Plastids are essential organelles in angiosperms and show non-Mendelian inheritance due to their evolution as endosymbionts. In approximately 80% of angiosperms, plastids are thought to be inherited from the maternal parent, whereas other species transmit plastids biparentally. Maternal inheritance can be generally explained by the stochastic segregation of maternal plastids after fertilization because the zygote is overwhelmed by the maternal cytoplasm. In contrast, biparental inheritance shows the transmission of organelles from both parents. In some species, maternal inheritance is not absolute and paternal leakage occurs at a very low frequency (∼10−5). A key process controlling the inheritance mode lies in the behavior of plastids during male gametophyte (pollen) development, with accumulating evidence indicating that the plastids themselves or their DNAs are eliminated during pollen maturation or at fertilization. Cytological observations in numerous angiosperm species have revealed several critical steps that mutually influence the degree of plastid transmission quantitatively among different species. This review revisits plastid inheritance from a mechanistic viewpoint. Particularly, we focus on a recent finding demonstrating that both low temperature and plastid DNA degradation mediated by the organelle exonuclease DEFECTIVE IN POLLEN ORGANELLE DNA DEGRADATION1 (DPD1) influence the degree of paternal leakage significantly in tobacco. Abstract Ancient hybridization leading to plastid capture is a likely cause of cytonuclear discordance, although discordance may also be attributed to other mechanisms. Cytonuclear discordance is ubiquitous in a clade of southern African Oxalis known as the Hirta clade, despite a notable lack of documented field hybrids for this genus. The aims of this study were to determine whether hybridization is possible between different Hirta clade species, what the fitness effects on seed set of possible hybridizations are, and to determine the mode of inheritance of plastid DNA in Oxalis. We tested these aims using manual interspecific and conspecific crosses, as well as self-pollination control crosses of species within this clade, conducted in a research collection context. Roughly 10% of interspecific crosses produced seed, most of which were confirmed as unambiguous hybrid plants using DNA sequencing of the Internal Transcribed Spacer (ITS). Low levels of self-compatibility were recorded in several species, while one accession showing very high levels of self-compatibility (accession MO1218). Interspecific crosses produced significantly less seed per capsule compared to conspecific crosses with respective parents, suggesting decreased fitness in the establishment of the F1 hybrids. Selfing seed set was significantly lower than interspecific and conspecific seed set upon removal of MO1218. Using plastid fragment length polymorphisms, Oxalis was found to have predominantly or exclusively maternally inherited plastid DNA. This study establishes the viability of the earliest stages of plastid capture in Oxalis , and provides context relevant to the lack of documented field hybrids in the southern African members of this genus. Future studies should focus on F1 survival and fertility and the maternal fitness effects of subsequent backcrosses to the maternal parent, as well as on hybridization rates under field conditions, to assess whether hybridization occurs at rates sufficient to affect Oxalis evolution in southern Africa. We applied a series of intra- and interspecific in situ cross-pollination experiments under greenhouse conditions to evaluate the breeding systems in four Fosterella species (Pitcairnioideae s.str.; Bromeliaceae). Viable hybrids were produced between each pair of the investigated species, suggesting that reproduction barriers may be low also under natural conditions. Seed germination rates proved to be high in each crossing treatment, indicating a high viability of the artificial hybrids. Large numbers of seeds were produced after both closed and open pollination treatments, suggesting that autogamy may be a major reproductive strategy in the genus. Our results support the concept that self-compatibility is an appropriate way to avoid natural hybridization in Bromeliaceae and could assist in maintaining species integrity in the presence of pollen flow. Paternity was verified in all crosses by genotyping parents and offspring with a set of polymorphic nuclear microsatellite markers. To study the mode of chloroplast inheritance, we developed a novel set of 24 chloroplast microsatellite markers using 454 pyrosequencing technology, and applied four of these markers for genotyping parents and offspring from all crosses. Plastid-encoded genes are maternally inherited in most plant species. Transgenes located on the plastid genome are thus within a natural confinement system, preventing their distribution via pollen. However, a low-frequency leakage of plastids via pollen seems to be universal in plants. Here we report that a very low-level paternal inheritance in Arabidopsis thaliana occurs under field conditions. As pollen donor an Arabidopsis accession ( Ler- Ely) was