There are currently few viable diagnostic techniques for in situ measurement of plasma facing component erosion. Digital holography is intended to fill this gap. Progress on the development of single and dual CO
2 laser digital holography diagnostics for in situ plasma facing component erosion is discussed. The dual laser mode's synthetic wavelength allows the measurable range to be expanded by a factor of ∼400 compared to single laser digital holography. This allows the diagnostic to measure surface height changes of up to 4.5 μm in single laser mode and up to 2 mm in dual laser mode. Results include ex situ measurements of plasma eroded targets and also dynamic measurements of nm and μm scale motion of a target mounted on a precision translation stage. Dynamic measurements have successfully been made with the system operating in both single and dual laser modes, from ∼50 nm to ∼4 μm in single laser mode and up to ∼400 μm in dual laser mode (limited only by the stage speed and camera acquisition duration). These results demonstrate the feasibility of using digital holography to characterize plasma facing component erosion dynamically, i.e., during plasma exposure. Results of proof-of-principle in situ digital holographic measurements of targets exposed to an electrothermal arc plasma source are presented. [ABSTRACT FROM AUTHOR]- Published

© 2017 The Author(s). Background: The fungal genus Aspergillus is of critical importance to humankind. Species include those with industrial applications, important pathogens of humans, animals and crops, a source of potent carcinogenic contaminants of food, and an important genetic model. The genome sequences of eight aspergilli have already been explored to investigate aspects of fungal biology, raising questions about evolution and specialization within this genus. Results: We have generated genome sequences for ten novel, highly diverse Aspergillus species and compared these in detail to sister and more distant genera. Comparative studies of key aspects of fungal biology, including primary and secondary metabolism, stress response, biomass degradation, and signal transduction, revealed both conservation and diversity among the species. Observed genomic differences were validated with experimental studies. This revealed several highlights, such as the potential for sex in asexual species, organic acid production genes being a key feature of black aspergilli, alternative approaches for degrading plant biomass, and indications for the genetic basis of stress response. A genome-wide phylogenetic analysis demonstrated in detail the relationship of the newly genome sequenced species with other aspergilli. Conclusions: Many aspects of biological differences between fungal species cannot be explained by current knowledge obtained from genome sequences. The comparative genomics and experimental study, presented here, allows for the first time a genus-wide view of the biological diversity of the aspergilli and in many, but not all, cases linked genome differences to phenotype. Insights gained could be exploited for biotechnological and medical applications of fungi.

21. Comparison of Verbal, Gesture and Skill Data Observed in Complex Motor Skill Acquisition. Study II.

- Abstract
This longitudinal field study performed on five female college freshmen attempted to verify a prior pilot study conducted on one subject to compare three verbal, four gesture, and three skill test variables to see which better discriminated between days, thereby providing the researcher a clearer picture of the subjects' changes during learning. Subjects were filmed over four daily one-half hour sessions by a professional team. Sixty tennis serves per subject (15 per day) were randomly selected for the analyses. Direct and RAO's discriminant analyses from SPSS using "day" as the group variable selected three gesture, one verbal, and one skill variable. Using these five variables, 39 percent of the serves were correctly classified. Compared to the pilot study, five instead of seven variables were selected. All the gesture variables were selected both times. One instead of three verbal variables were selected. Form took the place of speed among skill variables. Since gesture data were the most consistently discriminating, motor learning researchers should include gesture along with skill test and verbal data when studying the learner the first days of learning a complex motor skill. - Abstract
One novice subject's learning about the tennis serve was measured (over a 4-day period) by recording the subject's gestures and verbal remarks. This case study compared gestures, verbal remarks, skills, and form data to see which types best discriminated between days 1 to 4, and it also attempted to determine if discriminant analyses were suitable for such comparisons. Verbal data selected included: subject's evaluation of a serve, part of the serve described, and number of items the subject referenced. Gesture data selected included: body part used in the gesture, size of gesture, type of gesture, and number of gestures per serve. Results showed that generally gesture and verbal data appeared to better discriminate among days of early skill learning than did skill and form rating data. It was also found that the use of both contingency table and discriminant analyses is valuable. (Author/JMK)

The inclusive top quark pair (tt¯tt¯) production cross-section σtt¯σtt¯ has been measured in proton–proton collisions at s√=7 TeVs=7 TeV and s√=8 TeVs=8 TeV with the ATLAS experiment at the LHC, using tt¯tt¯ events with an opposite-charge eμeμ pair in the final state. The measurement was performed with the 2011 7 TeV dataset corresponding to an integrated luminosity of 4.6 fb−1fb−1 and the 2012 8 TeV dataset of 20.3 fb−1fb−1. The numbers of events with exactly one and exactly two bb-tagged jets were counted and used to simultaneously determine σtt¯σtt¯ and the efficiency to reconstruct and bb-tag a jet from a top quark decay, thereby minimising the associated systematic uncertainties. The cross-section was measured to be: σtt¯σtt¯=182.9±3.1±4.2±3.6±3.3 pb (s√=7 TeV)and=242.4±1.7±5.5±7.5±4.2 pb (s√=8 TeV), σtt¯=182.9±3.1±4.2±3.6±3.3 pb (s=7 TeV)andσtt¯=242.4±1.7±5.5±7.5±4.2 pb (s=8 TeV), where the four uncertainties arise from data statistics, experimental and theoretical systematic effects, knowledge of the integrated luminosity and of the LHC beam energy. The results are consistent with recent theoretical QCD calculations at next-to-next-to-leading order. Fiducial measurements corresponding to the experimental acceptance of the leptons are also reported, together with the ratio of cross-sections measured at the two centre-of-mass energies. The inclusive cross-section results were used to determine the top quark pole mass via the dependence of the theoretically predicted cross-section on mpoletmtpole giving a result of mpoletmtpole=172.9+2.5−2.6=172.9−2.6+2.5 GeV. By looking for an excess of tt¯tt¯ production with respect to the QCD prediction, the results were also used to place limits on the pair-production of supersymmetric top squarks t~1t~1 with masses close to the top quark mass, decaying via t~1→tχ~01t~1→tχ~10 to predominantly right-handed top quarks and a light neutralino χ~01χ~10, the lightest supersymmetric particle. Top squarks with masses between the top quark mass and 177 GeV are excluded at the 95 % confidence level.

