Macrophages are a persistent reservoir for HIV and may contribute to the rebound viremia observed after antiretroviral treatment is stopped. In this study, we further investigate the potential of poly(lactic-co-glycolic) acid (PLGA)-based nanocarriers as a new strategy to enhance penetration of therapeutic molecules into macrophages. We have prepared stable PLGA nanoparticles (NPs) and evaluated their capacity to transport an active molecule into the human monocyte/macrophage cell line THP-1 using bovine serum albumin (BSA) as a proof-of-concept compound. Intracellular localization of fluorescent BSA molecules encapsulated into PLGA NPs was monitored in live cells using confocal microscopy, and cellular uptake was quantified by flow cytometry. In vitro and in vivo toxicological studies were performed to further determine the safety profile of PLGA NPs including inflammatory effects. The size of the PLGA NPs carrying BSA (PLGA-BSA) in culture medium containing 10% serum was ~126 nm in diameter, and they were negatively charged at their surface (zeta potential =-5.6 mV). Our confocal microscopy studies and flow cytometry data showed that these PLGA-BSA NPs are rapidly and efficiently taken up by THP-1 monocyte-derived macrophages (MDMs) at low doses. We found that PLGA-BSA NPs increased cellular uptake and internalization of the protein in vitro. PLGA NPs were not cytotoxic for THP-1 MDM cells, did not modulate neutrophil apoptosis in vitro, and did not show inflammatory effect in vivo in the murine air pouch model of acute inflammation. In contrast to BSA alone, BSA encapsulated into PLGA NPs increased leukocyte infiltration in vivo, suggesting the in vivo enhanced delivery and protection of the protein by the polymer nanocarrier. We demonstrated that PLGA-based nanopolymer carriers are good candidates to efficiently and safely enhance the transport of active molecules into human MDMs. In addition, we further investigated their inflammatory profile and showed that PLGA NPs have low inflammatory effects in vitro and in vivo. Thus, PLGA nanocarriers are promising as a drug delivery strategy in macrophages for prevention and eradication of intracellular pathogens such as HIV and Mycobacterium tuberculosis.Keywords: PLGA nanoparticles, BSA delivery, inflammatory profile, neutrophil apoptosis, murine air pouch, HIV reservoir

Abstract

Background Diagnosing iron deficiency is challenging in the presence of systemic inflammation. Aim To investigate the relationship between plasma C-reactive protein (CRP), serum ferritin (SF) and transferrin saturation (TS), with the objective of establishing a straightforward ratio applicable in the presence of inflammatory syndrome. Design Test prospective cohort and validation retrospective cohort. Methods A prospective cohort of inpatients (n  = 140) assessed the correlation between CRP and SF/TS levels. The diagnostic performance of a determined ratio was evaluated for identifying iron deficiency (ID) using different definitions and in the presence of inflammation and/or chronic heart and/or kidney failure. A large validation cohort (n  = 795) further assessed the predictive power of this ratio. Results In a training cohort (median age 76 years [57–84]), a linear relation was observed between SF (µg/l) and CRP (mg/l), unlike with TS. The SF/CRP ratio accurately predicted ID, with receiver operating characteristic-area under the curve (ROC-AUC) values ranging from 0.85 to 0.92 for different ID definitions. A threshold of ≤6 demonstrated the highest Youden index (0.61). In the validation cohort (age 72 years [57–84]), the SF/CRP ratio exhibited an ROC-AUC of 0.88 [95% CI: 0.85–0.90], with an odds ratio of 37.9 [95% CI: 20.3–68.9] for the threshold of ≤6. Conclusion In this study, we demonstrated that the SF/CRP ratio, with a threshold of ≤6, is a simple and effective biomarker for ID, even in the presence of systemic inflammation or comorbidities. Abstract

