ABSTRACT: Besides health monitoring, a regular check of dairy heifers' growth rate is desirable, but it is rarely done because procedures that require restraint and handling can be associated with substantial stress for both animals and farmers. Inexperienced heifers, especially if they are highly responsive to humans, may find restraint and handling potentially aversive. This study investigated whether training heifers of different age and responsiveness toward humans (RTH), through operant conditioning, could reduce stress in animals, ease close contact and handling, and be feasible in terms of farmers' effort. We assessed 60 Holstein heifers of 2 age classes (young, n = 29, 291 ± 39 d; old, n = 31, 346 ± 62 d) according to the avoidance distance test and classified them as confident (n = 20), neutral (n = 21), or nonconfident (n = 19). Half of the heifers of each age and RTH class were trained (n = 29), whereas the other half was not (n = 31). The trained heifers were subjected to target training for 8 sessions and positively reinforced with feed to allow being touched on the muzzle, rump, and perineum. If a heifer refused positive reinforcement, the trainer stepped back as negative reinforcement. In the last week of the experiment, the effect of training on the reaction to handling was assessed in all heifers. We measured heart rate, root mean square of successive interbeat interval differences (RMSSD), and fecal cortisol metabolites (FCMet). The presence of behavioral distress signs was recorded as well. The avoidance distance test was performed a second time 24 h after the measuring session. All of the trained heifers, regardless of RTH class, successfully accomplished the target training task in 6 sessions, each spending on average 25.3 s per session. All of the trained heifers allowed touches on the rump and perineum at the end of the fourth session. Training nonconfident heifers required more time compared with the others. Trained heifers showed higher RMSSD than nontrained heifers (14.2 vs. 16.9 ms, respectively), indicating a lower vagal tone, and thus, a slightly lower stress level than nontrained heifers. Training did not lead to differences in HR, FCMet, or presence of stress behavioral signs. Nonconfident heifers had the highest mean baseline FCMet values compared with neutral and confident heifers (38.4 vs. 30.3 vs. 29.1 ng/g, respectively). Nonconfident heifers also showed the lowest value of FCMet 12 h after the measuring session (36.7 vs. 44.6 vs. 49.7 ng/g), likely due to a decreased responsiveness of the adrenal gland to a stressor. The average avoidance distance decreased between the beginning and the end of the experiment, especially for neutral and nonconfident heifers, regardless of whether they were trained or not. These results show how using operant conditioning on some heifers not only decreased their vagal tone, but also reduced the responsiveness to humans of all the animals, trained and not trained; in the latter case, this reduction was through nonassociative learning, such as habituation.

This article explores the personal interpretation of the moral world Aotearoa New Zealand newsroom leaders are guided by when shaping the news, or, in other words, it poses the question: Do personal moral values play a role in Aotearoa New Zealand newsroom? In this study, we examine whether newsroom leaders view morals as a driver in news-shaping, or whether the news values of objectivity, accuracy and fairness prevail over personal morals when it comes to informing the news. The research was conducted by interviewing six newsroom leaders from different media companies in Aotearoa New Zealand and the interview data were analysed within the theoretical and philosophical framework adopted from Lakoff (2002). The research suggests that working professionals in the media industry were not able to discern where their morals ended and where the professional news values started. Furthermore, the interviewees affiliated with public service media responded that upholding moral values would not have a financial impact on the news media organisation, whereas those affiliated with private media responded that it would.

The Global Navigation Satellite System (GNSS) is a well-known and fundamental tool for crustal monitoring projects and tectonic studies, thanks to its high coverage and the high quality of the data they provide. In particular, at slowly convergent margins, where deformation rates are of the order of a few millimetres per year, GNSS monitoring proves to be beneficial in detecting the diffuse deformation responsible for tectonic stress accrual. Its strength lies in the high precision achieved by GNSS permanent stations, especially when long-term data and stable structures are available at the stations. North-eastern Italy is a tectonically active region located in the northernmost sector of the Adria microplate, slowly converging with the Eurasia plate, characterized by low deformation rates and moderate seismicity. It greatly benefits from continuous and high-precision geodetic monitoring, since it has been equipped with a permanent GNSS network providing real-time data and daily observations over 2 decades. The Friuli Venezia Giulia Deformation Network (FReDNet) was established in the area in 2002 to monitor crustal deformation and contribute to the regional seismic hazard assessment. This paper describes GNSS time series spanning 2 decades of stations located in north-eastern Italy and surroundings as well as the outgoing velocity field. The documented dataset has been retrieved by processing the GNSS observations with the GAMIT/GLOBK software ver10.71, which allows calculation of high-precision coordinate time series, position and velocity for each GNSS station by taking advantage of the high-performance computing resources of the Italian High-Performance Computing Centre (CINECA) clusters. The GNSS observations (raw and standard RINEX – Receiver INdependent EXchange – formats) and the time series estimated with the same procedure are currently daily continued, collected and stored in the framework of a long-term monitoring project. Instead, velocity solutions are intended for annual updates. The time series and velocity field dataset documented here are available on Zenodo (https://doi.org/10.5281/zenodo.8055800, Tunini et al., 2024).

