricated by electrochemical anodization, which demonstrated a superhydrophobic wettability after modification with a self-assembled molecular layer. Subsequently, area-selective ultraviolet (UV) irradiation through a pre-designed pattern of water-based UV-resistant ink formed by an ink-jet technique was used to form a wettability difference. After removing the water-based ink, an oil-based ink was capable of depositing selectively on the superhydrophobic area to form the same pattern as the pre-designed pattern of water-based ink. This large-area patterning of an oil-based ink based on the photocatalytic wettability conversion is potentially applicable in offset printing.

The selective separation of chalcopyrite from galena (lead sulfide) through the flotation method is still a challenging task in the field of mineral engineering. Mercaptoacetic acid, though a common depressant of many gangue (commercially worthless) minerals, has shown to be less selective in the flotation separation of chalcopyrite and galena. This resent study therefore systematically investigated the selectivity of different mercapto acids (especially three types: 3-mercaptopropionic acid, 3-mercaptoisobutyric acid and mercaptoacetic acid) on the separation of chalcopyrite and galena by making the use of flotation experiments and first principle calculations. The calculation results demonstrated that the sulfhydryl and carboxyl groups existing on the molecular structure of three mercapto acids are the reactive and chelating centers to metal ions on sulfide mineral surfaces. Mercapto acids have higher binding energies to Cu2+ by 300−400 kJ/mol compared to Pb2+, indicating a higher affinity towards chalcopyrite. The order of reactivity and chelating ability noted was as follows: 3-mercaptopropionic acid > 3-mercaptoisobutyric acid > mercaptoacetic acid. Flotation results further showed that the selectivity of 3-mercaptopropionic acid or 3-mercaptoisobutyric acid was better than mercaptoacetic acid. The good agreement between the first principle calculations and the flotation results demonstrated that the former reagent could be served as a most selective depressant in the improved flotation separation of chalcopyrite and galena.

Styrene Butadiene Styrene (SBS) polymer-modified asphalt binders have become widely used in asphalt pavement because of their advantages in high- and low-temperature performance and fatigue resistance. Asphalt pavement is inevitably exposed to sunlight and ultraviolet (UV) radiation during its construction and service life. However, consideration of the aging effect of UV radiation is still limited in current pavement design and evaluation systems. In order to evaluate the impact of UV radiation on the aging properties of SBS-modified asphalt binders, UV aging tests were performed on Rolling Thin Film Oven Test (RTFOT)-aged samples with different UV radiation intensities and aging times. Sixteen different groups of tests were conducted to compare the rheological properties and functional group characteristics of SBS-modified asphalt binders. Dynamic Shear Rheometer (DSR), Bending Beam Rheometer (BBR), FTIR, and SEM tests were conducted to evaluate the aging mechanisms in various UV aging conditions. The results found that UV radiation seriously destroys the network structure formed by the cross-linking effect in SBS-modified asphalt binders, which aggravates the degradation of SBS and results in a great change of rheological properties after UV aging. The nature of SBS-modified asphalt binder aging resulted from the degradation of SBS and the changes of asphalt binder base composition, which lead to the transformation of colloidal structure and the deterioration of asphalt binder performance. The tests also found that continuous UV radiation can increase shrinkage stress in the asphalt binder surface and leads to surface cracking of the asphalt binder.

TiO2 pillared montmorillonite (T/M) modifiers have been studied to alleviate the aging of asphalt pavement and degrade automobile exhaust, but the photocatalytic activity of ordinary TiO2 is not good enough. In this study, in order to improve the photocatalytic performance of T/M, different metal (Ce, Cu, Fe) doped modifiers were prepared based on T/M. Metal doped TiO2 pillared montmorillonite was prepared by the sol-gel method. The modifier was characterized by X-ray diffraction (XRD) and an Ultraviolet visible (UV-Vis) spectrophotometer. The results show that TiO2 with different metal ions successfully entered into the layer of organic montmorillonite (OMMT) to form a pillared structure. Compared with the undoped TiO2 pillared montmorillonite (T/M), the optical absorption edge of the metal doped TiO2 pillared montmorillonite has an obvious red shift. In addition, the influences of the different content of modifiers on the properties of the original bitumen and catalytic capacities for automobile exhaust were also investigated. The results show that Ce doped TiO2 (Ce-T/M) pillared montmorillonite has the best improvement in high temperature performance and ultraviolet (UV) resistance of bitumen. In the experiment of automobile exhaust degradation, the degradation law of NO and HC showed Cu-T/M > Ce-T/M > Fe-T/M > T/M. These three kinds of metal ions can effectively improve the photocatalytic degradation efficiency of T/M.

Bitumen aging can lead to the deterioration of asphalt pavement performance, shortening the service life of road. In order to solve the problem that current studies on the ultraviolet (UV) aging of bitumen either ignore the effects of natural environmental conditions or only consider the effects of water. In this study, different aqueous media and UV coupled simulated aging tests were carried out on virgin bitumen and styrene butadiene styrene (SBS) modified bitumen in a UV environment chamber. The combination of macroscopic performance tests and microstructure tests was used to analyze the physical, rheological, and microstructure changes of virgin bitumen and SBS modified bitumen after The film oven test (TFOT) aging and UV aging in different environments (UV, UV + Water, UV + Acid, UV + Salt). Dynamic shear rheometer (DSR) results indicated that UV aging results in the increase of rutting factor and the improvement of rutting resistance at high temperature. The Fourier transform infrared spectrum (FTIR) results illustrated that the bitumen would be oxidized and SBS would be degraded under ultraviolet radiation. The four-component analysis test results showed that light component migrated to the heavy component during the aging process. Moreover, water will aggravate the UV aging of bitumen, and the presence of acid or salt worsens ultraviolet aging.

