ObjectivesWe aimed to assess the characteristics of adverse drug reactions (ADRs) collected in a university hospital.MethodsA retrospective analysis of ADRs spontaneously reported in the Hospital Pharmacovigilance Program database (RutiRAM) over a 13-year period was conducted. The analysis included a description of ADRs [System Organ Class (SOC)] and their seriousness, the drugs involved [level 1 of the Anatomical Therapeutic Chemical (ATC) Classification System], drug-drug interactions, medication errors, drugs ‘under additional monitoring’, positive rechallenge, and the ‘pharmacovigilance interest’ of ADRs. An ADR was considered of ‘pharmacovigilance interest’ when it was serious, and/or produced sequelae, and/or affected the paediatric population, and/or when the suspected drug was ‘under additional monitoring’. Additionally, an exploratory analysis for bivariate associations through an automated method was performed.ResultsA total of 2,148 spontaneous ADRs were registered in the RutiRAM database, with 92.5% recorded by medical doctors. The mean age of cases was 59.2 years (SD 20.9), range 1 day–99 years; 5.7% were paediatric, 46.2% adults, and 48.1% elderly. The drugs most often involved were anti-infectives (ATC group J), mainly amoxicillin-clavulanic acid. ‘Blood system disorders’ were the most frequent SOC ADRs, and skin rashes were the most frequent ADRs. The 63.2% of ADRs were considered of ‘pharmacovigilance interest’. Almost half of ADRs were hospital-acquired, and these were related to medication error; serious ADRs were related to drug-drug interactions and elderly patients, and involved drugs ‘under additional monitoring’ were related to younger ones.ConclusionThis is the first study to overview of ADRs reported in an HPVP over more than a decade. Almost two-thirds of the ADRs collected in the RutiRAM database are of sufficient quality to be classified as ‘pharmacovigilance interest’, and thus can contribute to signal detection and the issuing of drug alerts by pharmacovigilance systems. Analysing ADRs in hospitals contributes to patient safety by implementing relevant actions to prevent medication errors or ADRs, some of which can be applied to other centres.

Abstract There is a high interest on gut health in poultry with special focus on consequences of the intestinal diseases, such as coccidiosis and C. perfringens-induced necrotic enteritis (NE). We developed a custom gene expression panel, which could provide a snapshot of gene expression variation under challenging conditions. Ileum gene expression studies were performed through high throughput reverse transcription quantitative real-time polymerase chain reaction. A deep review on the bibliography was done and genes related to intestinal health were selected for barrier function, immune response, oxidation, digestive hormones, nutrient transport, and metabolism. The panel was firstly tested by using a nutritional/Clostridium perfringens model of intestinal barrier failure (induced using commercial reused litter and wheat-based diets without exogenous supplementation of enzymes) and the consistency of results was evaluated by another experiment under a coccidiosis challenge (orally gavaged with a commercial coccidiosis vaccine, 90× vaccine dose). Growth traits and intestinal morphological analysis were performed to check the gut barrier failure occurrence. Results of ileum gene expression showed a higher expression in genes involved in barrier function and nutrient transport in chickens raised in healthy conditions, while genes involved in immune response presented higher expression in C.perfringens-challenged birds. On the other hand, the Eimeria challenge also altered the expression of genes related to barrier function and metabolism, and increased the expression of genes related to immune response and oxidative stress. The panel developed in the current study gives us an overview of genes and pathways involved in broiler response to pathogen challenge. It also allows us to deep into the study of differences in gene expression pattern and magnitude of responses under either a coccidial vaccine or a NE.

Among natural freshwater pollutants, cyanotoxins, mycotoxins, and phytotoxins are the most important and less studied. Their identification in surface water is challenging especially cause of the lack of standards and established analytical parameters. Most target methods focus one or a single group of compounds with similar characteristics. Here we present an AIF fast method for the tentative identification of natural toxins in water. Respect to the previous method [1], it offers higher performances for the acquisition of unknown compounds at low levels for higher number of analytes.The key aspects of the method are:• The qualitative screening DIA-AIF workflow using Q Exactive Orbitrap. Both targeted and suspect screening bases have been combined with online databases and suspect list to retrieve candidates as suspect natural toxins and their metabolites or degradation products.• The in-silico analysis of mass spectrums allowed a fast structural characterization.• The workflow has been finally applied to real samples coming from the Czech Republic, Italy, and Spain allowing the determination of 17 suspect natural toxins, 4 of them confirmed. None toxin passed the limit of 1 µg/L taken from the legislation applied for microcystin LR and arbitrarily extended to all toxins.

