1. A Pilot Study Investigating Changes in the Human Plasma and Urine NAD+ Metabolome During a 6 Hour Intravenous Infusion of NAD+

Abstract

Accumulating evidence suggests that active maintenance of optimal levels of the essential pyridine nucleotide, nicotinamide adenine dinucleotide (NAD+) is beneficial in conditions of either increased NAD+ turnover or inadequate synthesis, including Alzheimer’s disease and other neurodegenerative disorders and the aging process. While studies have documented the efficacy of some NAD+ precursors such as nicotinamide riboside (NR) in raising plasma NAD+, no data are currently available on the fate of directly infused NAD+ in a human cohort. This study, therefore, documented changes in plasma and urine levels of NAD+ and its metabolites during and after a 6 h 3 μmol/min NAD+ intravenous (IV) infusion. Surprisingly, no change in plasma (NAD+) or metabolites [nicotinamide, methylnicotinamide, adenosine phosphoribose ribose (ADPR) and nicotinamide mononucleotide (NMN)] were observed until after 2 h. Increased urinary excretion of methylnicotinamide and NAD+ were detected at 6 h, however, no significant rise in urinary nicotinamide was observed. This study revealed for the first time that: (i) at an infusion rate of 3 μmol/min NAD+ is rapidly and completely removed from the plasma for at least the first 2 h; (ii) the profile of metabolites is consistent with NAD+ glycohydrolase and NAD+ pyrophosphatase activity; and (iii) urinary excretion products arising from an NAD+ infusion include NAD+ itself and methyl nicotinamide (meNAM) but not NAM.

Abstract

The increased resolution of numerical weather prediction models allows nowadays addressing more realistically urban meteorology and air pollution processes. This has triggered new interest in modelling and describing experimentally the specific features and processes of urban areas. Recent developments and results performed within the EU-funded project FUMAPEX on integrated systems for forecasting urban meteorology and air pollution are reported here. Sensitivity studies with respect to optimum resolution, parametrisation of urban roughness and surface exchange fluxes and the role of urban soil layers are carried out with advanced meso- or sub-meso meteorological models. They show that sensible improvements can be achieved by higher model resolution that is accompanied with better description of urban surface features. Recommendations, especially with respect to advanced urban air quality forecasting and information systems, are given together with an assessment of the needed further research and data.

Abstract

This qualitative study investigated how individuals managed their religious identity at work. Christians, non-Christians, and non-religious (n = 320) from across the United States took an online, open-ended questionnaire. Rich information emerged from the data concerning the why and how of religious disclosure, what happened after disclosure, and what strategies should be used for the most positive disclosure outcomes. The findings suggested a religiously respectful culture depends on its individual members' communicative practices. Religious conversations in the workplace were possible so long as individuals knew how to do so appropriately and effectively. Byline: J. P. Flori, P. Mestayer, C. Sacre The LOSTRAC software is devoted to real-time tracking of accidental atmospheric releases. It allows one to calculate, within a few minutes from the moment when the accident is reported and localised, displacement and diffusion of atmospheric pollution. It also provides short-term evolution forecasting. It should become one of the tools for assisting the authorities in charge of safety operations to take decisions. The core of the software consists of LINCOM and RIMPUFF codes. A data input interface and a results module capable of displaying detailed cartography have been developed around these two codes. The codes were elaborated by the Riso National Laboratory in Denmark, and the dialogue and cartography interfaces were developed at the Centre Scientifique et Technique du Batiment (CSTB). A team at the Dynamique de l'Atmosphere Habitee department at the Laboratoire de Mecanique des Fluides of the Ecole Centrale de Nantes has initiated and supervised the project for both interfaces. Future elaboration will cover all steps of the project from data input to the development of a short-term forecasting tool and to data setup, fitted for the software. The creation of a meteorological data management module is planned as priority.

