In this paper, we describe our solution to predict student STEM career choices during the 2017 ASSISTments Datamining Competition. We built a machine learning system that automatically reformats the data set, generates new features and prunes redundant ones, and performs model and feature selection. We designed the system to automatically find a model that optimizes prediction performance, yet the final model is a simple logistic regression that allows researchers to discover important features and study their effects on STEM career choices. We also compared our method to other methods, which revealed that the key to good prediction is proper feature enrichment in the beginning stage of the data analysis, while feature selection in a later stage allows a simpler final model.

The walnut (Juglans regia L.) is a typical and an economically important tree species for nut production with heterodichogamy. The absence of female and male flowering periods seriously affects both the pollination and fruit setting rates of walnuts, thereby affecting the yield and quality. Therefore, studying the characteristics and processes of flower bud differentiation helps in gaining a deeper understanding of the regularity of the mechanism of heterodichogamy in walnuts. In this study, a total of 3540 proteins were detected in walnut and 885 unique differentially expressed proteins (DEPs) were identified using the isobaric tags for the relative and absolute quantitation (iTRAQ)-labeling method. Among all DEPs, 12 common proteins were detected in all four of the obtained contrasts. GO and KEGG analyses of 12 common DEPs showed that their functions are distributed in the cytoplasm metabolic pathways, photosynthesis, glyoxylate and dicarboxylate metabolism, and the biosynthesis of secondary metabolites, which are involved in energy production and conversion, synthesis, and the breakdown of proteomes. In addition, a function analysis was performed, whereby the DEPs were classified as involved in photosynthesis, morphogenesis, metabolism, or the stress response. A total of eight proteins were identified as associated with the morphogenesis of stamen development, such as stamen-specific protein FIL1-like (XP_018830780.1), putative leucine-rich repeat receptor-like serine/threonine-protein kinase At2g24130 (XP_018822513.1), cytochrome P450 704B1-like isoform X2 (XP_018845266.1), ervatamin-B-like (XP_018824181.1), probable glucan endo-1,3-beta-glucosidase A6 (XP_018844051.1), pathogenesis-related protein 5-like (XP_018835774.1), GDSL esterase/lipase At5g22810-like (XP_018833146.1), and fatty acyl-CoA reductase 2 (XP_018848853.1). The pomegranate fruit is valued for its nutritional and medicinal properties, and the composition and content of primary and secondary metabolites are the main factors impacting its nutritional and medicinal properties. However, a deep understanding of metabolites in different parts of fruit is still lacking. Here, the peel, aril, and seed of mature pomegranate fruits were analyzed separately to compare metabolic component differences using UPLC/MS–MS. A total of 858 metabolites belonging to 11 classes were identified, of which flavonoids, such as delphinidin‐3‐O‐glucoside and cyanidin‐3‐O‐glucoside; tannins, such as ellagic acid and punicalin; and terpenoids, such as corosolic acid and madasiatic acid, were upregulated in the peel. Lipids, such as punicic acid, methyl linolenate, and linoleic acid; alkaloids, such as indole and choline; nucleotides and derivatives mainly including 2‐deoxyribose‐1‐phosphate and 9‐(arabinosyl)‐hypoxanthine, were upregulated in seeds. Phenolic acids, such as 1‐O‐galloyl‐4,6‐(S)‐HHDP‐β‐D‐glucose and 1,7‐di‐O‐galloyl‐D‐sedoheptulose, and flavonoids, such as cyanidin‐3‐O‐glucoside, cyanidin‐3‐O‐(2″‐O‐xylosyl) galactoside, and delphinidin‐3‐O‐glucoside, were upregulated in aril. The flavone and flavonol biosynthesis (ko00944) pathways were significantly enriched between the peel and seed, as were the anthocyanin biosynthesis (ko00944) pathways between the aril and seed and the flavonoid biosynthesis (ko00941) pathways between the peel and aril. Additionally, functional antioxidants, such as 10,16‐dihydroxypalmitic acid, 3‐O‐methylellagic acid, and 3,3’‐O‐dimethylellagic acid, were first identified in pomegranate fruits. Our results revealed the composition and abundance of primary and secondary metabolites in pomegranate fruit, which can lay the foundation for further elucidation of its nutritional and medicinal properties. For the current coal mine directional drilling MWD equipment and system are independent of each other, resulting in different forms of MWD data at the bottom of the hole, forming data islands; Based on the B/S system architecture mode, by comparing the measurement while drilling mode and measurement data format, the requirement module of data comprehensive acquisition system is analyzed; the background database of the integrated collection system is set up to develop the development of the integrated collection system of directional drilling while drilling data in coal mine. The verification shows that the data acquisition system can meet the unified collection and processing of various MWD data, the overall analysis and evaluation function, and achieve real-time data upload. It provides data network transmission guarantee for directional drilling data supervision platform in coal mine, and improves the intelligent level of coal mine drilling.

