Air-source heat pumps (ASHPs), functioning as thermally controlled loads, possess significant adjustable capabilities and controllability when aggregated, establishing them as premium resources for demand-response engagement. This paper proposes a control strategy for the aggregation of ASHP loads to participate in demand response across multiple scenarios, framed within a three-tier architecture: electric power system, Load Aggregator (LA), and thermal load. Load Aggregators, considering the user-comfort temperature ranges and the thermal storage characteristics of buildings, aim to minimize heating costs through time-of-use electricity pricing, while assessing the adjustability of the load. Upon receiving control directives from the power system's dispatch department, the strategy allocates load adjustments by considering user comfort and system regulatory needs, thereby addressing issues like aggregated power oscillations and significant rebound loads. The effectiveness of the proposed strategy is corroborated through simulation, demonstrating its potential to enhance demand-response participation and ameliorate associated power stability challenges. The study aims to analyze genetic mutation and clinical characteristics and study their correlation with survival prognosis of patients with myelodysplastic syndromes (MDS). Moreover, the differential DNA methylation profiles between TET2 mutated (Mut)/ASXL1 wild-type (WT) and TET2-Mut/ASXL1-Mut MDS samples were investigated to explore the mechanism of MDS patients with TET2/ASXL1 mutations. The clinical data of 195 patients diagnosed with MDS were selected and statistically analyzed. The DNA methylation sequencing data set was obtained from the GEO and bioinformatics analyzed. Of the 195 MDS patients, 42 (21.5%) carried TET2 mutations. 81% of TET2-Mut patients could detect comutated genes. The most commonly comutated gene in MDS patients with TET2-Mut was ASXL1, which had a tendency towards poorer prognosis (P = 0.08). GO analysis showed that highly methylated differentially methylated genes (DMGs) was mainly enriched in biological processes such as cell surface receptor signal pathway and cell secretion. Hypomethylated DMGs was mainly enriched in cell differentiation and cell development. KEGG analysis showed that hypermethylated DMGs was mainly enriched in Ras signal pathway and MAPK signal pathway. Hypomethylated DMGs was mainly enriched in extracellular matrix receptor interaction and focal adhesion. PPI network analysis identified 10 hub genes of hypermethylated and hypomethylated DMGs that may be associated with patients with TET2-Mut/ASXL1-Mut respectively. Our results illustrate the interrelationships between genetic mutations and clinical phenotypes and disease outcomes, with substantial potential for clinical application. Real-time fault detection in power distribution networks has become a popular issue in current power systems. However, the low power and computational capabilities of edge devices often fail to meet the requirements of real-time detection. To overcome these challenges, this paper proposes a lightweight algorithm, named Comprehensive-YOLOv5, for identifying defects in distribution networks. The proposed method focuses on achieving rapid localization and accurate identification of three common defects: insulator without loop, cable detachment from the insulator, and cable detachment from the spacer. Based on the You Only Look Once version 5 (YOLOv5) algorithm, this paper adopts GhostNet to reconstruct the original backbone of YOLOv5; introduces Bidirectional Feature Pyramid Network (BiFPN) structure to replace Path Aggregation Network (PANet) for feature fusion, which enhances the feature fusion ability; and replaces Generalized Intersection over Union GIOU with Focal Extended Intersection over Union (Focal-EIOU) to optimize the loss function, which improves the mean average precision and speed of the algorithm. The effectiveness of the improved Comprehensive-YOLOv5 algorithm is verified through a "morphological experiment", while an "algorithm comparison experiment" confirms its superiority over other algorithms. Compared with the original YOLOv5, the Comprehensive-YOLOv5 algorithm improves mean average precision (mAP) from 88.3% to 90.1% and increases Frames per second (FPS) from 20 to 52 frames. This improvement significantly reduces false positives and false negatives in defect detection. Consequently, the proposed algorithm enhances detection speed and improves inspection efficiency, providing a viable solution for real-time detection and deployment at the edge of power distribution networks. Background: Ovarian cancer (OC) is the most lethal gynecological malignancy worldwide. Increasing evidence indicates that TBC domain family is implicated in various cellular events contributing to initiation and development of different cancers, including OC. However, the role of TBC1D2, a crucial member of TBC domain family, remains unclear in OC. Methods: IHC and qRT-PCR were employed to determine TBC1D2 expression in OC tissues and cells. In vitro and in vivo assays involving proliferation, migration, invasion were performed to explore the role of TBC1D2 in OC development. The underlying mechanism by which TBC1D2 promotes OC metastasis were elucidated using bioinformatics analysis, western blotting and