This article delineates a study conducted within a psychiatric day hospital in France, adhering to the principles of institutional psychotherapy and therapeutic clubs that originated from the anti-psychiatric movement of the 1970s. The study specifically explores two activities within this framework: a music club and a video club. The primary objective of each club was to establish activities involving both the patients and the care staff to dissolve the boundaries between them. Additionally, these activities aimed to engage both patients and care staff in the conceptualization and execution processes, thereby fostering a collaborative approach within the psychiatric care setting.

Lameness is a widespread multifactorial condition affecting the health and performance of dairy cows. Despite the growing support by precision farming technologies, farmers still lack reliable data-driven tools to early identify lame cows. This study used a synthesis of reviews to identify cow’s behavioural and productive parameters most related to lameness and estimate their deviation due to a lameness event. The methodological approach used reviews as starting point to identify the most pertinent studies with the intention of extracting and analysing data from these primary studies. The final dataset used information collected from 31 research papers, cited in 15 reviews, and involved more than 25,000 dairy cows. Five parameters were suitable for the meta-analysis: one about eating behaviour (eating time), three regarding activity and resting behaviour (lying bouts, lying bout duration and lying time) and milk yield. The meta-analysis revealed that all parameters had a significant deviation in cows affected by lameness. The calculation of the pooled means allowed to quantify a mean value for the deviation imposed by a severe lameness event from the value recorded on nonlame cows. Compared to a nonlame animal, a lame cow had a significant negative deviation for eating time (−39 min/day), number of lying bouts (−0.5/day), and milk yield (−3 kg/day). Lame cows had positive deviations for lying bout duration (+12 min/bout) and daily lying time (+42 min/day). The individual or combined use of these mean deviation values as alarm reference thresholds could improve the accuracy of the current automated lameness detection systems.

Abstract Neurodegenerative disorders are characterized by the brain deposition of insoluble amyloidogenic proteins, such as α-synuclein or Tau, and the concomitant deterioration of cell functions such as the autophagy-lysosomal pathway (ALP). The ALP is involved in the degradation of intracellular macromolecules including protein aggregates. ALP dysfunction due to inherited defects in lysosomal or non-lysosomal proteins causes a group of diseases called lysosomal storage disorders (LSD) because of abnormal accumulation of lysosomal degradation substrates. Supporting the contribution of ALP defects in neurodegenerative diseases, deposition of amyloidogenic proteins occurs in LSD. Moreover, heterozygous mutations of several ALP genes represent risk factors for Parkinson’s disease. The reciprocal contribution of α-synuclein accumulation and lysosomal dysfunction have been extensively studied. However, whether this adverse crosstalk also embraces Tau pathology needs more investigation. Here, we show in human primary fibroblasts that Tau seeds isolated from the brain of Alzheimer’s disease induce Tau accumulation in acidic degradative organelles and lysosomal stress. Furthermore, inhibition of glucocerebrosidase, a lysosomal enzyme mutated in Gaucher’s disease and a main risk for Parkinson’s disease, causes lysosomal dysfunction in primary fibroblasts and contributes to the accumulation of Tau. Considering the presence of Tau lesions in Parkinson’s disease as well as in multiple neurodegenerative disorders including Alzheimer’s disease, our data call for further studies on strategies to alleviate ALP dysfunction as new therapeutic opportunity for neurodegenerative diseases and LSD.

Abstract Tau (MAPT) is a microtubule-associated protein causing common neurodegenerative diseases or rare inherited frontotemporal lobar degenerations. Emerging evidence for non-canonical functions of Tau in DNA repair and P53 regulation suggests its involvement in cancer. To bring new evidence for a relevant role of Tau in cancer, we carried out an in-silico pan-cancer analysis of MAPT transcriptomic profile in over 10000 clinical samples from 32 cancer types and over 1300 pre-clinical samples from 28 cancer types provided by the TCGA and the DEPMAP datasets respectively. MAPT expression associated with key cancer hallmarks including inflammation, proliferation, and epithelial to mesenchymal transition, showing cancer-specific patterns. In some cancer types, MAPT functional networks were affected by P53 mutational status. We identified new associations of MAPT with clinical outcomes and drug response in a context-specific manner. Overall, our findings indicate that the MAPT gene is a potential major player in multiple types of cancer. Importantly, the impact of Tau on cancer seems to be heavily influenced by the specific cellular environment.