used, which carried a plastid-localized atrazine resistance due to a point mutation in the psbA gene. The frequency of pollen transmission into F1 plants, based on their ability to express the atrazine resistance was 1.9 × 10. We extended our analysis to another cruciferous species, the world-wide cultivated crop Brassica napus. First, we isolated a fertile and stable plastid transformant (T36) in a commercial cultivar of B. napus (cv Drakkar). In T36 the aadA and the bar genes were integrated in the inverted repeat region of the B. napus plastid DNA following particle bombardment of hypocotyl segments. Southern blot analysis confirmed transgene integration and homoplasmy of plastid DNA. Line T36 expressed Basta resistance from the inserted bar gene and this trait was used to estimate the frequency of pollen transmission into F1 plants. A frequency of <2.6 × 10 was determined in the greenhouse. Taken together, our data show a very low rate of paternal plastid transmission in Brassicacea. Plastids and mitochondria, the DNA-containing cytoplasmic organelles, are maternally inherited in the majority of angiosperm species. Even in plants with strict maternal inheritance, exceptional paternal transmission of plastids has been observed. Our objective was to detect rare leakage of plastids via pollen in Nicotiana sylvestris and to determine if pollen transmission of plastids results in co-transmission of paternal mitochondria. As father plants, we used N. sylvestris plants with transgenic, selectable plastids and wild-type mitochondria. As mother plants, we used N. sylvestris plants with Nicotiana undulata cytoplasm, including the CMS-92 mitochondria that cause cytoplasmic male sterility (CMS) by homeotic transformation of the stamens. We report here exceptional paternal plastid DNA in approximately 0.002% of N. sylvestris seedlings. However, we did not detect paternal mitochondrial DNA in any of the six plastid-transmission lines, suggesting independent transmission of the cytoplasmic organelles via pollen. When we used fertile N. sylvestris as mothers, we obtained eight fertile plastid transmission lines, which did not transmit their plastids via pollen at higher frequencies than their fathers. Abstract: Cultivated chickpea (Cicer arietinum) was crossed with its wild relatives from the genus Cicer to transfer favorable genes from the wider gene pool into the cultivar. Post-hybridization barriers led to yellowing and subsequent senescence from as early as 5 days after fertilization, however, the ovules of hybrid embryos could be rescued in vitro. Hybrids were classified as green, partially green or albino. The hybrid status of regenerated plantlets in vitro was confirmed by amplification of nuclear DNA markers. To check whether chloroplast development correlated with plastid DNA inheritance in these crosses, primers were designed using conserved plastid gene sequences from wild and cultivated species. All three possible plastid inheritance patterns were observed: paternal, maternal and biparental. This is the first report of biparental inheritance of plastid DNA in Cicer. No correlation was observed between parental origin of the plastid genome and degree of albinism, indicating that chloroplast development in hybrid genotypes was mostly influenced by nuclear factors. Albinism is a common problem encountered in interspecific crosses and tissue culture experiments including anther culture and generation of doubled haploids. It is characterized by partial or complete loss of chlorophyll pigments and incomplete differentiation of chloroplast membranes. This in turn impairs photosynthesis and the plants eventually die at a young stage without reaching maturity. Environmental conditions such as light, temperature, media composition and culture conditions play some role in determining the frequency of albino plant formation. Genetic factors are even more important, and are major determinants in albinism. Genetic studies in different crops show that it is a recessive trait governed by many loci. Both the nuclear and chloroplast genomes affect albinism and incompatibilities between the two are a probable cause of many pigment defects in hybrid progenies. Such incompatibility has been reported in a large number of angiosperms. The mechanisms behind these incompatibilities are poorly understood. Studies of plastid DNA inheritance together with observations using electron microscopy have established that the transmission of plastids can be maternal, paternal or biparental, even within the same genus, especially following wide crosses; contrary to the widespread belief that plastids are almost always transmitted from the maternal parent. Albinism has been overcome in some crop species through somatic hybridization and development of cybrids (cytoplasmic hybrids). However, the strict requirement of efficient protoplast regeneration is a major limitation of these techniques. This review focuses on albinism following interspecific crosses or development of doubled haploids facilitated by tissue culture experiments, underlying mechanisms, and the possibilities for dealing with this important biotechnological limitation. Plastid