co-immunoprecipitation. Results: Upregulation of TBC1D2 was found in OC and was associated with a poor prognosis. Meanwhile, TBC1D2 promoted OC cell proliferation, migration, and invasion in vitro and facilitated tumor growth and metastasis in vivo. Moreover, TBC1D2 contributed to OC cell invasion by E-cadherin degradation via disassembling Rac1-IQGAP1 complex. In addition, miR-373-3p was screened out and identified to inhibit OVCAR3 invasion via negative regulation of TBC1D2. Conclusion: Our findings indicated that TBC1D2 is overexpressed in OC and contributes to tumor metastasis via E-cadherin degradation. Aiming at the power sharing among converter stations and the frequency stability of weak AC system connected by converter stations in multi‐terminal voltage source converter high‐voltage direct current (VSC‐HVDC) system, a coordinated control strategy without communication is proposed, which can realise the bidirectional active power support between DC system and weak AC system. The strategy divides the voltage of DC system and the frequency of weak AC system into open‐time operation area and time‐limited operation area, respectively. When DC voltage and AC frequency are in the open‐time operation area, respectively, unbalanced power in the system can be flexibly allocated according to the power margins of converter stations, which reduces DC voltage fluctuation and the influence of DC voltage errors (line length and other factors) among the converter stations on power regulation. When the AC frequency is in the time‐limited operation area, the DC system can provide power support for the weak AC system to maintain frequency stability. When the DC voltage is in the time‐limited operation area, the weak AC system loses part of the frequency quality to provide short‐term power support for the DC system to maintain the stability of VSC‐HVDC system as far as possible. Cidea and Cidec are two members of Cell death‐inducing DNA fragmentation factor‐alpha‐like effector family proteins, which could be involved in lipid or fat metabolism. To better understand the roles of Cidea and Cidec in fatty liver hemorrhagic syndrome (FLHS), 150 healthy 155‐day‐old Hyline Brown laying hens were randomly divided into control group (fed with basic diet) and experimental group (fed with high‐energy low‐protein [HELP] diet). Analysis of the liver by tissue sectioning and hematoxylin and eosin staining showed that the HELP diet induced micro‐vesicular steatosis in laying hens. Subsequently, based on the liver color scores and the range of lipid accumulation observed in histological examination, we classified livers with <50% vacuolization as mild FLHS and >50% as severe FLHS. The results showed that the levels of Cidea and Cidec mRNA expression were markedly elevated in the liver and adipose tissues with FLHS and the levels of Cidea and Cidec mRNA expression in the liver with severe FLHS were significantly higher than that in the liver with mild FLHS. The extended black-start strategy is new exploration for black-start restoration of power systems, in which more than one non-black-start unit can be restored simultaneously during the black-start stage. This paper presents a multiobjective optimization method coordinating unit restarting with load restoration for extended black-start decision making. The objective functions of the black-start restoration optimization are defined as restoring non-black-start units and unserved loads as much as possible under steady-state and transient system operation constraints. A hybrid algorithm combining the lexicographic method with the improved bacterial foraging algorithm is used to solve the defined optimization model. The proposed method has been tested on the IEEE 118-bus system and the southern Hebei power system of China, and the test results verify the effectiveness of the proposed decision-making method. In order to improve the performance of a doubly fed induction generator (DFIG) system, we put forward a high performance nonlinear passivity-based control (PBC) method on DFIG. Firstly, we build a PBC mathematical model for DFIG. We design the passive controller for the inner loop in the control system based on passivity theory. Then we calculate the rotor’s control voltages which are modulated afterwards to pulse to control the rotor side converter. The maximal wind energy capture is effectively realized. The rotor speed and DFIG currents fast track their expected values. The independent regulation of the stator active power and reactive power is achieved. Finally we perform simulations to verify the effectiveness of the proposed method. Furthermore, we employ the Wigner-Ville distribution (WVD) and continuous wavelet transform (CWT) as two time-frequency representation methods to indicate that the proposed method in the paper performs well from the perspective of energy distribution in time and frequency domain. To solve the problems of DC voltage control and power allocation in the hybrid multi-terminal high voltage direct current system effectively, a DC voltage adaptive droop control strategy based on DC voltage-current characteristics is proposed. Based on adjustment of the droop coefficient of the converter station, the proposed control strategy introduces the influence factor of the droop coefficient, which considers the dynamic power margin of the converter station