Background: Gingivitis in response to biofilm formation may exhibit different trajectories. The purposes of the present study were to characterize the composition of the supragingival microbiota and salivary cytokine and protein levels in healthy individuals with different gingivitis patterns, to test the hypothesis that manifestations of gingivitis associate with specific profiles in terms of supragingival microbiota, salivary cytokines, and proteins. Methods: Forty orally and systemically healthy individuals refrained from all oral hygiene procedures for a period of 14 days, followed by a resolution period of 14 days with regular oral care. Supragingival plaque level and bleeding on probing (BOP) were recorded, and supragingival plaque as well as saliva samples were collected at baseline, day 14, and day 28. Based on change in BOP% from baseline to day 14, rapid (n = 15), moderate (n = 10), and slow (n = 15) responders were identified. Supragingival microbiota composition, salivary cytokine, and protein levels were compared between groups at baseline, day 14, and day 28. Results: A significantly higher baseline abundance of Capnocytophaga, Eikenella, and Campylobacter species were recorded in rapid responders, whereas a significantly higher baseline abundance of Streptococcus species were detected in slow responders. Slow responders expressed a high degree of resilience, with minimal difference in microbial composition at baseline and after 14 days of resolution (day 28). On the contrary, significant differences in relative abundance of members of the core microbiota, Streptococcus, Actinomyces, and Rothia species, was noted in baseline samples versus day 28 samples in rapid responders. Comparable baseline cytokine and protein levels were recorded in all groups. Conclusion: Supragingival microbiota composition, but not saliva cytokine and protein profiles, seems to influence the extent of the inflammatory response during development of gingivitis in systemically healthy individuals. Background: Social isolation and feelings of loneliness are very prevalent in people with neurodegenerative diseases and are associated with a lower quality of life and other negative outcomes. These problems were increased during the COVID-19 pandemic resulting in initiatives to address social isolation. Given the potential benefits of nature-based and animal-assisted intervention strategies (NATAIS), it is crucial to further investigate if and how these strategies might minimize negative effects of social isolation and feelings of loneliness in this population. Therefore, the aim of this project was to develop a research agenda for NATAIS in people with neurodegenerative diseases, especially during challenging times, such as pandemics. Methods: This article outlines the process and results of a group concept mapping procedure aimed at developing a research agenda based on a logic model. In total, 19 work group members participated through a combination of in-person and online group meetings. Additionally, face-to-face group sessions were held at two international scientific conferences, during which feedback was solicited from 12 experts in the field of NATAIS and psychogeriatrics. Results: The group concept mapping procedure resulted in 14 clusters describing various future research topics, which were further refined and detailed during group discussions. The remaining eleven clusters, encompassing important research themes within the field of NATAIS, were organized into a logic model and summarized into the research agenda. The overarching cluster 'ethical issues, possible risk factors, and their solutions' was considered the most relevant during times of increased social isolation, such as during a pandemic, along with the necessity for more accessible NATAIS. Conclusions: This project resulted in a research agenda, directing future research and fostering collaboration between practitioners and researchers in the field of NATAIS. Such an enhanced partnership between science and practice has the potential to significantly contribute to the well-being of people with neurodegenerative diseases, in their daily lives and also during pandemics. Data from systematic reviews and meta-analyses show that probiotics positively impact clinical parameters of oral diseases such as gingivitis, dental caries, and periodontitis. However, the working mechanism of probiotics is not fully understood, but is hypothesized to be mediated by direct and indirect interactions with the oral microbiota and the human host. In the present narrative review, we focused on the microbiological effect of probiotic supplements based on data retrieved from randomized clinical trials (RCTs). In addition, we assessed to what extent contemporary molecular