IntroductionConductive hearing loss (CHL) attenuates the ability to transmit air conducted sounds to the ear. In humans, severe hearing loss is often accompanied by alterations to other neural systems, such as the vestibular system; however, the inter-relations are not well understood. The overall goal of this study was to assess vestibular-related functioning proxies in a rat CHL model.MethodsMale Sprague–Dawley rats (N=134, 250g, 2months old) were used in a CHL model which produced a >20dB threshold shift induced by tympanic membrane puncture. Auditory brainstem response (ABRs) recordings were used to determine threshold depth at different times before and after CHL. ABR threshold depths were assessed both manually and by an automated ABR machine learning algorithm. Vestibular-related functioning proxy assessment was performed using the rotarod, balance beam, elevator vertical motion (EVM) and Ferris-wheel rotation (FWR) assays.ResultsThe Pre-CHL (control) threshold depth was 27.92dB±11.58dB compared to the Post-CHL threshold depth of 50.69dB±13.98dB (mean±SD) across the frequencies tested. The automated ABR machine learning algorithm determined the following threshold depths: Pre-CHL=24.3dB, Post-CHL same day=56dB, Post-CHL 7 days=41.16dB, and Post-CHL 1 month=32.5dB across the frequencies assessed (1, 2, 4, 8, 16, and 32kHz). Rotarod assessment of motor function was not significantly different between pre and post-CHL (~1week) rats for time duration (sec) or speed (RPM), albeit the former had a small effect size difference. Balance beam time to transverse was significantly longer for post-CHL rats, likely indicating a change in motor coordination. Further, failure to cross was only noted for CHL rats. The defection count was significantly reduced for CHL rats compared to control rats following FWR, but not EVM. The total distance traveled during open-field examination after EVM was significantly different between control and CHL rats, but not for FWR. The EVM is associated with linear acceleration (acting in the vertical plane: up-down) stimulating the saccule, while the FWR is associated with angular acceleration (centrifugal rotation about a circular axis) stimulating both otolith organs and semicircular canals; therefore, the difference in results could reflect the specific vestibular-organ functional role.DiscussionLess movement (EVM) and increase time to transverse (balance beam) may be associated with anxiety and alterations to defecation patterns (FWR) may result from autonomic disturbances due to the impact of hearing loss. In this regard, vestibulomotor deficits resulting in changes in balance and motion could be attributed to comodulation of auditory and vestibular functioning. Future studies should manipulate vestibular functioning directly in rats with CHL.

(1) Background: The forest of Pinus sylvestris var. mongolica is an important semi-arid ecosystem in Hulunbuir sandy land that plays a key role in the carbon cycle and wind erosion control. It is crucial to explore the main factors affecting the radial growth of trees of P. sylvestris var. mongolica. (2) Methods: The study established the tree-ring chronology of P. sylvestris var. mongolica and analyzed the relationships among the radial growth, competition index, and climate variables using correlation analysis and a linear mixed effect model to explore the influence of competition and climate on radial growth of P. sylvestris var. mongolica. (3) Results: The results indicated that tree growth is mainly affected by the maximum average temperature (Tmax) and precipitation in June and July of the current year and that tree growth significantly decreased with increasing competition pressure. Analysis of the linear mixed effect model showed that tree age, competition intensity, self-calibrating Palmer drought severity index (scPDSI) from May to July, and vapor pressure deficit (VPD) have a significant impact on radial growth. (4) Conclusions: The competition plays a dominant role in radial growth of P. sylvestris var. mongolica compared to climate factors. This study helps to understand the growth mechanism of P. sylvestris var. mongolica forests under climate change and provides a scientific basis for effective management of semi-arid forests.