Aim: To explore the clinical presentation, course, treatment and impact of early treatment in patients with remethylation disorders from the European Network and Registry for Homocystinurias and Methylation Defects (E-HOD) international web-based registry. Results: This review comprises 238 patients (cobalamin C defect n = 161; methylenetetrahydrofolate reductase deficiency n = 50; cobalamin G defect n = 11; cobalamin E defect n = 10; cobalamin D defect n = 5; and cobalamin J defect n = 1) from 47 centres for whom the E-HOD registry includes, as a minimum, data on medical history and enrolment visit. The duration of observation was 127 patient years. In 181 clinically diagnosed patients, the median age at presentation was 30 days (range 1 day to 42 years) and the median age at diagnosis was 3.7 months (range 3 days to 56 years). Seventy-five percent of pre-clinically diagnosed patients with cobalamin C disease became symptomatic within the first 15 days of life. Total homocysteine (tHcy), amino acids and urinary methylmalonic acid (MMA) were the most frequently assessed disease markers; confirmatory diagnostics were mainly molecular genetic studies. Remethylation disorders are multisystem diseases dominated by neurological and eye disease and failure to thrive. In this cohort, mortality, thromboembolic, psychiatric and renal disease were rarer than reported elsewhere. Early treatment correlates with lower overall morbidity but is less effective in preventing eye disease and cognitive impairment. The wide variation in treatment hampers the evaluation of particular therapeutic modalities. Conclusion: Treatment improves the clinical course of remethylation disorders and reduces morbidity, especially if started early, but neurocognitive and eye symptoms are less responsive. Current treatment is highly variable. This study has the inevitable limitations of a retrospective, registry-based design. © 2018 SSIEM

Abstract

Objectives To study the responsiveness of a combined power Doppler and greyscale ultrasound (PDUS) score for assessing synovitis in biologic-naive patients with rheumatoid arthritis (RA) starting abatacept plus methotrexate (MTX). Methods In this open-label, multicentre, single-arm study, patients with RA (MTX inadequate responders) received intravenous abatacept (∼10 mg/kg) plus MTX for 24 weeks. A composite PDUS synovitis score, developed by the Outcome Measures in Rheumatology–European League Against Rheumatism (OMERACT–EULAR)-Ultrasound Task Force, was used to evaluate individual joints. The maximal score of each joint was added into a Global OMERACT–EULAR Synovitis Score (GLOESS) for bilateral metacarpophalangeal joints (MCPs) 2–5 (primary objective). The value of GLOESS containing other joint sets was explored, along with clinical efficacy. Results Eighty-nine patients completed the 24-week treatment period. The earliest PDUS sign of improvement in synovitis was at week 1 (mean change in GLOESS (MCPs 2–5): −0.7 (95% CIs −1.2 to −0.1)), with continuous improvement to week 24. Early improvement was observed in the component scores (power Doppler signal at week 1, synovial hyperplasia at week 2, joint effusion at week 4). Comparable changes were observed for 22 paired joints and minimal joint subsets. Mean Disease Activity Score 28 (C reactive protein) was significantly reduced from weeks 1 to 24, reaching clinical meaningful improvement (change ≥1.2) at week 8. Conclusions In this first international prospective study, the composite PDUS score is responsive to abatacept. GLOESS demonstrated the rapid onset of action of abatacept, regardless of the number of joints examined. Ultrasound is an objective tool to monitor patients with RA under treatment. Abstract

Objective: To compare the clinical presentation and outcome of giant cell arteritis (GCA)-related aortitis according to the results of temporal artery biopsy (TAB).Method: Patients with GCA-related aortitis diagnosed between 2000 and 2017, who underwent TAB, were retrospectively included from a French multicentre database. They all met at least three American College of Rheumatology criteria for the diagnosis of GCA. Aortitis was defined by aortic wall thickening > 2 mm on computed tomography scan and/or an aortic aneurysm, associated with an inflammatory syndrome. Patients were divided into two groups [positive and negative TAB (TAB+, TAB-)], which were compared regarding aortic imaging characteristics and aortic events, at aortitis diagnosis and during follow-up.Results: We included 56 patients with TAB+ (70%) and 24 with TAB- (30%). At aortitis diagnosis, patients with TAB- were significantly younger than those with TAB+ (67.7 ± 9 vs 72.3 ± 7 years, p = 0.022). Initial clinical signs of GCA, inflammatory parameters, and glucocorticoid therapy were similar in both groups. Coronary artery disease and/or lower limb peripheral arterial disease was more frequent in TAB- patients (25% vs 5.3%, p = 0.018). Aortic wall thickness and type of aortic involvement were not significantly different between groups. Diffuse arterial involvement from the aortic arch was more frequent in TAB- patients (29.1 vs 8.9%, p = 0.03). There were no differences between the groups regarding overall, aneurism-free, relapse-free, and aortic event-free survival.Conclusion: Among patients with GCA-related aortitis, those with TAB- are characterized by younger age and increased frequency of diffuse arterial involvement from the aortic arch compared to those with TAB+, without significant differences in terms of prognosis. Abstract