inheritance was followed during sexual reproduction in the raphid pennate diatom Pseudo-nitzschia delicatissima, using rbcL haplotypes as plastid identification tools. Pseudo-nitzschia species are dioecious and show functional anisogamy with ‘male’ mating type+(PNd+) cells and ‘female’ PNd− cells. Vegetative cells possess two plastids. In P. delicatissima, meiosis results in two gametes that both contribute two plastids to the zygote. The latter initially contains four plastids, but during auxospore development two of these four seem to disappear, and the initial cell emerging from the auxospore appears to contain only two. Here we assessed if the plastids are inherited strictly unipaternally, strictly biparentally, or randomly. We traced the source of the plastids in the F1 generation by using PNd+ and PNd− parental strains with different rbcL genotypes, here denoted AA (homoplastidial, with two plastids of rbcL haplotype A) and BB (homoplastidial; two plastids of haplotype B). Results showed that 16 out of 96 strains raised each from single F1 cells had retained two paternal (PNd+) plastids, 20 had two maternal (PNd−) plastids and the remaining 60 had one maternal and one paternal plastid. This pattern is in accordance with the hypothesis that either two of the four plastids are eliminated during auxospore formation, or that all plastids are retained in the auxospore and segregate in pairs joining at random during the first mitotic division of the initial cell. Heteroplastidic F1-strains retained the AB genotype throughout the vegetative phase of their life cycle. The finding that 60 out of 96 F1 strains were heteroplastidial contrasts with an absence of such genotypes in our strains raised from single cells sampled in the Gulf of Naples. Plastid DNA is absent in pollen or sperm cells of Arabidopsis thaliana. Accordingly, plastids and mitochondria, in a standard genetic cross, are transmitted to the seed progeny by the maternal parent only. Our objective was to test whether paternal plastids are transmitted by pollen as an exception. The maternal parent in our cross was a nuclear male sterile ( ms1-1/ ms1-1), spectinomycin-sensitive L er plant. It was fertilized with pollen of a male fertile RLD-Spc1 plant carrying a plastid-encoded spectinomycin resistance mutation. Seedlings with paternal plastids were selected by spectinomycin resistance encoded in the paternal plastid DNA. Our data, in general, support maternal inheritance of plastids in A. thaliana. However, we report that paternal plastids are transmitted to the seed progeny in Arabidopsis at a low (3.9 × 10−5) frequency. This observation extends previous reports in Antirrhinum majus, Epilobium hirsutum, Nicotiana tabacum, Petunia hybrida, and the cereal crop Setaria italica to a cruciferous species suggesting that low-frequency paternal leakage of plastids via pollen may be universal in plants previously thought to exhibit strict maternal plastid inheritance. The genetic tools employed here will facilitate testing the effect of Arabidopsis nuclear mutations on plastid inheritance and allow for the design of mutant screens to identify nuclear genes controlling plastid inheritance. Patterns of inheritance of the chloroplast genome in Passiflora were analyzed by examining the progeny from both interspecific and intraspecific crosses. Artificial crosses of field-collected material were performed in greenhouses at The University of Texas at Austin. DNA from fresh leaf material was analyzed by Southern blot techniques to identify the donor of the chloroplast genome. Initially, single progeny were analyzed for 11 crosses; two intraspecific crosses demonstrated maternal inheritance, whereas the nine interspecific crosses had paternal inheritance. Subsequently, the donor of the chloroplast genome was determined for multiple progeny in seven crosses. Passiflora oerstedii X P. retipetala showed strict paternal inheritance in all of 17 progeny. A series of five crosses and backcrosses between P. oerstedii and P. menispermifolia demonstrated strictly paternal inheritance. Finally, when 15 progeny were analyzed for the P. costaricensis X P. costariceosis cross, 12 of the 15 showed maternal inheritance, whereas the remaining three were biparental. Interestingly, all interspecific crosses had primarily paternal inheritance, whereas all intraspecific crosses had primarily maternal inheritance. In alfalfa ( Medicago sativa L.), plastids are inherited biparentally. Patterns of plastid transmission vary according to the genotypes involved, but there is a strong bias in favor of male plastid transmission. Previous cytological studies on the male gametophyte of this species have not provided an adequate explanation for the differences in plastid transmission frequencies among genotypes. In the present study, we compared egg cells from genotypes classified as strong or weak plastid transmitters to determine whether plastid transmission strength is correlated with egg cell structure before fertilization. We found that plastids in the mature egg cells of the strong female (genotype 6-4) are significantly larger than in mature eggs of the weak female (genotype CUF-B), and that