Grape white rot is a devastating fungal disease caused by Coniella diplodiella. The pathogen delivers effectors into the host cell that target crucial immune components to facilitate its infection. Here, we examined a secreted effector of C. diplodiella , known as CdE1, which has been found to inhibit Bax-triggered cell death in Nicotiana benthamiana plants. The expression of CdE1 was induced at 12–48 h after inoculation with C. diplodiella , and the transient overexpression of CdE1 led to increased susceptibility of grapevine to the fungus. Subsequent experiments revealed an interaction between CdE1 and Vitis davidii cysteine-rich receptor-like kinase 10 (VdCRK10) and suppression of VdCRK10-mediated immunity against C. diplodiella , partially by decreasing the accumulation of VdCRK10 protein. Furthermore, our investigation revealed that CRK10 expression was significantly higher and was up-regulated in the resistant wild grapevine V. davidii during C. diplodiella infection. The activity of the VdCRK10 promoter is induced by C. diplodiella and is higher than that of Vitis vitifera VvCRK10 , indicating the involvement of transcriptional regulation in CRK10 gene expression. Fluorite and calcite have similar surface properties and natural floatability, so their flotation separation has always been a problem faced by the beneficiation industry. The key to flotation separation is the choice of depressants. Sodium hexametaphosphate (SHMP) has a good effect on fluorite calcite selective inhibition. In this paper, the effects of Zn2+ on the selective inhibition of SHMP in the flotation process of fluorite and calcite were studied through single mineral and artificial mixed mineral flotation experiments. Solution chemical calculation, X-ray photoelectron spectroscopy (XPS), Fourier Transform Infrared spectroscopy (FT-IR) and Time of Flight Secondary Ion Mass Spectrometry (ToF-SIMS) analyses investigated the mechanism of action of Zn2+, which had the most significant negative effect on the selective inhibition of SHMP. The results show that the main components of SHMP inhibiting minerals are HPO42- and H2PO4-, which can react with Ca active sites on the mineral surface to form hydrophilic Ca(H2PO4)2 and CaHPO4, while Zn2+ The presence of HPO42- in solution resulted in the formation of stable ZnHPO4 complexes, thereby weakening the inhibitory effect of SHMP on minerals. For industrial flotation foam image processing, accurate bubble size measurement and feature extraction are very important to optimize the flotation process and to improve the recovery of mineral resources. This paper presents an improved algorithm to investigate mineral flotation foam image segmentation for mineral processing. Several libraries implemented for the Python programming language are used for image enhancement and compensation, quantitative analysis of factors influencing the image segmentation accuracy, and suggestions for improvement of the flotation foam image processing. The bubble characteristics-size and morphology-and the influence of the flotation conditions on the flotation foam image are analyzed. A Python implementation of the Retinex image compensation method-region-adaptive and multiscale-is proposed to address known issues of uneven illumination and shadows affecting flotation foam images, thereby improving brightness uniformity. Finally, an improved version of the watershed segmentation algorithm included in the Python Open Source Computer Vision library is used for segmentation analysis. The accuracy of the flotation foam image segmentation is 3.3% higher than for the standard watershed algorithm and the segmentation time is 9.9% shorter. [ABSTRACT FROM AUTHOR]

This brief considers the Direction of Arrival (DOA) estimation for Distributed mmWave Radar System (DmmWRS) in the presence of coherent sources and a single snapshot. To this end, the large virtual aperture is firstly derived assuming that DmmWRS works in time-sharing mode. Then, a Fast Local Iterative Search Algorithm (FLISA) resorting to Iterative Adaptive Approach (IAA) and prior DOA information obtained by a single radar, is developed to accurately estimate DOA within a small azimuth region of interest. Finally, numerical simulations and actual measurements show the superiority of the proposed approach than some counterparts providing better angle measurement performance, more importantly, FLISA has lower computational complexity and faster convergence than IAA. Grinding is an important link in the process of mineral processing. It plays a vital role in mineral processing by optimizing the grinding process, improving the quality