Leprosy is the second most prevalent mycobacterial disease globally. Despite the existence of an effective therapy, leprosy incidence has consistently remained above 200,000 cases per year since 2010. Numerous host genetic factors have been identified for leprosy that contribute to the persistently high case numbers. In the past decade, genetic epidemiology approaches, including genome-wide association studies (GWAS), identified more than 30 loci contributing to leprosy susceptibility. However, GWAS loci commonly encompass multiple genes, which poses a challenge to define causal candidates for each locus. To address this problem, we hypothesized that genes contributing to leprosy susceptibility differ in their frequencies of rare protein-altering variants between cases and controls. Using deep resequencing we assessed protein-coding variants for 34 genes located in GWAS or linkage loci in 555 Vietnamese leprosy cases and 500 healthy controls. We observed 234 nonsynonymous mutations in the targeted genes. A significant depletion of protein-altering variants was detected for the IL18R1 and BCL10 genes in leprosy cases. The IL18R1 gene is clustered with IL18RAP and IL1RL1 in the leprosy GWAS locus on chromosome 2q12.1. Moreover, in a recent GWAS we identified an HLA-independent signal of association with leprosy on chromosome 6p21. Here, we report amino acid changes in the CDSN and PSORS1C2 genes depleted in leprosy cases, indicating them as candidate genes in the chromosome 6p21 locus. Our results show that deep resequencing can identify leprosy candidate susceptibility genes that had been missed by classic linkage and association approaches.

Leprosy is a chronic disease caused by Mycobacterium leprae. Worldwide, more than 200,000 new patients are affected by leprosy annually, making it the second most common mycobacterial disease after tuberculosis. The MHC/HLA region has been consistently identified as carrying major leprosy susceptibility variants in different populations at times with inconsistent results. To establish the unambiguous molecular identity of classical HLA class I and class II leprosy susceptibility factors, we applied next-generation sequencing to genotype with high-resolution 11 HLA class I and class II genes in 1,155 individuals from a Vietnamese leprosy case-control sample. HLA alleles belonging to an extended haplotype from HLA-A to HLA-DPB1 were associated with risk to leprosy. This susceptibility signal could be reduced to the HLA-DRB1*10:01~ HLA-DQA1*01:05 alleles which were in complete linkage disequilibrium (LD). In addition, haplotypes containing HLA-DRB3~ HLA-DRB1*12:02 and HLA-C*07:06~ HLA-B*44:03~ HLA-DRB1*07:01 alleles were found as two independent protective factors for leprosy. Moreover, we replicated the previously associated HLA-DRB1*15:01 as leprosy risk factor and HLA-DRB1*04:05~HLA-DQA1*03:03 as protective alleles. When we narrowed the analysis to the single amino acid level, we found that the associations of the HLA alleles were largely captured by four independent amino acids at HLA-DRβ1 positions 57 (D) and 13 (F), HLA-B position 63 (E) and HLA-A position 19 (K). Hence, analyses at the amino acid level circumvented the ambiguity caused by strong LD of leprosy susceptibility HLA alleles and identified four distinct leprosy susceptibility factors.

Leprosy is a chronic infectious disease of the skin and peripheral nerves with a strong genetic predisposition. Recent genome-wide approaches have identified numerous common variants associated with leprosy, almost all in the Chinese population. We conducted the first family-based genome-wide association study of leprosy in 622 affected offspring from Vietnam, followed by replication in an independent sample of 1181 leprosy cases and 668 controls of the same ethnic origin. The most significant results were observed within the HLA region, in which six SNPs displayed genome-wide significant associations, all of which were replicated in the independent case/control sample. We investigated the signal in the HLA region in more detail, by conducting a multivariate analysis on the case/control sample of 319 GWAS-suggestive HLA hits for which evidence for replication was obtained. We identified three independently associated SNPs, two located in the HLA class I region (rs1265048: OR = 0.69 [0.58-0.80], combined p-value = 5.53x10-11; and rs114598080: OR = 1.47 [1.46-1.48], combined p-value = 8.77x10-13), and one located in the HLA class II region (rs3187964 (OR = 1.67 [1.55-1.80], combined p-value = 8.35x10-16). We also validated two previously identified risk factors for leprosy: the missense variant rs3764147 in the LACC1 gene (OR = 1.52 [1.41-1.63], combined p-value = 5.06x10-14), and the intergenic variant rs6871626 located close to the IL12B gene (OR = 0.73 [0.61-0.84], combined p-value = 6.44x10-8). These results shed new light on the genetic control of leprosy, by dissecting the influence of HLA SNPs, and validating the independent role of two additional variants in a large Vietnamese sample.