However, this method has several limitations, such as the passive role assumed by students and the loss of their attention. In order to improve the use of lectures, some authors have suggested the use of minicases. These are short texts that include several questions, which are related with the contents of the lecture and students are asked to answer them at its end. The present paper describes an experience of using minicases in the teaching of pharmacology. It offers ten examples to illustrate how they are built. Most of them are related with the real world, they permit to put knowledge of basic sciences in context of clinical problems and stimulate the understanding of key concepts of the discipline.

General elections and regional head elections are vital expressions of popular sovereignty and democratic representation within the framework of the rule of law. These elections underscore that sovereignty and power reside with the people. However, the electoral process often leads to disputes, which can arise from administrative issues, criminal activities, and the results of vote counting. Consequently, an effective election dispute resolution mechanism is essential, emphasizing the principle of equality before the law for all parties involved, to ensure the integrity of the electoral process. This research employs normative methods to examine the laws and regulations governing regional head elections. It analyzes relevant data and case examples in relation to the principle of equality before the law in both legislative and regional head elections. The novelty of this research lies in its exploration of the equal standing of parties before the law, particularly in the context of disputes arising during regional head elections.

Soil nitrogen isotopic composition is used to drive the IsoTONE model, which is constrained with measurements of tropospheric nitrous oxide isotopic composition. The model results reveal causes of rising mean global nitrous oxide emission factor.

We apply methods of the so-called\inverse problem of the calculus of variations" to the stabilization of an equilibrium of a class of two-dimensional controlled mechanical systems. The class is general enough to include, among others, the inverted pendulum on a cart and the inertia wheel pendulum. By making use of a condition that follows from Douglas' classification, we derive feedback controls for which the control system is variational. Devising agricultural management schemes that enhance food security and soil carbon levels is a high priority for many nations. However, the coupling between agricultural productivity, soil carbon stocks and organic matter turnover rates is still unclear. Archived soil samples from four decades of a long-term crop rotation trial were analyzed for soil organic matter (SOM) cycling-relevant properties: C and N content, bulk composition by nuclear magnetic resonance (NMR) spectroscopy, amino sugar content, short-term C bioavailability assays, and long-term C turnover rates by modeling the incorporation of the bomb spike in atmospheric 14C into the soil. After > 40 years under consistent management, topsoil carbon stocks ranged from 14 to 33 Mg C ha−1 and were linearly related to the mean productivity of each treatment. Measurements of SOM composition demonstrated increasing amounts of plant- and microbially derived SOM along the productivity gradient. Under two modeling scenarios, radiocarbon data indicated overall SOM turnover time decreased from 40 to 13 years with increasing productivity – twice the rate of decline predicted from simple steady-state models or static three-pool decay rates of measured C pool distributions. Similarly, the half-life of synthetic root exudates decreased from 30.4 to 21.5 h with increasing productivity, indicating accelerated microbial activity. These findings suggest that there is a direct feedback between accelerated biological activity, carbon cycling rates and rates of carbon stabilization with important implications for how SOM dynamics are represented in models.

In this article, the full description of a heart failure with reduced ejection fraction (HF_REF) cohort of 192 patients is provided. Tables with the baseline demographic, prior history, ECG parameters, echocardiographic parameters, laboratory values and pharmacological treatment of these patients are included. Also, the quartile values of the analyzed circulating biomarkers: high sensitivity Troponin T (hs-TnT), galectin-3 (Gal-3), C-terminal propeptide of type I procollagen (CICP), soluble AXL (sAXL) and Brain Natriuretic Peptide (BNP) are given. The main demographic and clinical features of the patients’ subgroups that have hs-TnT, Gal-3, CICP or BNP above the third quartile are described. Tables with Pearson correlation analysis of the HF_REF patients’ biomarker levels are included. And Pearson correlation analysis of the HF_REF patients’ hs-TnT, Gal-3, CICP levels with patients’ biochemical parameters, blood count and inflammation parameters are also described. These data are related to the research articles (AXL receptor tyrosine kinase is increased in patients with heart failure (M. Batlle, P. Recarte-Pelz, E. Roig, M.A. Castel, M. Cardona, M. Farrero, et al., 2014) [1] and Use of serum levels of high sensitivity troponin T, galectin-3 and C-terminal propeptide of type I procollagen at long term follow-up in Heart Failure patients with reduced ejection fraction: comparison with soluble AXL and BNP (M. Batlle, B. Campos, M. Farrero, M. Cardona, B. González, M.A. Castel, et al., 2016) [2].

