We present 60 years of Δ14CO2 measurements from Wellington, New Zealand (41° S, 175° E). The record has been extended and fully revised. New measurements have been used to evaluate the existing record and to replace original measurements where warranted. This is the earliest direct atmospheric Δ14CO2 record and records the rise of the 14C bomb spike and the subsequent decline in Δ14CO2 as bomb 14C moved throughout the carbon cycle and increasing fossil fuel CO2 emissions further decreased atmospheric Δ14CO2. The initially large seasonal cycle in the 1960s reduces in amplitude and eventually reverses in phase, resulting in a small seasonal cycle of about 2 ‰ in the 2000s. The seasonal cycle at Wellington is dominated by the seasonality of cross-tropopause transport and differs slightly from that at Cape Grim, Australia, which is influenced by anthropogenic sources in winter. Δ14CO2 at Cape Grim and Wellington show very similar trends, with significant differences only during periods of known measurement uncertainty. In contrast, similar clean-air sites in the Northern Hemisphere show a higher and earlier bomb 14C peak, consistent with a 1.4-year interhemispheric exchange time. From the 1970s until the early 2000s, the Northern and Southern Hemisphere Δ14CO2 were quite similar, apparently due to the balance of 14C-free fossil fuel CO2 emissions in the north and 14C-depleted ocean upwelling in the south. The Southern Hemisphere sites have shown a consistent and marked elevation above the Northern Hemisphere sites since the early 2000s, which is most likely due to reduced upwelling of 14C-depleted and carbon-rich deep waters in the Southern Ocean, although an underestimate of fossil fuel CO2 emissions or changes in biospheric exchange are also possible explanations. This developing Δ14CO2 interhemispheric gradient is consistent with recent studies that indicate a reinvigorated Southern Ocean carbon sink since the mid-2000s and suggests that the upwelling of deep waters plays an important role in this change.

A regional atmospheric inversion method has been developed to determine the spatial and temporal distribution of CO2 sinks and sources across New Zealand for 2011–2013. This approach infers net air–sea and air–land CO2 fluxes from measurement records, using back-trajectory simulations from the Numerical Atmospheric dispersion Modelling Environment (NAME) Lagrangian dispersion model, driven by meteorology from the New Zealand Limited Area Model (NZLAM) weather prediction model. The inversion uses in situ measurements from two fixed sites, Baring Head on the southern tip of New Zealand's North Island (41.408° S, 174.871° E) and Lauder from the central South Island (45.038° S, 169.684° E), and ship board data from monthly cruises between Japan, New Zealand, and Australia. A range of scenarios is used to assess the sensitivity of the inversion method to underlying assumptions and to ensure robustness of the results. The results indicate a strong seasonal cycle in terrestrial land fluxes from the South Island of New Zealand, especially in western regions covered by indigenous forest, suggesting higher photosynthetic and respiratory activity than is evident in the current a priori land process model. On the annual scale, the terrestrial biosphere in New Zealand is estimated to be a net CO2 sink, removing 98 (±37) Tg CO2 yr−1 from the atmosphere on average during 2011–2013. This sink is much larger than the reported 27 Tg CO2 yr−1 from the national inventory for the same time period. The difference can be partially reconciled when factors related to forest and agricultural management and exports, fossil fuel emission estimates, hydrologic fluxes, and soil carbon change are considered, but some differences are likely to remain. Baseline uncertainty, model transport uncertainty, and limited sensitivity to the northern half of the North Island are the main contributors to flux uncertainty.

Transport of animals is a stressful procedure often resulting in significant losses for the slaughter plant. This study aimed to determine whether or not pigs would benefit from a loading density (low density (LD)) (179 kg/m2) below the normal EU standard loading density (normal density (ND)) (235 kg/m2). Eight similar, 550-km-long road journeys, were followed in which fattening pigs were transported across Germany from farm to slaughter plant. During each journey all pigs were transported at LD (n=4) or ND (n=4). Twelve female pigs per journey (total n=96) were randomly selected for measurement and monitoring of body temperature, behaviour, heart rate and blood parameters. Betulinic acid (BA) is a naturally occurring pentacyclic triterpenoid with anti-inflammatory, antiviral and anti-cancer properties. Beneficial cardiovascular effects such as increased nitric oxide (NO) production through enhancement of endothelial NO synthase (eNOS) activity and upregulation of eNOS expression have been demonstrated for this compound. In the present study, immortalized human EA.hy 926 endothelial cells were incubated for up to 1 h with 1-100 µM BA and with the phosphatidylinositol-3-kinase (PI3K) inhibitors LY294002 and wortmannin, or the estrogen receptor (ER) antagonist ICI 182,780. Phosphorylation status of eNOS and total eNOS protein were analyzed by Western blotting using a serine 1177 phosphosite-specific antibody. Bioactive NO production was assessed by determination of cGMP content in rat lung fibroblasts (RFL-6) reporter cells. Short-term incubation of EA.hy 926 cells with BA resulted in eNOS phosphorylation at the serine 1177 residue in a concentration- and time-dependent manner with a half-maximal effective concentration of 0.57 µM. This was associated with an enhanced production of NO. BA-induced eNOS phosphorylation and NO production was completely blocked by pretreatment with ICI 182,780, and was attenuated by pretreatment with the PI3K inhibitors wortmannin and LY294002. These results indicate that fast non-genomic effects of ER with downstream signaling through the PI3K/Akt pathway and consecutive eNOS phosphorylation at serine 1177 are involved in BA-induced eNOS activation.