34. Energy and water budgets of asphalt concrete pavement under simulated rain events.

Abstract

Urban areas are subject to high human pressure and forthcoming enhanced hydrologic and climatic risks due to both city development and climate change. An asphalt concrete parking lot was instrumented in Nantes, France, to quantify the energy and hydrological responses of the surface to simulated rainfalls. The surface fluxes (precipitation, evaporation, radiation exchanges, sensible heat convection and conduction, runoff) were measured in situ and used to close the water budget with residual closure errors lower than 10%, depending on the surface evaporation retrieval method. The latent heat flux estimated from scintillometry measurements provided a better water budget closure than the direct eddy-correlation measurements. Runoff was the primary component of the water budget and represented around 80% of the total precipitation, compared to 17% for surface evaporation. The scintillometry method provided water evaporation time series at a 1-min time scale during the experiment. These series were used to characterize the rapid changes in the hydrological and energetic budgets of the asphalt surface after a precipitation event. During the drying phase the surface evaporation was significantly active, yielding 80% of the turbulent fluxes with a Bowen ratio of 0.25. [ABSTRACT FROM AUTHOR]

International audience; The Eval-PDU research, funded by French National Research Agency' program Sustainable City, aims at testing methodologies for Strategic Environmental Assessment of Urban Mobility Master Plans with an application to Nantes Metropolis. The general approach used for this interdisciplinary research consists in combining several modelling of flows: first vehicles flows in the city, then the flows of atmospheric pollutants and noise pollution resulting from the traffic. In front of phenomena that are quite impossible to be precisely measured considering their micro-local diversity, modelling seems the only solution to estimate the effect of transport policies on the urban environment (in comparison with a reference situation where they are not applied) and thus to enlighten collective action in order to minimize negative externalities.

International audience; The FluxSAP 2010 is the first of two experimental campaigns aiming at quantitatively assessing the contribution of urban vegetation in the sensible heat and water vapour fluxes, over an heterogeneous area including buildings, semi-impervious surfaces, low and high vegetation. The 2010 experiment objective was primarily the feasibility of measurements with five different methods, and their analysis with respect to the surface cover mode variability of their footprints.

Abstract

International audience; The project objective is to develop an optimized methodology to assess the environmental impacts of urban mobility plans (UMP, in French PDU), taking into account their social and economic consequences. The main proposed methodology is based on a systemic approach: multi-factor (air quality, noise, energy consumption, greenhouse gas emission) numerical simulations with a chain of physically-based models, based on alternative and comparative scenarios. The social and economic consequences of these alternative simulations are assessed by means of econometric models. Two alternative approaches are explored: (i) the use of composite environmental indicators to correlate the sources to the impacts, especially health impacts, and (ii) the analysis of sample surveys on what makes inhabitants' quality of life, well-being and territorial satisfaction and on citizens' behavioral changes linked to transport offer changes.

Abstract

The specific problems of determining and simulating the surface energy balance (SEB) and the mixing height (MH) over urban areas are examined. The SEB and MH are critical components of algorithms and numerical models for the urban boundary layer, though the constituent parts of the SEB and the MH are not routinely measured by national weather services. Parameterisations are thus needed in applications. In this investigation, several recently developed algorithms and models for estimating the SEB and MH were applied to new datasets and assessed. Results are discussed in terms of the need for spatial resolution and the parameters needed to describe the urban atmosphere. Limitations of models are identified and recommendations for further development and observations are given. Having identified gaps in knowledge, key findings from new urban experiments and numerical modelling for the SEB and MH are given. The diurnal cycle for the SEB is significantly different from rural conditions-urban heat storage is needed in urban parameterisations. The urban MH is increased over the rural MH, as shown by several numerical schemes and careful sodar analyses. This work has been carried out within the COST-715 Action 'Meteorology applied to urban air pollution problems (1998-2004). COST 715 reached a consensus proposing representatively sited measurements of meteorological parameters and turbulent fluxes above roof-tops, and recognised that such data are needed to improve numerical models of the urban surface processes. © Springer Science+Business Media, B.V. 2007.

Abstract

International audience; The large eddy simulations employing the SUBMESO model with the urban soil layer model SM2-U were performed for the model domain covering the Danish Island of Sealand and including the Copenhagen metropolitan area. Monthly and diurnal cycle variability were studied for the net radiation, sensible and storage heat fluxes, surface's temperatures, and others. These were evaluated for selected urban vs. non urban related types of covers/surfaces and urban districts such as city center, high buildings, industrial, and residential. Results showed strong effects of urban features on temporal and spatial variability. They are useful and applicable for verification of numerical weather prediction models and development of urban canopy parameterizations.
