We propose a new method for the extraction of cellulose nanocrystals (CNCs) from post-consumer cotton textiles through surface functionalization followed by mechanical treatment. Cotton-based textiles were esterified using an 85 wt% solution of citric acid at 100 °C, then further fibrillated in a microfluidizer. The final product, citrated cellulose nanocrystals (CitCNCs), was a dispersion of needle-like nanoparticles with high crystallinity. Up to 78 wt% of the cotton fabric was converted to CitCNCs that exhibited higher yields and a higher surface group content than CNCs extracted through H2SO4 hydrolysis, although CitCNCs showed a broader size distribution and decreased thermal stability. Experimental data supported by DFT calculations showed that the carboxyl groups on the CitCNC surface are bonded to cellulose by mono or diester linkages. An early-stage life cycle assessment (LCA) was performed to evaluate the environmental impact of using discarded textiles as a source of cellulose and analyze the environmental performance of the production of CitCNCs. Our work showed a significant reduction in the environmental burden of CNC extraction using post-consumer cotton instead of wood pulp, making clothing a good feedstock. The environmental impact of CitCNC production was mainly dominated by citric acid. As a proof of concept, around 58 wt% of the citric acid was recovered through evaporation and subsequent crystallization, which could reduce climate impact by 40%. With this work, we introduce a catalyst-free route to valorize textiles with the extraction of CitCNCs and how conducting LCA in laboratory-scale processes might guide future development and optimization. Maritime archaeological investigations of the wreck of the medieval warship Gribshunden (1495), flagship of King Hans of Denmark and Norway, have revealed diverse artifacts including exotic spices imported from far distant origins: saffron, ginger, clove, peppercorns, and almond. The special circumstances of the vessel's last voyage add unique context to the assemblage. Gribshunden and an accompanying squadron conveyed the king, courtiers, noblemen, and soldiers from Copenhagen to a political summit in Kalmar, Sweden. At that conference, Hans expected the Swedish Council to elect him king of Sweden, and thereby fulfill his ambition to reunify the Nordic region under a single crown. To achieve this, Hans assembled in his fleet and particularly aboard his flagship the people and elite cultural signifiers that would convince the Swedish delegation to accept his rule. Along the way, the ships anchored near Ronneby, Blekinge. Written sources record that an explosion and fire caused Gribshunden to sink off Stora Ekön (Great Oak Island). Exotic spices were status markers among the aristocracy in Scandinavia and around the Baltic Sea during the Middle Ages (1050–1550 CE). Until the Gribshunden finds, these extravagances have rarely or never been represented archaeologically. Evidence of their use and consumption in medieval Scandinavia has been limited to sparse written references. We present here the botanical remains from the Gribshunden shipwreck and compare them to previous archaeobotanical finds from the medieval Baltic region. These opulent status symbols traveled with a medieval king en route to a major historical event. The combination of textual and archaeological evidence allows a novel analytical view of the social environment in which these luxurious foods were consumed. [ABSTRACT FROM AUTHOR]

Aims  Fibroblast growth factor (FGF) 21, a key regulator of energy metabolism, is currently evaluated in humans for treatment of type 2 diabetes and non-alcoholic steatohepatitis. However, the effects of FGF21 on cardiovascular benefit, particularly on lipoprotein metabolism in relation to atherogenesis, remain elusive. Methods and results   Here, the role of FGF21 in lipoprotein metabolism in relation to atherosclerosis development was investigated by pharmacological administration of a half-life extended recombinant FGF21 protein to hypercholesterolaemic APOE*3-Leiden.CETP mice, a well-established model mimicking atherosclerosis initiation and development in humans. FGF21 reduced plasma total cholesterol, explained by a reduction in non-HDL-cholesterol. Mechanistically, FGF21 promoted brown adipose tissue (BAT) activation and white adipose tissue (WAT) browning, thereby enhancing the selective uptake of fatty acids from triglyceride-rich lipoproteins into BAT and into browned WAT, consequently accelerating the clearance of the cholesterol-enriched remnants by the liver. In addition, FGF21 reduced body fat, ameliorated glucose tolerance and markedly reduced hepatic steatosis, related to up-regulated hepatic expression of genes involved in fatty acid oxidation and increased