Average body temperature was lower (P<0.05) for pigs transported at LD (38.0±0.07°C) than those transported at ND (38.3±0.06°C). During loading heart rate increased in both ND and LD pigs and declined after the vehicle had been closed before departure but remained slightly elevated in ND pigs. Pigs transported at ND displayed signs of stress (elevated HR and body temperatures) during the drivers' break. Blood parameters were only slightly (not significant) effected by loading density. Results indicate that pigs are more capable of adapting to long (550 km) transport conditions when loaded at a density below the present EU requirement.

The purported effects of "circulation-improving" herbs used in traditional Chinese medicine (TCM) show striking similarities with the vascular actions of nitric oxide (NO) produced by the endothelial NO synthase (eNOS). We have previously reported that Salviae miltiorrhizae radix and Zizyphi spinosae semen upregulate eNOS expression. In the present study, we studied the effect on eNOS gene expression of 15 Chinese herbs with potential effects on the vasculature, and identified Prunella vulgaris L. (PVL) (flowering spike) as a potent eNOS-upregulating agent. In EA.hy 926 cells, a cell line derived from human umbilical vein endothelial cells (HUVEC), an aqueous extract of PVL increased eNOS promoter activity, eNOS mRNA and protein expressions, as well as NO production in concentration- and time-dependent manners. We have previously shown that ursolic acid (a constituent of Salviae miltiorrhizae radix), betulinic acid (a compound present in Zizyphi spinosae semen), luteolin and cynaroside (ingredients of artichoke, Cynara scolymus L.) are capable of enhancing eNOS gene expression. These compounds are also present in significant quantities in PVL. Thus, PVL contains active principles that stimulate human eNOS gene expression, and such compounds may have therapeutic potential against cardiovascular diseases.

Danshen, the dried root of Salvia miltiorrhiza Bunge (Lamiaceae), is one of the most commonly used traditional Chinese medicines for cardiovascular indications. In EA.hy 926 cells, a cell line derived from human umbilical vein endothelial cells (HUVEC), an aqueous extract of danshen, and also a methanol extract of the plant, increased eNOS promoter activity, eNOS mRNA and protein expression, as well as endothelial NO production. A dichloromethane extract, in contrast, did not change eNOS gene expression. Thus, the active danshen constituent(s) responsible for eNOS upregulation is (are) hydrophilic and/or alcohol-soluble. One such compound is ursolic acid that significantly increased eNOS expression in EA.hy 926 cells and native HUVEC, and enhanced bioactive NO production measured in terms of its cGMP increasing activity. Other tested hydrophilic and alcohol-soluble compounds isolated from danshen had no effect on eNOS expression. Interestingly, ursolic acid also reduced the expression of the NADPH oxidase subunit Nox4 and suppressed the production of reactive oxygen species in human endothelial cells. Upregulation of eNOS and a parallel downregulation of Nox4 lead to an increase in bioactive NO. This in turn could mediate some of the beneficial effects of danshen. Ursolic acid is a prototypical compound responsible for this effect of the plant.

Nitric oxide (NO) produced by endothelial NO synthase (eNOS) is a protective principle in the vasculature. Many cardiovascular diseases are associated with reduced NO bioactivity and eNOS uncoupling due to oxidative stress. Compounds that reverse eNOS uncoupling and increase eNOS expression are of therapeutic interest. Zizyphi Spinosi semen (ZSS) is one of the most widely used traditional Chinese herbs with protective effects on the cardiovascular system. In human umbilical vein endothelial cells (HUVEC) and HUVEC-derived EA.hy 926 cells, an extract of ZSS increased eNOS promoter activity, eNOS mRNA and protein expression, and NO production in a concentration- and time-dependent manner. Major ZSS constituents include saponins, such as jujuboside A and B, and pentacyclic triterpenes, such as betulin and betulinic acid. Jujuboside A, jujuboside B, or betulin had no significant effect on eNOS expression, whereas betulinic acid increased eNOS mRNA and protein expression in HUVEC and EA.hy 926 cells. Interestingly, betulinic acid also attenuated the expression of NADPH oxidase subunits Nox4 and p22phox, thereby reducing oxidative stress and improving eNOS function. Consequently, betulinic acid-treated endothelial cells showed an increased production of bioactive NO (as indicated by a higher efficacy in stimulating cGMP generation in RFL-6 reporter cells). Thus, betulinic acid possesses combined properties of eNOS up-regulation and NADPH oxidase down-regulation. Compounds such as betulinic acid may have a therapeutic potential in cardiovascular disease.