Acute lung injury (ALI) induced by intestinal ischemia/reperfusion (II/R) has high incidence and mortality, in which IL-1β was essential for the full development of ALI. However, the detailed regulating mechanism for this phenomenon remains to be unclear. The purpose of this study was to investigate whether inhibition of P38 MAPK could downregulate the expression of IL-1β to protect lung from acute injury in II/R rats. Here, we found that the level of pulmonary edema at 16 hours after operation (hpo) was obviously enhanced compared to that in 8hpo and sham groups. Immunofluorescent staining demonstrated that IL-1β and P38 MAPK were detected in lung tissues. And rats with II/R have the highest translation level for IL-1β and phosphorylation of P38 MAPK in lung tissues at 16hpo compared with 8hpo and sham groups. Moreover, administration of SB239063, an inhibitor of P38 α and β, could effectively downregulate the expressions of IL-1β and protects lung tissues from injury in II/R rats. Our findings indicate that the inhibition of P38 α and β may downregulate the expression of IL-1β to protect lung from acute injury in II/R, which could be used as a potential target for reducing ALI induced by II/R in the future clinical trial.

A novel nanocomposite ZnFe2O4-TiO2/MCM-41 (ZTM) was synthesized by a sol-gel method and characterized through X-ray diffraction (XRD), high-resolution transmission electron microscopy (HRTEM), N2 adsorption-desorption, Raman spectroscopy, and ultraviolet visible (UV-vis) spectrophotometry. The results confirmed the incorporation of ZnFe2O4-TiO2 nanoparticles inside the pores of the mesoporous MCM-41 host without destroying its integrity. ZnFe2O4 nanoparticles can inhibit the transformation of anatase into rutile phase of TiO2. Incorporation of ZnFe2O4-TiO2 within MCM-41 avoided the agglomeration of nanoparticles and reduced the band gap energy of TiO2 to enhance its visible light photocatalytic activity. UV-vis absorption edges of ZTM nanocomposites redshifted with the increase of Zn/Ti molar ratio. The nanocomposite approach could be a potential choice for enhancing the photoactivity of TiO2, indicating an interesting application in the photodegradation and photoelectric fields.

We report improved whole-genome shotgun sequences for the genomes of indica and japonica rice, both with multimegabase contiguity, or almost 1,000-fold improvement over the drafts of 2002. Tested against a nonredundant collection of 19,079 full-length cDNAs, 97.7% of the genes are aligned, without fragmentation, to the mapped super-scaffolds of one or the other genome. We introduce a gene identification procedure for plants that does not rely on similarity to known genes to remove erroneous predictions resulting from transposable elements. Using the available EST data to adjust for residual errors in the predictions, the estimated gene count is at least 38,000-40,000. Only 2%-3% of the genes are unique to any one subspecies, comparable to the amount of sequence that might still be missing. Despite this lack of variation in gene content, there is enormous variation in the intergenic regions. At least a quarter of the two sequences could not be aligned, and where they could be aligned, single nucleotide polymorphism (SNP) rates varied from as little as 3.0 SNP/kb in the coding regions to 27.6 SNP/kb in the transposable elements. A more inclusive new approach for analyzing duplication history is introduced here. It reveals an ancient whole-genome duplication, a recent segmental duplication on Chromosomes 11 and 12, and massive ongoing individual gene duplications. We find 18 distinct pairs of duplicated segments that cover 65.7% of the genome; 17 of these pairs date back to a common time before the divergence of the grasses. More important, ongoing individual gene duplications provide a never-ending source of raw material for gene genesis and are major contributors to the differences between members of the grass family.

Conjoint analysis of multiple protein biomarkers can improve the accuracy of disease analysis. Rolling circle amplification (RCA) generates different products by designing circular templates, which can subsequently bind with specific probes to generate various fluorescence signals; thus, it has potential for application in the analysis of various protein biomarkers. Current RCA approaches to detect proteins commonly follow an indirect primer-controlled RCA mode. And the molecular beacon probe combines with RCA products through free collision to generate signals, resulting in lower reaction efficiency. Herein, we propose a direct template-controlled RCA mode using nanosheets as carriers and quenchers for fluorescent probes to simultaneously detect two protein biomarkers. A dual functional magnetic bead was first designed to recognize and capture two proteins while releasing two templates for subsequent RCA. RCA products compete with probes loaded on two-dimensional metal–organic framework nanosheets for hybridization, completing the transition from single-stranded to double-stranded DNA. Double-stranded DNA is far from the nanosheets, and the recovered fluorescence signal can be used to evaluate the concentration of target proteins. This method exhibits excellent analytical performance and can successfully achieve the analysis of Tau and AβO in Alzheimer's disease clinical cerebrospinal fluid samples. [ABSTRACT FROM AUTHOR]