hepatic VLDL-triglyceride secretion. Ultimately, FGF21 largely decreased atherosclerotic lesion area, which was mainly explained by the reduction in non-HDL-cholesterol as shown by linear regression analysis, decreased lesion severity, and increased atherosclerotic plaque stability index. Conclusion  FGF21 improves hypercholesterolaemia by accelerating triglyceride-rich lipoprotein turnover as a result of activating BAT and browning of WAT, thereby reducing atherosclerotic lesion severity and increasing atherosclerotic lesion stability index. Abstract Background Oat, Avena sativa is the sixth most important cereal in the world. Presently oat is mostly used as feed for animals. However, oat also has special properties that make it beneficial for human consumption and has seen a growing importance as a food crop in recent decades. Increased demand for novel oat products has also put pressure on oat breeders to produce new oat varieties with specific properties such as increased or improved β-glucan-, antioxidant- and omega-3 fatty acid levels, as well as modified starch and protein content. To facilitate this development we have produced a TILLING (Targeting Induced Local Lesions IN Genomes) population of the spring oat cultivar SW Belinda. Results Here a population of 2600 mutagenised M2 lines, producing 2550 M3 seed lots were obtained. The M2 population was initially evaluated by visual inspection and a number of different phenotypes were seen ranging from dwarfs to giants, early flowering to late flowering, leaf morphology and chlorosis. Phloroglucinol/HCl staining of M3 seeds, obtained from 1824 different M2 lines, revealed a number of potential lignin mutants. These were later confirmed by quantitative analysis. Genomic DNA was prepared from the M2 population and the mutation frequency was determined. The estimated mutation frequency was one mutation per 20 kb by RAPD-PCR fingerprinting, one mutation per 38 kb by MALDI-TOF analysis and one mutation per 22.4 kb by DNA sequencing. Thus, the overall mutation frequency in the population is estimated to be one mutation per 20-40 kb, depending on if the method used addressed the whole genome or specific genes. During the investigation, 6 different mutations in the phenylalanine ammonia-lyase (AsPAL1) gene and 10 different mutations in the cellulose synthase-like (AsCslF6) β-glucan biosynthesis gene were identified. Conclusion The oat TILLING population produced in this work carries, on average, hundreds of mutations in every individual gene in the genome. It will therefore be an important resource in the development of oat with specific characters. Abstract Background Inbreeding and loss of genetic diversity are expected to increase the extinction risk of small populations, but detailed tests in natural populations are scarce. We combine long-term population and fitness data with those from two types of molecular markers to examine the role of genetic effects in a declining metapopulation of southern dunlins Calidris alpina schinzii, an endangered shorebird. Results The decline is associated with increased pairings between related individuals, including close inbreeding (as revealed by both field observations of parentage and molecular markers). Furthermore, reduced genetic diversity seems to affect individual fitness at several life stages. Higher genetic similarity between mates correlates negatively with the pair's hatching success. Moreover, offspring produced by related parents are more homozygous and suffer from increased mortality during embryonic development and possibly also after hatching. Conclusions Our results demonstrate strong genetic effects in a rapidly declining population, emphasizing the importance of genetic factors for the persistence of small populations.

In this paper, we examine the exchange of crops and livestock through the application of strontium (87Sr/86Sr) isotope analysis on cereal grains and faunal tooth enamel from the regional center of Uppåkra and three nearby settlements in Scania, southern Sweden, dating to the first millennium AD. Around a third of the fauna have non-local 87Sr/86Sr values, indicating the import of livestock from several different regions. After cleaning, almost all of the cereal grains have non-local 87Sr/86Sr values, which is surprising given the nearby abundance of fertile agricultural soils. We therefore suggest considering non-locally grown crops to be those whose 87Sr/86Sr values fall outside the normal distribution; if this approach is used, around 20% of the analyzed crop samples are interpreted as having grown non-locally. This study demonstrates the potential of combining strontium isotopic data of archaeobotanical and zooarchaeological material for gaining insights into the movement of agricultural products in prehistory.