Leprosy is a human infectious disease caused by Mycobacterium leprae. A strong host genetic contribution to leprosy susceptibility is well established. However, the modulation of the transcriptional response to infection and the mechanism(s) of disease control are poorly understood. To address this gap in knowledge of leprosy pathogenicity, we conducted a genome-wide search for expression quantitative trait loci (eQTL) that are associated with transcript variation before and after stimulation with M. leprae sonicate in whole blood cells. We show that M. leprae antigen stimulation mainly triggered the upregulation of immune related genes and that a substantial proportion of the differential gene expression is genetically controlled. Indeed, using stringent criteria, we identified 318 genes displaying cis-eQTL at an FDR of 0.01, including 66 genes displaying response-eQTL (reQTL), i.e. cis-eQTL that showed significant evidence for interaction with the M. leprae stimulus. Such reQTL correspond to regulatory variations that affect the interaction between human whole blood cells and M. leprae sonicate and, thus, likely between the human host and M. leprae bacilli. We found that reQTL were significantly enriched among binding sites of transcription factors that are activated in response to infection, and that they were enriched among single nucleotide polymorphisms (SNPs) associated with susceptibility to leprosy per se and Type-I Reaction, and seven of them have been targeted by recent positive selection. Our study suggested that natural selection shaped our genomic diversity to face pathogen exposure including M. leprae infection.

Leprosy Type-1 Reactions (T1Rs) are pathological inflammatory responses that afflict a sub-group of leprosy patients and result in peripheral nerve damage. Here, we employed a family-based GWAS in 221 families with 229 T1R-affect offspring with stepwise replication to identify risk factors for T1R. We discovered, replicated and validated T1R-specific associations with SNPs located in chromosome region 10p21.2. Combined analysis across the three independent samples resulted in strong evidence of association of rs1875147 with T1R (p = 4.5x10-8; OR = 1.54, 95% CI = 1.32-1.80). The T1R-risk locus was restricted to a lncRNA-encoding genomic interval with rs1875147 being an eQTL for the lncRNA. Since a genetic overlap between leprosy and inflammatory bowel disease (IBD) has been detected, we evaluated if the shared genetic control could be traced to the T1R endophenotype. Employing the results of a recent IBD GWAS meta-analysis we found that 10.6% of IBD SNPs available in our dataset shared a common risk-allele with T1R (p = 2.4x10-4). This finding points to a substantial overlap in the genetic control of clinically diverse inflammatory disorders.

After sustained exposure to Mycobacterium leprae, only a subset of exposed individuals develops clinical leprosy. Moreover, leprosy patients show a wide spectrum of clinical manifestations that extend from the paucibacillary (PB) to the multibacillary (MB) form of the disease. This "polarization" of leprosy has long been a major focus of investigation for immunologists because of the different immune response in these two forms. But while leprosy per se has been shown to be under tight human genetic control, few epidemiological or genetic studies have focused on leprosy subtypes. Using PubMed, we collected available data in English on the epidemiology of leprosy polarization and the possible role of human genetics in its pathophysiology until September 2015. At the genetic level, we assembled a list of 28 genes from the literature that are associated with leprosy subtypes or implicated in the polarization process. Our bibliographical search revealed that improved study designs are needed to identify genes associated with leprosy polarization. Future investigations should not be restricted to a subanalysis of leprosy per se studies but should instead contrast MB to PB individuals. We show the latter approach to be the most powerful design for the identification of genetic polarization determinants. Finally, we bring to light the important resource represented by the nine-banded armadillo model, a unique animal model for leprosy.

