Variants of concern (VOCs) of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS‐CoV‐2) exhibit high infectivity due to mutations, particularly in the spike protein, that facilitate enhanced binding of virus to human angiotensin‐converting enzyme 2 (hACE2). The D614G mutation, situated in S1‐domain, promotes the open conformation of spike protein, augmenting its interaction with hACE2. Activated water neutralizes pathogens by damaging biological molecules; however, its effect on mutated SARS‐CoV‐2 or VOCs requires further exploration. Here, the efficacy of nitric oxide (NOx)‐plasma activated water (PAW) in inhibiting infections by SARS‐CoV‐2 pseudovirus expressing D614G‐mutated spike protein is investigated, which serves as a model for mutated SARS‐CoV‐2. Results demonstrated high prevalence of D614G mutation in SARS‐CoV‐2 and its VOCs. NOx‐PAW is non‐toxic to cells at high concentration, inhibiting infection by 71%. Moreover, NOx‐PAW induced structural changes in S1‐domain of spike protein, reducing its binding affinity and lowering clathrin‐mediated endocytosis‐related gene expression. Additionally, in silico analysis revealed NOx species in NOx‐PAW played key role in impairing S1‐domain function of the mutated SARS‐CoV‐2 pseudovirus by interacting directly with it. Collectively, these findings reveal the potent inactivation ability of PAW against mutated SARS‐CoV‐2 and suggest its potential application in combating emerging variants of SARS‐CoV‐2 and other viral threats. The vaccine is a premier healthcare intervention strategy in the battle against infectious infections. However, the development and production of vaccines present challenges in terms of complexity, cost, and time consumption. Alternative methodologies, such as nonthermal plasma and plant‐based technologies, have emerged as potential alternatives for conventional vaccine manufacturing processes. While plasma‐based approaches offer a rapid and efficient pathogen inactivation method devoid of harsh reagents, plant‐based techniques present a more economically viable and scalable avenue for vaccine production. The imperative urges these approaches to address pressing global health challenges posed by emerging and recurring infectious diseases, surpassing the limitations of traditional vaccine fabrication methods. The primary goal of this review is to provide a comprehensive overview of the current research landscape, covering conceptualization, production, and potential advantages of plasma‐based and plant‐based vaccines. Furthermore, exploring the obstacles and opportunities intrinsic to these strategies is undertaken, elucidating their potential impact on vaccination strategies. The aggressiveness and limited treatment options make triple‐negative breast cancer (TNBC) a challenging condition to treat. Immune checkpoint blockade therapy, explicitly targeting the programmed death‐ligand 1 (PD‐L1) pathway, is a promising therapeutic approach. However, the effectiveness of PD‐L1 inhibitors like atezolizumab depends on adequate PD‐L1 expression within cancer cells. To this end, this study used nonthermal plasma (NTP) to modulate PD‐L1 in TNBC cells. The current investigation revealed that NTP treatment leads to an upregulation of PD‐L1; this might increase the availability of PD‐L1 to PD‐L1 inhibitors. The developmental rapidity of nanotechnology poses higher risks of exposure to humans and the environment through manufactured nanomaterials. The multitude of biological interfaces, such as DNA, proteins, membranes, and cell organelles, which come in contact with nanoparticles, is influenced by colloidal and dynamic forces. Consequently, the ensued nano-bio interface depends on dynamic forces, encompasses many cellular absorption mechanisms along with various biocatalytic activities, and biocompatibility that needs to be investigated in detail. Addressing the issue, the study offers a novel green synthesis strategy for antibacterial AgNPs with higher biocompatibility and elucidates the mechanistic in vivo biocompatibility of silver nanoparticles (AgNPs) at the cellular and molecular levels. The analysis ascertained the biosynthesis of G-AgNPs with the size of 25 ± 10 nm and zeta potential of −29.2 ± 3.0 mV exhibiting LC50 of 47.2 μg mL−1 in embryonic zebrafish. It revealed the mechanism as a consequence of abnormal physiological metabolism in oxidative stress and neutral lipid metabolism due to dose-dependent interaction with proteins such as he1a, sod1, PEX protein family, and tp53 involving amino acids such as arginine, glutamine and leucine leading to improper apoptosis. The abounding use of cobalt oxide nanoparticles (Co3O4) requires a detailed understanding of