of grinding products and ensuring the follow-up operation indicators. In this paper, the Python language, intelligent theoretical control technology and mineral processing were combined to solve the problem of ore feeding control in mineral processing. Using error factor analysis, an extended control algorithm was designed. The NumPy library and data collected from the Yuan YangMou concentrator in China were used to quantitatively analyze the factors affecting the error of electronic belt scales. This paper introduces the use of Kalman filtering for electronic belt scale weight data to reduce the effect of noise and hence reduce errors. The factors affecting the process of mill feeding are also analyzed. The core ideas and methods of fuzzy control theory are summarized, and a Python-based fuzzy controller suitable for the mill feeding process that improves the overall robustness and accuracy of feeding system is implemented. Methyl jasmonate (MeJA) plays a vital role in plant disease resistance and also induces the expression of disease resistance genes in plants. In this study, a transcriptome analysis was performed on grapevine leaves after 12, 24 and 48 h of MeJA-100 μM treatment. A total of 1242 differentially expressed genes (DEGs) were identified from the transcriptome data, and the analysis of the DEGs showed that genes related to phytohormone signal transduction, jasmonic acid-mediated defense, Mitogen-activated protein kinase (MAPK), and flavonoid biosynthetic pathways were upregulated. As Pathogenesis-related gene 1 (PR1) is an important marker gene in plant defense also upregulated by MeJA treatment in RNA-seq data, the VvPR1 gene was selected for a promoter analysis with β-glucuronidase (GUS) through transient expression in tobacco leaves against abiotic stress. The results showed that the region from −1837 bp to −558 bp of the VvPR1 promoter is the key region in response to hormone and wound stress. In this study, we extended the available knowledge about induced defense by MeJA in a grapevine species that is susceptible to different diseases and identified the molecular mechanisms by which this defense might be mediated. Video highlight detection (VHD) has emerged as a pressing task due to the unprecedentedly increasing amount of video data, such as those from e-commerce live-broadcasting platforms. Many approaches focus on exploiting text data, in the form of video description or time-sync comments, to facilitate the VHD task. Despite the promising results, they have largely overlooked the noises inherent in the text data and have mostly relied on isolating the feature of text and video. In this paper, we introduce a novel model to handle VHD, termed Coevolution Network (CoEvo-Net), that allows us to account for joint learning of the language and video features explicitly via a coevolution paradigm, in which features from the two data modalities progressively refine each other. This is achieved by a dedicated CoEvo-Cell that takes language and video together as inputs, extracts cross-modality, and filters the undesired parts of the input, such as words in a sentence. Furthermore, we release a large-scale dataset of e-commerce for VHD, in which each video is coupled with a sentence for description, to benchmark the sentence-based VHD approaches. Existing wideband beampattern synthesis methods usually require multiple beamformers, each corresponding to one frequency bin, which results in excessive resource consumption in engineering practice. To alleviate the resource consumption of wideband array beamforming, we propose a novel time-domain wideband digital beamformer that can control the beam shape over the whole bandwidth for interference mitigation by using only one set of amplitude–phase weights and integer time delay filters. To tackle the resultant optimization problem, two effective algorithms are introduced, and both can solve the problem within polynomial time. One is an alternating optimization algorithm that decomposes the original problem into multiple tractable subproblems iteratively. The other is the convex approximation algorithm that transforms the original problem into a series of second-order cone programming problems iteratively. The two algorithms exhibit their respective advantages in terms of convergence and computational complexity. Extensive numerical examples are presented to evaluate the proposed time-domain wideband beamforming strategy, highlighting that it can form a deep notch in any interested spatial–spectral region to suppress the strong interferences. This letter considers a distributed cascade of two millimeter-wave (mmWave) radars