Social enrichment or social isolation affects a range of innate behaviors, such as sex, aggression, and sleep, but whether there is a shared mechanism is not clear. Here, we report a neural mechanism underlying social modulation of spontaneous locomotor activity (SoMo-SLA), an internal-driven behavior indicative of internal states. We find that social enrichment specifically reduces spontaneous locomotor activity in male flies. We identify neuropeptides Diuretic hormone 44 (DH44) and Tachykinin (TK) to be up- and down-regulated by social enrichment and necessary for SoMo-SLA. We further demonstrate a sexually dimorphic neural circuit, in which the male-specific P1 neurons encoding internal states form positive feedback with interneurons coexpressing doublesex (dsx) and Tk to promote locomotion, while P1 neurons also form negative feedback with interneurons coexpressing dsx and DH44 to inhibit locomotion. These two opposing neuromodulatory recurrent circuits represent a potentially common mechanism that underlies the social regulation of multiple innate behaviors. [ABSTRACT FROM AUTHOR]

Abstract The N-methyl-d-aspartate receptors (NRs) in hippocampal CA3 are crucial for the synaptic transmission and plasticity within the CA3 recurrent circuit, which supports the hippocampal functions, such as pattern completion, and reverberatory association of sensory inputs. Previous study showed that synchronous activation of distinct cell populations in CA3, which correspond to distinct events, associated independent events, suggesting that the recurrent circuit expressing NRs in CA3 mediates the artificial association of memory events stored in CA3 ensembles. However, it is still unclear whether CA3 NRs are crucial for the artificial association of memory events stored in the CA3 ensembles. Here we report that the triple transgenic mice (cfos-tTA/KA1-Cre/NR1 flox/flox), which specifically lack NRs in the CA3 cell ensembles, showed impairment in artificial association between two events, which in control mice triggered artificial association. This result indicates that NRs in the hippocampal CA3 are required for the artificial association of memory events stored in the CA3 cell ensembles.

The stress response necessitates an immediate boost in vital physiological functions from their homeostatic operation to an elevated emergency response. However, the neural mechanisms underlying this state-dependent change remain largely unknown. Using a combination of in vivo and ex vivo electrophysiology with computational modeling, we report that corticotropin releasing hormone (CRH) neurons in the paraventricular nucleus of the hypothalamus (PVN), the effector neurons of hormonal stress response, rapidly transition between distinct activity states through recurrent inhibition. Specifically, in vivo optrode recording shows that under non-stress conditions, CRHPVN neurons often fire with rhythmic brief bursts (RB), which, somewhat counterintuitively, constrains firing rate due to long (~2 s) interburst intervals. Stressful stimuli rapidly switch RB to continuous single spiking (SS), permitting a large increase in firing rate. A spiking network model shows that recurrent inhibition can control this activity-state switch, and more broadly the gain of spiking responses to excitatory inputs. In biological CRHPVN neurons ex vivo, the injection of whole-cell currents derived from our computational model recreates the in vivo-like switch between RB and SS, providing direct evidence that physiologically relevant network inputs enable state-dependent computation in single neurons. Together, we present a novel mechanism for state-dependent activity dynamics in CRHPVN neurons.

Abstract Previous gain-of-function studies using an optogenetic technique showed that manipulation of the hippocampal dentate gyrus or CA1 cell ensembles is important for memory reactivation and to generate synthetic or false memory. However, gain-of-function study manipulating CA3 cell ensembles has not been reported. The CA3 area of the hippocampus comprises a recurrent excitatory circuit, which is thought to be important for the generation of associations among the stored information within one brain region. We investigated whether the coincident firing of cell ensembles in one brain region, hippocampal CA3, associates distinct events. CA3 cell ensembles responding to context exploration and during contextual fear conditioning were labeled with channelrhodopsin-2 (ChR2)-mCherry. The synchronous activation of these ensembles induced freezing behavior in mice in a neutral context, in which a foot shock had never been delivered. The recall of this artificial associative fear memory was context specific. In vivo electrophysiological recordings showed that 20-Hz optical stimulation of ChR2-mCherry-expressing CA3 neurons, which is the same stimulation protocol used in behavioral experiment, induced long-term potentiation at CA3-CA3 synapses. Altogether, these results demonstrate that the synchronous activation of ensembles in one brain region, CA3 of the hippocampus, is sufficient for the association of distinct events. The results of our electrophysiology potentially suggest that this artificial association of memory events might be induced by the strengthening of synaptic efficacy between CA3 ensembles via recurrent circuit.