Using the Sahel as an example, and more specifically the situation in the Lake Chad region, this article looks at how the growing recognition of climate change in scientific and political-institutional agendas is changing the way we think about the relationship between environmental crises, agriculture and violence. By recalling how this discussion is part of long-standing reflections on the relationship between resources and conflicts, he analyses the crystallisation of the debate into two camps - neo-determinist quantitative approaches at continental scales of study, on the one hand, and critical social science approaches such as Political ecology, which are more local and territorialized, on the other; as well as the role of think tanks as mediators of scientific work towards the political sphere. It explains the seduction of neo-determinist analyses despite the refutations fed by field research.

In Africa, although the first land observatory projects emerged at the end of the 1980s, it was from the mid-2000s onwards that they multiplied with the implementation of land observatories in Madagascar, Cameroon, Burkina Faso, but also, in South Africa, Uganda and Senegal. These land observatory projects, led by very different actors, aim to improve land governance for the benefit of better management of agricultural and natural areas and their interfaces. However, despite the high visibility and attractiveness of the concept, many land observatories do not manage to engage in effective information production and knowledge dissemination activities. This article aims to shed light on the main factors that explain the implementation gaps between projects on ‘paper’ and projects in the ‘field’, paying particular attention to the socio-economic and political constraints on access to, and production and sharing of data in the field of land.

Purpose: To evaluate, in vitro, the efficiency of a novel apparatus to test the adherence and penetration of bacteria on different membranes for guided regeneration. Methodology: To create the 3D device, Computer Aided Design/Computer Aided Manufacturing (CAD/CAM) systems were used. Three types of biomaterials were tested (n = 6): (DT) a collagen membrane; (DS) a polymer membrane; and (LP) a dense polytetrafluoroethylene barrier. The biomaterials were adapted to the apparatuses and challenged with two different monospecies bacterial culture of A. actinomycetemcomitans b and S. mutans. After 2 h, bacterial adherence and penetration were quantified by counting the number of colony-forming units (CFUs). Two specimens from each group were used for image analysis using Confocal Laser Scanning Microscopy. Statistical analysis was performed. Findings: The DS group had a higher adherence of S. mutans compared to A. actinomycetemcomitans b (p = 0.05). There was less adherence of A. actinomycetemcomitans b in the DS group, compared to the LP (p = 0.011) and DT (p < 0.001) groups. Only the membranes allowed penetration, which was blocked by barriers. The DT group allowed a greater penetration of S. mutans to occur compared to A. actinomycetemcomitans b (p = 0.009), which showed a higher penetration into the DS membranes compared to S. mutans (p = 0.016). The penetration of A. actinomycetemcomitans b through DS was higher compared to its penetration through DT and LP (p < 0.01 for both). DT and DS allowed a greater penetration of S. mutans to occur compared to LP, which prevented both bacterial species from penetrating. Conclusion: The apparatus allowed for the settlement and complete sealing of the biomaterials, enabling standardization.

Southern Europe’s mutating weather conditions and the European environmental agenda have suggested the cropping of maize (Zea mays L.) after winter cereal cultivation, even if shortening the growing period could result in an immature harvesting stage, limiting its silage quality. The experimental design investigated the effects of four harvesting dry matter (DM) classes (DMvl, 23.9%; DMl, 25.3%; DMm, 26.2%; DMh, 30.4%) in two inoculant types (heterofermentative (HE) vs. homofermentative (HOM) on fermentative quality, DM losses, and aerobic stability. The early harvested DMvl and DMl classes had the lowest silage density (−3) and resulted in an organic acids profile lowering the fermentative quality and increasing the DM losses, while no differences were detected following the use of the inoculants. The aerobic stability was more susceptible to further adverse fermentation via opportunistic microorganisms in the DMm and DMh classes, probably due to the lower moisture content, but the use of both HE and HOM lactic acid bacteria seemed to contain this silage surface damage. In summary, a shortening of the maize growing period might limit the achievement of the maturity stage ideal for high-quality silage, hampering the positive effects of both HOM and HE inoculants in the ensiling process of early harvested maize.