methods have been employed in clinical trials in the field of oral probiotics. Multiple RCTs have been performed studying the potential effect of probiotics on gingivitis, dental caries, and periodontitis, as evaluated by microbial endpoints. In general, results are conflicting, with some studies reporting a positive effect, whereas others are not able to record any effect. Major differences in terms of study designs and sample size, as well as delivery route, frequency, and duration of probiotic consumption, hamper comparison across studies. In addition, most RCTs have been performed with a limited sample size using relatively simple methods for microbial identification, such as culturing, qPCR, and DNA–DNA checkerboard, while high-throughput methods such as 16S sequencing have only been employed in a few studies. Currently, state-of-the-art molecular methods such as metagenomics, metatranscriptomics, and metaproteomics have not yet been used in RCTs in the field of probiotics. The present narrative review revealed that the effect of probiotic supplements on the oral microbiota remains largely uncovered. One important reason is that most RCTs are performed without studying the microbiological effect. To facilitate future systematic reviews and meta-analyses, an internationally agreed core outcome set for the reporting of microbial endpoints in clinical trials would be desirable. The aim was to test if probiotics counteract oral dysbiosis during 14 days of sugar stress and subsequently help restore oral homeostasis. Eighty healthy individuals received either probiotics (n = 40) or placebo lozenges (n = 40) for 28 days and rinsed with a 10% sucrose solution 6–8 times during the initial 14 days of the trial. Saliva and supragingival samples were collected at baseline, day 14, and day 28. Saliva samples were analyzed for levels of pro-inflammatory cytokines, albumin, and salivary enzyme activity. The supragingival microbiota was characterized according to the Human Oral Microbiome Database. After 14 days of sugar stress, the relative abundance of Porphyromonas species was significantly higher (p = 0.03) and remained significantly elevated at day 28 in the probiotic group compared to the placebo group (p = 0.004). At day 28, the relative abundance of Kingella species was significantly higher in the probiotic group (p = 0.03). Streptococcus gordinii and Neisseria elongata were associated with the probiotic group on day 28, while Streptococcus sobrinus was associated with the placebo group on day 14 and day 28. On day 28, the salivary albumin level was significantly lower in the probiotic group. The present study demonstrates a potential stabilizing effect on the supragingival microbiota mediated by consumption of probiotics during short-term sugar stress. [ABSTRACT FROM AUTHOR]

The present study aims to test whether probiotics protect against experimental gingivitis incited by 14 days of oral hygiene neglect and/or subsequently support the restoration of oral homeostasis. Eighty systemically and orally healthy participants refrained from oral hygiene procedures for 14 days, followed by 14 days with regular oral hygiene procedures. Additionally, participants consumed either probiotics (n = 40) or placebo (n = 40) throughout the trial. At baseline, day 14, and day 28, supragingival plaque score and bleeding-on-probing percentage (BOP %) were registered, and supragingival plaque and saliva samples were collected. The supragingival microbiota was characterized using 16S sequencing, and saliva samples were analyzed for levels of pro-inflammatory cytokines and proteases. At day 28, the relative abundance of Lautropia (p = 0.014), Prevotella (p = 0.046), Fusobacterium (p = 0.033), and Selenomonas (p = 0.0078) genera were significantly higher in the placebo group compared to the probiotics group, while the relative abundance of Rothia (p = 0.047) species was associated with the probiotics group. Streptococcus sanguinis was associated with the probiotics group, while Campylobacter gracilis was associated with the placebo group. No difference was observed in salivary cytokines, albumin, or any enzyme activity. In the last few decades, the number of people living in fire-prone ecosystems has increased, placing more people and private property at risk to future fire events. Substantial research has demonstrated consistent public support for the use of prescribed fires in fuel-reduction efforts; however, continuing public concern regarding smoke emissions and negative air quality impacts exists. To date, limited research has specifically examined public attitudes toward smoke emissions. In this study, we use a mail-back or internet survey to assess citizen information seeking behaviors regarding smoke emissions in four