Depending on the epidemiological setting, a variable proportion of leprosy patients will suffer from excessive pro-inflammatory responses, termed type-1 reactions (T1R). The LRRK2 gene encodes a multi-functional protein that has been shown to modulate pro-inflammatory responses. Variants near the LRRK2 gene have been associated with leprosy in some but not in other studies. We hypothesized that LRRK2 was a T1R susceptibility gene and that inconsistent association results might reflect different proportions of patients with T1R in the different sample settings. Hence, we evaluated the association of LRRK2 variants with T1R susceptibility.An association scan of the LRRK2 locus was performed using 156 single-nucleotide polymorphisms (SNPs). Evidence of association was evaluated in two family-based samples: A set of T1R-affected and a second set of T1R-free families. Only SNPs significant for T1R-affected families with significant evidence of heterogeneity relative to T1R-free families were considered T1R-specific. An expression quantitative trait locus (eQTL) analysis was applied to evaluate the impact of T1R-specific SNPs on LRRK2 gene transcriptional levels.A total of 18 T1R-specific variants organized in four bins were detected. The core SNP capturing the T1R association was the LRRK2 missense variant M2397T (rs3761863) that affects LRRK2 protein turnover. Additionally, a bin of nine SNPs associated with T1R were eQTLs for LRRK2 in unstimulated whole blood cells but not after exposure to Mycobacterium leprae antigen.The results support a preferential association of LRRK2 variants with T1R. LRRK2 involvement in T1R is likely due to a pathological pro-inflammatory loop modulated by LRRK2 availability. Interestingly, the M2397T variant was reported in association with Crohn's disease with the same risk allele as in T1R suggesting common inflammatory mechanism in these two distinct diseases.

Leprosy is a persistent infectious disease caused by Mycobacterium leprae that still affects over 200,000 new patients annually. The host genetic background is an important risk factor for leprosy susceptibility and the PARK2 gene is a replicated leprosy susceptibility candidate gene. The protein product of PARK2, Parkin, is an E3 ubiquitin ligase that is involved in the development of various forms of Parkinsonism. The human macrophage is both a natural host cell of M. leprae as well as a primary mediator of natural immune defenses, in part by secreting important pro-inflammatory cytokines and chemokines. Here, we report that down-regulation of Parkin in THP-1 macrophages, human monocyte-derived macrophages and human Schwann cells resulted in a consistent and specific decrease in interleukin-6 (IL-6) and monocyte chemoattractant protein 1 (MCP-1/CCL2) production in response to mycobacteria or LPS. Interestingly, production of IL-6 at 6 hours by THP-1 cells stimulated with live M. leprae and M. bovis BCG was dependent on pretreatment with 1,25-dihydroxyvitamin D(3) (VD). Parkin knockdown in VD-treated cells blocked IL-6 induction by mycobacteria. However, IκB-α phosphorylation and levels of IκB-ξ, a nuclear protein required for IL-6 expression, were not affected by Parkin silencing. Phosphorylation of MAPK ERK1/2 and p38 was unaffected by Parkin silencing while JNK activation was promoted but did not explain the altered cytokine production. In a final set of experiments we found that genetic risk factors of leprosy located in the PARK2 promoter region were significantly correlated with M. leprae sonicate triggered CCL2 and IL6 transcript levels in whole blood assays. These results associated genetically controlled changes in the production of MCP-1/CCL2 and IL-6 with known leprosy susceptibility factors.

Leprosy reversal reactions type 1 (T1R) are acute immune episodes that affect a subset of leprosy patients and remain a major cause of nerve damage. Little is known about the relative importance of innate versus environmental factors in the pathogenesis of T1R. In a retrospective design, we evaluated innate differences in response to Mycobacterium leprae between healthy individuals and former leprosy patients affected or free of T1R by analyzing the transcriptome response of whole blood to M. leprae sonicate. Validation of results was conducted in a subsequent prospective study. We observed the differential expression of 581 genes upon exposure of whole blood to M. leprae sonicate in the retrospective study. We defined a 44 T1R gene set signature of differentially regulated genes. The majority of the T1R set genes were represented by three functional groups: i) pro-inflammatory regulators; ii) arachidonic acid metabolism mediators; and iii) regulators of anti-inflammation. The validity of the T1R gene set signature was replicated in the prospective arm of the study. The T1R genetic signature encompasses genes encoding pro- and anti-inflammatory mediators of innate immunity. This suggests an innate defect in the regulation of the inflammatory response to M. leprae antigens. The identified T1R gene set represents a critical first step towards a genetic profile of leprosy patients who are at increased risk of T1R and concomitant nerve damage.

