Chagas disease (CD) is a neglected illness affecting approximately seven million individuals, with vector transmission occurring via triatomine bugs. The Rhodniini tribe comprises 24 species, grouped into the Rhodnius and Psammolestes genera. Given the importance of accurately identifying CD vectors, the taxonomy of Psammolestes spp. was revisited using morphological and morphometric data. Specimens of P. tertius, P. coreodes, and P. arthuri were collected, and the morphological characteristics of the head, thorax, abdomen, and eggs were analyzed. Morphometric studies of eggs were also conducted. Dichotomous keys allowing for the differentiation of Psammolestes spp. were elaborated based on adult insect and egg morphological characteristics. Through these studies, it was possible to differentiate the three Psammolestes species and confirm that this genus should not be classified under the Rhodnius genus, contributing to Rhodniini taxonomy. Abstract Background The evolutionary history of biodiversity in South America has been poorly studied in the seasonal dry tropical forest (SDTF). Species diversification in this ecosystem may have a twofold explanation. First, intermittent connections in the middle and late Pleistocene promoted species dispersal and/or genetic connectivity between lineages isolated in disjunct patches of forest. Second, allopatric speciation proceeded immediately after the formation and colonization of the SDTF in the Neogene. Here we studied the diversification of Psammolestes, a genus endemic of the SDTF and naturally infected with Trypanosoma cruzi (agent of Chagas disease), using a combination of phylogenetic, population genetics and niche model methods, and evaluated the reliability of the three morphospecies currently recognized. Results Our multilocus analyses recovered P. coreodes and P. tertius in a monophyletic clade sister to P. arthuri. Species delimitation tests recovered these lineages as different species despite the shared genetic variation observed between P. coreodes and P. tertius in five genes. Also, genetic variation of the genus clustered in three groups that were consistent with the three morphospecies. Our demographic model predicted a scenario of divergence in absence of gene flow, suggesting that mixed haplotypes may be the result of shared ancestral variation since the divergence of the subtropical-temperate species P. coreodes and P. tertius. In contrast, the tropical species P. arthuri was highly differentiated from the other two in all tests of genetic structure, and consistently, the Monmonier’s algorithm identified a clear geographical barrier that separates this species from P. coreodes and P. tertius. Conclusions We found three genetically structured lineages within Psammolestes that diverged in absence of gene flow in the late Miocene. This result supports a scenario of species formation driven by geographical isolation rather than by divergence in the face of gene flow associated with climatic oscillations in the Pleistocene. Also, we identified the Amazon basin as a climatic barrier that separates tropical from subtropical-temperate species, thus promoting allopatric speciation after long range dispersion. Finally, each species of Psammolestes occupies different climatic niches suggesting that niche conservatism is not crucial for species differentiation. These findings influence the current vector surveillance programs of Chagas disease in the region.