communities in high fire risk areas. Path analysis was used to apply the risk information seeking and processing (RISP) model to examine factors that motivate people to seek information relevant to smoke emissions. We find that residents were concerned about smoke emissions and believed that they needed more information. Residents' intentions to seek information were influenced by information (in)sufficiency, the number of sources used, and smoke acceptability, among other factors. Findings suggest that currently available information resources on smoke may not be sufficient to meet residents' information needs, particularly for those most motivated to learn more about emissions. D'importants travaux de recherche ont demontre que l'utilisation du brulage dirige pour reduire les combustibles recoit un appui constant du public. Cependant, les preoccupations du public concernant les impacts sur les emissions de fumee et la mauvaise qualite de l'air restent presentes. A ce jour, peu d'etudes ont specifiquement ete consacrees aux attitudes du public face aux emissions de fumee. Dans cette etude, nous avons effectue un sondage en envoyant un questionnaire par la poste a retourner dans le courrier ou a completer via Internet pour evaluer l'information que possedent les citoyens a la recherche d'un comportement face aux emissions de fumee dans quatre communautes situees dans des regions a haut risque de feu. L'analyse des pistes causales a ete utilisee pour appliquer le modele de traitement de l'information et de recherche sur le risque (RISP) afin d'etudier les facteurs qui motivent les gens a chercher de l'information au sujet des emissions de fumee. Nous avons constate que les residents sont preoccupes par les emissions de fumee et croient qu'ils ont besoin de plus d'information. Les intentions des residents de chercher plus d'information ont ete influencees entre autres par l'insuffisance ou non d'information, le nombre de sources utilisees et l'acceptabilite de la fumee. Les resultats indiquent que les ressources documentaires actuellement disponibles ne sont peut-etre pas suffisantes pour satisfaire les besoins d'information des residents, particulierement ceux qui sont les plus motives a en apprendre davantage au sujet des emissions. Frequent intake of free sugars is a major risk factor for dental caries, but the immediate influence of sugar intake on the supragingival microbiota remains unknown. We aim to characterize the effect of 14 days of sugar rinsing on the supragingival microbiota. Forty orally and systemically healthy participants rinsed their mouth with a 10% sucrose solution, 6–8 times a day, for 14 days, followed by 14 days without sugar stress. Supragingival plaque samples were collected at baseline, and after 14, and 28 days. The supragingival microbiota was analyzed using 16S rDNA sequencing. Taxonomic classification was performed using the Human Oral Microbiome Database. After 14 days of sugar stress induced by the daily sugar rinses, a significant loss of α-diversity (p = 0.02) and a significant increase in the relative abundance of Actinomyces (6.5% to 9.6%, p = 0.006) and Corynebacterium (6.2% to 9.1%, p = 0.03) species were recorded. In addition, a significant decrease in Streptococcus (10.3% to 6.1%, p = 0.001) species was observed. Sugar-mediated changes returned to baseline conditions 14 days after the last sugar rinse. The present study shows that temporary sugar stress induces loss of diversity and compositional changes to the supragingival microbiota, which are reversible if oral care is maintained. A current focus of our research is dedicated to investigating tools that can be used to couple pancreas‐on‐a‐chip systems on‐line with mass spectrometry. To enable on‐line‐cleavage of insulin´s disulfide bonds, we coupled an inter‐chain cleavage reactor with a nano liquid chromatography‐mass spectrometry system. The reactor featured a high intensity 254 nm photochemical mercury quartz lamp radiating upon an ultraviolet transparent tubing where acetone and isopropanol form hydroxyalkyl radicals which cleave insulin disulfide bridges. The cleavage allowed for robust fragmentation mass spectra of insulin using nanospray/Q‐Exactive mass spectrometry. A nano liquid chromatography separation column was placed downstream to the reactor. We found that for the nano liquid chromatography format, a silica monolith provided substantially less carry‐over and improved chromatographic performance to the investigated particle‐packed columns. The photochemical reactor/silica monolithic nano liquid chromatography/nanospray/Q‐Exactive mass spectrometry was compatible with cell culture medium, and will be further investigated for on‐line coupling with pancreas‐on‐a‐chip systems. Objectives: Sleep disturbances