The Group on Earth Observations (GEO) established the Forest Carbon Tracking (FCT) task in 2008 to provide satellite-based observation support to countries wishing to establish a national system for forest carbon monitoring. The main goal of the GEO FCT task is to support countries on the path towards the establishment of national Monitoring, Reporting and Verification (MRV) systems. To have a full national accounting system supporting the REDD mechanism, reliable methods for estimation of changes in forest carbon emissions are needed. To reduce the possibility of leaking in such a system wall-to-wall coverage is desirable and methods to separate the anthropogenic caused changes from the natural ones must be in place. A set of general recommendations is defined within the task, like national wall-to-wall coverage with annual update at the spatial resolution of 25-30 meter. Based on the above recommendations two products, Forest/Non-Forest and Land-use type maps, are considered highest priority (Horizon-1) with a set of more diverse and trend related Horizon-2 products to come later. There is not a single method or kind of method that has been identified as the most effective across a wide range of environments and types of imagery, so to achieve consistency through space and time a set of guiding principles has been established. In general, automated analysis (computer-based) is preferable to visual interpretation of imagery and changes are more efficient monitored by comparing images as opposed to comparing maps derived from images. The mapping of forest information products, when possible, should be based on a time series of satellite observations. Optical images, with wealth of developed methods and historically rich data archive, especially the Landsat data series, constitutes a base source for the information products. However, due to the vulnerable of optical sensors to clouds, development of methods for efficient interoperable use of optical and SAR data is a goal for the task. If reporting should be based on emission reference levels the MRV-system must be able to measure a proxy such as above ground forest biomass with reliable confidence. This is a challenge with wide-area optical or SAR sensors. Valuable solutions have been extensive modeling of biomass changes trough time from a stack of images combined with ancillary data and growth functions and methods where disturbance can be identified by the change patterns that occur when a measure of forest cover is plotted as a trajectory through time. Such a trajectory approach can also be used to smooth the annual variations in biomass estimation caused by noise in the images and thereby produce a more stable and reliable estimate than possible from single date images. Even small biomass changes caused by degradation or forest regrowth has been successfully monitored with such approaches.

Diatoms are photosynthetic secondary endosymbionts found throughout marine and freshwater environments, and are believed to be responsible for around one-fifth of the primary productivity on Earth. The genome sequence of the marine centric diatom Thalassiosira pseudonana was recently reported, revealing a wealth of information about diatom biology. Here we report the complete genome sequence of the pennate diatom Phaeodactylum tricornutum and compare it with that of T. pseudonan to clarify evolutionary origins, functional significance and ubiquity of these features throughout diatoms. In spite of the fact that the pennate and centric lineages have only been diverging for 90 million years, their genome structures are dramatically different and a substantial fraction of genes (~40%) are not shared by these representatives of the two lineages. Analysis of molecular divergence compared with yeasts and metazoans reveals rapid rates of gene diversification in diatoms. Contributing factors include selective gene family expansions, differential losses and gains of genes and introns, and differential mobilization of transposable elements. Most significantly, we document the presence of hundreds of genes from bacteria. More than 300 of these gene transfers are found in both diatoms, attesting to their ancient origins, and many are likely to provide novel possibilities for metabolite management and for perception of environmental signals. These findings go a long way towards explaining the incredible diversity and success of the diatoms in contemporary oceans.

Mycosphaerella is one of the largest genera of plant pathogenic fungi with more than 1,000 named species, many of which are important pathogens causing leaf spotting diseases in a wide variety of crops including cereals, citrus, banana, eucalypts, soft fruits, and horticultural crops. A few species of Mycosphaerella cause disease in humans and other vertebrates. An international project was initiated to sequence the genomes of M. graminicola and M. fijiensis, two of the most economically important pathogens of wheat and banana, respectively, along with 40,000 ESTs from M. fijiensis and the related maize pathogen Cercospora zeae-maydis, through the Community Sequencing Program sponsored by the U.S. DOE-Joint Genome Institute. The 9x M. graminicola sequencing is complete and was made public November 1, 2006 following automated and manual annotation. Due to the very good assembly statistics as well as a >2000-marker DArT linkage map that was aligned to the genome, JGI decided to finish the M. graminicola genome at the Stanford Human Genome Center. The majority of chromosomes have been sequenced completely including both telomeres. These data indicate that M. graminicola has both the largest chromosome number and the smallest chromosome sizes recorded among filamentous ascomycetes. The M. fijiensis EST sequencing has resulted in more than 30,000 ESTs and the genome sequencing is currently at approximately 2.5x, which enabled us to revise the genome size estimate of M. fijiensis to approximately 68 Mb, which is 70% larger than that for M. graminicola. The current status of both sequencing projects will be discussed.