to achieve a large virtual aperture for high-resolution angle measurement. In order to explore the high sidelobe problem caused by distributed radars, we derive the general Bartlett spatial spectrum expression of the virtual array. Furthermore, we introduce four cases to describe the relationship between the radar spacing, the number of transmitters/receivers, and the sidelobe position of the spatial spectrum. Distributed phased Multiple-Input Multiple-Output (phased-MIMO) radar system consisting of separated subarrays possesses a high angle resolution but with sharing high sidelobes. This paper therefore focuses on the design of a virtual low-sidelobe beampattern in distributed phased-MIMO radar. An iterative framework with respect to subarray layout and receive weighting coefficients is proposed to minimize beampattern Peak Sidelobe Level (PSL) accounting for practical position constraints as well as the mainlobe level restriction. In each iteration, an efficient cyclic optimization algorithm based on the Coordinate Descent (CD) framework is developed to seek for the subarray layout through splitting high dimensional layout optimization problem into multiple one-dimensional optimization problems, and the SemiDefinite Relaxation (SDR) technique and rank one approximation are explored to design weighting coefficients. This letter deals with a shaped beampattern synthesis problem through controlling the mainlobe ripple and the output filter energy based on the minimization of the peak sidelobe level in the spatial region of interest. To tackle the challenging optimization problem with nonconvex constraints, a novel first-order iterative convex approximation (FOICA) procedure that handles the nonconvex constraints with their convex upper bound functions resorting to the first-order Taylor expansion is developed. Hence, in each iteration, the original problem is approximated through a convex problem globally solved via a polynomial time method. Additionally, a nonnegative slack variable to ensure a decreasing trend in the objective function during the iteration is introduced. Grape white rot caused by Coniella diplodiella (Speg.) affects the production and quality of grapevine in China and other grapevine-growing countries. Despite the importance of C. diplodiella as a serious disease-causing agent in grape, the genome information and molecular mechanisms underlying its pathogenicity are poorly understood. To bridge this gap, 40.93 Mbp of C. diplodiella strain WR01 was de novo assembled. A total of 9,403 putative protein-coding genes were predicted. Among these, 608 and 248 genes are potentially secreted proteins and candidate effector proteins (CEPs), respectively. Additionally, the transcriptome of C. diplodiella was analyzed after feeding with crude grapevine leaf homogenates, which reveals the transcriptional expression of 9,115 genes. Gene ontology enrichment analysis indicated that the highly enriched genes are related with carbohydrate metabolism and secondary metabolite synthesis. Forty-three putative effectors were cloned from C. diplodiella , and applied for further functional analysis. Among them, one protein exhibited strong effect in the suppression of BCL2-associated X (BAX)-induced hypersensitive response after transiently expressed in Nicotiana benthamiana leaves. Background. CD147/basigin (Bsg), a transmembrane glycoprotein, activates matrix metalloproteinases and promotes inflammation. Objective. The aim of this study is to explore the clinical significance of CD147 in the pathogenesis of inflammatory bowel disease (IBD). Results. In addition to monocytes, the clinical analysis showed that there is no significance obtained in leucocyte, neutrophil, eosinophil, basophil, and erythrocyte between IBD and controls. Immunohistochemistry analysis showed that CD147 was increased in intestinal tissue of patients with active IBD compared to that in the control group. What is more, CD147 is involved in intestinal barrier function and intestinal inflammation, which was attributed to the fact that it has an influence on MCT4 expression, a regulator of intestinal barrier function and intestinal inflammation, in HT-29 and CaCO2 cells. Most importantly, serum level of CD147 content is higher in active IBD than that in inactive IBD or healthy control, which could be a biomarker of IBD. Conclusion. Background. Pyroptosis, a novel form of inflammatory programmed cell death, was recently found to be a cause of mucosal barrier defect. In our pervious study, CD147 expression was documented to increase in intestinal tissue of inflammatory bowel disease (IBD). Objective. The aim