This study looks at how welding intensity, speed, voltage, and stick-out affect the structural and mechanical characteristics of metal inert gas (MIG) welding on SUS 201 stainless steel and C20 steel. The Taguchi method is used to optimize the study's experiment findings. The results show that the welding current has a more significant effect on the tensile test than the welding voltage, stick-out, and welding speed. Welding voltage has the lowest influence. In addition to the base metals' ferrite, pearlite, and austenite phases, the weld bead area contains martensite and bainite microstructures. The optimal parameters for the ultimate tensile strength (UTS), yield strength, and elongation values are a 110 amp welding current, 15 V of voltage, a 500 mm.min−1 welding speed, and a 10 mm stick-out. The confirmed UTS, yield strength, and elongation values are 452.78 MPa, 374.65 MPa, and 38.55%, respectively, comparable with the expected value derived using the Taguchi method. In the flexural test, the welding current is the most critical element affecting flexural strength. A welding current of 110 amp, an arc voltage of 15 V, a welding speed of 500 mm.min−1, and a stick-out of 12 mm are the ideal values for flexural strength. The flexural strength, confirmed at 1756.78 MPa, is more than that of the other samples. The study's conclusions can offer more details regarding the dissimilar welding industry. This study examines the effects of stick-out, welding current, welding speed, and voltage on the mechanical characteristics and microstructure of MIG welding on SUS 304 stainless steel and S20C steel. The Taguchi method was used to maximize the experiment's outcomes. Fine columnar dendrites formed at fusion sites, and δ-ferrite phases with dark lines and shapes accumulated between the fusion line and the austenite phases. A welding current of 110 A, voltage of 15 V, welding speed of 500 mm/min, and stick-out of 12 mm were the optimal settings for the ultimate tensile strength (UTS). The UTS value confirmation was 469.4 MPa, which agrees with the estimated value determined using the Taguchi technique. The tensile test revealed that welding current had a far greater impact on mechanical qualities than welding voltage, speed, and stick-out distance. The ideal welding parameters for flexural strength were as follows: stick-out of 12 mm, arc voltage of 15 V, welding speed of 450 mm/min, and welding current of 110 amp. The Taguchi method is useful, as evidenced by the validation of the flexure strength of 1937.45 MPa, which is much greater than the other samples. The impact of the thermal annealing process on the mechanical characteristics of the dissimilar weld joints could be the subject of future research. In this study, we present a novel approach to injection molding, focusing on the strength of weld lines in polyamide 6 (PA6) composite samples. By implementing a mold temperature significantly higher than the typical molding practice, which rarely exceeds 100°C, we assess the effects of advanced mold temperature management. The research introduces a newly engineered mold structure specifically designed for localized mold heating, distinguishing it as the 'novel cavity.' This innovative design is compared against traditional molding methods to highlight the improvements in weld line strength at elevated mold temperatures. To optimize the molding parameters, we apply an Artificial Neural Network (ANN) in conjunction with a Genetic Algorithm (GA). Our findings reveal that the optimal ultimate tensile strength (UTS) and elongation values are achieved with a filling time of 3.4 seconds, packing time of 0.8 seconds, melt temperature of 246°C, and a novel high mold temperature of 173°C. A specific sample demonstrated the best molding parameters at a filling time of 3.4 seconds, packing time of 0.4 seconds, melt temperature of 244°C, and mold temperature of 173°C, resulting in an elongation value of 582.6% and a UTS of 62.3 MPa. The most influential factor on the PA6 sample's UTS and elongation at the weld line was found to be the melt temperature, while the filling time had the least impact. SEM analysis of the fracture surfaces revealed ductile fractures with rough surfaces and grooves, indicative of the weld line areas' bonding quality. These insights pave the way for significant improvements in injection molding conditions, potentially revolutionizing the manufacturing process by enhancing the structural integrity of the weld lines in molded PA6 samples. Weld line defects, commonly occurring during the plastic product manufacturing process, are caused by the merging