Motion sickness (MS) is caused by mismatch between conflicted motion perception produced by motion challenges and expected “internal model” of integrated motion sensory pattern formed under normal condition in the brain. Stroboscopic light could reduce MS nausea symptom via increasing fixation ability for gaze stabilization to reduce visuo-vestibular confliction triggered by distorted vision during locomotion. This study tried to clarify whether MS induced by passive motion could be alleviated by stroboscopic light with emitting rate and intensity synchronized to acceleration–deceleration phase of motion. We observed synchronized and unsynchronized stroboscopic light (SSL: 6 cycle/min; uSSL: 2, 4, and 8 cycle/min) on MS-related gastrointestinal symptoms (conditioned gaping and defecation responses), motor disorders (hypoactivity and balance disturbance), and central Fos protein expression in rats receiving Ferris wheel-like rotation (6 cycle/min). The effects of color temperature and peak light intensity were also examined. We found that SSL (6 cycle/min) significantly reduced rotation-induced conditioned gaping and defecation responses and alleviated rotation-induced decline in spontaneous locomotion activity and disruption in balance beam performance. The efficacy of SSL against MS behavioral responses was affected by peak light intensity but not color temperature. The uSSL (4 and 8 cycle/min) only released defecation but less efficiently than SSL, while uSSL (2 cycle/min) showed no beneficial effect in MS animals. SSL but not uSSL inhibited Fos protein expression in the caudal vestibular nucleus, the nucleus of solitary tract, the parabrachial nucleus, the central nucleus of amygdala, and the paraventricular nucleus of hypothalamus, while uSSL (4 and 8 cycle/min) only decreased Fos expression in the paraventricular nucleus of hypothalamus. These results suggested that stroboscopic light synchronized to motion pattern might alleviate MS gastrointestinal symptoms and motor disorders and inhibit vestibular-autonomic pathways. Our study supports the utilization of motion-synchronous stroboscopic light as a potential countermeasure against MS under abnormal motion condition in future.

Background: Late embryogenesis abundant (LEA) proteins were reported to be related to adversity stress and drought tolerance. Lea-3 from Arachis hypogaea L. (AhLea-3) was previously found to be related to salt tolerance according to the result of transcriptome profiling and digital gene expression analysis. So, AhLea-3 was cloned and the salt tolerance was validated by transgenic peanut plants. Results: AhLea-3 was isolated from M34, a salt-resistant mutant of peanut, with its cDNA as the template. AhLea-3 contains one intron and two extrons, and the full-length cDNA sequence contains 303 bp. AhLea-3 was ligated to pCAMBIA1301 to obtain the overexpression vector pCAMBIA1301-AhLea-3, which was then transferred into peanut variety Huayu23. The expression level of AhLea-3, as determined by qRT-PCR analysis, was >10 times higher in transgenic than in non-transgenic plants. Five days after they were irrigated with 250 mM NaCl, the transgenic plants showed less severe leaf wilting, higher activities of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase), and lower malonic dialdehyde content than non-transgenic plants. Relative to non-transgenic plants, the transgenic plants had a higher photosynthetic net rate, stomatal conductance, and transpiration rate, and a lower intercellular CO2 concentration after salt stress treatment (250 mM NaCl). Conclusions: These results indicate that overexpression of AhLea-3 increased the salt tolerance of transgenic peanut plants. Vestibular damage can induce locomotor abnormalities in both animals and humans. Rodents with bilateral vestibular loss showed vestibular deficits syndrome such as circling, opisthotonus as well as locomotor and exploratory hyperactivity. Previous studies have investigated the changes in the dopamine system after vestibular loss, but the results are inconsistent and inconclusive. Numerous evidences indicate that the orexin system is implicated in central motor control. We hypothesized that orexin may be potentially involved in vestibular loss-induced motor disorders. In this study, we examined the effects of arsanilate- or 3, 3′-iminodipropionitrile (IDPN)-induced vestibular lesion (AVL or IVL) on the orexin-A (OXA) labeling in rat hypothalamus using immunohistochemistry. The vestibular lesion-induced locomotor abnormalities were recorded and verified using a histamine H4 receptor antagonist JNJ7777120 (20 mg/kg, i.p.). The effects of the orexin receptor type 1 antagonist SB334867 (16 μg, i.c.v.) on these behavior responses were also investigated. At 72 h post-AVL and IVL, animals exhibited vestibular deficit syndrome and locomotor hyperactivity in the home cages. These responses were significantly alleviated by JNJ7777120 which also eliminated AVL-induced increases in exploratory behavior in an open field. The numbers of OXA-labeled neurons in the hypothalamus were significantly increased in the AVL animals at 72 h post-AVL and in the IVL animals at 24, 48 and 72 h post-IVL. SB334867 significantly attenuated the vestibular deficit syndrome and locomotor hyperactivity at 72 h post-AVL and IVL. It also decreased exploratory behavior in the AVL animals. These results suggested that the alteration of OXA expression might contribute to locomotor abnormalities after acute vestibular lesion. The orexin receptors might be the potential therapeutic targets for vestibular disorders.