of this study was to determine the function of serum CD147 in pyroptosis. Methods. The study group consisted of 96 cases. The centration of CD147, IL-1β, and IL-18 levels in serum was assessed by ELISA. Real-time PCR and WB were performed to analyze the effect of CD147 on pyroptosis. Results. In this study, our results showed that CD147 induced cell pyroptosis in intestinal epithelial cells (IECs) by enhancement of IL-1β and IL-18 expression and secretion in IECs, which is attributed to activation of inflammasomes, including caspase-1 and GSDMD as well as GSDME, leading to aggregate inflammatory reaction. Mechanically, CD147 promoted phosphorylation of NF-κB p65 in IECs, while inhibition of NF-κB activity by the NF-κB inhibitor BAY11-7082 reversed the effect of CD147 on IL-1β and IL-18 secretion. Most importantly, serum CD147 level is slightly clinically correlated with IL-1β, but not IL-18 level. Conclusion. These findings revealed a critical role of CD147 in the patients with IBD, suggesting that blockade of CD147 may be a novel therapeutic strategy for the patients with IBD. [ABSTRACT FROM AUTHOR]

Cancer is always the greatest threat to human health. In this work, a split-ring resonator (SRR)-based sensor was simulated for label-free detection on biomarkers in microwave. The model of biosensor was simulated by Computer Simulation Technology 2017 (CST2017) and some influences of feature parameters on resonance frequency and Q-factor were also discussed. From the simulation results, the model of sensor in our work showed ultrahigh Q-factor and sensitivity under different permittivities and also exhibited a trend of better performance when it works in higher frequency. As a common depressant, acidified water glass (AWG) has strong inhibitory effects on fluorite and calcite. The inhibited fluorite is difficult to be recovered, thus resulting in the waste of resources and low flotation efficiency. In this study, the interaction of fluoride ions with fluorite and calcite surfaces was investigated, and its effects of AWG adsorption on mineral surfaces were evaluated. Micro-flotation experiments indicated that the flotation recovery of fluorite is 88.72% after fluoride ion treatment, that is, approximately fourfold with respect to that without fluoride ion modification. The results of solution chemical calculations showed that SiO(OH)3− is the main component to inhibit fluorite, and Si(OH)4 is the main component to inhibit calcite in AWG solution. XPS and ICP-MS results showed that fluoride ions can improve the floatability of fluorite by converting CaSiO3 on the surface of fluorite into CaF2, but the conversion ability of Si(OH)4 on the surface of calcite is weak, which increases the difference in floatability between fluorite and calcite. The above results were further verified by the analysis of flotation foam image and contact angle measurement. After fluorine ion treatment, the contact angle of fluorite increased, and it was more easily adsorbed on the foam. Therefore, the interaction of the fluoride ion with the fluorite surfaces eliminated the adsorption of AWG on fluorite, thereby resulting in the enhanced hydrophobicity of fluorite. Hub vane diffusers can obtain higher pressure recovery and efficiency compared with the vaneless diffuser and a wider operating range compared with the vane diffuser. To improve the performance of high pressure ratio centrifugal compressors it is necessary to understand the flow phenomena in the hub vane diffusers. Although some research work on hub vane diffuser flow has been published, many unknown flow physics and disputed questions still remain. A computational analysis of the flowfield in a high-speed centrifugal compressor with a backswept unshrouded impeller and wedge vane channel diffuser is presented. In order to obtain the optimum hub vane height, the pressure and velocity fields were numerically simulated for eight kinds of hub vane, which are varied as follows: h/b = 0 (vaneless), 0.2, 0.3, 0.4, 0.5, 0.6, 0.8 and 1 (vane). The advanced Navier-Stokes solver EURANUS/TURBO is applied with a Baldwin-Lomax model for closure. The calculations are performed with a central space discretization scheme, with second- and fourth-order artificial dissipation terms. The numerical procedure applied a four-stage explicit Runge-Kutta scheme, coupled to local time stepping. The good agreement of the computed stage performance and the experimental data of the stage with the vane diffuser shows that the solver is effective and reasonable. Numerical results show that the maximum flowrate attained by the stage with the vane diffuser is about 12.5 per