of two opposing streams of molten plastic. The presence of weld lines harms the product's aesthetic appeal and durability. This study uses artificial neural networks to forecast the ultimate tensile strength of a PA6 composite incorporating 30% glass fibers (GFs). Data were collected from tensile strength tests and the technical parameters of injection molding. The packing pressure factor is the one that significantly affects the tensile strength value. The melt temperature has a significant impact on the product's strength as well. In contrast, the filling time factor has less impact than other factors. According to the scanning electron microscope result, the smooth fracture surface indicates the weld line area's high brittleness. Fiber bridging across the weld line area is evident in numerous fractured GF pieces on the fracture surface, which enhances this area. Tensile strength values vary based on the injection parameters, from 65.51 MPa to 73.19 MPa. In addition, the experimental data comprise the outcomes of the artificial neural networks (ANNs), with the maximum relative variation being only 4.63%. The results could improve the PA6 reinforced with 30% GF injection molding procedure with weld lines. This study investigates the mechanical properties of coconut sawdust powder combined with polypropylene (PP). The effect of compatibility content, wood powder (WP) content, and injection molding parameters on the properties of coconut wood powder composite (WPC) is evaluated. The results could be used to figure out the optimal mechanical properties such as tensile strength, elongation, elastic modulus, and flexural strength by selecting suitable parameters and composition. The bonding between the WP particles and the PP matrix is good, and the WP is uniformly distributed across the composite matrix, as indicated in the scanning electron microscopy (SEM) results. Interestingly, with the presence of the compatibilizer oleamide, increasing the WP content from 20 wt.% to 40 wt.% did not result in WP accumulation in the composite matrix. Notably, at 20 wt.% WP, the elongation is the highest (at 7.40 wt.%), while at 30 wt.% WP, the elastic modulus reaches the highest value. The maximum ultimate tensile strength (UTS) value is obtained at 35 wt.% WP. Higher WP mostly results in greater flexural strength and shore D hardness. At 40 wt.% WP, the WPC achieves its peak shore D hardness of 77.6. The Taguchi results suggest that WP content is the most critical factor in the UTS value of coconut WPCs. The filling pressure ranks second, followed by the packing pressure. Finally, unlike the other characteristics, the melt temperature has a minimal impact on the UTS value. [ABSTRACT FROM AUTHOR]

This paper investigates the effects of heating movement techniques on the properties of low-carbon steel samples that are 3D printed using S20C lamination object manufacturing (LOM). A Tungsten iner gas (TIG) machine and a computer numerical control (CNC) machine were used together to join the steel sheet. The LOM samples were created with a straight-profile, short-profile, cross-profile, and curved-profile. The results indicate that the majority of the samples had a grain size number of 7–9. The samples exhibited an isotropy grain shape. The LOM samples exhibited dimples, which suggests ductility fractures. Pore flaws showed up in the microstructure of the cross-profile and short-profile samples during the LOM process. The samples with curved- and straight-profiles had a better microstructure. In comparison to samples with a short profile and a cross-profile, the samples with a straight-profile and a curved-profile had a superior combination of ultimate tensile strengths (UTSs) and elongation value. The straight- and curved-profiles' greater elongation and tensile strength can be attributed to their improved microstructure and finer grain size. A straight-profile sample with an elongation value of 25.6% and a UTS value of 430 MPa was the ideal LOM sample. Conversely, the weakest sample was the LOM sample with a cross-profile, which had an elongation value of 10.8% and a UTS value of 332.5 MPa. This research could provide further information about the LOM method and the best straight-profile movement strategy. A suitable TIG gun movement strategy could produce a good LOM sample with a good microstructure, tensile strength, and ductility. This study surveys the impacts of injection parameters on the deformation rate of the injected flexure hinge made from ABS, PP, and HDPE. The flexure hinges are generated with different filling time, filling pressure, filling speed, packing