significantly more plastids are positioned in the apical portion of the mature egg cell of genotype 6-4 than in CUF-B. Immature eggs in the two genotypes show the same pattern as mature eggs with regard to plastid number and polarization. Since only the apical portion of the egg cell/zygote gives rise to the functional embryo, these results indicate that the potential input of female plastids, in terms of plastid size and number, may be an important factor in determining the inheritance patterns of these organelles in alfalfa. Cultivar variability for levels of plastid DNA (cpDNA) in the germ cell line of germinated pea pollen has suggested the possibility of biparental plastid transmission. In order to examine this possibility further, RFLP markers were used to follow the transmission of cpDNA from parents to their F offspring. Results from these inheritance studies clearly indicate the presence of only maternal plastid markers in the F progeny of each cross examined, irrespective of the pollen cpDNA levels of the paternal parent. The same result is obtained for F progeny produced from crosses using pollen characterized by comparatively high cpDNA content, even when offspring are sampled at early developmental stages. Thus, there appears to be little correspondence between pollen cytological data indicating potential paternal plastid transmission and data from molecular marker studies confirming that P. sativum generally follows a uni-parental-maternal mode of plastid inheritance. Prior cytological observations using DAPI/epifluorescence microscopy have suggested that the method could be used to rapidly screen plant species for their potential mode of plastid DNA transmission. Cytoplasmic DAPI-DNA aggregates were observed in generative cells of germinated pollen of Medicago sativa (alfalfa), a species known genetically to display biparental transmission, but not in Antirrhinum majus (snapdragon), a species known to be maternal for plastid transmission. If, as suggested, these aggregates are plastid DNA nucleoids, then M. sativa pollen should contain plastid DNA detectable by molecular biology methods and A. majus pollen should not. Total DNA was isolated from germinated pollen and analyzed by Southern blot hybridization. A clone containing part of the rbcL gene from the garden pea plastome was used as a probe for plastid DNA. This probe hybridized with a restriction fragment from M. sativa pollen DNA, but not detectably with A. majus pollen DNA, thereby corroborating the identification of the cytoplasmic DAPI-DNA aggregates in M. sativa pollen as plastid DNA, and confirming the cytologically determined absence of plastic DNA in A. majus pollen. The mode of plastid transmission in the garden pea ( Pisum sativum L.) was analyzed cytologically using the DNA-fluorochrome 4′,6-diamidino-2-phenylindole (DAPI) in conjunction with epifluorescence microscopy. The reproductive cells of mature pollen obtained from 12 inbred lines and cv 'Early Alaska' were examined for the presence or absence of DAPI-stained plastid DNA aggregates. Plastid DNA was detected in all 13 pea lines examined, although there was variability with regard to the percentage of pollen graines showing plastid DNA aggregates of generative cells (ranging from 3% in accession 82-12r to 65% in accession 82-14n). Interspecific hybrids of various Epilobium species have been produced in order to analyse plastid inheritance using restriction fragment polymorphisms of plastid DNA as markers. This analysis reveals that interspecific hybrids exhibit only the fragment pattern of the maternal plastome. Southern hybridization experiments using cloned species-specific plastid DNA fragments as markers confirm the maternal type of plastid inheritance in Epilobium, while providing at least a tenfold increase of sensitivity to detect restriction polymorphisms. Within the limit of detection even young seedlings contain no plastid DNA from the paternal parent. However, investigations of plastomes of large populations have provided evidence that a very low frequency of paternal plastid transmission can occur. Thus, the mechanism which ensures the elimination of paternal plastids is not 100% efficient. This suggestion is also supported by intraspecific reciprocal crosses between plants carrying mutant white and normal green plastids. Chloroplast DNA (cpDNA) restriction site diversity was assessed by 21 enzyme/probe combinations in 30 accessions of six Lens species, including the recently recognized L. lamottei and L. tomentosus. A total of 118 fragments were scored and 26 restriction site mutations were identified. The cpDNA restriction pattern supports circumscribing L. lamottei and L. tomentosus as independent species. The value of the data for reconstructing phylogeny in the genus is discussed. The cpDNA of all 13 accessions of the lentil’s wild progenitor, L. culinaris subsp. orientalis, differed from that of the single lentil cultivars used in this study. This diversity indicates that other populations of this subspecies from Turkey and Syria examined by Mayer and Soltis (1994) are potentially the founder members of lentil. DNA sequence analysis of chloroplast genomes has