cent less than with the hub vane diffusers. The stage efficiency reaches its maximum value for the hub vane diffuser with an h/b ratio of 0.5 at lower flowrate and for the hub vane diffuser with an h/b ratio of 0.4 at higher flowrate. The hub vane diffusers have a wider operating range than the vane diffuser. The hub vane diffuser with an h/b ratio of 0.3 shows a higher mass-averaged static pressure coefficient up to a flow coefficient φ = 0.16. The hub vane diffuser with an h/b ratio of 0.4 shows a slightly higher mass-averaged static pressure coefficient over an operating range of φ = 0.18-0.28, but it falls beyond φ = 0.3 below that of the vaneless diffuser. The performance of the centrifugal compressor with the shroud vane diffuser is better than that with the hub vane diffuser for the same height of the vane. [ABSTRACT FROM AUTHOR]

The electrochemical sensing of dopamine is of great significance for studying and treating neurochemical diseases due to its potential feasibility for in vivo diagnostics. The commonly used sensors suffer from low sensitivity, the interference of ascorbic acid, and poor flexibility. In this paper, the function of electrode substrates including polyolefin, polystyrene, and polyethylene terephthalate films were investigated for their ability to improve electrochemical performances and provide favorable flexibility. The interference from ascorbic acid was cut down to a minimum by reducing the electrochemical resistance and the ascorbic acid diffusion current. The results demonstrate that gold electrodes prepared on polyolefin films exhibit a low charge transfer resistance of about 20 Ω, high sensitivity of dopamine detection (7.8 μA/μM), which is about 312 folds that of silicon electrode (0.025 μA/μM) and excellent flexibility. Having regulated the fabrication process of graphene by altering self-assembly layers and modification area, the sensor shows a dopamine detection limit of 0.11 μM in the presence of 500 μM ascorbic acid, and a sensitivity of 0.33 μA/μM. This work is valuable for the further improvement of the sensitivity and selectivity of the electrochemical sensor. • This paper presents a flexible micro sensor on a polyolefin substrate exhibits a high sensitivity of dopamine detection (7.8 μA/μM), about 312 folds of silicon-based sensor (0.025 μA/μM). • This proposed sensor shows a considerably low detection limit of dopamine (0.11 μM) in the existence of 500 μM ascorbic acid, compared with the most pure graphene-modified dopamine sensor. • A simple model of voltammetry curve simulation is reported to identify the interference mechanism of ascorbic acid for enhancing performance of the proposed microsensors. • Layer-by-layer self-assembly of graphene minimizes the interference from ascorbic acid by reducing electrochemical resistance and enhancing ascorbic acid diffusion. Objective: Inflammatory bowel disease (IBD) is a disorder intestinal inflammation and impaired barrier function, associated with increased epithelial expression of monocarboxylate transporter 4 (MCT4). However, the specific non‐metabolic function and clinical relevance of MCT4 in IBD remain to be fully elucidated. Methods: Lentivirus‐mediated overexpression of MCT4 was used to assess the role of MCT4 in transcriptionally regulating ZO‐1 and IL‐6 expression by luciferase assays, WB and ChIP. IP was used to analyse the effect of MCT4 on the interaction NF‐κB‐CBP or CREB‐CBP, and these MCT4‐mediated effects were confirmed in vivo assay. Results: We showed that ectopic expression of MCT4 inhibited ZO‐1 expression, while increased pro‐inflammatory factors expression, leading to destroy intestinal epithelial barrier function in vitro and in vivo. Mechanistically, MCT4 contributed NF‐κB p65 nuclear translocation and increased the binding of NF‐κB p65 to the promoter of IL‐6, which is attributed to MCT4 enhanced NF‐κB‐CBP interaction and dissolved CREB‐CBP complex, resulting in reduction of CREB activity and CREB‐mediated ZO‐1 expression. In addition, treatment of experimental colitis with MCT4 inhibitor α‐cyano‐4‐hydroxycinnamate (CHC) ameliorated mucosal intestinal barrier function, which was due to attenuation of pro‐inflammation factors expression and enhancement of ZO‐1 expression. Baicalin is a traditional Chinese herb purified from the root of Scutellaria baicalensis Georgi. In this study, we further analyzed the molecular mechanism behind the anti-tumor activity of Baicalin in colorectal cancer (CRC). The establishment of circular RNA (circRNA)/microRNA (miRNA)/messenger RNA (mRNA) axis was predicted by bioinformatic databases and verified by dual-luciferase reporter