α-Synuclein is present in Lewy bodies of patients with both sporadic and familial Parkinson's disease. However, pathogenic mutations Ala30Pro and Ala53Thr in α-synuclein are rare causes of disease. Synphilin-1 has been demonstrated to associate with α-synuclein and promote the formation of cytosolic inclusions in vitro. Two-point genetic linkage analysis of a dinucleotide repeat within the synphilin-1 gene initially implicated this locus as a cause of Parkinson's disease in three of nine families. However, subsequent haplotype, sequencing, and association analyses in these three families and an independent case-control series suggest that variability within the locus does not confer susceptibility to Parkinson's disease.

Abstract The circadian clock is an internal molecular oscillator and coordinates numerous physiological processes through regulation of molecular pathways. Tissue‐specific clocks connected by mobile signals have previously been found to run at different speeds in Arabidopsis thaliana tissues. However, tissue variation in circadian clocks in crop species is unknown. In this study, leaf and tuber global gene expression in cultivated potato under cycling and constant environmental conditions was profiled. In addition, we used a circadian‐regulated luciferase reporter construct to study tuber gene expression rhythms. Diel and circadian expression patterns were present among 17.9% and 5.6% of the expressed genes in the tuber. Over 500 genes displayed differential tissue specific diel phases. Intriguingly, few core circadian clock genes had circadian expression patterns, while all such genes were circadian rhythmic in cultivated tomato leaves. Furthermore, robust diel and circadian transcriptional rhythms were observed among detached tubers. Our results suggest alternative regulatory mechanisms and/or clock composition is present in potato, as well as the presence of tissue‐specific independent circadian clocks. We have provided the first evidence of a functional circadian clock in below‐ground storage organs, holding important implications for other storage root and tuberous crops.

Nowadays, synthetic cathinones (SCs) is the second more representative subclass of New Psychoactive Substances, accounting for 104 analogues in the illegal market. Since its first report in 2011, α-pyrrolidinovalerophenone (α-PVP) gained popularity among drug users, provoking an increased number of intoxications. Nonetheless, pharmacokinetics data is still limited in the literature. An observational non-controlled naturalistic study on 8 healthy volunteers was conducted to assess the α-PVP and β-OH-α-PVP concentrations in OF and urine, after snorting 10 mg or 20 mg of α-PVP. A multi-analytical approach based on GC-EI-MS/MS and LC-HESI-HRMS/MS was developed and fully validated for the analytes quantification, while four untargeted LC-HESI-HRMS/MS methods in full-MS and ddMS 2 were set up for unknown metabolites characterization in urine samples assisted by a dedicated data mining software. In OF, α-PVP reached a mean C max of 762 ± 323 ng/mL at 1 h after 10 mg administration, while a C max of 2,900 ± 1,373 ng/mL at 47 min after 20 mg dose. In urine, a total α-PVP mean amount of 179.2 ± 94.9 µg was accumulated after 10 mg dose, (27.2 ± 9.8 µg between 0-2 h and 152.0 ± 98.2 µg between 2-5 h), while a total amount of 122.9 ± 44.0 µg, of (36.2 ± 16.5 and 86.7 ± 28.3 µg between 0-2 and 2-5 h, respectively) was detected after 20 mg dose. Among the 10 identified metabolites, β-OH-α-PVP was a minor metabolite (total amount: 56.4 ± 27.1 and 69.1 ± 38.1 µg after 10 mg and 20 mg). The N-butanoic acid metabolite was the most abundant, detected also as glucuronide. In conclusion, α-PVP showed a later time peak than non-pyrrolidine SCs, with comparable C max. Background: Psychotherapy for Prolonged Grief Disorder (PGD), a condition characterized by an intense and persistent grief response, has received increased attention over the past decades. Evidence-based approaches to prevent PGD are currently scarce, and not always effective. This paper introduces a protocol for a clinical trial exploring the effectiveness of a Meaning Reconstruction psychotherapy approach (MR) assisted with ayahuasca, a traditional indigenous medicine., Method: The outlined protocol is a three-arm, non-randomized controlled trial focused on reducing normal and pathological grief symptoms, comparing the effectiveness of Ayahuasca-assisted MR therapy (A-MR), MR therapy alone (MR) and No Treatment (NT). At least 69 people who lost a first-degree relative during the prior year, and with a Texas Revised Inventory of Grief score up 39 (TRIG ≥ 40), will participate in the trial. Participants will be allocated to an A-MR (n ≥ 23), MR (n ≥ 23) or NT (n ≥ 23) group. Those from the A-MR and MR therapy groups will undergo a therapeutic process involving 9 sessions of online psychotherapy. In addition, the A-MR condition involves 2 group sessions of ayahuasca. The primary outcomes will be normal and pathological grief severity as measured by the TRIG and Traumatic Grief Inventory Self-Report (TGI-SR), administered at baseline, post-treatment, and 3-month follow up. Measures of quality of life, post-traumatic growth, meaning-made, psychological flexibility, and self-belief consistency will be also included. In addition, subjective effects of ayahuasca and acceptance-avoidance promoting effects will be assessed following ayahuasca administration. Tripartite ATP-independent periplasmic (TRAP) transporters are widespread in prokaryotes and are responsible for the transport of a variety of different ligands, primarily organic acids. TRAP transporters can be divided into two subclasses; DctP-type and TAXI type, which share the same overall architecture and substrate-binding protein requirement. DctP-type transporters are very well studied and have been shown to transport a range of compounds including dicarboxylates, keto acids, and sugar acids. However, TAXI-type transporters are relatively poorly understood. To address this gap in our understanding, we have structurally and biochemically characterized VC0430 from Vibrio cholerae. We show it is a monomeric, high affinity glutamate-binding protein, which we thus rename VcGluP. VcGluP is stereoselective, binding the L-isomer preferentially, and can also bind L-glutamine and L-pyroglutamate with lower affinity. Structural characterization of ligand-bound VcGluP revealed details of its binding site and biophysical characterization of binding site mutants revealed the substrate binding determinants, which differ substantially from those of DctP-type TRAPs. Finally, we have analyzed the interaction between VcGluP and its cognate membrane component, VcGluQM (formerly VC0429) in silico, revealing an architecture hitherto unseen. To our knowledge, this is the first transporter in V. cholerae to be identified as specific to glutamate, which plays a key role in the osmoadaptation of V. cholerae, making this transporter a potential therapeutic target., (© 2024 Davies et al.)

Studying when and where gross genomic rearrangements occurred during evolution is key to understanding changes in genome structure with functional consequences that might eventually lead to speciation. Here we identified chromosome rearrangements in ruminants, a clade characterized by large chromosome differences. Using 26 genome assemblies, we reconstructed five ancestral karyotypes and classified the rearrangement events occurring in each lineage. With these reconstructions, we then identified evolutionary breakpoints regions (EBRs) and synteny fragments. Ruminant karyotype evolution is characterized by inversions, while interchromosomal rearrangements occurred preferentially in the oldest ancestor of ruminants. We found that EBRs are depleted of protein coding genes, including housekeeping genes. Similarly, EBRs are not enriched in high GC regions, suggesting that meiotic double strand breaks might not be their origin. Overall, our results characterize at fine detail the location of chromosome rearrangements in ruminant evolution and provide new insights into the formation of EBRs., (© 2023 The Authors. Introduction: Genetic variations impact drug response, driving the need for personalised medicine through pre-emptive pharmacogenetic testing. However, the adoption of pre-emptive pharmacogenetic testing for commonly prescribed drugs, such as statins, outside of tertiary hospitals is limited due to a lack of pharmacoeconomic evidence to support widespread implementation by healthcare policy-makers. The Spanish Consortium for the Implementation of Pharmacogenetics (iPHARMGx Consortium) addresses this by developing a clinical trial master protocol that will govern multiple nested adaptive clinical trials that compare genotype-guided treatments to standard care in specific drug-gene-population triads, asses their cost-efficacy and identify novel biomarkers through advanced sequencing techniques. The first of these studies aims to assess whether a pre-emptive statin therapy genotyping scheme reduces the incidence of statin-associated muscle symptoms (SAMS) in a population at risk of cardiovascular disease susceptible of receiving high-intensity or moderate-intensity doses of statins: The PREVESTATGx trial., Methods and Analysis: the PREVESTATGX trial is a multicentre, adaptive randomised controlled pragmatic phase IV clinical trial nested to the iPHARMGx master protocol with two parallel arms, aiming for superiority. Randomisation will be conducted on an individual basis with a centralised approach and stratification by centre. After inclusion in the trial and genotyping has been performed, subjects will be randomly allocated to experimental group (pharmacogenetic genotype-guided statin prescription) or standard-of-care statin prescription (as deemed by attending physician). The main objective is to assess the efficacy of a statin pre-emptive genotyping strategy in reducing the incidence of SAMS. A total of 225 subjects will be recruited among the 10 participating centres if no futility/efficacy boundary is reached in the prespecified interim analyses. Recruitment will be carried out during a 12-month period and subjects will be followed for a 9-month period., Ethics and Dissemination: The PREVESTATGx trial received ethical approval on 24 April 2024. The use of "crumb rubber" coming from recycling materials in outdoor floors like playgrounds has been a frequent practice during the last years. However, these surfaces are object of abrasion and weathering being a potential source of micro and nanoplastics (MNPLs) to the atmosphere and a potential source of human exposure to them. Our main goal has been to expose different crumb rubber materials to summer weathering effects. The released inhalable fractions were sampled for two months with passive samplers and the composition of MNPLs and plastic additives (organic and inorganic) were evaluated. The ecotoxicological effects of leached materials