5-Fluorouracil (5-FU) is widely used in the treatment of gastric cancer, and the emergence of drug resistance and toxic effects has limited its application. Therefore, there is an urgent need for safe and effective novel drugs or new therapies. β-Ionone (BI) is found in vegetables and fruits and possesses an inhibitory proliferation of tumor cells in vitro and in vivo. In this study, we investigated whether BI could enhance the inhibitory effects of 5-FU on the proliferation of gastric adenocarcinoma cells and the growth of gastric cancer cell xenografts in a mouse model. The effects of BI and 5-FU alone or their combination on the cell viability, apoptosis, and mitochondrial membrane potential, the cell cycle, and its related proteins—Cyclin D1, and CDK4 as well as PCNA and GSK-3β were evaluated in SGC-7901 cells and MKN45 cells by MTT, MB, flow cytometry and Western blot. In addition, the effects of BI and 5-FU alone or their combination on the growth of SGC-7901 cell xenografts in nude mice were investigated. The results showed that BI significantly enhanced the sensitivity of gastric adenocarcinoma cells to 5-FU in vitro and in vivo, i.e. proliferation inhibited, apoptosis induced and GSK-3β protein activated. Therefore, our results suggest that BI increases the antitumor effect of 5-FU on gastric adenocarcinoma cells, at least partly from an activated GSK-3β signaling pathway. [ABSTRACT FROM AUTHOR]

This paper demonstrates a five-layer InAs/InP quantum-dash semiconductor optical amplifier (QDash-SOA), which will be integrated into microwave-photonic on-chip devices for millimeter-wave (mmWave) over fibre wireless networking systems. A thorough investigation of the QDash-SOA is conducted regarding its communication performance at different temperatures, bias currents, and input powers. The investigation shows a fibre-to-fibre (FtF) small-signal gain of 18.79 dB and a noise figure of 6.3 dB. In a common application with a 300 mA bias current and 25 °C temperature, the peak FtF gain is located at 1507.8 nm, which is 17.68 dB, with 3 dB gain bandwidth of 56.6 nm. Furthermore, the QDash-SOA is verified in a mmWave radio-over-fibre link with QAM (32 Gb/s 64-QAM 4-GBaud) and OFDM (250 MHz 64-QAM) signals. The average error vector magnitude of the QAM and OFDM signals after a 2 m wireless link could be as low as 8.29% and 6.78%, respectively. Triple‐negative breast cancer (TNBC) has greater infiltration of M2‐like macrophages (TAMs), which enhances cancer cell invasion and leads to a poor prognosis. TNBC progression is mediated by both tumor cells and the tumor microenvironment (TME). Here we elucidate the mechanism of the interaction between TNBC cells and TAMs. In this study, we confirmed that CD44v5 is highly expressed in TNBC, which drives TNBC cell metastasis and promotes TAM polarization by co‐localizing with IL4Rα and inhibiting its internalization and degradation, thereby promoting activation of the STAT3/IL6 signaling axis. At the same time, TAMs also facilitate TNBC cell metastasis by secreting IL‐4, IL‐6, and other cytokines, in which the IL‐4/IL‐4R/STAT3/IL‐6 signaling axis plays the same role for TNBC cells responding to TAMs. Moreover, we found that the above progress could be suppressed when the CD44v5 domain was blocked. We demonstrated that the CD44v5/IL‐4R/STAT3/IL‐6 signaling pathway plays a key role in TNBC cell metastasis, and in TNBC cells inducing TAM polarization and responding to TAMs, promoting metastasis. Magnesium alloys have been used to manufacture biodegradable implants, bone graft substitutes, and cardiovascular stents. WE43 was the most widely used magnesium alloy. The degradation process begins when the magnesium alloy stent is implanted in the body and comes into contact with body fluid. The degradation products include hydrogen, Mg2+, local alkaline environment, and unsoluble products. A large number of studies focused on Mg2+ and pH in vitro, and in vivo of magnesium alloys, but few studies on unsoluble corrosion products (UCPs). In this study, UCPs of WE43 alloy were prepared by immersion in vitro, and their effects on macrophages were investigated. The results showed that the unsoluble corrosion products were Mg24Y5, Mg12YNd, and MgCO3·3H2O, which were dose‐dependent on the apoptosis and necrosis of