Under natural conditions, crops typically suffer from severe challenges due to the increasing of abiotic and biotic stresses which severely affect plant growth and reduc crop yield. The present study investigated the single and combined impacts of Sclerotinia sclerotiorum and salinity stress on common bean (Phaseolus vulgaris L.) seedling which is scarcely studied. The study evaluated the in vitro and in vivo influence of two salinity tolerant Trichoderma isolates, T. koningii and T. harzianum against S. sclerotiorum under salinity stress. The results showed the ability of T. koningii and T. harzianum to grow and sporulate at high levels of salinity, 80 mM NaCl, without significantly impacting their ability to produce cell wall degrading enzymes, cellulase and chitinase. Amylase and proteinase (Prb1) genes were detected in T. harzianum. The in vitro assay revealed that both isolates could inhibit the growth of S. sclerotiorum under high salinity concentrations. In a greenhouse experiment, both Trichoderma isolates ameliorated the damaging impacts of S. sclerotiorum under salinity stress on common bean seedlings' germination and growth characteristics compared to their untreated control. Both bioagents significantly attenuated the damping-off and collar/stem rot percentages of infected common bean under salinity stress. Salinity stress intensified the effect of S. sclerotiorum on photosynthetic pigments, induced oxidative and nitrative stress, hampered ionic homeostasis, and deactivated antioxidants and defense-related molecules. On the other hand, Trichoderma isolates restrained the reduction of chlorophylls and carotenoids, ascorbate, reduced glutathione, flavonoids, phenolics, and various antioxidant enzymes, especially for single stresses and T. harzianum. All these upregulations reflected in keeping the cell membranes of common beans seedling more stable where the levels of lipid peroxidation and methylglyoxal due to the reduction of reactive oxygen species and upregulation of nitric oxide, which expressed better growth under pathogen attack or/and saline. The tested isolates, T. koningii and T. harzianum could be used as effective biological control against S. sclerotiorum on common beans in saline soils or areas irrigated with saline water.

Nowadays, the use of nanoparticles (NPs) has become useful in the different application fields. The aim of this study was to investigate the in vitro antimicrobial potential of metalZnO nanoparticles (ZnO NPs) against several bacterial and fungal strains including; Escherichia coli (ATCC 25922), Bacillus cereus (ATCC 13753), Staphylococcus aureus (ATCC 8095), Pseudomonas aeruginosa (ATCC10662), Candida albicans (ATCC10231) and Aspergillus niger (AUMC3663). Results obtained by X-ray diffraction analysis (XRD) showed that the NPs size was in the range of 35.1- 43.7 nm. Images of the scanning electron microscopy (SEM) demonstrated the rod shape nature of the ZnO NPs, and the semi-spherical shapes of the Zn9.7TM0.3O NPs. The effect of different concentrations of ZnO NPs on the in vitro growth of the bacterial and fungal strains was evaluated using the agar well diffusion assay. Current results showed that Cd-ZnO NP recorded the highest antimicrobial potency; expressing inhibition zones diameter range of 12- 45 mm, while ZnO NPs demonstrated the least activity exhibiting inhibition zones diameter that ranged from 0- 36 mm. Among all the examined ZnO-NPs, treatment of E. coli and Staph. aureus cells with Cd-ZnO proved to be the most effective in causing membrane leakage of reducing sugars, protein and DNA recording; 0.41 µg/ ml and 0.38 µg/ ml; 14.91 µg/ ml and 15.98 µg/ ml; 0.81 µg/ ml and 0.96 µg/ ml, respectively. This study emphasized that ZnO NPs could be used as alternative antimicrobial agents to control the bacterial and fungal pathogens. Manipulation of ZnO NPs is ecofriendly; as it reduces the use of the synthetic pesticides and chemical therapeutic agents, which pollute the environment. In the future, in vivo application of these NPs necessitates the proof that they have no phytotoxicity and\or cytotoxicity.

Background: Ischemic stroke is one of the serious complications of diabetes. Non-coding RNAs are established as promising biomarkers for diabetes and its complications. The present research investigated the expression profiles of serum TUG1, LINC00657, miR-9, and miR-106a in diabetic patients with and without stroke.Methods: A total of 75 diabetic patients without stroke, 77 patients with stroke, and 71 healthy controls were recruited in the current study. The serum expression levels of TUG1, LINC00657, miR-9, and miR-106a were assessed using quantitative real-time polymerase chain reaction assays.Results: We observed significant high expression levels of LINC00657 and miR-9 in the serum of diabetic patients without stroke compared to control participants. At the same time, we found marked increases of serum TUG1, LINC00657, and miR-9 and a marked decrease of serum miR-106a in diabetic patients who had stroke relative to those without stroke. Also, we revealed positive correlations between each of TUG1, LINC00657, and miR-9 and the National Institutes of Health Stroke Scale (NIHSS). However, there was a negative correlation between miR-106a and NIHSS. Finally, we demonstrated a negative correlation between LINC00657 and miR-106a in diabetic patients with stroke.Conclusion: Serum non-coding RNAs, TUG1, LINC00657, miR-9, and miR-106a displayed potential as novel molecular biomarkers for diabetes complicated with stroke, suggesting that they might be new therapeutic targets for the treatment of diabetic patients with stroke.