their environmental and biomedical nanotoxicity and an eminent solution to the associated hazards; molecular and atomic aspects of the subject are poorly understood. This study reconnoiters the in vitro and in vivo nanotoxicity of Co3O4 nanoparticles using human colon cell lines and the embryonic zebrafish model. The synthesis of Co3O4 nanoparticles (G-CoONP) is delineated via the deployment of a medicinal herb, Calotropis gigantea, as an alternative greener solution; stable G-CoONP with a size of 41 ± 15 nm are attainable. Gas chromatography-mass spectroscopy (GCMS) analysis revealed the role of floral extract biomolecules in G-CoO NP synthesis. The in vitro and in vivo effects are accompanied by dose-dependent exposure at the molecular level by eliciting Sod1 and P53 genes up to 8.2 and 5.2 fold leading to a significant change in the reactive oxygen species and apoptosis level. It unraveled the toxicity of the cobalt oxide NP as increased apoptosis elicited by higher oxidative stress due to the accumulation and internalization of nanoparticles in cells and embryos. Green synthesized G-CoONP exhibited higher biocompatibility than commercial C-CoONP with reduced apoptosis and ROS in both human colon cell lines and zebrafish embryos. In silico analysis portrayed the intrinsic atomic interaction of Co3O4 NP with cysteine, arginine, and histidine of oxidative stress (SOD1/sod1) and apoptosis (TP53/tp53) proteins leading to dysregulation of their structural and functional integrity in human and zebrafish, respectively. A proximal discrepancy in intrinsic atomic interaction due to the H-bonding and hydrophobic interaction at the differential in vitro and in vivo vicinage served as a key determinant factor for the cellular biocompatibility of Co3O4 nanoparticles. Materials & methods: Synthesis of NP was achieved using a high-energy ball milling method. Following characterization, its antibacterial activity and cellular and molecular biocompatibility were evaluated in vitro by experimental and computational approaches. Results: A total of 15 h of milling pepper produced NP with a size of 44 ± 12 nm and zeta potential of -22 ± 12 mV. Bulk pepper and NP showed antibacterial activity and an LC50 of 1.9 μM and 2.1 μM in HCT116 colon cells. Components of pepper, piperine and β-caryophyllene were found to interact with superoxide dismutase [Cu-Zn] and apoptotic protease-activating factor-1-caspase-9 through different amino acids via H-bonds. Conclusion: NP exhibits significant antibacterial activity with cellular biocompatibility due to intrinsic atomic interaction. Aim: To synthesize biocompatible and ecofriendly silver nanoparticles (AgNPs) and elucidate their in vivo molecular and cellular nanotoxicity at an atomic level. Materials & methods: AgNPs were biosynthesized using Andrographolide and their in vivo cellular and molecular biocompatibility was evaluated using zebrafish embryos. Results: AgNPs with a size of 80 nm and ζ potential of -52 mV were obtained. The LC50 for the AgNPs embryonic zebrafish was found to be 125 μg/ml. AgNPs induced reactive oxygen species production and elicit apoptosis mechanistically, then nanoparticles were shown to interact with caspase-3 and caspase-9 proteins through proline, cysteine, glycine and histidine amino acid residues via H-bond of corresponding bond energies. Glioblastoma (GBM) remains a formidable clinical challenge, with cancer stem cells (CSCs) contributing to treatment resistance and tumor recurrence. Conventional treatments often fail to eradicate these CSCs characterized by enhanced resistance to standard therapies through metabolic plasticity making them key targets for novel treatment approaches. Addressing this challenge, this study introduces a novel combination therapy of dichloroacetate (DCA), a metabolic modulator and nonthermal plasma to induce oxidative stress in glioblastomas. Our results demonstrate that DCA and nonthermal plasma (NTP) synergistically increase ROS production, resulting in endoplasmic reticulum (ER) stress and mitochondrial reprogramming, key factors in the initiation of programmed cell death. Furthermore, the combination downregulated key stemness markers indicating effective CSCs suppression. Upregulation of pro-apoptotic proteins and downregulation of anti-apoptotic factors highlight the induction of apoptosis in glioma cells. Recently nanotechnology has evolved as one of the most revolutionary technologies in the world. It has now become a multi-trillion-dollar business that covers the production of physical, chemical, and biological systems at scales ranging from atomic and molecular levels to a wide range of industrial applications, such as electronics, medicine, and