Rhodnius includes 21 species that can transmit Trypanosoma cruzi (the etiological agent of Chagas disease, also known as American trypanosomiasis) and Trypanosoma rangeli. The Rhodnius prolixus complex comprises seven species, including Rhodnius marabaensis, Rhodnius prolixus, and Rhodnius robustus, which occur in the northern region of Brazil. Since both adults and immatures can carry T. cruzi, in this study the five nymphal instars of the three species mentioned were dorsally characterized. Methods Using microscopy, morphometrics, and geometric morphometrics, the present work measures and describes the morphological characters of the five nymphal instars of R. marabaensis, R. prolixus, and R. robustus. Results The study enabled the characterization of all five nymphal instars, as well as the distinction between the three species in each of their instars. Conclusions The morphological, morphometrics of the head, thorax, and abdomen and geometric morphometrics studies of the head enabled the specific distinction of these three species in all five instars. In Latin America, Chagas disease has been mostly transmitted to humans by contact with the feces or urine of triatomine species infected with the protozoan Trypanosoma cruzi. There are currently 156 species in the subfamily Triatominae, distributed in 18 genera and five tribes. The prolixus group of the genus Rhodnius is composed of 11 species. Rhodnius marabaensis was the last species described and considered in this grouping of vectors. Knowledge about the biology, ecology, and behavior of these vectors is of great epidemiological importance, and in order to expand the knowledge of the biology of R. marabaensis, this paper describes the biological cycle and emergence rates of the species under laboratory conditions. The experiment was carried out at temperatures ranging from 15.5 to 29 °C (average of 24 °C) and humidity ranging from 51.4 to 72.2 (average of 63). For each of the fifteen couples, the egg emergence rate was calculated throughout the oviposition period. The oviposition period lasted from February to September, and the emergence rate varied between 13.9 and 53.3%. R. marabaensis presented an emergence rate of 46.7% and a total biological cycle of 193 days (the mean time required for emergence (25.1 days), 1st nymphal instar (19.4 days), 2nd nymphal instar (22.1 days), 3rd nymphal instar (26.2 days), 4th nymphal instar (29.3 days), and 5th nymphal instar (70.9 days)). Based on the biological cycle of R. marabaensis and 14 other Rhodnius species already described in the literature, it was also possible to calculate the averages for the groups prolixus, pictipes, and pallescens and, mainly, for the genus Rhodnius, contributing to the knowledge of this important group of Chagas disease vectors. Rhodnius prolixus is the principal vector of Trypanosoma cruzi, the aetiological agent of Chagas disease in American countries. This insect is haematophagous during all life cycles and, to antagonize its haemostatic, inflammatory and immune systems, it secretes saliva while feeding on the vertebrate host's blood. Here, we investigated characteristic changes of the salivary glands (SG) that occur during insect development. Two pairs of lobules and ducts comprise the SG of R. prolixus. The organ's size increases over time, but the microanatomical structures are preserved during insect development. Both lobules have a single layer epithelium formed by binucleated cells, which surrounds the saliva reservoir. The principal lobule presents higher polysaccharide and total protein contents than the accessory lobe. A network of external muscle layers is responsible for organ contraction and saliva release. Apocrine, merocrine and holocrine secretion types occur in the secretory epithelium. Dopamine, serotonin and tyrosine-hydroxylase are neural-related molecules that regulate SG function both during and after feeding.

Several species can transmit the protozoan parasite Trypanosoma cruzi, the causative agent of Chagas disease, significantly impacting public health in Latin American countries. The Triatominae can be classified into five tribes, of which the Rhodniini is very important because of its large vector capacity and wide geographical distribution. The Rhodniini tribe comprises 23 (without R. taquarussuensis) species and although several studies have addressed their taxonomy using morphological, morphometric, cytogenetic, and molecular techniques, their evolutionary relationships remain unclear, resulting in inconsistencies at the classification level. Conflicting hypotheses have been proposed regarding the origin, diversification, and identification of these species in Latin America, muddying our understanding of their dispersion and current geographic distribution. Clarifying these factors can help for the design of vector control strategies. The aim of this review is to depict the different approaches used for taxonomy of the Rhodniini and to shed light on their evolution and biogeography.

Rhodnius barretti , a new triatomine species, is described based on adult specimens collected in rainforest environments within the Napo ecoregion of western Amazonia (Colombia and Ecuador). R. barretti resembles Rhodnius robustus s.l. , but mitochondrial cytochrome b gene sequences reveal that it is a strongly divergent member of the “robustus lineage”, i.e., basal to the clade encompassing Rhodnius nasutus , Rhodnius neglectus , Rhodnius prolixus and five members of the R. robustus species complex. Morphometric analyses also reveal consistent divergence from R. robustus s.l. , including head and, as previously shown, wing shape and the length ratios of some anatomical structures. R. barretti occurs, often at high densities, in Attalea butyracea and Oenocarpus bataua palms. It is strikingly aggressive and adults may invade houses flying from peridomestic palms. R. barretti must therefore be regarded as a potential Trypanosoma cruzi vector in the Napo ecoregion, where Chagas disease is endemic.

Psammolestes tertius Lent & Jurberg, 1965 is a sylvatic triatomine bug species that occurs in thornbird nests in Brazil. We studied infestation of 527 nests of Phacellodomus rufifrons Wied, 1821 and Phacellodomus ruber Vieillot, 1817 by P. tertius in eight Brazilian states across the Cerrado and Caatinga ecoregions. Psammolestes tertius occurred in 44% of the nests, mainly in those of P. ruber. Overall, 3,261 specimens (52% nymphs) were captured, with an average of 14 bugs per infested nest (range 1-75). While P. tertius occurrence and age structure (percentage of nymphs) were similar in Cerrado and Caatinga nests, infestation intensity (bugs/infested nest) was higher in the Cerrado. Last-stage nymphs and adults were predominant in P. tertius populations from both P. ruber and P. rufifrons nests. The results indicate that Phacellodomus nests are favourable habitats for P. tertius in the open/dry ecoregions of Brazil. The percentage of adult P. tertius specimens (48%) was higher than that observed in other triatomine species (usually less than 5% under either experimental or natural conditions), suggesting that adult bugs tend to remain in thornbird nests. The stable blood supply provided by resident/breeding birds would favor steady P. tertius infestation of thornbird nests across Brazilian Cerrado and Caatinga ecoregions.

