Abstract

Summary. Grapevine Pinot gris virus (GPGV) is a recently identified trichovirus infecting grapevines. Despite wide distribution, there is limited available information on epidemiology, transmission, and associated symptoms of grapevine leaf mottling and deformation. Occurrence and genetic diversity of GPGV variants were surveyed in an organically cultivated Hungarian vineyard that was planted between 1996 and 2014. Sequence analysis demonstrated the widespread presence and high variability of GPGV, and according to phylogenetic analyses, the Hungarian virus isolates were clas- sified into three groups. Most of the identified variants clustered with the representa- tive asymptomatic isolates, but all isolates from one grapevine cultivar grouped with representative isolates of clade B. 20. Detection of ' Candidatus Phythoplasma prunorum' in Apricot Trees and its Associated Psyllid Samples.

Abstract

'Candidatus Phytoplasma prunorum' is causing ever increasing economic losses through the decline of apricot trees in European countries, e.g., Hungary. In this study, the pathogen was identified from plant tissues and insects by nested-PCR. The insect species were identified via morphology and molecular methods. The incidence of the pathogen was 29.6% in randomly selected apricot trees. Most of the infected trees with symptoms died within a year. These results show that phytoplasma is significantly present and causes damage in the investigated plantations. The only known insect vector of this phytoplasma is the plum psyllid, Cacopsylla pruni, which was regularly encountered in the sampled apricot orchards and in their surroundings. In a two-year study, several adults among the sampled specimens were observed to be infected by the pathogen. This observation further confirms the role of the plum psyllid in vectoring the phytoplasma. All the sampled plum psyllid adults belonged to the 'B' biotype. Besides C. pruni, Cacopsylla crataegi was abundant in the samples. Several adults of the latter species were also infected by the pathogen 'Ca. Phytoplasma prunorum'. The rates of occurrence of this phytoplasma in male and female adults of the two psyllid species appeared to be similar. Abstract

The paper discusses the identification of the empirical, partially intelligible white noise processes generated by deterministic numerical algorithms. The introduced fuzzy‐random complementary approach can identify the inner hidden correlational patterns of the empirical white noise process if the process has a real hidden structure of this kind. We have shown how the characteristics of autocorrelated white noise processes change as the order of autocorrelation increases. Based on this approach, the original empirical white noise process transformed by the autocorrelation operator can be considered to be random data series (randomlikeness), and at the same time, it has function‐like characteristics (functionlikeness), as well. We approach the analysis of the mentioned complementarity by modeling the autocorrelation functions of the empirical white noise processes using tensor product (TP) model transformation. Abstract

International audience; Impact of "bois noir" disease (BN) on performance of 'Chardonnay' grown in Eger (Hungary) is reported from plant (vegetative and reproductive parameters, fruit composition) to wine (wine quality, sensory analysis). BN-associated chlorophyll degradation and leaf rolling resulted in a loss in functioning surface of the canopy. Lack of lignification and non-viable buds evoked unpredictable crop production. Severity and extent of reduction in production varied between years, but the crop loss was always above 53%. In the case of BN-affected grapevines the reduced quantity resulted in a low quality of grape bunch composition, which enhanced unfavourable outcomes in wines. Differences in fruit composition and wine quality between healthy and BN affected grapevines were most pronounced in years with optimal weather conditions and the negative effect was covered up in poor years. In wine regions where mechanical harvesting is applied, harvesting of phytoplasma diseased vineyards can therefore not be selective and qualitative. According to the three years study presented BN induce severe loss, and BN-affected plants compromise the yield and wine quality, and therefore the overall profitability of a vineyard.

Abstract

International audience; Little is known about the mechanisms of insects - phytoplasma interactions which are driving the ecological diversification of phytoplasmas. Phytoplasma surface proteins play an important role in phytoplasma life cycle and polymorphism of this protein might determine the transmission ability of different insect species. Strains of Candidatus Phytoplasma solani' from several European countries, based on variable membrane protein gene (stamp), were classified into four distinct phylogenetic clusters which clusters are present in different geographical areas. This may reflect the distribution of the different known insect vectors and the adaptation of the various 'Ca. P. solani' strain to a specific vector. To detect STAMP protein, monoclonal antibody 2A10 is available. This antibody was produced against StolburC, which is a representative strain of stamp clusters I. In this work we performed heterologous expression antigenic membrane proteins STAMP from different 'Ca. P. solani' genetic clusters and tested their recognition by the 2A10 monoclonal antibody.