time, packing pressure, cooling time, and melt temperature. The amplification ratio of the samples between different injection parameters and different plastic types is measured and compared to figure out the optimal one with a high amplification ratio. The results show that the relationship between the input and output data of the ABS, PP, and HDPE flexure hinges at different injection molding parameters is a linear relation. Changing the material or many injection molding parameters of the hinge could lead to a great impact on the hinge's performance. However, changing each parameter does not lead to a sudden change in the input and output values. Each plastic material has different optimal injection parameters and displacement behaviors. With the ABS flexure hinge, the filling pressure case has the greatest amplification ratio of 8.81, while the filling speed case has the lowest value of 4.81. With the optimal injection parameter and the input value of 105 µm, the ABS flexure hinge could create a maximum average output value of 736.6 µm. With the PP flexure hinge, the melt temperature case achieves the greatest amplification ratio of 6.73, while the filling speed case has the lowest value of 4.1. With the optimal injection parameter and the input value of 128 µm, the PP flexure hinge could create a maximum average output value of 964.8 µm. The average amplification ratio values of all injection molding parameters are 6.85, 5.41, and 4.01, corresponding to ABS, PP, and HDPE flexure hinges. Generally, the ABS flexure hinge has the highest amplification ratios, followed by the PP flexure hinge. The HDPE flexure hinge has the lowest amplification ratios among these plastic types. With the optimal injection parameter and the input value of 218 µm, the HDPE flexure hinge could create a maximum average output value of 699.8 µm. The results provide more insight into plastic flexure hinges and broaden their applications by finding the optimal injection parameters and plastic types. [ABSTRACT FROM AUTHOR]

The influence of welding angle, welding current, travel speed, pulse time, and torch height on the geometry, macrostructure, and mechanical properties of Tungsten Inert Gas (TIG) orbital welding on an SUS 304 stainless steel pipe is investigated in this study. The results show that an electrode angle of 45° produces better weld joints than angles of 30°, 60°, 90°, and 120°. Furthermore, the electrode angle of 30° results in an acceptable weld width but a low depth of penetration (DOP) value. Welding current and weld speed have a significant impact on heat dispersion during TIG welding of an SUS 304 stainless steel pipe. The high welding current may result in blow-hole flaws, particularly near the conclusion of the welding process when heat is accumulated. A long torch height of 2 mm causes unevenness in the weld joints because the arc may be distorted when compared to shorter torch height cases. The pulse time of 0.2 s is too lengthy for a low-welding current situation because it will generate a small weld pool. As a result, the weld pool solidification process speeds up, and porosity emerges in the weld bead. A pulse time of 0.1 s results in a better weld joint. To avoid blow-hole creation, the welding current should be gradually reduced during the process. In addition, the Taguchi results demonstrate that the welding current has the greatest effect on the ultimate tensile strength (UTS) value, followed by welding speed, pulse time, electrode angle, and torch height. Furthermore, the ideal parameters for the UTS value are an electrode angle of 45°, a torch height of 2.0 mm, a welding current of 174 A, a welding speed of 72 mm/min, and a pulse time of 0.3 s. Material additive or 3D printing technology has been recognized as a future manufacturing technique. The mechanical properties of products manufactured using 3D printing technology depend on many factors, especially the printing angle. Resin Castable Wax can be used as a matrix or scaffold for sensor devices in 3D printing. In this study, we explore the effects of printing angle on the mechanical properties of Resin Castable Wax V1 using the 3D stereolithography printing method. The samples are printed at three different printing angles of 0, 45, and 90°. Increasing the printing angle from 0 to 90° improves the tensile and compressive strengths. The sample printed at the angle of 90° shows the highest ultimate tensile and compressive strengths of 23.13 and 124.97 MPa, respectively, and vice versa for the sample printed at 0°. Because of the gradual inclination of the material toward the direction of load