assay and RNA immunoprecipitation (RIP) assay. Baicalin dose-dependently reduced the expression of circRNA myosin heavy chain 9 (circMYH9) in CRC cells. Baicalin exposure suppressed the malignant phenotypes of CRC cells, which were largely reversed by the overexpression of circMYH9. CircMYH9 functioned as a molecular sponge for miR-761. CircMYH9 overexpression protected CRC cells from Baicalin-induced injury partly through down-regulating miR-761. MiR-761 interacted with the 3′ untranslated region (3' UTR) of hepatoma-derived growth factor (HDGF) mRNA. CircMYH9 up-regulated HDGF expression partly through sponging miR-761 in CRC cells. MiR-761 silencing counteracted the anti-tumor activity of Baicalin partly through up-regulating HDGF in CRC cells. Baicalin suppresses xenograft tumor growth in vivo , and this suppressive effect was partly reversed by the overexpression of circMYH9. Objective: To investigate the impact of IGJ on the proliferation, inflammation, and motility of rheumatoid arthritis (RA) fibroblast-like synoviocytes and elucidate the underlying mechanism., Methods: The expression of IGJ RA fibroblast-like synoviocytes was assessed using immunoblot and qPCR. Cell growth was evaluated using CCK-8 and FCM assays. The effects on inflammatory response were determined by ELISA and immunoblot assays. Cell motility was assessed using transwell and immunoblot assays. The mechanism was further confirmed using immunoblot assays., Results: IGJ expression was found to be elevated in fibroid synovial cells of RA. IGJ ablation inhibited the growth of MH7A cells and suppressed the inflammatory response. Knockdown of IGJ also blocked cell motility. Grape white rot, a devastating disease of grapevines caused by Coniella diplodiella (Speg.) Sacc., leads to significant yield losses in grape. Breeding grape cultivars resistant to white rot is essential to reduce the regular use of chemical treatments. In recent years, Chinese grape species have gained more attention for grape breeding due to their high tolerance to various biotic and abiotic factors along with changing climatic conditions. In this study, we employed whole-genome resequencing (WGR) to genotype the parents of 'Manicure Finger' ( Vitis vinifera , female) and '0940' ( Vitis davidii , male), along with 101 F 1 mapping population individuals, thereby constructing a linkage genetic map. The linkage map contained 9337 single-nucleotide polymorphism (SNP) markers with an average marker distance of 0.3 cM. After 3 years of phenotypic evaluation of the progeny for white rot resistance, we confirmed one stable quantitative trait locus (QTL) for white rot resistance on chromosome 3, explaining up to 17.9% of the phenotypic variation. For this locus, we used RNA-seq to detect candidate gene expression and identified PR1 as a candidate gene involved in white rot resistance. Salicylic acid (SA) is a well-studied phenolic plant hormone that plays an important role in plant defense against the hemi-biothrophic and biothrophic pathogens and depends on the living cells of host for the successful infection. In this study, a pathogenesis test was performed between Vitis davidii and V. vinifera cultivars against grape white rot disease ( Coniella diplodiella ). V. davidii was found to be resistant against this disease. SA contents were found to be higher in the resistant grape cultivar after different time points. RNA-seq analysis was conducted on susceptible grapevine cultivars after 12, 24, and 48 h of SA application with the hypothesis that SA may induce defense genes in susceptible cultivars. A total of 511 differentially expressed genes (DEGs) were identified from the RNA-seq data, including some important genes, VvWRKY1/2, VvNPR1 , VvTGA2 , and VvPR1, for the SA defense pathway. DEGs related to phytohormone signal transduction and flavonoid biosynthetic pathways were also upregulated. The quantitative real-time PCR (qRT-PCR) results of the significantly expressed transcripts were found to be consistent with the transcriptome data, with a high correlation between the two analyses. The pathogenesis-related gene 1 ( VvPR1 ), which is an important marker gene for plant defense, was selected for further promoter analysis. The promoter sequence showed that it contains some important cis-elements (W-box, LS7, as-1, and TCA-element) to recruit the transcription factors VvWRKY, VvNPR1 , and VvTGA2 to express the VvPR1 gene in response to SA treatment. Furthermore, the VvPR1 promoter was serially deleted into different fragments (-1,837, -1,443, -1,119, -864, -558, -436, and -192 ) bp and constructed vectors with the GUS reporter gene. Deletion