In the past few years, mass spectrometry imaging (MSI) has brought many new inspirations to plant research. However, current MSI experiments usually include only a single batch of samples, casting doubts on the reproducibility of phytochemical distribution across different batches. Consequently, MSI has seldom been applied to conduct species discrimination. In this experiment, MSI was employed to discriminate between two taxonomically similar plants, Scutellaria baicalensis Georgi and Scutellaria rehderiana Diels. A new concept termed a "spatial marker" was proposed in this article, which referred to the phytochemical marker that presented both intraspecies similarity and interspecies dissimilarity. Multiple batches of S. baicalensis and S. rehderiana were analyzed using MSI, proving that the authentication protocol using spatial markers was reliable and reproducible. The observed spatial markers were further identified using on-tissue tandem mass spectrometry and liquid chromatography coupled with mass spectrometry. Additionally, the spectral data collected from MSI were utilized to set up algorithm models for species discrimination. External validation confirmed that the established random forest model was extrapolated well to unknown samples. Radix isatidis (Banlangen) is a widely used traditional Chinese medicine for treating fever and removing toxic heat. Pharmacological studies have indicated that (R)-goitrin (epigoitrin) is one of the main constituents accounting for its antiviral activity, while (S)-goitrin (goitrin) is known as an antithyroid factor. To better control the quality of radix isatidis and its formulations, it is imperative to enantiomerically determine the contents of R- and S-goitrin. In this study, an enantioselective method based on reversed phase chromatography was developed for the assay of (R, S)-goitrin enantiomers. Optimum separation was obtained on an S-Chiral A column (4.6 mm × 250 mm, 5 μm) using methanol/water (30 : 70, v/v) as the mobile phase. After validation, the method was applied to quantify the enantiomers in Banlangen granules, which is the most prescribed commercial formulation of radix isatidis. Compared to enantioselective resolution approaches based on normal phase chromatography, the new method could be conveniently performed using regular reversed phase high-performance liquid chromatography (RP-HPLC) equipment and was found to be more suitable for the enantioselective quality control of water-extracted and soluble medicines. The accuracy of near-infrared quantitative models for traditional Chinese medicine is restricted by the matrix, low-concentration markers, and overlapping spectral bands. Competitive adaptive reweighted sampling, a recently developed algorithm, selects an optimal combination of multicomponent spectral data. In this article, the performance of this method was evaluated through the analysis of traditional Chinese medicine. The near-infrared spectra of the pharmaceutics were obtained and the concentration of puerarin was determined. After optimization of spectral pretreatment methods, competitive adaptive reweighted sampling was performed in each dataset to select key wavenumbers. Sixty-eight, thirty, and eight variables were selected for the raw material, intermediate product, and final product, respectively. Partial least squares models were constructed based on the selected variables. Enhanced accuracy was obtained using the competitive adaptive reweighted sampling-coupled models. The results indicated that competitive adaptive reweighted sampling improved the accuracy and simplified calibration while offering a new approach for the rapid and nondestructive analysis of traditional Chinese medicine. Like chemical drugs, research and development of herbal medicine also have a need to resolve enantiomers. To help illustrating the antiviral bioactivity of Isatidis Radix, a widely used traditional Chinese medicine (TCM), supercritical fluid chromatography (SFC) was used for analytical and preparative separation of (R,S)-goitrin, which was reported as the active ingredient of the herbal. Improved resolution was achieved on Chiralpak IC column, using acetonitrile as the organic modifier, representing a tenfold increase in speed, compared to the previous normal phase HPLC (NPLC) method. The newly developed chromatographic method was validated in terms of linearity, precision, limit of detection (LOD), and limit of quantitation (LOQ). Scale-up purification of (R)-goitrin and (S)-goitrin was performed on a preparative column with >90% total recovery. The absolute stereochemical assignment of the purified isomers was determined through optical rotation study. Semi-transparent reactive microgel in nanosize has been prepared by modified microemulsion polymerization using a common emulsifier, crosslinking agent and functional monomer. The microgels are translucent reactive nanoparticles, with the size of 40–100 nm, consisting of inner-crosslinked polymer up to 40%. FT-IR proved the functional groups, such as epoxy and hydroxy, are on the surface of the microgel nanoparticles. Rheological detection demonstrated the apparent pseudoplasticity of the non-aqueous microgel dispersion prepared by the phase transfering from the O/W microlatexes. [ABSTRACT FROM AUTHOR]

Perilla leaf oil (PLO) is a global premium vegetable oil with abundant nutrients and substantial economic value, rendering it susceptible to potential adulteration by unscrupulous entrepreneurs. The addition of cinnamon oil (CO) is one of the main adulteration avenues for illegal PLOs. In this study, new and real-time ambient mass spectrometric methods were developed to detect CO adulteration in PLO. First, atmospheric solids analysis probe tandem mass spectrometry combined with principal component analysis and principal component analysis-linear discriminant analysis was employed to differentiate between authentic and adulterated PLO. Then, a spectral library was established for the instantaneous matching of cinnamaldehyde in the samples. Finally, the results were verified using the SRM mode of ASAP-MS/MS. Within 3 min, the three methods successfully identified CO adulteration in PLO at concentrations as low as 5% v/v with 100% accuracy. The proposed strategy was successfully applied to the fraud detection of CO in PLO. • Detect CO-adulterated PLO by Radian ASAP with pattern recognition and library matching. • First time ASAP-MS/MS was used for accuracy verification. • As low as 5% cinnamon oil adulteration in perilla leaf oil could be detected. • Three ASAP methods were validated against each other with consistent results. • High specificity and reliability were achieved by the new assay.