force and the larger printing boundary at a higher printing angle, the mechanical properties are improved when the printing angle is increased. This research focuses on the fabrication of plastic flexure hinges from diverse plastics such as ABS, PP, HDPE, and LDPE. To enhance hinge efficiency, the recycling ratios are also investigated. The amplification ratio of different recycle ratios and plastic types are measured. The results show that the input and output displacements of all PP, ABS, and HDPE hinges are linear. The presence of recycled plastics has no impact on this basis. The pure PP, ABS, and HDPE flexure hinges achieve the highest amplification ratios of 5.728, 8.249, and 5.668. The addition of recycled plastics reduces the amplification ratio. This decrease in the amplification ratio, however, is small. At a 25% recycle ratio, the PP, ABS, and HDPE flexure hinges have 12%, 13.3%, and 21.7% lower amplification ratios than the pure plastic hinges. Furthermore, the utilization of recycled plastics may lessen the need for new plastic made from raw materials. With the PP flexure hinge, a maximum input value of 157 µm could result in an output value of 886 µm. At a maximum input value of 115 µm, the ABS flexure hinge could achieve an output value of 833 µm. Finally, a maximum input value of 175 µm might result in an output value of 857 µm when using the HDPE flexure hinge. The average amplification ratio values for all recycling ratios for PP, ABS, and HDPE flexure hinges are, respectively, 5.35, 7.60, and 5.02. The ABS flexure hinge frequently outperforms the PP and HDPE flexure hinge in terms of amplification ratios. Among these plastics, HDPE flexure hinges have the lowest amplification ratio. In general, increasing the thermoplastic polyurethane (TPU) content of the LDPE/TPU blend increases the amplification ratio. The cause could be the TPU's high compatibility with the LDPE polymer. The LDPE/TPU blend hinge offers a broader range of the amplification ratio of 2.85–10.504 than the PP, ABS, and HDPE flexure hinges. It is interesting that changing the blend percentage has a much greater impact on the amplification ratio than changing the recycling ratio. The findings broaden the range of applications for plastic flexure hinges by identifying optimal plastic types. This study examines the impact of injection parameters on the weld line strength of the polyamide 6 and 30% fiberglass (PA6 + 30% FG) composite samples. The effects of filling time, packing time, packing pressure, melt temperature, and mold temperature on the ultimate tensile strength (UTS) and the elongation value of the weld line are investigated. The results reveal that the filling time factor has the lowest influence rate. On the contrary, the packing pressure has the most considerable value of UTS standard deviation, indicating that this factor has a high impact rate. The melt temperature factor has the highest elongation standard deviation, pointing out the strong impact of melt temperature on the elongation value. In reverse, the filling time factor has the lowest elongation standard deviation, showing the low impact of this factor on the elongation value. Increasing the mold temperature enhances the elongation value greatly because a higher temperature generates a better connection in the weld line area. Although the UTS value improves modestly when the mold temperature control system is used, the elongation result from the mold temperature parameter is better than expected. The UTS result from all parameters presents a minor deviation; therefore, it is lower than expected. The optimal strength result from artificial neural networks with genetic algorithm optimization is 85.1 MPa, which is higher than the best experiment result of 76.8 MPa. Scanning electron microscopy (SEM) results show that the interface between the fiberglass and the PA matrix has high adherence. The fracture surface is smooth, indicating that the PA6 + 30% FG composite sample has a high fragility level. Mold heating is an essential process in plastic injection molding. Raising the temperature of the mold before injecting liquefied plastic can ease the mold-filling process. A cooling channel can be used to transport high-temperature fluids for this purpose, such as hot water or oil. This dual purpose is a cost-effective solution for heating the mold because the target temperature is easily achieved using this method. In addition, a conformal cooling channel (CCC) can provide more efficient mold heating than a straight cooling channel. This study used the response surface methodology to determine the optimum