This article comprehensively addresses the problems associated with the structure of recurrent circuits of military aircraft with metal fuselage structure. The authors describe the issues concerning the usage of the fuselage as a recurrent circuit of electric current and with negative conductors connecting the fuselage with negative terminals of the receiver or the source. The military aircraft are frequently operated under harsh conditions of increased humidity, dustiness and variable temperatures. All these factors as mentioned above contribute to the occurrence of intensive electrochemical corrosion at the contact points of airframe's elements or electrical conductors' with the fuselage. Recurrent connections are thought to be a common feature of the neural circuits that encode memories, but how memories are laid down in such circuits is not fully understood. Here we present evidence that courtship memory in Drosophila relies on the recurrent circuit between mushroom body gamma (MBγ), M6 output, and aSP13 dopaminergic neurons. We demonstrate persistent neuronal activity of aSP13 neurons and show that it transiently potentiates synaptic transmission from MBγ>M6 neurons. M6 neurons in turn provide input to aSP13 neurons, prolonging potentiation of MBγ>M6 synapses over time periods that match short-term memory. These data support a model in which persistent aSP13 activity within a recurrent circuit lays the foundation for a short-term memory.

Current thought envisions dopamine neurons conveying the reinforcing effect of the unconditioned stimulus during associative learning to the axons of Drosophila mushroom body Kenyon cells for normal olfactory learning. Here, we show using functional GFP reconstitution experiments that Kenyon cells and dopamine neurons from axoaxonic reciprocal synapses. The dopamine neurons receive cholinergic input via nicotinic acetylcholine receptors from the Kenyon cells; knocking down these receptors impairs olfactory learning revealing the importance of these receptors at the synapse. Blocking the synaptic output of Kenyon cells during olfactory conditioning reduces presynaptic calcium transients in dopamine neurons, a finding consistent with reciprocal communication. Moreover, silencing Kenyon cells decreases the normal chronic activity of the dopamine neurons. Our results reveal a new and critical role for positive feedback onto dopamine neurons through reciprocal connections with Kenyon cells for normal olfactory learning.

Abstract: The ongoing steady nature of consciousness in everyday life implies that the underlying neural activity possesses a high level of stability. The prolonged cognitive events of sustained attention, imagery, and working memory also imply high stability of underlying neural activity. This paper proposes that stabilization of neural activity is produced by apical dendrite activity in pyramidal neurons within recurrent corticothalamic circuits, and proposes that the wave activities of apical dendrites that stabilize ongoing activity constitute the subjective impressions of an attended object and the entire sensory background. The cortical minicolumn, as the functional unit of the cortex, is separated into an axis consisting of layer 5 pyramidal neurons and a surrounding shell consisting of layer 2/3 pyramidal neurons. It is proposed that apical dendrites of the axis generate sensory impressions, and basal dendrites of the shell process the brief-lasting input–output identifications of objects that give rise to ideas. Abstract: Recurrent neural activity is a pervasive mode of cortical operations and is believed to underlie cognitive functions of working memory, attention, and the generation of spontaneous activity during sleep . It is proposed here that activity in corticothalamic recurrent circuits underlies the sustaining of attention, and that extended durations of attention are made possible by the stabilizing effects of electrical activity in long apical dendrites of pyramidal neurons. Using the cue–target delay task as a framework, the present paper describes sustained attention during the cue–target delay as activity in recurrent circuits involving layer 5/6 pyramidal neurons. At target onset, persistent activity in apical dendrites of layer 2/3 pyramidal neurons (projected from the recurrent circuits) can enhance the processing of incoming pulse trains at basal dendrites. Apical dendrite activity is assumed to modulate the soma processing of layer 2/3 and layer 5/6 pyramidal neurons at subthreshold voltage levels. The variability of successive soma depolarizations from the apical dendrite strongly influences the stability of activity in the corticothalamic recurrent circuit. Lower variability promotes higher stability. According to the present model of apical dendrite function, soma depolarizations can be reduced in variability and maintained within subthreshold levels by increasing the distance that EPSPs propagate along the apical dendrite. The close relationship between sustained attention and the electrical field potentials produced by repeated EPSP propagations in apical dendrites is supported in a brief review of sustained attention experiments that have employed measures of EEG, ERS/ERD, ERP, and LFP.