emulating runoff events was evaluated in freshwater micro crustacean Daphnia magna and the green algae Chlorella vulgaris. The analysis of MNPLs showed the presence of polyethylene, polypropylene, polybutadiene, polysiloxanes and polybutylene at concentrations up to 30,426 ng/m 3 . In the same fraction, we also identified up to 56 plastic additives, including antioxidants, pigments, copolymers, flame retardants, fungicides, lubricants, plasticizers, UV filters and metal ions. Climate change impacts the Mediterranean region, transforming it from region with a semi-arid climate to a region with an arid climate. Under this situation, while groundwater is an essential hydric resource, its existence is in danger due to anthropogenic pressures. Persistent mobile organic compounds (PMOCs) have recently been recognised as an emerging problem; however, PMOCs in groundwater need to be better characterised. Here, we present a new analytical method to characterise the profile of PMOCs in groundwater based on two parallel solid-phase extraction (SPE), using weak anion exchange and weak cation exchange. Extracts were analysed by ultraperformance liquid chromatography (UPLC) using mix-mode chromatography for those compounds analysed under negative ionisation conditions and hydrophilic interaction liquid chromatography (HILIC) under positive conditions coupled to high-resolution mass-spectrometry (HRMS) using a Q-Exactive Orbitrap™ analyser. For the suspect screening of PMOCs in groundwater, the acquisition mode was in full scan (FS) by "independent scan of all ion fragmentation". For the tentative identification, different online databases such as Environmental and Food Safety (EFS) HRAM Compound database, PFAS NIST database, ChemSpider for chemical structural information, MzCloud as a mass spectral database, and an in-house list with 1280 PMOC structures have been used. The performance of the method was assessed with 29 representative PMOCs which were selected based on the previous literature. The recovery rates have been between 63 and 110 % for 90 % of the target compounds and method limits of quantification (MLQ) between 0.3 and 10.5 ng/L. The optimised approach was applied to assess PMOCs in the Besòs River aquifer, NE Spain, showing 148 tentatively identified structures at confidence levels 1-3. Among them, 66 suspects were tentatively identified at level 3, 54 at level 2, and 28 confirmed at level 1. Most of these compounds were polar and highly polar compounds which are difficult to retain with other extraction approaches. Major detected compounds were pharmaceuticals and personal care products (46), followed by perfluoroalkyl and polyfluoroalkyl substances (PFAS) (32), industrial additives (27), and pesticides (23), among other groups. Urban aquifers are at risk of contamination from persistent and mobile organic compounds (PMOCs), especially per- and polyfluoroalkyl substances (PFAS), which are artificial organic substances widely used across various industrial sectors. PFAS are considered toxic, mobile and persistent, and have therefore gained significant attention in environmental chemistry. Moreover, precursors could transform into more recalcitrant products under natural conditions. However, there is limited information about the processes which affect their behaviour in groundwater at the field-scale. In this context, the aim of this study is to assess the presence of PFAS in an urban aquifer in Barcelona, and identify processes that control their evolution along the groundwater flow. 21 groundwater and 6 river samples were collected revealing the presence of 16 PFAS products and 3 novel PFAS. Short and ultra-short chain PFAS were found to be ubiquitous, with the highest concentrations detected for perfluorobutanesulfonic acid (PFBS), trifluoroacetic acid (TFA) and trifluoromethanesulfonic acid (TFSA). Long chain PFAS and novel PFAS were found to be present in very low concentrations (<50 ng/L). It was observed that redox conditions influence the behaviour of a number of PFAS controlling their attenuation or recalcitrant behaviour. Most substances showed accumulation, possibly explained by sorption/desorption processes or transformation processes, highlighting the challenges associated with PFAS remediation. In addition, the removal processes of different intensities for three PFAS were revealed. In this study we report on efforts to develop an enantioselective method for the detection of the drug of abuse clephedrone (1-(4-chlorophenyl)-2-(methylamino)-1-propanone (4-chloromethcathinone, also known as 4-CMC or para-chloro-methcathinone)) and its phase-1 metabolites in human biological fluids. The major goal is not to only report results, but primarily to emphasize the various challenges encountered when developing a reliable analytical method for the detection and quantification of novel psychoactive substances (NPS) and their metabolites in the matrix of interest. Such challenges start with the lack of chemical stability of some NPS in biological matrices. Additionally, most often metabolites are unavailable in pure form to serve as analytical standards, just as deuterated standards for native drugs and metabolites are frequently not commercially available. Furthermore, if the NPS is chiral, enantiomerically pure standards with known absolute stereochemistry are required, as well as a stereochemical stability of a drug and its metabolites becomes an issue. In addition, the chirality of a NPS significantly increases