Stand structure indices were examined in this study to clarify stand structure and interaction among individual trees. This analysis provides a greater understanding of density-dependent interactions among intraspecific trees with different vitality using a point pattern analysis method. This study was undertaken in Hulunbuir Sandland, Inner Mongolia, China, in natural Pinus sylvestris L. var. mongolica Litv. stands which have different densities and relatively little human interference Results for Regeneration seedlings CR-seedlings) (31.07% ±15.5), Suppressed trees (S-trees) (55.0% ±13.3) and Dominant trees (D-trees) (13. 3% ±2.6) indicated that this forest was under a period of renewal and growth. Apart for R-seedlings, growth dynamics differed depending on the density gradient in all plots, and obvious significant differences were recorded between S-trees growth and density gradient. The spatial distribution of S-trees was also found to have a significant impact on germination and growth of R-seedlings in these forests. However, D-trees did not directly affect germination and survival ofR-seedlings. Many studies have demonstrated sex and age differences in motion sickness, but the underlying physiological basis is still in controversy. In the present study, we tried to investigate the potential correlates of endocrine and/or neuronal activity with sex and age differences in rats with motion sickness. LiCl-induced nausea symptom was evaluated by conditioned gaping. Motion sickness was assessed by measurement of autonomic responses (i.e., conditioned gaping and defecation responses), motor impairments (i.e., hypoactivity and balance disturbance) after Ferris wheel-like rotation, and blood hormone levels and central Fos protein expression was also observed. We found that rotation-induced conditioned gaping, defecation responses and motor disorders were significantly attenuated in middle-aged animals (13- and 14-monthage) compared with adolescents (1- and 2-month-age) and young-adults (4- and/or 5-month-age). LiCl-induced conditioned gapings were also decreased with age, but was less pronounced than rotation-induced ones. Females showed greater responses in defecation and spontaneous locomotor activity during adolescents and/or young-adult period. Blood adrenocorticotropic hormone and corticosterone significantly increased in 4-month-old males after rotation compared with static controls. No significant effect of rotation was observed in norepinephrine, epinephrine, β-endorphin and argininevasopressin levels. The middle-aged animals (13-month-age) also had higher number of rotation-induced Fos-labeled neurons in the spinal vestibular nucleus, the parabrachial nucleus (PBN), the central and medial nucleus of amygdala (CeA and MeA) compared with adolescents (1-month-age) and young-adults (4-month-age) and in the nucleus of solitary tract (NTS) compared with adolescents (1-month-age). Sex difference in rotation-induced Fos-labeling was observed in the PBN, the NTS, the locus ceruleus and the paraventricular hypothalamus nucleus at 4 and/or 13 months of age. These results suggested that the sex and age differences in motion sickness may not correlate with stress hormone responses and habituation. The age-dependent decline in motion sickness susceptibility might be mainly attributed to the neuronal activity changes in vestibulo-autonomic pathways contributing to homeostasis regulation during motion sickness. According to the extended Huygens-Fresnel integral, the expressions for the on-axis spectrum of partially coherent Gaussian Schell-model (GSM) rectangular array beams passing through ABCD optical systems have been derived. The generalized Fresnel number of the system, the spatial coherent parameter of array beamlets and the array beam parameters including the number of beamlets and the separation distance between beamlets have been taken as the characteristic parameters to compare the spectral shifts of GSM array beams for the two types of the superposition, i.e. the correlated superposition and the uncorrelated superposition. In particular, the effect of characteristic parameters on the on-axis relative spectral shifts has been discussed in detail. The results show that the spectral intensity of GSM array beams for the two types of the superposition passing through ABCD systems depends on the source spectral density S0(ω), the spatial coherent parameter of array beamlets β, the generalized Fresnel number of the system F and the array beam parameters. Furthermore, for the uncorrelated superposition, the spectrum of GSM array beams only exhibit the blue-shift, whereas for the correlated superposition, GSM array beams exhibit the spectral switch and the number of spectral switches increases with the increase of array beam parameters in the near field due to the interference between beamlets. This paper introduces a new portable intelligent electronic nose system developed especially for measuring and analysing livestock and poultry farm odours. It can be used in both laboratory and field. The sensor array of the proposed electronic nose consists of 14 gas sensors, a humidity sensor, and a temperature sensor. The gas sensors were especially selected for the main compounds from the livestock farm odours. An expert system called “Odour Expert” was developed to support researchers’ and farmers’ decision making on odour control strategies for livestock and poultry operations. “Odour Expert” utilises several advanced artificial intelligence technologies tailored to livestock and poultry farm odours. It can provide more advanced odour analysis than existing commercially available products. In addition, a rank of odour generation factors is provided, which refines the focus of odour control research. Field experiments were conducted downwind from the barns on 14 livestock and poultry farms. Experimental results show that the predicted odour strengths by the electronic nose yield higher consistency in comparison to the perceived odour intensity by human panel.