macrophages. After phagocytosis of UCPs, macrophages mainly metabolize in lysosome, and autophagy also participates in the metabolism of UCPs. It also decreases mitochondrial membrane potential and increases lysosomes, endoplasmic reticulum stress, and P2X7 receptor activation. These will increase reactive oxygen species (ROS) in cells, activating NLRP3 inflammatory corpuscles, activating the downstream pro‐IL18 and pro‐IL1β, and converting it to IL‐18, and IL‐1β. However, its pro‐inflammatory effect is far lower than that of the classical Lipopolysaccharide (LPS) pro‐inflammatory pathway. Enzyme biofuel cells (EBFCs) can convert chemical or biochemical energy in fuel into electrical energy, and therefore have received widespread attention. EBFCs have advantages that traditional fuel cells cannot match, such as a wide range of fuel sources, environmental friendliness, and mild reaction conditions. At present, research on EBFCs mainly focuses on two aspects: one is the use of nanomaterials with excellent properties to construct high-performance EBFCs, and the other is self-powered sensors based on EBFCs. This article reviews the applied nanomaterials based on the working principle of EBFCs, analyzes the design ideas of self-powered sensors based on enzyme biofuel cells, and looks forward to their future research directions and application prospects. This article also points out the key properties of nanomaterials in EBFCs, such as electronic conductivity, biocompatibility, and catalytic activity. And the research on EBFCs is classified according to different research goals, such as improving battery efficiency, expanding the fuel range, and achieving self-powered sensors. We present here a performance comparison of quantum-dash (Qdash) semiconductor amplifiers (SOAs) with three, five, eight, and twelve InAs dash layers grown on InP substrates. Other than the number of Qdash layers, the structures were identical. The eight-layer Qdash SOA gave the highest amplified spontaneous emission power (4.3 dBm) and chip gain (26.4 dB) at 1550 nm, with a 300 mA CW bias current and at 25 °C temperature, while SOAs with fewer Qdash layers (for example, three-layer Qdash SOA), had a wider ASE bandwidth (90 nm) and larger 3 dB gain saturated output power (18.2 dBm) in a shorter wavelength range. The noise figure (NF) of the SOAs increased nearly linearly with the number of Qdash layers. The longest gain peak wavelength of 1570 nm was observed for the 12-layer Qdash SOA. The most balanced performance was obtained with a five-layer Qdash SOA, with a 25.4 dB small-signal chip gain, 15.2 dBm 3 dB output saturated power, and 5.7 dB NF at 1532 nm, 300 mA and 25 °C. These results are better than those of quantum well SOAs reported in a recent review paper. The high performance of InAs/InP Qdash SOAs with different Qdash layers shown in this paper could be important for many applications with distinct requirements under uncooled scenarios. Diode optical frequency comb lasers are promising compact solutions to generate high-speed optical pulses for applications in high spectral efficiency wavelength division multiplexing transmission with advanced modulation formats. In this paper, an InAs/InP quantum dot (QDot) C-band single-section passively mode-locked laser (MLL) based broadband optical frequency comb source with a free spectral range of 28.4 GHz is presented. The device exhibits less than 1.5 MHz optical linewidth (phase noise) over 56 channels and 2.1 fs pulse-to-pulse timing jitter with a central wavelength of 1550 nm. Using this comb, we demonstrate an aggregate data transmission capacity of 12.5 Terabit/s over 100 km of standard single mode fiber by employing dual-polarization with 16 QAM modulation format. This investigation shows the viability for semiconductor QDot MLLs to be used as low-cost optical source in Terabit/s or higher optical networks. [ABSTRACT FROM AUTHOR]