the number of species to be detected in the sample and thus challenges the development of an adequate separation method. Microplastics (MPLs) are contaminants of emerging concern (CECs) ubiquitous in aquatic environments, which can be bioaccumulated along the food chain. In this study, the accumulation of polyethylene (PE), polystyrene (PS) and polyethylene terephthalate (PET) microplastics (MPLs) of sizes below 63 μm was assessed in Mediterranean mussels (Mytilus galloprovincialis spp). Moreover, the potential of mussels to uptake and bioaccumulate other organic contaminants, such as triclosan (TCS) and per- and polyfluoroalkyl substances (PFASs), was evaluated with and without the presence of MPLs. Then, the modulation of MPLs in the human bioaccessibility of co-contaminants was assessed by in vitro assays that simulated the human digestion process. Exposure experiments were carried out in 15 L marine microcosms. The bioaccumulation and bioaccessibility of PE, PS, PET, and co-contaminants were assessed by means of liquid chromatography -size exclusion chromatography-coupled to high-resolution mass spectrometry (LC(SEC)-HRMS). Our outcomes confirm that MPL bioaccumulation in filter-feeding organisms is a function of MPL chemical composition and particle sizes. Finally, despite the lower accumulation and bioaccumulation of PFASs in the presence of MPLs, the bioaccessibility assays revealed that PFASs bioaccessibility was favoured in the presence of MPLs. Substance use disorders (SUD), also named addiction when it is severe, is a chronic brain disorder with serious impact on individual who suffer, the public health and with high burden of disease. They are multitude of mechanisms/factors involved in addiction: from individual characteristics of the person (from genetic to impacts of stress, sex, and age) to social and environmental situation (availability and accessibility of substances, cultural and legal aspects, socio-economical situation) and type of substance of use (pharmacological characteristics) Then, research on Addiction must include different, complementary, and translational perspectives. In this review, we explore the neurobiological, psychosocial, and epidemiological knowledge of substance addiction, and the main role played by pharmacology in the research in this field. In Spain, since 2002, collaborative networks have emerged for comprehensive research on addictions, with the creation of the Addictive Disorders Network (RTA), currently redefined as the Research Network for Primary Care in Addictions (RIAPAd) with the support of the Carlos III Health Institute (Instituto de Salud Carlos III). Background: Management of syphilis, a sexually transmitted infection (STI) with increasing incidence, is challenged by drug shortages, scarcity of randomised trial data, an absence of non-penicillin alternatives for pregnant women with penicillin allergy (other than desensitisation), extended parenteral administration for neurosyphilis and congenital syphilis, and macrolide resistance. Linezolid was shown to be active against Treponema pallidum, the causative agent of syphilis, in vitro and in the rabbit model. We aimed to assess the efficacy of linezolid for treating early syphilis in adults compared with the standard of care benzathine penicillin G (BPG)., Methods: We did a multicentre, open-label, non-inferiority, randomised controlled trial to assess the efficacy of linezolid for treating early syphilis compared with BPG. We recruited participants with serological or molecular confirmation of syphilis (either primary, secondary, or early latent) at one STI unit in a public hospital and two STI community clinics in Catalonia (Spain). Participants were randomly allocated in a 1:1 ratio using a computer-generated block randomisation list with six participants per block, to receive either oral linezolid (600 mg once per day for 5 days) or intramuscular BPG (single dose of 2·4 million international units) and were assessed for signs and symptoms (once per week until week 6 and at week 12, week 24, and week 48) and reagin titres of non-treponemal antibodies (week 12, week 24, and week 48). The primary endpoint was treatment response, assessed using a composite endpoint that included clinical response, serological response, and absence of relapse. Clinical response was assessed at 2 weeks for primary syphilis and at 6 weeks for secondary syphilis following treatment initiation. Serological cure was defined as a four-fold decline in rapid plasma reagin titre or seroreversion at any of the 12-week, 24-week, or 48-week timepoints. The absence of relapse was defined as the presence of different molecular sequence types of T pallidum in recurrent syphilis. Non-inferiority was shown if the lower limit of the two-sided 95% CI for the difference in rates of treatment response was higher than -10%. The primary analysis was done in the per-protocol population. The trial is registered at ClinicalTrials.gov (NCT05069974) and was stopped for futility after interim analysis., Findings: Between Oct 20, 2021, and Sept 15, 2022, 62 patients were assessed for eligibility, and 59 were randomly assigned to linezolid (n=29) or BPG (n=30). In the per-protocol population, after 48 weeks' follow-up, 19 (70%) of 27 participants (95% CI 49·8 to 86·2) in the linezolid group had responded to treatment and 28 (100%) of 28 participants (87·7 to 100·0) in the BPG group (treatment difference -29·6, 95% CI -50·5 to -8·8), which did not meet the