Abstract The subterranean termite Psammotermes hypostoma Desneux is considered as an important pest that could cause severe damage to buildings, furniture, silos of grain and crops or any material containing cellulose. This species of termites is widespread in Egypt and Africa. The lower termite’s ability to digest cellulose depends on the association of symbiotic organisms gut that digest cellulose (flagellates and bacteria). In this study, 33 different bacterial isolates were obtained from the gut of the termite P. hypostoma which were collected using cellulose traps. Strains were grown on carboxymethylcellulose (CMC) as a sole source of carbon. Cellulolytic strains were isolated in two different cellulose medium (mineral salt medium containing carboxymethylcellulose as the sole carbon source and agar cellulose medium). Five isolates showed significant cellulolytic activity identified by a Congo red assay which gives clear zone. Based on biochemical tests and sequencing of 16s rRNA genes these isolates were identified as Paenibacillus lactis, Lysinibacillus macrolides, Stenotrophomonas maltophilia, Lysinibacillus fusiformis and Bacillus cereus, that deposited in GenBank with accession numbers MG991563, MG991564, MG991565, MG991566 and MG991567, respectively.

The present investigation was carried out using 51 diverse bitter gourd accessions as material for studying genetic diversity and relatedness using morphological and SSR markers. A wide variation was observed for morphological traits like the number of days to the first female flower anthesis (37.33–60.67), the number of days to the first fruit harvest (47.67–72.00), the number of fruits/plant (12.00–46.67), fruit length (5.00–22.23 cm), fruit diameter (1.05–6.38 cm), average fruit weight (20.71–77.67 g) and yield per plant (513.3–1976 g). Cluster analysis for 10 quantitative traits grouped the 51 accessions into 6 clusters. Out of 61 SSR primers screened, 30 were polymorphic and highly informative as a means to differentiate these accessions. Based on genotyping, a high level of genetic diversity was observed, with a total of 99 alleles. The polymorphic information content (PIC) values ranged from 0.038 for marker BG_SSR-8 to 0.721 for S-24, with an average of 0.429. The numbers of alleles ranged from 2 to 5, with an average of 3.3 alleles per locus. Gene diversity ranged from 0.04 for BG_SSR-8 to 0.76 for S-24, showing a wide variation among 51 accessions. The UPGMA cluster analysis grouped these accessions into 3 major clusters. Cluster I comprised 4 small, fruited accessions that are commercially cultivated in central and eastern India. Cluster II comprised 35 medium- to long-sized fruited accessions, which made up an abundant and diverse group. Cluster III comprised 11 long and extra-long fruited accessions. The polymorphic SSR markers of the study will be highly useful in genetic fingerprinting and mapping, and for association analysis in Momordica regarding several economic traits.

Background: Polymorphisms of long noncoding RNAs are lately documented as hazardous factors for the development of numerous tumors. Furthermore, the evaluation of noncoding RNAs has emerged as a novel detector of breast cancer patients. We aimed to genotype the HOXA transcript at the distal tip (HOTTIP) rs1859168 and assess its relationship with the levels of the serum HOTTIP and its target miR-615-3p in patients with breast cancer (BC). Methods: One hundred and fifty-one patients with BC, 139 patients with fibroadenoma (FA), and 143 healthy participants were incorporated into the current study. The genotyping of rs1859168 and the measurements of the HOTTIP and miR-615-3p levels were assessed using quantitative real-time PCR. Results: We revealed a significant association between each of the CC genotypes, C allele, dominant and recessive models, and the increased risk of BC (p = 0.013, p < 0.001, p < 0.001, and p < 0.001, respectively) relative to the healthy controls. Similarly, the CC genotype, C allele, and recessive model were observed to be related to the increased incidence of BC with respect to FA (p < 0.001 for all). A significant upregulation of HOTTIP and a marked decrease of miR-615-3p were verified in patients with BC compared to each of the healthy individuals, patients with FA, and the non-BC group (healthy subjects + FA) (p < 0.001 for all). A significant negative correlation was demonstrated between the expression of HOTTIP and miR-615-3p in the serum of patients with BC. The HOTTIP expression was upregulated, while that of miR-615-3p was downregulated in patients with BC who carried the CC genotype with respect to those who carried the AA or AC genotypes (p < 0.05 for all). Conclusions: The genetic variants of rs1859168 are linked to an increased susceptibility to BC. Moreover, HOTTIP and miR-615-3p may be used as novel indicators and targets for the treatment of patients with BC.

BackgroundNeonatal sepsis is a serious condition. Recent clinical studies have indicated that microRNAs (miRNAs) are key players in the pathogenesis of sepsis, which could be used as biomarkers for this condition.Patients and methodsA total of 90 neonates with sepsis and 90 healthy neonates were enrolled in this study. qRT-PCR was performed to measure the expression levels of serum miR-34a-5p and miR-199a-3p.ResultsmiR-34a-5p and miR-199a-3p serum levels were significantly reduced in neonates with sepsis compared with those in healthy neonates (P = 0.006 and P = 0.001, respectively). Significant correlations of miR-34a-5p and miR-199a-3p with each of TLC, RDW, RBS, and C-reactive protein (CRP) as well as SNAPII were observed, indicating their associations with the severity of neonatal sepsis.ConclusionmiR-34a-5p and miR-199a-3p may be useful as novel biomarkers in neonatal sepsis and may provide a new direction for its treatment.

Genetic variants in microRNAs (miRNAs) can alter the miRNAs expression and/or function, accordingly, affecting the related biological pathways and disease risk. Dysregulation of miR-155 and miR-146a expression levels has been well-described in viral hepatitis B (HBV). In the current study, we aimed to assess rs767649 T/A and rs57095329 A/G polymorphisms in miR-155, and miR-146a genes, respectively, as risk factors for Chronic HBV (CHBV) in the Egyptian population. Also, we aimed to do in silico analysis to investigate the molecules that primarily target these miRNAs. One hundred patients diagnosed as CHBV and one hundred age and sex-matched controls with evidence of past HBV infection were genotyped for miR-155 (rs767649) and miR-146a (rs57095329) using real-time polymerase chain reaction.
