Asplenium jiulianshanense, a new tetraploid fern species of the A. normale complex (Aspleniaceae) from Jiulianshan National Nature Reserve, southern Jiangxi, China is described and illustrated. We inferred the phylogenetic position of the new species based on sequences from seven plastid markers (atpB, rbcL, rps4, rps4-trnS, trnL, trnL-F, and trnG) and one low-copy nuclear gene, pgiC. The plastid phylogeny supported a close relationship among the new species A. jiulianshanense, A. minutifolium, and A. kiangsuense, while the nuclear phylogeny differed in topology from the plastid tree. The new species may be due to hybridization between A. kiangsuense and A. boreale. Morphologically, the new species can easily be distinguished from other members of the A. normale complex by rachises bearing a gemma near the apex, pinna margins entire to sparsely crenate, and (1‒)3‒4(‒6) sori per pinna. The proper screw geometry and pilot-hole size remain controversial in current biomechanical studies. Variable results arise from differences in specimen anatomy and density, uncontrolled screw properties and mixed screw brands, in addition to the use of different tapping methods. The purpose of this study was to evaluate the effect of bone density and pilot-hole size on the biomechanical performance of various pedicle screw geometries. Six screw designs, involving three different outer/inner projections of screws (cylindrical/conical, conical/conical and cylindrical/cylindrical), together with two different thread profiles (square and V), were examined. The insertional torque and pullout strength of each screw were measured following insertion of the screw into test blocks, with densities of 20 and 30 pcf, predrilled with 2.7-mm/3.2-mm/3.7-mm pilot holes. The correlation between the bone volume embedded in the screw threads and the pullout strength was statistically analyzed. Our study demonstrates that V-shaped screw threads showed a higher pullout strength than S-shaped threads in materials of different densities and among different pilot-hole sizes. The configuration, consisting of an outer cylindrical shape, an inner conical shape and V-shaped screw threads, showed the highest insertional torque and pullout strength at a normal and higher-than-normal bone density. Ilex L., the exclusive genus of Aquifoliaceae, encompasses over 600 dioecious wood species with a highly irregular distribution, predominantly found in South America and Asia. The phylogeny and classification of this genus remain enigmatic due to significant early extinctions, constrained morphological diversity, recent hybridization/introgression, and conflicting signals from previously utilized markers. This study presents phylogenetic reconstructions based on complete chloroplast genome sequences and single nucleotide polymorphisms (SNPs) derived from genome resequencing data. A total of 116 accessions of Ilex, representing approximately 108 taxa, were included as the ingroup, with five accessions of two species serving as outgroups. Analysis of the chloroplast genome and nuclear SNP data individually resulted in two robust phylogenetic trees, revealing substantial discrepancies between the chloroplast genome and nuclear SNP phylogenies at both the species and clade levels. The chloroplast genome sequences successfully resolved relationships within this genus into eight strongly supported major clades, while the nuclear SNPs resolved relationships into seven highly supported major clades. Our nuclear SNP phylogenetic tree, in comparison to the chloroplast genome tree, aligns more closely with the recently updated classification of Ilex in multiple instances. The Ilex L. (hollies) genus of Aquifoliaceae shows high species diversity in tropical and subtropical regions of Asia and South America. Throughout the range of the genus, Ilex species have been widely used in beverage and medicine production and as ornamentals. Here, we assembled a high-quality, chromosome-level genome of Ilex latifolia , which has extremely high economic value because of its useful secondary metabolite production and the high ornamental value of its decorative red berries. The 99.8% genome sequence was anchored to 20 pseudochromosomes, with a total length of 766.02 Mb and a scaffold N50 of 33.45 Mb. Based on the comparative genomic analysis of 14 angiosperm species, we recovered I. latifolia as the sister group to all other campanulids. Two whole-genome duplication (WGD) events were identified in hollies: one shared ancient WGD in the ancestor of all eudicots and a recent and independent WGD in hollies. We performed a genome-wide search to screen candidate genes involved in the biosynthesis of pentacyclic triterpenoid saponins in I. latifolia . Three subfamilies of CYP450 (CYP71A, CYP72A, and CYP716A) appear to have expanded. The transcriptomic analysis of I. latifolia leaves at five developmental stages revealed that two CYP716A genes and one CYP72A gene probably play important roles in this biosynthetic pathway. In addition, we totally identified 12 genes in the biosynthesis pathways of pelargonidin and cyanidin and observed their differential expression in green and red fruit pericarps, suggesting an association between pelargonidin and cyanidin biosynthesis and fruit pericarp color change. The accumulation of pelargonidin and cyanidin is expected to play an important role in the ornamental value of I. latifolia .