according to the direction of DC current variation in the converter station. When changes in the hybrid multi-terminal high voltage direct current system power flow occur, the droop coefficient of the converter station can be adjusted by the influence factor of the droop coefficient, so that the converter station can participate in power regulation according to its own power regulating ability. Consequently, the proposed control strategy can reasonably allocate the active power and minimize the deviation of the DC voltage. Besides, the stability analysis of the proposed control strategy is also carried out. Simulation results have verified the feasibility and effectiveness of the proposed control strategy. The Energy Internet is an inevitable trend of the development of electric power system in the future. With the development of microgrids and distributed generation (DG), the structure and operation mode of power systems are gradually changing. Energy routers are considered as key technology equipment for the development of the Energy Internet. This paper mainly studies the control of the LAN-level energy router, and discusses the structure and components of the energy router. For better control of the power transmission of an energy router, the energy routing control strategy for an integrated microgrid, including photovoltaic (PV) energy, battery-energy storage and electric vehicles (EVs) is studied. The front stage DC/DC converter of the PV system uses maximum power point tracking (MPPT) control. The constant current control is used by the bidirectional DC/DC converter of the battery-energy storage system and the EV system when they discharge. The DC/AC inverters adopt constant reactive power and constant DC voltage control. Constant current constant voltage control is adopted when an EV is charged. The control strategy model is simulated by Simulink, and the simulation results verify the feasibility and effectiveness of the proposed control strategy. Density of renewable energy sources in distribution systems has developed a novel structure, called microgrid (MG), which consists of small-scale power grids with controllable and uncontrollable loads. MGs are a combination of different distributed generation resources (DGRs) which act as a controllable system at the distribution voltage level and supply power or heat to a group of local loads. Due to the high fluctuations of available power at the distribution voltage level, MGs may fail to supply major consumers. Therefore, several MGs are used by dividing consumers into smaller units; each unit is supplied by one MG. In this study, renewable sources such as wind turbine (WT), photovoltaic (PV) cell, and hydrogen storage system are considered as MGs. A novel energy management strategy is proposed using hydrogen storage system and considering uncertainties of renewable sources in the MG. This strategy aims to minimize the operating costs of batteries and hydrogen storage systems as well as unsupplied and surplus energy costs considering load supply constraints. Various resource constraints are employed in the proposed strategy. Moreover, demand response (DR) program is applied for MG optimal operation. The proposed model is implemented on a system using MATLAB software and meta-heuristic gray wolf optimization (GWO) algorithm. Objective: Animals will experience energy deprivation processes such as moulting, clutching, migration and long-distance transportation under natural survival conditions and in production practices, and the body will trigger a series of adaptive metabolic changes during these processes. Fasting and refeeding after fasting can induce remodeling of nutrients and energy metabolism. This study aims to investigate the mechanisms by which the gut microbiota and liver of poultry respond to energy deprivation under specific conditions., Methods: Ninety 252-day-old laying hens were randomly divided into 3 groups: (1) fed ad libitum (control group); (2) fasted from day 13 to day 17 (fasting group); (3) fasted from day 1 to day 5, then refed on a specific feeding way (refeeding group). After that, the serum, liver, jejunum tissues, and cecum contents were sampled and sent for metabolome, transcriptome, morphology, and 16S rDNA sequencing analyses, respectively., Results: Results showed that food deprivation not only observably decreased the body weight, liver index, and the villus height and villus/crypt ratio of jejunum, but also significantly changed the gut microbiota compositions, serum metabolic profiles, and the hepatic gene expression patterns of laying hens, whereas these changes were effectively reversed by the following refeeding operation. At the same time, metabolome combined transcriptome analysis revealed that both serum differential metabolites and hepatic differential expressed genes (DEGs) were consistently enriched in the lipid and amino metabolism pathways, and strong correlations were synchronously found between the differential metabolites and both of the differential gut microbial genera and DEGs, suggesting the crosstalks among gut, liver and their resulting serum metabolic products., Conclusion: The results suggested that the organism might coordinate to maintain metabolic homeostasis under energy deprivation through a combination of changes in gut microbial composition and hepatic gene expression.