are common in people with dementia and increase with the severity of the disease. Sleep disturbances are complex and caused by several factors and are difficult to treat. There is a need for more robust and systematic studies dealing with sleep disturbances in older people with dementia. The aim of this study was to investigate effects from robot-assisted (Paro) group activity on sleep patterns in nursing home (NH) residents with dementia.Design: A cluster randomized controlled trial.Setting: Special care units in 10 NH in Norway.Participants: A total of 60 participants over 65 years with dementia were recruited. Thirty participants were recruited to the intervention group and 30 participants to the control group.Intervention: Participants participated in group activity with Paro for 30 minutes twice a week over 12 weeks or in control group (treatment as usual).Measurements: Sleep-wake patterns were assessed objectively by 7 days of wrist actigraphy before and after the intervention. Data were collected between March 2013 and September 2014. Data were analyzed using mixed models.Results: Positive effects on change in sleep were found in the intervention group as compared with the control group. The intervention group increased percentage of sleep efficiency, increased the amount of total sleep time and reduced number of nocturnal awakenings. In addition, a significant effect was found in reduced awakenings after sleep onset.Conclusions: Social stimulation through engaging group activity could contribute to improved sleep in people with dementia in NH. Increased sleep efficiency and total sleep time, in addition to fewer night awakenings, affect central indicators of good sleep quality across the life span. Improved sleep quality will also affect quality of life and comorbidities in vulnerable groups. We believe group activity with Paro to be an accessible and feasible non-pharmacological treatment for those who enjoy Paro. [ABSTRACT FROM AUTHOR]

Nano liquid chromatography (nanoLC), with columns having an inner diameter (ID) of ≤100 μm, can provide enhanced sensitivity and enable analysis of limited samples. NanoLC has become an established tool in omics research, and is gaining ground in other applications as well. There are several variants and formats of nanoLC columns, including packed columns, monoliths, open tubular columns, and the pillar array format. Most applications are done with packed columns, while e.g. the monolith and open tubular columns are still less established as routine tools. The pillar array format is a new variant with excellent resolution and low backpressure, and has recently been commercialized and used for bio-applications. The need for meaningful activities that enhance engagement is very important among persons with dementia (PWDs), both for PWDs still living at home, as well as for PWDs admitted to a nursing home (NH). In this study, we systematically registered behaviours related to engagement in a group animal-assisted activity (AAA) intervention for 21 PWDs in NHs and among 28 home-dwelling PWDs attending a day care centre. The participants interacted with a dog and its handler for 30 minutes, twice a week for 12 weeks. Video-recordings were carried out early (week 2) and late (week 10) during the intervention period and behaviours were categorized by the use of an ethogram. AAA seems to create engagement in PWDs, and might be a suitable and health promoting intervention for both NH residents and participants of a day care centre. The increase in area burned by wildfire has simultaneously brought increased concern about smoke impacts, both from wildfires and fires intentionally set to manage landscapes. Public concern about the potential health and other impacts of smoke can cause apprehension among managers who are considering prescribed burns, some to the point that they choose not to burn. Yet, very little is known about public acceptance of smoke. Using results from public surveys in four states, this paper explores the factors that influence public acceptance of smoke from six types of fire (wildfire, prescribed fire, agricultural burns, managed fire, pile burns and burns on private lands) as well as aggregate acceptance across all fire types. Overall, the public is generally accepting of smoke, particularly for fire types that are either viewed as uncontrollable (wildfire) or providing benefits to broader society rather than individuals. However, a sizable minority does not accept smoke. Factors that influence acceptance of smoke vary by fire type, but several coincided for multiple types of fire, including both health and non-health related risk, confidence in managing agencies, beliefs about benefits of prescribed fire use and rural living, among others.