BACKGROUND: Lingonberry (Vaccinium vitis-idaea L.), as an important natural and wild plant resource in the world, has high economic and nutritional values. Many researchers have focused on the effect of antioxidant and enzyme inhibitors. OBJECTIVE: The present study aimed to evaluate the active ingredients, in vitro antioxidant and enzyme-inhibitory activity from different parts (root, stem, leaf, and fruit) of wild lingonberry. METHODS: The active ingredients of lingonberry were determined by ultra-performance liquid chromatography-triple quadrupole-mass spectrometry (UPLC-TQ-MS/MS). Antioxidant activities were measured by DPPH, ABTS, FRAP and CUPRAC assays. Principal component analysis (PCA) and agglomerated hierarchical clustering (AHC) were used to analyze the relationship between active ingredients, antioxidant and enzyme-inhibitory activity. RESULTS: Phenolic compounds were significantly higher in leaf and stem. The enzyme inhibitory of the extracts varied observably according to the plant parts. Fruit had the highest acetylcholinesterase (317.67 mg GALAEs/g) and butyrylcholinesterase (346.04 mg GALAEs/g) inhibitory activity, while leaf had the most potent activity on α-amylase (256.59 mg ACAEs/g), α-glucosidase (186.70 mg ACAEs/g) and tyrosinase (42.87 mg KAEs/g). Tyrosinase had strong correlation and similarity with phenolic acids and flavonoids in the correlation analysis and PCA. CONCLUSIONS: 29 active ingredients were detected, including phenolic acids, flavonoids, anthocyanins, and triterpenes. Lingonberry sample to inhibit the activity of tyrosinase was associated with five flavonoids (kaempferol-3-O-galactoside, kaempferol-3-O-β-D-glucosyl (1 ⟶ 2) galactoside, biorobin,,quercetin 3-O-glucoside-7-O-rhamnoside, rutinum) and phenolic acid content (arbutin). We demonstrate real-time broadband multi-Gb/s electrical RF synthesizer-free millimeter-wave (MMW) signals generation and wireless transmission at the 5G new radio (NR) frequency band of 47 GHz based on analog radio-over-fiber (A-RoF) fronthaul. This is enabled by a low noise, highly correlated, monolithic C-band semiconductor InAs/InP quantum-dash (QDash) dual-wavelength distributed feedback (DW-DFB) laser. One laser mode is encoded using 4-/6-GBaud multilevel quadrature amplitude modulation (M-QAM) (16-/32-/64-QAM) baseband data signals, the other lasing mode is used as an optical local oscillator for optical-heterodyne remote up-conversion to a MMW carrier of 47.27 GHz. Consequently, optical baseband modulated data signals with data capacity up to 36 Gb/s (6-GBaud × 64-QAM) are transmitted through back-to-back (BtB) and 25-/50-km of standard single mode fiber (SSMF) before the MMW carrier is optically synthesized remotely for free space wireless data transmission and detection over up to 9-m. The end-to-end MMW-over-fiber (MMWoF) wireless link is thoroughly characterized exhibiting promising error-vector-magnitude (EVM) and bit-error-rate (BER) values. The 4-/6-GBaud 16-QAM MMWoF wireless links achieve EVMs down to 6.32%/7.33%, 6.71%/7.78%, and 7.35%/8.91% through BtB, 25-km, and 50-km SSMF, respectively. Similarly, the EVM for 32-QAM and 64-QAM links is observed to be 5.56%/6.56% and 6.05%/6.62%, respectively. Moreover, in each case, the calculated BER is below the forward error correction (FEC) limit of 3.8 × 10−3. The results corroborate the potential and viability of the QDash DW-DFB laser as a simple, efficient and cost-effective alternative to individual laser sources for deployment in broadband photonic MMWoF fronthaul systems of 5G wireless networks. Heart failure (HF) is the typical terminal stage of cardiac diseases involving inflammatory states. The function of microRNAs (miRNAs) in the progress of HF remains poorly understood. In this study, real-time PCR results showed a decreased expression of miRNA-181b (miR-181b) in HF patients compared with healthy individuals. Besides, miR-181b expressions were negatively correlated with hypersensitive C-reactive protein (hsCRP) levels in the serum of HF patients. Receiver operator characteristic (ROC) curve analysis showed that miR-181b was a diagnostic predictor of HF, and the area under the curve was 0.970 (DCM-induced HF group) and 0.962 (ICM-induced HF group). Strikingly, in HF rats induced by isoproterenol (ISO), the expression of miR-181b of heart tissue was suppressed before tumor necrosis factor-alpha (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) increase, as revealed by western blot and real-time PCR. Besides, the overexpression of miR-181b also decreased the expression of TNF-α, IL-1β, and IL-6 in lipopolysaccharide- (LPS-) induced neonatal cardiomyocytes. The determination of timing jitter