Fusarium endophytes damage cereal crops and contaminate produce with mycotoxins. Those fungi overcome the main chemical defence of host via detoxification by a malonyl‐CoA‐dependent enzyme homologous to xenobiotic metabolizing arylamine N‐acetyltransferase (NAT). In Fusarium verticillioides (teleomorph Gibberella moniliformis, GIBMO), this N‐malonyltransferase activity is attributed to (GIBMO)NAT1, and the fungus has two additional isoenzymes, (GIBMO)NAT3 (N‐acetyltransferase) and (GIBMO)NAT2 (unknown function). We present the crystallographic structure of (GIBMO)NAT1, also modelling other fungal NAT homologues. Monomeric (GIBMO)NAT1 is distinctive, with access to the catalytic core through two "tunnel‐like" entries separated by a "bridge‐like" helix. In the quaternary arrangement, (GIBMO)NAT1 monomers interact in pairs along an extensive interface whereby one entry of each monomer is covered by the N‐terminus of the other monomer. Although monomeric (GIBMO)NAT1 apparently accommodates acetyl‐CoA better than malonyl‐CoA, dimerization changes the active site to allow malonyl‐CoA to reach the catalytic triad (Cys110, His158 and Asp173) via the single uncovered entry, and anchor its terminal carboxyl‐group via hydrogen bonds to Arg109, Asn157 and Thr261. Lacking a terminal carboxyl‐group, acetyl‐CoA cannot form such stabilizing interactions, while longer acyl‐CoAs enter the active site but cannot reach catalytic Cys. Other NAT isoenzymes lack such structural features, with (GIBMO)NAT3 resembling bacterial NATs and (GIBMO)NAT2 adopting a structure intermediate between (GIBMO)NAT1 and (GIBMO)NAT3. Biochemical assays confirmed differential donor substrate preference of (GIBMO)NAT isoenzymes, with phylogenetic analysis demonstrating evolutionary separation. Given the role of (GIBMO)NAT1 in enhancing Fusarium pathogenicity, unravelling the structure and function of this enzyme may benefit research into more targeted strategies for pathogen control. The current study used a mixed-methods approach to examine how low-income mothers managed their household economies, their experiences of economic pressure, and the consequences for family and child functioning. Qualitative analyses (N = 32 families) revealed that experiences of economic pressure were associated with an inability to afford "both" basic needs and some modest but highly valued "extras." To meet demands, mothers reported using a variety of strategies, including instrumental support from friends and family members and other financial strategies. Results from the quantitative analyses (N = 516 families; 800 children, ages 6-15) were generally consistent with patterns observed in the qualitative analyses and extended the findings to include effects on parenting practices and children's behavioral functioning.

Research on working poor families participating in an anti-poverty initiative in Milwaukee found 60% of families had at least one child with significant problems (learning, school achievement and/or behavior, retardation, or other disabilities). Comparison with other families with children with troubles found the program families adapted less specifically to the troubled child and more to economic problems. (Contains references.) Pragmatic approaches can effectively address questions of cultural dynamics—how culture is renewed, contested, or changed in historical situations. The practice-oriented direction of much of contemporary cognitive science offers an emerging opportunity to more fully integrate thought and emotion into anthropological pragmatism. Here I urge attention to a realm of enacted culture that illustrates how certain interactive processes bring cultural conventions into a communicative environment where they are renewed, transformed, created. As an example, I discuss depictions, improvisational performances in which speakers use material resources such as their hands or voices to formulate and portray thoughts. In a close analysis of three short narratives, I show that depictions are used by speakers to help to create an affect-rich environment in which cultural conventions are introduced into interactional space, whence they become subject to negotiation. It is not only in these spontaneous and improvisational practices that cognition and culture unfold in time as a dialectical interplay of mind and environment; this also occurs in stylized formats such as public rituals and narratives.