cosmetics. Nanobiomaterials synthesis are promising approaches produced from various biological elements be it plants, bacteria, peptides, nucleic acids, etc. Owing to the better biocompatibility and biological approach of synthesis, they have gained immense attention in the biomedical field. Moreover, due to their scaled-down sized property, nanobiomaterials exhibit remarkable features which make them the potential candidate for different domains of tissue engineering, materials science, pharmacology, biosensors, etc. Miscellaneous characterization techniques have been utilized for the characterization of nanobiomaterials. Currently, the commercial transition of nanotechnology from the research level to the industrial level in the form of nano-scaffolds, implants, and biosensors is stimulating the whole biomedical field starting from bio-mimetic nacres to 3D printing, multiple nanofibers like silk fibers functionalizing as drug delivery systems and in cancer therapy. The contribution of single quantum dot nanoparticles in biological tagging typically in the discipline of genomics and proteomics is noteworthy. This review focuses on the diverse emerging applications of Nanobiomaterials and their mechanistic advancements owing to their physiochemical properties leading to the growth of industries on different biomedical measures. Alongside the implementation of such nanobiomaterials in several drug and gene delivery approaches, optical coding, photodynamic cancer therapy, and vapor sensing have been elaborately discussed in this review. Foodborne infection is one of the leading sources of infections spreading across the world. Foodborne pathogens are recognized as multidrug-resistant (MDR) pathogens posing a significant problem in the food industry and healthy consumers resulting in enhanced economic burden, and nosocomial infections. The continued search for enhanced microbial detection tools has piqued the interest of the CRISPR-Cas system and Nanoparticles. CRISPR-Cas system is present in the bacterial genome of some prokaryotes and is repurposed as a theragnostic tool against MDR pathogens. Nanoparticles and composites have also emerged as an efficient tool in theragnostic applications against MDR pathogens. The diagnostic limitations of the CRISPR-Cas system are believed to be overcome by a synergistic combination of the nanoparticles system and CRISPR-Cas using nanoparticles as vehicles. In this review, we have discussed the diagnostic application of CRISPR-Cas technologies along with their potential usage in applications like phage resistance, phage vaccination, strain typing, genome editing, and antimicrobial. we have also elucidated the antimicrobial and detection role of nanoparticles against foodborne MDR pathogens. Thermal plasma and non-thermal (or cold atmospheric) plasma (NTP) are two types of plasma. Atoms and heavy particles are both available at the same temperature in thermal plasma. Cold atmospheric plasma (CAP) is intended to be non-thermal since its electrons are hotter than the heavier particles at ambient temperature. Direct barrier discharge (DBD), atmospheric plasma pressure jet (APPJ), etc. methods can be used to produce plasma, however, all follow a basic concept in their generation. This review focuses on the anticipated uses of cold atmospheric plasma in dentistry, such as its effectiveness in sterilizing dental instruments by eradicating bacteria, its advantage in dental cavity decontamination over conventional methods, root canal disinfection, its effects on tooth whitening, the benefits of plasma treatment on the success of dental implant placement, and so forth. Mitochondrial bioenergetics is vital for the proper functioning of cellular compartments. Impairments in mitochondrial DNA encoding the respiratory chain complexes and other assisting proteins, accumulation of intracellular reactive oxygen species, an imbalance in cellular calcium transport, or the presence of organic pollutants, high fat-ketogenic diets or toxins, and advancing age can result in complex disorders, including cancer, metabolic disease, and neurodegenerative disorders. Such manifestations are distinctly exhibited in several age-related neurodegenerative diseases, such as in Parkinson's disease (PD). Defects in complex I along with perturbed signaling pathways is a common manifestation of PD. Impaired oxidative phosphorylation could increase the susceptibility to PD. Therefore, unraveling the mechanisms of mitochondrial complexes in clinical scenarios will assist in developing potential early biomarkers and standard tests for energy failure diagnosis and assist to pave a new path for targeted therapeutics against PD.