Poster Designing agricultural systems targeting an increase of natural regulations and a decrease of pest populations should be based on the optimization of various ecological services and biodiversity by modifying deeply the composition, structure and organization of agroecosystems. An agroforestry system combining fruit trees, vegetables and wild trees and shrubs is one possible option. Key challenges are to find out (i) a compromise between economic/technical constraints and ecological processes stimulating natural regulations and (ii) to test spatial combinations of vegetation. We explored the basic ecological principles necessary for the functioning of such cropping systems and elaborated an approach enabling the design and monitoring of complex horticultural agroforestry systems, at the level of a production unit. We present a co-design research project for a 4 ha pilot farm unit in the periurban area of Avignon, France (La Durette). We elaborated over two years different possible spatial arrangements and prototypes through participative discussions involving scientists, advisors and farmers. We progressively evaluated different combinations and ratios of fruit and vegetable production through calculation of expected yield, operational costs and income over ten years and through an expert evaluation of the labour time requirements. We also studied different possible spatial arrangements by modelling the impact of canopies on the available light for annual crops . This led to proposals for a sustainable and viable crop production system managed by 2 farmers. The different calculation tools could be further used to elaborate whole farm planning guides for horticultural agroforestry farms under temperate climate.

Abstract

Remote sensing, surveillance applications or telecommunications require optronic sensors, onboard satellite or airborne platform. Performances limitation of these sensors may be caused by cloud presence in the field of view. To take into account cloud presence, a Monte Carlo method is used. Geometrical and optical clouds properties necessary to build the model are obtained from CALIOP measurements that enable one to deal with the optically thinnest clouds. Different viewing geometries are presented. They correspond to surveillance missions by an airborne sensor with close-to-the- horizon viewing and to optical link between an aircraft and a satellite. Results obtained are compared to a previous study and improvements reached with our new method are discussed.

Abstract

International audience; For a few years, Onera has developed static Fourier transform spectrometers. These spectrometers are napshot and benefit from a high étendue, thus making them well suited for measuring fast moving scenes, such as rocket plumes. In particular, Onera has developed two cryogenic infrared instruments, Mistere and Microspoc, which can measure spectra at a high repetition rate (few hundred of Hertz), on a wide spectral range ([2.9μm ; 9.6μm] and [1.5μm ; 5μm]), and with a moderate spectral resolution (15cm-1 and 30cm-1). The design of these spectrometers will be presented, as well as examples of experimental results.

Abstract

International audience; BACKGROUND: Apixaban, an oral factor Xa inhibitor administered in fixed doses, may simplify the treatment of venous thromboembolism. METHODS: In this randomized, double-blind study, we compared apixaban (at a dose of 10 mg twice daily for 7 days, followed by 5 mg twice daily for 6 months) with conventional therapy (subcutaneous enoxaparin, followed by warfarin) in 5395 patients with acute venous thromboembolism. The primary efficacy outcome was recurrent symptomatic venous thromboembolism or death related to venous thromboembolism. The principal safety outcomes were major bleeding alone and major bleeding plus clinically relevant nonmajor bleeding. RESULTS: The primary efficacy outcome occurred in 59 of 2609 patients (2.3%) in the apixaban group, as compared with 71 of 2635 (2.7%) in the conventional-therapy group (relative risk, 0.84; 95% confidence interval [CI], 0.60 to 1.18; difference in risk [apixaban minus conventional therapy], -0.4 percentage points; 95% CI, -1.3 to 0.4). Abstract