Summary: Cell wall peroxidases and plasma membrane‐localized NADPH oxidases are considered to be the main sources of the apoplastic oxidative burst in plants attacked by microbial pathogens. In spite of this established doctrine, approaches attempting a comparative, side‐by‐side analysis of the functions of extracellular reactive oxygen species (ROS) generated by the two enzymatic sources are scarce. Previously, we have reported the role of Arabidopsis NADPH oxidase RBOHD (respiratory burst oxidase homologue D) in plants challenged with the necrotrophic fungus Alternaria brassicicola. Here, we present results on the activity of apoplastic class III peroxidases PRX33 (At3g49110) and PRX34 (At3g49120) investigated in the same Arabidopsis–Alternaria pathosystem. ROS generated by Arabidopsis peroxidases PRX33 and PRX34 increase the necrotic symptoms and colonization success of A. brassicicola. In addition, the knockdown of PRX33 and PRX34 transcript levels leads to a reduced number of host cells showing an extracellular burst of ROS after inoculation with A. brassicicola. Our results also reveal an age‐dependent transcript distribution of ROS‐producing peroxidase and NADPH oxidase enzymes, and some potential new components of the RBOHD, PRX33 and PRX34 signalling networks. 41. First Occurence of Fire Blight on Apricot (Prunus armeniaca) in Hungary.

Abstract

During July 2012, a severe unusual disease symptom was observed on young shoots on apricot (Prunus armeniaca 10/13 hybrid) in the city of Pomáz, near Budapest. The naturally infected shoots showed typical symptoms of fire blight including terminal shoots with brown to black necrotic lesions. Symptoms were the same as fire blight symptoms reported from other hosts and locations. The first occurrence of fire blight on an apricot tree in Europe was recorded in Czech Republic in 2011. Samples of the leaves and shoots with symptoms were macerated and spread on King's medium B. After 24 hours of incubation at 26 °C, bacteria morphologically similar to E. amylovora were detected. Isolate induced hypersensitive reaction on tobacco (Nicotiana tabacum L. 'White Burley') leaves. Biochemical test was also used for identification, and the result of API 20E kit (Biomérieux, Marcy l'Etoile, France), demonstrate that the bacterium belongs to Enterobacteriaceae family. A pathogenicity tests were positive on young apricot shoots and immature fruits. For molecular identification of the pathogen the 16S rDNA region was amplified from isolate Ea-ApricotPo1 with a general bacterial primer pair (63f forward and 1389r reverse). The PCR products were cloned into a pGEM T-Easy plasmid vector (Promega, Madison, WI USA) and were transformed into Escherichia coli DH5α cells. A recombinant plasmid (2A2.5) was sequenced by M13 forward and reverse primers. The sequence was deposited in GenBank (Accession No. HF546214) and showed 99-100% sequence homology with a number of E. amylovora isolates, including type strain FN666575 with 100% similarity. On the basis of the symptoms, colony morphology, biochemical tests, and 16S rDNA sequence homology, the pathogen was identified as E. amylovora. It is still a difficult problem to solve for a suspension system simultaneously holding the body of the car in comfort and executing requirements imposed for other safety systems like ABS, ESP, steer-by-wire etc. Passive suspension systems are unlikely to provide a solution, and therefore the introduction of semi-active suspensions in practical use is necessary. A possible solution could be a permanent magnetic (PM) synchronous tube generator that can operate as a controllable shock absorber parallel with energy recuperative operation. Abstract

In Hungary, from the beginning of the 19th century, horse-chestnut trees have been planted widely and are popular ornamental trees in public parks, along streets, and in gardens. In the summer of 2015, longitudinal cracks on the trunk and branches and the intensive oozing of brown liquid were observed from a wound in a horse-chestnut tree in a park in Budapest. Some years later, in 2018 and 2019, the same symptoms were found in trees in other locations in Budapest. Several bacteria were reported that induce similar symptoms, including cracks and cankers on the bark of trunks and branches and sticky, white, red, brown, or black oozing. These pathogens belong to the genera Brenneria and Lonsdalea. Bark and exudate samples were taken with the aim of identifying the causal agent by conventional and molecular methods. Our results confirmed that the bacteria isolated from Aesculus hippocastanum trees belong to the genus Brenneria and phylogenetic analysis of the 16S rRNA gene region proved to have the closest phylogenetic relation with the Brenneria nigrifluens strains. Abstract