non-inferiority criterion. The number of drug-related adverse events (all mild or moderate) was similar in both treatment groups (five [17%] of 29, 95% CI 5·8 to 35·8 in the linezolid group vs five [17%] of 30, 5·6 to 34·7, in the BPG group). This study investigated the effects of xylanase and stimbiotic (fiber fermentation enhancer) on the response of broiler chickens fed different corn varieties and determine correlations between variables of interest. Four corn genetic varieties were selected based on their range in nutrient composition. Diets containing 600 g/kg of each corn were supplemented with 0 or 100 g/ton of xylanase or stimbiotic. A total of 1,152 one-day-old male broiler chicks (Ross 308) were divided into 12 treatments, each with 8 pens and 12 birds per pen, for a 21-day study. On d 21, performance parameters were measured, and the ileal energy and organic matter (OM) digestibility and cecal xylanase activity determined. Stimbiotic supplementation improved mFCR compared with all other treatments. There was a treatment by corn variety interaction for body weight (BW), BW gain and coefficient of variation (CV) of BW (P ≤ 0.05). Birds fed corn Variety 1 (highest neutral dietary fiber, protein and soluble arabinoxylan content) supplemented with stimbiotic had the highest BW, while Variety 2 control diet had the lowest. The BW CV in corn Variety 2 was the highest, which improved with stimbiotic supplementation. The BW CV in corn Variety 1 responded better to stimbiotic than xylanase. There were no treatment differences on BW CV in corn Varieties 3 and 4. The lowest OM digestibility was observed in birds fed corn Variety 1 with xylanase, and the highest value was associated with corn Variety 3 with xylanase (highest total arabinoxylan). Xylanase and stimbiotic supplementation increased the endogenous xylanase activity regardless of the corn variety (P ≤ 0.05). Positive correlations between corn fiber contents and phytic acid and the arabinose:xylose ratio were seen, while nonstarch polysaccharide content was negatively correlated with apparent metabolizable energy. In conclusion, corn variety influenced nutrient digestibility and broiler chicken growth. The increasing consumption of pharmaceuticals, alongside their limited removal in wastewater treatment plants (WWTPs), have led to their ubiquitous occurrence in receiving aquatic environments. This study addresses the occurrence of 68 pharmaceuticals (PhACs) in the Ebro River Delta region (NE Spain), as well as their distribution in different environmental compartments, including surface water, sediments, biota (river biofilm and fish tissues), and field-collected plastic litter. In addition, their concentrations in serving WWTPs, as possible sources of environmental contamination, were also determined. Our study confirmed the widespread occurrence of PhACs in riverine and, to a more limited extent, coastal environments. Most frequently detected PhACs belonged to analgesics/anti-inflammatories (e.g., ibuprofen) and psychiatric drugs (e.g., venlafaxine) therapeutic groups, followed by antihypertensives (e.g., valsartan) and antibiotics (e.g., azithromycin). Seasonal differences in cumulative levels of PhACs were reported for water and sediments (winter>summer). Despite spatial gradients were not clear along the river, a non-negligible contribution of upstream Ebro sites (reference area) was highlighted, which was unexpected based on the low anthropogenic pressure. Sediments represented a minor attenuation pathway for the selected PhACs, whereas they were more heavily accumulated in biota: fish liver (up to 166 ng/g dw), river biofilms (up to 108 ng/g dw), fish plasma (up to 63 ng/mL), and fish muscle (up to 31 ng/g dw). These findings highlight the importance of biomonitoring in the characterization of polluted areas and prioritization of hazardous substances (e.g., psychiatric drugs) in aquatic systems, and a particular interest of fish plasma as non-destructive biomonitoring matrix. PhACs were also detected on plastic litter, demonstrating their role as environmental sinks for certain PhACs (e.g., analgesics/anti-inflammatories, psychiatric drugs). Despite potential health implications, data on the presence of Glyphosate (GLY) and other non-GLY herbicides in human matrices remain scarce. This study aimed to develop a simple and cost-effective methodology for detecting and quantifying GLY, its primary biodegradation product; aminomethylphosphonic acid (AMPA); and glufosinate (GLU) in plasma and urine of environmentally and occupationally exposed populations from the province of Córdoba (Argentina). Different alternatives of pre-treatment, derivatization with FMOC-Cl, solid phase extraction, and final sample conditioning steps were evaluated to improve the quantification of the herbicides by a high-performance liquid chromatography system coupled to a triple-quadrupole mass spectrometer. Recoveries ranged from 39 to 84% in both matrices, while limits of quantification were 3, 1, and 0.3 ng/mL and 3.6, 5.1, and 0.3 ng/mL for AMPA, GLY, and GLU in plasma and urine, respectively. In plasma samples, GLY was the most frequently detected analyte (32%), followed by GLU (10%). In urine samples, GLU was the most frequently detected herbicide (13%), followed by GLY (6%). No differences between group or matrix correlations were found. This study is the first report of GLU in human biological matrices and should be used to establish baseline values for future surveillance systems.