CCC shape for heat distribution in a mold, and the simulation results confirmed its optimization. Electrical discharge machining (EDM) is a highly regarded method for producing ultra-precise mechanical parts. In this study, the process parameters of die-sinking EDM using copper electrodes and American Iron and Steel Institute (AISI) P20 tool steel workpieces are optimized for various output responses. The study surveys three input parameters, including Current (I), Pulse on Time (Ton), and Pulse Off Time (Toff). Some statistical methods, such as Taguchi and Analysis of Variance (ANOVA), are applied to find the optimal set of parameters for the output responses, consisting of Material Removal Rate (MRR), Electrode Wear Rate (EWR), and Surface Roughness (SR), and determine the most influential input factor. With the L9 Orthogonal Array (OA), the analytical results demonstrate the optimal parameter set for MRR is I = 6 A, Ton = 120 µs, and Toff = 30 µs, while those optimal values for EWR and SR are I = 2 A, Ton = 120 µs, and Toff = 90 µs and I = 2 A, Ton = 60 µs, and Toff = 30 µs, respectively. The study also indicates that input factor I has the most effect on the output responses, followed by Ton and Toff. Moreover, Grey relational analysis in the Taguchi method is also employed for multi-response optimization. The optimal parameter set for the three output factors is I = 6 A, Ton = 120 µs, and Toff = 60 µs, respectively. This study presents an innovative approach to enhancing weld line strength in advanced polymer injection molding through applying gas-assisted mold temperature control, significantly increasing mold temperature beyond typical values observed in conventional processes. We investigate the effects of various heating times and frequencies on the fatigue strength of Polypropylene (PP) samples and the tensile strength of Acrylonitrile Butadiene Styrene (ABS) composite samples at different Thermoplastic Polyurethane (TPU) percentages and heating times. Using gas-assisted mold heating, mold temperatures exceeding 210 °C are achieved, which represents a significant advancement compared to the standard mold temperatures of less than 100 °C. As a result, the fatigue strength of the PP sample with mold heating at 15 s shows a remarkable increase of up to 5.4 times at 5 Hz compared to the sample without mold temperature control. Moreover, ABS/TPU blends with 15 wt.% TPU exhibit the highest ultimate tensile strength (UTS) value of 36.8 MPa, while blends with 30 wt.% TPU have the lowest UTS value of 21.3 MPa. This advancement demonstrates the potential for improved welding line bonding and fatigue strength in manufacturing. Our findings reveal that increasing the mold temperature before injection results in higher fatigue strength in the weld line, with the TPU percentage having a more significant influence on the mechanical properties of ABS/TPU blends than heating time. This study aims to explore the effects of Thermoplastic Polyurethane (TPU) content on the weld line properties of Polypropylene (PP) and Acrylonitrile Butadiene Styrene (ABS) blends. In PP/TPU blends, increasing the TPU content results in a significant decrease in the PP/TPU composite's ultimate tensile strength (UTS) and elongation values. Blends with 10 wt%, 15 wt%, and 20 wt% TPU and pure PP outperform blends with 10 wt%, 15 wt%, and 20 wt% TPU and recycled PP in terms of UTS value. The blend with 10 wt% TPU and pure PP achieves the highest UTS value of 21.85 MPa. However, the blend's elongation decreases due to the poor bonding in the weld line area. According to Taguchi's analysis, the TPU factor has a more significant overall influence on the mechanical properties of PP/TPU blends than the recycled PP factor. Scanning electron microscope (SEM) results show that the TPU area has a dimple shape on the fracture surface due to its significantly higher elongation value. The 15 wt% TPU sample achieves the highest UTS value of 35.7 MPa in ABS/TPU blends, which is considerably higher than other cases, indicating good compatibility between ABS and TPU. The sample containing 20 wt% TPU has the lowest UTS value of 21.2 MPa. Furthermore, the elongation-changing pattern corresponds to the UTS value. Interestingly, SEM results present that the fracture surface of this blend is flatter than the PP/TPU blend due to a higher compatibility rate. The 30 wt% TPU sample has a higher rate of dimple area than the 10 wt% TPU sample. Moreover, ABS/TPU blends gain a higher UTS value than PP/TPU blends. Increasing the TPU ratio mainly reduces the elastic modulus of both ABS/TPU blends and PP/TPU blends.