analysis revealed that the VvPR1 promoter between -1837 bp to -558 bp induced significant GUS expression with respect to the control. On the basis of these results, the -558 bp region was assumed to be an important part of the VvPR1 promoter, and this region contained the important cis-elements related to SA, such as TCA-element (-1,472 bp), LS7 (-1,428 bp), and as-1 (-520 bp), that recruit the TFs and induce the expression of the VvPR1 gene. Early cancer diagnosis and treatment are crucial research fields of human health. One method that has proven efficient is biomarker detection which can provide real-time and accurate biological information for early diagnosis. This review presents several biomarker sensors based on electrochemistry, surface plasmon resonance (SPR), nanowires, other nanostructures, and, most recently, metamaterials which have also shown their mechanisms and prospects in application in recent years. Compared with previous reviews, electrochemistry-based biomarker sensors have been classified into three strategies according to their optimizing methods in this review. This makes it more convenient for researchers to find a specific fabrication method to improve the performance of their sensors. Besides that, as microfabrication technologies have improved and novel materials are explored, some novel biomarker sensors-such as nanowire-based and metamaterial-based biomarker sensors-have also been investigated and summarized in this review, which can exhibit ultrahigh resolution, sensitivity, and limit of detection (LoD) in a more complex detection environment. The purpose of this review is to understand the present by reviewing the past. Ras-association domain family protein 6 (RASSF6) is a member of tumor suppressor RASSFs family with a wide range of function from RAS interaction, Hippo signaling involvement to cell cycle and apoptosis regulation. RASSF6 is reported inactivated in various types of cancer. However, whether RASSF6 is associated with colorectal cancer and the underlying mechanisms have yet to be investigated. In our previous exome sequencing study, we found a somatic loss-of-function (LoF) mutation in RASSF6 in one sporadic colorectal cancer (sCRC) patient, and two missense mutations in deep sequencing group of sCRC samples, implying the possibility that RASSF6 may be involved in the pathogenesis of sCRC. In this study, we demonstrate that RASSF6 acts as a tumor suppressor in colon cancer cells. Decreased level of RASSF6 was observed in adenocarcinoma compared to normal tissues, especially in advanced tumor cases. Further experiments showed exogenous introduction of RASSF6 into LoVo cells suppressed cell proliferation, migration, invasion, and induced apoptosis in vitro as well as tumor growth in vivo. In contrast, knockdown of RASSF6 in HT-29 cells showed the opposite effects. Taken together, our results suggest, in addition to epigenetics changes, functional somatic mutations may also contribute to the downregulation of RASSF6 and further participate in the pathogenesis of sCRC. RASSF6 may serve as a novel candidate against tumor growth for sCRC.

Haplotype analysis has been increasingly used to study the genetic basis of human diseases, but models for characterizing genetic interactions between haplotypes from different chromosomal regions have not been well developed in the current literature. In this article, we describe a statistical model for testing haplotype-haplotype interactions for human diseases with a case-control genetic association design. The model is formulated on a contingency table in which cases and controls are typed for the same set of molecular markers. By integrating well-established quantitative genetic principles, the model is equipped with a capacity to characterize physiologically meaningful epistasis arising from interactions between haplotypes from different chromosomal regions. The model allows the partition of epistasis into different components due to additive × additive, additive × dominance, dominance × additive, and dominance × dominance interactions. We derive the EM algorithm to estimate and test the effects of each of these components on differences in the pattern of genetic variation between cases and controls and, therefore, examine their role in the pathogenesis of human diseases. The method was further extended to investigate gene-environment interactions expressed at the haplotype level. The statistical properties of the models were investigated through simulation studies and its usefulness and utilization validated by analyzing the genetic association of sarcoidosis from a human genetics project.