Human exposure to micro (nano)plastics (MNPLs) has become a significant concern as a potential health threat. Exposure routes include ingestion, inhalation, and dermal contact, being food and drinking water the primary sources of oral exposure. Here we present the quantification of polymers of MNPLs particles from 700 nm to 20 μm in bottled water commercialised in Spain, including an estimation of the potential risk for daily consumers. We evaluated samples from 20 popular brands in 0.5 and 1.5 L plastic bottles. A double-suspect screening approach developed and validated in our research group for drinking water was adapted for bottled water samples. The identification and quantification of MNPLs-polymers in mass units and the tentative identification of plastic additives (PA) until the second level of confidence was carried out based on high-performance liquid chromatography coupled to high-resolution mass spectrometry (HPLC-HRMS). The results showed the presence of polypropylene (PP), polyethylene (PE) and polypropylene terephthalate (PET) in the samples. Among them, PE was the most frequently detected and quantified polymer (55% of samples) followed by PET which was detected in 33% of the samples and showing the highest concentration (4700 ng L -1 ). The median value of the sum of polymer concentrations was 359 ng L -1 . In addition, 28 plastic additives were detected, where at least one of them was present in 100% of the samples. Stabilizers and plasticisers were the most frequently identified. A prioritisation study was performed using a multi-QSAR modelling software, where bis(2-ethylhexyl) adipate and bis(2-ethylhexyl) phthalate were estimated as the most potentially harmful compounds for human health. Motivation: The rapid increase in the number of chromosome-scale genome assemblies has renewed interest in chromosome evolution studies. The visualization of syntenic relationships between genomes is a crucial initial step in the study of chromosome rearrangements and evolution. There are few tools available that serve this purpose, and they can be difficult to learn. Moreover, these tools are limited in the number of species comparisons that can be visualized and the size of chromosome rearrangements identified. Thus, the development of novel visualization tools is in strong need., Results: Here, we present syntenyPlotteR, an R package developed to visualize homologous synteny blocks in a pairwise or multispecies manner. Bioplastics arise as an alternative to plastic production delinked from fossil resources. However, as their demand is increasing, there is a need to investigate their environmental fingerprint. Here we study the toxicity of microplastics (MPLs) of two widely used materials, the polylactic acid (PLA) and the polyhydroxybutyrate (PHB) on the environmental aquatic model species Daphnia magna. The study was focused on sublethal behavioural and feeding endpoints linked to antipredator scape responses and food intake. The study aimed to test that MPLs from single-use household comercial items and among them bioplastics should be more toxic than those obtained from standard plastic polymers and fossil plastic materials due to the greater amount of plastic additives, and that MPLs should be more toxic than plastic extracts due to the contribution of both particle and plastic additive toxicity. MPLs were obtained by cryogenic grinding and sea-sand erosion to obtain irregular particles. MPL included standard polymers and nine comercial items of PLA and PHB and one fossil-based material of high-density polyethylene (HDPE). The additive content in commercial items was characterised by liquid chromatography coupled with high-resolution mass spectrometry. D. magna juveniles were exposed for 24 h to particles and their plastic extracts. Results indicated that the toxicity of bioplastic particles was five times higher than the effects produced by exposure to the content of the additives alone, that bioplastic particles were more toxic than fossil ones and that particles obtained from commercial items were more toxic than those obtained from PLA, PHB or HDPE polymer standards. Predicted toxicity from the measured plastic additives in the studied commercially available household items, however, was poorly related with the observed behavioural and feeding effects.