obtained from optical phase noise measurements is investigated in InAs/InP quantum dash Fabry–Pérot mode-locked coherent comb lasers with different pulse repetition rates. The results are compared with those determined through a direct measurement of the first harmonic of the RF power spectrum. Very good agreement is achieved. The ability to measure the timing jitter from optical phase noise is not restricted by the repetition rate of the laser being studied, allowing it to be applied to extremely high repetition rate lasers. Using these lasers we demonstrate 5.4 Tbit/s (PAM-4 48 × 28 GBaud PDM) and 10.3 Tbit/s (16 QAM 56 × 23 GBaud PDM) aggregate data transmission capacity. A three-degree-of-freedom flight simulator platform with a translational and two rotational movement capabilities is studied. Based on the Lagrange method the dynamic model is established and then simplified by eliminating the sub-diagonal elements of the matrixes. A washout algorithm is employed to qualify the platform for flight simulation in limited space. In order to improve the modeling and experimental accuracies further when some dynamic parameters of an actual platform are unknown, the stepwise identification method is proposed to solve this problem. By dividing the driving torque into the load torque, reducer driving torque, screw driving toque and integrated friction torque, and main dynamic parameters are identified by applying proper trajectories and estimated by the weighed least square method. We demonstrated automatic cross carrier-envelope phase locking between two synchronized pulse trains at different wavelengths generated within a single InAs/InP quantum-dot diode laser cavity. The resulting waveforms, verified by optical auto-/cross-correlation and explained by a theoretical model, show multipeaks or quasi-periodic subpulses with a central-subpulse pulse duration of about 183 fs. We compare the timing jitter of a mode-locked InAs/InP quantum dash (QD) coherent comb laser (CCL) with and without an external cavity self-injection feedback locking (SIFL) system. The jitter is determined through a measurement of the first harmonic of the RF power spectrum of the laser output detected using a fast photodiode. A significant reduction in timing jitter is observed in the laser with the external cavity SIFL. A pulse to pulse root-mean-square time jitter of 1.56 fs is achieved for the laser with the external cavity SIFL. This low timing jitter is a promising light source for the next generation high-speed coherent networking systems and all-optical signal processing. We demonstrate results of a double polarization 16-QAM data format transmission system at a symbol rate of 25 GBd using more than 30 individual channels of the QD CCL. An error vector magnitude of 5.8% and bit error rate of 3.0 × 10–9 were obtained. And, 6.0 Tb/s (16QAM 30 × 25 Gbd PDM) coherent transmission is achieved. [ABSTRACT FROM AUTHOR]

Abstract The red raspberry (Rubus idaeus L.) is a common fruit worldwide and its extract has been found to inhibit the growth of many types of tumors, mainly because it is rich in bioactive phytochemicals. However, the mechanism underlying its anticancer activity in hepatocellular carcinoma (HCC) is not well understood. Herein, the aim of this study was to determine the effects of red raspberry phytochemicals on the proliferation of hepatocellular carcinoma cells and to elucidate its biochemical and molecular targets. CCK8 and colony formation, as well as flow cytometry assays, were employed to determine the effects of red raspberry extract (RRE) on cell proliferation and cell cycle distribution in HCC cells. Our results showed that RRE significantly inhibited cell proliferation and arrested cell cycle progression at the S phase in HCC cells. RRE increased the expression of phosphatase and tensin homologue deleted on chromosome 10 (PTEN) by reducing the methylation status of the PTEN gene promoter and inhibiting DNMT1 expression and regulated AKT signaling pathway. These findings show that red raspberry phytochemicals inhibit the proliferation of HCC cells by regulating PTEN/AKT signaling pathway, providing evidence that RRE may be used as a potential auxiliary therapy for patients with HCC. Background: Continuous damage from oxidative stress and apoptosis are the