revealed many short nucleotide repeats analogous to nuclear microsatellites, or simple sequence repeats (SSRs). We designed PCR primers flanking five of these regions identified in the chloroplast sequence from Pinus thunbergii and tested them for amplification in Pinus radiata, P. elliotii, P. taeda, P. strobus, Pseudotsuga menziesii, Cupressus macrocarpa, four New Zealand native conifer species ( Podocarpus totara, Podocarpus hallii, Podocarpus nivalis, Agathis australis), and four angiosperms ( Vitex lucens, Nestegis cunninghamii, Actinidia chinensis, and Arabidopsis thaliana). A PCR product in the expected size range was amplified from all species and interspecific polymorphism was detected at all five loci. Intraspecific polymorphism was detected in P. radiata with four of the five primer pairs. One of these polymorphic chloroplast SSR (cpSSR) was then used to determine the inheritance of chloroplasts in 206 progeny from four control-pollinated, full-sibling P. radiata families. Approximately 99% of the progeny had the cpSSR variant of the pollen parent indicating that in Pinus radiata, like most other conifers, chloroplasts are typically inherited from the paternal parent. These results suggest that polymorphic chloroplast SSRs will be a valuable tool for studying chloroplast diversity, cyto-nuclear disequilibrium, and plastid inheritance in a range of species, and for the analysis of gene flow via pollen and paternity in species with paternal transmission of chloroplasts. Zonal pelargoniums exhibit biparental plastid inheritance. After G x W plastid crosses the progeny are a mixture of green, variegated and white embryos corresponding to a maternal, biparental or paternal inheritance of plastids, respectively. There are two patterns of segregation: type-I females have families in which the majority of embryos are green, variegated are of intermediate frequency and white are the least frequent. Type-II females have families in which green and white embryos are present at about the same frequency and variegated are the least common. The results of many selfs and crosses made within and between 8 type-I and 8 type-II plants led us to conclude that the type of female was determined by its genotype with respect to a pair of complementary genes. Plants giving rise to the type-II pattern contained one or two copies of the dominant alleles of both genes, whereas in the absence of either one or both dominant alleles the plants were type I. The genes were called Pr1/pr1 and Pr2/pr2, an adaptation of symbolism used previously. All 8 type IIs were double heterozygotes Pr1pr1, Pr2pr2, whereas we found 3 genotypes among the type Is, Pr1Pr1, pr2pr2; pr1pr1, Pr2Pr2 and pr1pr1, Pr2pr2. In unrelated experiments we found type IIs of which some were again double heterozygotes and others single heterozygotes Pr1pr1, Pr2Pr2 or Pr1Pr1, Pr2pr2. The ultrastructure of egg cells in Abies alba was examined to elucidate the lack of maternal inheritance of plastids. Before fertilization, maternal plastids are absent in the perinuclar zone containing mainly mitochondria and smooth endoplasmic reticulum. During egg cell development the maternal plastids are transformed into large inclusions which are situated mostly towards the periphery of the egg cell, and finally disintegrate. As a consequence, they do not participate in zygote formation. RFLP analysis of cpDNA of parental trees and their F1 interspecific hybrids ( A. alba× A. numidica, A. alba× A. nordmanniana, A. nordmanniana× A. Alba) using HindIII and BamHI showed a paternal mode of cpDNA inheritance. Genetic studies have demonstrated biparental inheritance of plastids in alfalfa. The ratio of paternal to maternal plastids in the progeny varies according to the genotypes of the parents, which can be classified as strong or weak transmitters of plastids. Previous cytological investigations of generative cells and male gametes have provided no consistent explanation for plastid inheritance patterns among genotypes. However, plastids in the mature egg cells of a strong female genotype (6-4) were found to be more numerous and larger than in mature eggs of a weak female genotype (CUF-B), and the plastids in 6-4 eggs are positioned equally around the nucleus. In CUF-B, the majority of plastids are positioned below (toward the micropyle) the mid level of the nucleus, which is the future division plane of the zygote. Since only the apical portion of the zygote produces the embryo proper, plastids in the basal portion were predicted to become included in the suspensor cells and not be inherited. In the present study, we examined zygotes and a two-celled proembryo from a cross between CUF-B and a strong male genotype (301), a cross that results in over 90% of the progeny possessing paternal plastids only. Our results indicate that the distribution of plastids observed in the CUF-B egg cell is maintained through the first division of the zygote. Further, paternal plastids are similarly distributed; however, within the apical portion of the zygote and in the apical cell of the two-celled proembryo, the number of paternal plastids is typically much greater than the number of maternal plastids.