The paper presents an experimental system for detection and prevention of the unintended lane-departure of road vehicles. The system is realised by a co-operation of a drive stability system and a computer vision system. The detection algorithm does not depend on the type of the lane-boundaries (lane-marks, paved lane-boundary, transition from asphalt to grass) and filters out information marks which do not signalise lane-boundaries. If lane-departure is predicted, then intervention to the vehicle's motion is applied by unilateral braking. Abstract

This paper presents a computer-controlled suspension design for tractors and semi-trailers cabs and driver's seats. Two separate models representing the vehicle with a cab suspension and the seat with an operator modeled as a lumped mass system are presented. The vehicle is represented by an 8 degrees-of-freedom (DOF) model, which includes a 3-axle tractor semi-trailer and a separated cab with suspensions. The seat/operator system is modeled by a 6 DOF model to simulate the vertical dynamic response of the driver including the seat suspension. The cab is supported by a set of controllable suspensions at both of the front and rear ends of the cab. The seat is attached to the floor of the cab by another controllable suspension. Each controllable suspension is modeled by a set of linear spring and non-linear damper whose coefficient is time dependent. First, a sky-hook based semi-active controller was designed to seek optimum cab suspension. Then, the second state controller was used with the cab's velocity and displacement as input to minimize the parameters of the seat/operator model. The seat suspension parameters were optimized to yield the best ride quality under the prescribed input road conditions. Abstract

Cucumber mosaic virus (CMV, Cucumovirus, Bromoviridae) is an economically significant virus infecting important horticultural and field crops. Current knowledge regarding the specific functions of its movement protein (MP) is still incomplete. In the present study, potential post-translational modification sites of its MP were assayed with mutant viruses: MP/S28A, MP/S28D, MP/S120A and MP/S120D. Ser28 was identified as an important factor in viral pathogenicity on Nicotiana tabacum cv. Xanthi, Cucumis sativus and Chenopodium murale. The subcellular localization of GFP-tagged movement proteins was determined with confocal laser-scanning microscopy. The wild type movement protein fused to green fluorescent protein (GFP) (MP-eGFP) greatly colocalized with callose at plasmodesmata, while MP/S28A-eGFP and MP/S28D-eGFP were detected as punctate spots along the cell membrane without callose colocalization. These results underline the importance of phosphorylatable amino acids in symptom formation and provide data regarding the essential factors for plasmodesmata localization of CMV MP. 50. Role of Oxalis corniculata L. as plant virus reservoir with special regard to Tomato Spotted Wilt Virus (TSWV-RB) strain occurrence in rock-wool cultivation in Hungary

Abstract

The creeping wood sorrel (Oxalis corniculata L.) in Central Europe occurs mainly in anthropogenic areas, where it grows in parks and landfill sites or in protected cultivation sites like greenhouses and nurseries. In Hungary O. corniculata has been spreading since the 19th century. In glass- and greenhouses it is the only weed species that was able to settle and flourish on hydroponic rock-wool and coconut fibre growing media. Among virus pathogens of forced tomato (LYPES) and paprika crops (CPSAN) in Hungary the Tomato Spotted Wilt Virus (TSWV) has become an important one, severely affecting the plants and on occasions causing 100% yield losses. The spread of the virus was largely assisted by its effective vector the Western Flower Thrips (Frankliniella occidentalis Pergande). Initially the control of the disease was based on the control of the thrips, later, virus-resistant varieties were introduced. This however proved insufficient to fight the disease as it was discovered, quite recently, that in certain white pepper (CPSAN) varieties the virus was able to break up the resistance. According to data of the year 2012, those varieties that were considered resistant, showed up to 50% virus infection. Considering the wide spread of O. corniculata and increasing occurrence of the resistance-breaking TSWV isolates, surveys in greenhouses were conducted to examine whether the weed could serve as reservoir for TSWV. Samples were collected of the virus infected crop plants and O. corniculata growing in the same coconut fibre cubes. The samples were examined using test-plants, serological and RT-PCR methods. In result it was found that the symptoms were indeed caused by resistance-breaking TSWV isolates. However, no virus was found in the suspected O. corniculata samples. Therefore it was concluded that in this particular case the O. corniculata cannot be considered reservoir for the virus, in the hydroponic culture.