Background: Daratumumab is widely used in multiple myeloma (MM) and light chain amyloidosis (AL amyloidosis). The purpose of this study was to identify adverse event (AE) signals for daratumumab through the FDA Adverse Event Reporting System (FAERS) database to assess its safety in a large sample of people., Methods: Based on data from the FAERS database, three disproportionality analysis methods were used to mine AE signals for daratumumab, including reporting odd ratio (ROR), proportional reporting ratio (PRR), and bayesian configuration promotion neural network (BCPNN)., Results: A total of 9220 AE reports with daratumumab as the primary suspect drug were collected, containing 23,946 AEs. Within these reports, 252 preferred terms (PT) levels, 73 high level term (HLT) levels and 11 system organ class (SOC) levels of AE signals were detected, along with some new AEs. Most AEs occurred within the first month after drug administration., Conclusion: Our findings were consistent with the results of established studies that daratumumab has a good safety profile. The newly identified AEs are of concern and prospective clinical studies are needed to confirm whether they are causally related to daratumumab. This study provided an early warning for the safe use of daratumumab and also provided guidance for further safety studies.

Background: The high clinical and molecular heterogeneity of acute myeloid leukemia (AML) has led to an unsatisfactory clinical prognosis, thus we sought to incorporate both clinical features and molecular abnormalities to construct a new prognostic model., Methods: A database search of the Gene Expression Omnibus (GEO) revealed 238 cases of adult AML. The independent risk factors were assessed using both univariate and multivariate Cox regression, as well as least absolute shrinkage and selection operator (LASSO) regression. The predictive accuracy, discriminatory power and clinical applicability of the nomogram were determined by the consistency index (C-index), calibration curves and decision curve analysis (DCA). In addition, a single-centre cohort of 135 cases was used for external validation., Results: Multivariate Cox regression analysis showed that the independent influences on overall survival (OS) were age, type of disease, DNMT3A , IDH2 and TP53 mutations. The area under the curve (AUC) values for the training set were 0.755, 0.745 and 0.757 at 1, 2 and 3 years respectively; the AUC for the validation set were 0.648, 0.648 and 0.654 at 1, 2 and 3 years; and the AUC for the northwest China set were 0.692, 0.724 and 0.689 at 1, 2 and 3 years. The calibration and DCA indicated good consistency and clinical utility of the nomogram. Background: Somatic mutations are widespread in patients with Myelodysplastic Syndrome (MDS) and are associated with prognosis. However, a practical prognostic model for MDS that incorporates somatic mutations urgently needs to be developed., Methods: A cohort of 201 MDS patients from the Gene Expression Omnibus (GEO) database was used to develop the model, and a single-center cohort of 115 MDS cohorts from Northwest China was used for external validation. Kaplan-Meier analysis was performed to compare the effects of karyotype classifications and gene mutations on the prognosis of MDS patients. Univariate and multivariate Cox regression analyses and Lasso regression were used to screen for key prognostic factors. The shinyapps website was used to create dynamic nomograms with multiple variables. The time-dependent receiver operating characteristic (ROC) curves, calibration plots, and decision curve analysis (DCA) were used to evaluate the model's discrimination, accuracy and clinical utility., Results: Six risk factors (age, bone morrow blast percentage, ETV6, TP53, EZH2, and ASXL1) were considered as predictor variables in the nomogram. The nomogram showed excellent discrimination, with respective the area under the ROC curve (AUC) values of 0.850, 0.839, 0.933 for the training cohort at 1 year, 3 years and 5 years; 0.715, 0.802 and 0.750 for the testing cohort at 1 year, 3 years and 5 years; and 0.668, 0.646 and 0.731 for the external validation cohort at 1 year, 3 years and 5 years. The calibration curves and decision curve showed that the nomogram had good consistency and clinical practical benefit.