Abstract Tau gene mutations cause a progressive dementia and neurotoxic Tau forms deposited in neurofibrillary tangles are hallmarks of neurodegenerative tauopathies. Loss of non-canonical Tau functions may contribute to disease. In fact, Tau depletion affects the cellular response to DNA damage and tauopathies exhibit the accumulation of DNA lesions. Moreover, Tau modulates P53 activity and cell fate. Considering that MDM2 is the main antagonist of P53, we investigated, using orthogonal assays, if Tau interacts with MDM2. We report the existence in cells and brain of a Tau-MDM2 complex that, in vitro, exhibits reduced P53 ubiquitination activity in a manner sensitive to a Tau mutation. The Tau-MDM2 interaction involves the microtubule-binding domain of Tau and the acidic domain of MDM2, reminiscent of the binding of Tau to negatively charged microtubules. Notably, MDM2 accumulates aberrantly in neurofibrillary tangles. Aging-associated insults may expose a novel loss-of-function of Tau in neurodegeneration and cancer.

Abstract Background The most common subtype of ovarian cancer (OC) showing immunogenic potential is represented by the high-grade serous ovarian cancer (HGSOC), which is characterized by the presence of tumor-infiltrating immune cells able to modulate immune response. Because several studies showed a close correlation between OC patient’s clinical outcome and expression of programmed cell death protein-1 or its ligand (PD-1/PD-L1), the aim of our study was to investigate if plasma levels of immunomodulatory proteins may predict prognosis of advanced HGSOC women. Patients and methods Through specific ELISA tests, we analyzed plasma concentrations of PD-L1, PD-1, butyrophilin sub-family 3A/CD277 receptor (BTN3A1), pan-BTN3As, butyrophilin sub-family 2 member A1 (BTN2A1), and B- and T-lymphocyte attenuator (BTLA) in one hundred patients affected by advanced HGSOC, before surgery and therapy. The Kaplan–Meier method was used to generate the survival curves, while univariate and multivariate analysis were performed using Cox proportional hazard regression models. Results For each analyzed circulating biomarker, advanced HGSOC women were discriminated based on long (≥ 30 months) versus short progression-free survival (PFS 0.42 ng/mL), PD-1 (> 2.48 ng/mL), BTN3A1 (> 4.75 ng/mL), pan-BTN3As (> 13.06 ng/mL), BTN2A1 (> 5.59 ng/mL) and BTLA (> 2.78 ng/mL). Furthermore, a lower median PFS was associated with peritoneal carcinomatosis, age at diagnosis > 60 years or Body Mass Index (BMI) > 25. A multivariate analysis also suggested that plasma concentrations of PD-L1 ≤ 0.42 ng/mL (HR: 2.23; 95% CI: 1.34 to 3.73; p = 0.002), age at diagnosis ≤ 60 years (HR: 1.70; 95% CI: 1.07 to 2.70; p = 0.024) and absence of peritoneal carcinomatosis (HR: 1.87; 95% CI: 1.23 to 2.85; p = 0.003) were significant prognostic marker for a longer PFS in advanced HGSOC patients. Conclusions The identification of high-risk HGSOC women could be improved through determination of the plasma PD-L1, PD-1, BTN3A1, pan-BTN3As, BTN2A1 and BTLA levels.

Introduction: Progressive Tau deposition in neurofibrillary tangles and neuropil threads is the hallmark of tauopathies, a disorder group that includes Alzheimer’s disease. Since Tau is a microtubule-associated protein, a prevalent concept to explain the pathogenesis of tauopathies is that abnormal Tau modification contributes to dissociation from microtubules, assembly into multimeric β-sheets, proteotoxicity, neuronal dysfunction and cell loss. Tau also localizes in the cell nucleus and evidence supports an emerging function of Tau in DNA stability and epigenetic modulation.Methods: To better characterize the possible role of Tau in regulation of chromatin compaction and subsequent gene expression, we performed a bioinformatics analysis of transcriptome data obtained from Tau-depleted human neuroblastoma cells.Results: Among the transcripts deregulated in a Tau-dependent manner, we found an enrichment of target genes for the polycomb repressive complex 2. We further describe decreased cellular amounts of the core components of the polycomb repressive complex 2 and lower histone 3 trimethylation in Tau deficient cells. Among the de-repressed polycomb repressive complex 2 target gene products, IGFBP3 protein was found to be linked to increased senescence induction in Tau-deficient cells.Discussion: Our findings propose a mechanism for Tau-dependent epigenetic modulation of cell senescence, a key event in pathologic aging.