important mechanisms that facilitate chronic heart failure (CHF). Molecular hydrogen (H2) has potentiality in the aspects of anti-oxidation. The objectives of this study were to investigate the possible mechanism of H2 inhalation in delaying the progress of CHF. Methods and Results: A total of 60 Sprague-Dawley (SD) rats were randomly divided into four groups: Sham, Sham treated with H2, CHF and CHF treated with H2. Rats from CHF and CHF treated with H2 groups were injected isoprenaline subcutaneously to establish the rat CHF model. One month later, the rat with CHF was identified by the echocardiography. After inhalation of H2, cardiac function was improved vs. CHF (p < 0.05), whereas oxidative stress damage and apoptosis were significantly attenuated (p < 0.05). In this study, the mild oxidative stress was induced in primary cardiomyocytes of rats, and H2 treatments significantly reduced oxidative stress damage and apoptosis in cardiomyocytes (p < 0.05 or p < 0.01). Finally, as a pivotal transcription factor in reactive oxygen species (ROS)-apoptosis signaling pathway, the expression and phosphorylation of p53 were significantly reduced by H2 treatment in this rat model and H9c2 cells (p < 0.05 or p < 0.01). Conclusion: As a safe antioxidant, molecular hydrogen mitigates the progression of CHF via inhibiting apoptosis modulated by p53. Glioblastoma (GBM) is one of the most lethal brain tumors with a short survival time. EGFR amplification and mutation is the most significant genetic signature in GBM. About half of the GBMs with EGFR amplification express a constitutively autophosphorylated variant of EGFR, known as EGFRvIII. Our in vitro data demonstrated further enhanced EGFRvIII activity and tumor cell invasion in the tumor microenvironment of hypoxia plus extracellular matrix (ECM) vitronectin, in which EGFRvIII and integrin β3 tended to form complexes. The treatment with ITGB3 siRNA or the integrin antagonist cilengetide preferentially interrupted the EGFRvIII/integrin β3 complex, effectively reduced tumor cell invasion and activation of downstream signaling effectors. Cilengitide is recently failed in Phase III CENTRIC trial in unselected patients with GBM. However, we found that cilengitide demonstrated efficacious tumor regression via inhibition of tumor growth and angiogenesis in EGFRvIII orthotopic xenografts. Bioinformatics analysis emphasized key roles of integrin β3, hypoxia and vitronectin and their strong correlations with EGFRvIII expression in malignant glioma patient samples in vivo. In conclusion, we demonstrate that EGFRvIII/integrin β3 complexes promote GBM progression and metastasis in the environment of hypoxia and vitronectin-enrichment, and cilengitide may serve as a promising therapeutics for EGFRvIII-positive GBMs.

Tocotrienols have been shown to inhibit proliferation and induce apoptosis in cancer cells. However, the molecular mechanisms involved in tocotrienol-induced apoptosis are still unclear. In the present study, ??-tocotrienol induced apoptosis in human gastric adenocarcinoma SGC-7901 cell line through down regulation of the extracellular signal-regulated kinase (ERK) signalling pathway. Furthermore, ??-tocotrienol-induced apoptosis was accompanied by down regulation of Bcl-2, up regulation of Bax, activation of caspase-3, and subsequent poly (ADP-ribose) polymerase cleavage. These results indicated that up or down regulation of Bcl-2 family proteins play a major role in the initiation of ??-tocotrienol-induced apoptosis as an activator of caspase-3.??-Tocotrienol also down regulated the activation of the Raf-ERK signalling pathway, and down regulated c-Myc by decreasing the expressions of Raf-1 and p-ERK1/2 proteins. The results suggest that key regulators in tocotrienol-induced apoptosis may be Bcl-2 families and caspase-3 in SGC-7901 cells through down regulation of the Raf-ERK signalling pathway. Abstract: 1-to-8 and 1-to-4 optical waveguide power splitters were fabricated using the focused 800nm-amplified femtosecond laser to directly write a series of 1-to-2 Y-junction splitters in a fused silica glass sample. A shifted multi-scan technique was developed to substantially reduce the excessive insertion loss of the involved Y junctions and the polarization dependent loss of such integrated splitters.