In Martinique (French West Indies), because of the susceptibility of the insular environment, the demand from society and policy-makers is high for sustainable and environmentally friendly fruit production systems. In tropical wet areas, weed management is crucial but difficult to control without herbicides. Herbicide becomes the most important pesticide input in banana plantations and orchards, thus developing herbicide-free alternatives are a priority. Cover crop intercropping constitutes the best option. Besides weed control, we considered their ability to reduce runoff and soil erosion, to compete with the main crop for water and nutrients, to host pests and natural enemies, and to preserve biodiversity. Thus, cover crops turn into global services crops. A multi-step and multi-criteria grid was built to select the ideal cover crop. We tested it on banana fields and citrus orchards. First, cover crop pre-selection was common and checked for climate suitability, seed availability, height, status of noninvasive species and capacity to be perennial. Second, we assessed the agronomic behaviour of seven pre-selected cover crops under field conditions. We measured cover crop specific traits (germination rate, covering rate, perenniality, biomass production, root depth, nutrient uptake or recycling, regrowth after mowing, adaptation to shade). Last, according to each cropping system specifications, we designed prototypes with the most relevant cover crops. In citrus orchards, grasses characterized by a high covering index associated with a low biomass production were selected, but natural enemies hosting services were poor. In banana fields, grasses were intercropped too, but nitrogen competi¬tion, was too high and led to loss in banana yield. As a conclusion, we showed that the concept of an ideal cover plant has to be implemented and combined in a more efficient mixed cover system (grasses mixed with nitrogen fixing plants).

Abstract

The incorporation of landscape management into Conservation Biological Control (CBC) strategies is a priority area of research but is hindered by a lack of harmonisation of the means to describe and measure the effectiveness of CBC, the organisms under focus and the landscape. This paper provides a set of recommendations that represents the consensus amongst experts of the ENDURE network. The most important data values that were identified were: pest population level: natural enemy population or % parasitism/predation; crop damage; estimate of mobility of study organisms (dispersal function) and non-explicit spatial measurements such as the proportion of the landscape offering resources and the connectivity between resource patches. For all these measurements. careful consideration should be given to the appropriate spatial and temporal scale of assessment. For analysis, we advocate an iterative use of modeling tools, particularly individual-based models, and statistical approaches: the former to understand mechanisms underlying the population dynamics of pest and their natural enemies in landscapes and the latter to characterize the observed patterns of these populations in a given landscape.

Organic agriculture in Citrus cultivation in Martinique seems to be very difficult regarding high pests and diseases pressure in the tropical area. A comparison between standard and organic cultivation of Tahiti Lime shows that the main constraints are located in the weed control and also in the high price of nutritional input approved for organic farming. The yield cumulated during the four first years is similar for both crop managements.

Abstract

A wide variety of optronic sensors, on board a satellite or airborne platform, are used for remote sensing, surveillance applications, and telecommunications. Cloud presence in the field of view is one of the key factors limiting the performances of these sensors. Consequently, cloud presence must be taken into account in order to evaluate their performances. To that end, a Monte Carlo method is used. Geometrical and optical cloud properties necessary to build the model are obtained from Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP) measurements that enable one to deal with the optically thinnest clouds. Different viewing geometries are presented, corresponding to surveillance missions by an airborne sensor with close-to-the-horizon viewing and to an optical link between an aircraft and a satellite. Abstract

Aphids are major pests in apple orchards, debilitating the crop and spreading disease. We investigated whether early-season predation by canopy spiders may be effectively controlling aphid numbers in three organic orchards. For this purpose, we monitored the aphid population dynamics from the winter eggs to colony stages and compared this to spider abundances and rates of predation on aphids detected by diagnostic polymerase chain reaction. For the latter, we applied existing general aphid primers. We found that spiders ate colony fundatrices and that aphid numbers were negatively related to spider abundance. Spiders were the main active predators within the orchards when the first colony fundatrices were present, indicating their importance in the early control of aphid populations.