In the present work, we investigated whether it is possible to detect morphometric changes in Rhodnius neglectus Lent, 1954 (a candidate vector of Chagas disease in Central Brazil) populations in the transition from sylvatic to laboratory conditions. We analyzed size and shape variation in wings of sylvatic parents and their laboratory descendents (first, third and fifth generations) using geometric morphometric techniques. Sexual size dimorphism and shape of wings were maintained, but wing size decreased from sylvatic specimens to their laboratory generations. Size variation in R. neglectus should reflect the expected morphometric changes between sylvatic and domestic populations and can be applied to analyze the level of adaptation of R. neglectus to domestic habitats. This information might be useful to detect persistent infestations in dwellings after insecticide application, or new infestations from the sylvatic environment, and is therefore important to guide vector surveillance strategies for Chagas disease.

Abstract: Chagas disease incidence has sharply declined over the last decade. Long-term disease control will, however, require extensive, longitudinal surveillance systems capable of detecting (and dealing with) reinvasion-reinfestation of insecticide-treated dwellings by non-domiciliated triatomines. Sound surveillance design calls for reliable data on vector ecology, and these data must cover different spatial scales. We conducted a multi-scale assessment of ecological and evolutionary trends in members of the tribe Rhodniini, including (i) a macroscale analysis of Rhodniini species richness and composition patterns across the Americas, and (ii) a detailed, mesoscale case-study of ecological and behavioural trends in Rhodnius neglectus and R. nasutus. Our macroscale overview provides some comprehensive insights about key mechanisms/processes probably underlying ecological and genetic diversification in the Rhodniini. These insights translate into a series of testable hypotheses about current species distributions and their likely causes. At the landscape scale, we used geometric morphometrics to identify dubious specimens as either R. neglectus or R. nasutus (two near-sibling species), and studied palm tree populations of these two vector taxa in five geographical areas. The data suggest that deforestation and the associated loss of habitat and host diversity might increase the frequency of vector–human contact (and perhaps Trypanosoma cruzi infection rates in vectors). Surveillance in central-northeastern Brazil should prioritise deforested landscapes where large palm trees (e.g., Attalea, Mauritia, Copernicia, Acrocomia or Syagrus) occur near houses. We anticipate that, by helping define the distribution patterns and ecological preferences of each species, multi-scale research will significantly strengthen vector surveillance systems across Latin America. Abstract: Of the currently known 140 species in the family Reduviidae, subfamily Triatominae, those which are most important as vectors of the aetiologic agent of Chagas disease, Trypanosoma cruzi, belong to the tribes Triatomini and Rhodniini. The latter not only transmit T. cruzi but also Trypanosoma rangeli, which is considered apathogenic for the mammalian host but can be pathogenic for the vectors. Using different molecular methods, two main lineages of T. cruzi have been classified, T. cruzi I and T. cruzi II. Within T. cruzi II, five subdivisions are recognized, T. cruzi IIa–IIe, according to the variability of the ribosomal subunits 24Sα rRNA and 18S rRNA. In T. rangeli, differences in the organization of the kinetoplast DNA separate two forms denoted T. rangeli KP1+ and KP1−, although differences in the intergenic mini-exon gene and of the small subunit rRNA (SSU rRNA) suggest four subpopulations denoted T. rangeli A, B, C and D. The interactions of these subpopulations of the trypanosomes with different species and populations of Triatominae determine the epidemiology of the human-infecting trypanosomes in Latin America. Often, specific subpopulations of the trypanosomes are transmitted by specific vectors in a particular geographic area. Studies centered on trypanosome–triatomine interaction may allow identification of co-evolutionary processes, which, in turn, could consolidate hypotheses of the evolution and the distribution of T. cruzi/T. rangeli-vectors in America, and they may help to identify the mechanisms that either facilitate or impede the transmission of the parasites in different vector species. Such mechanisms seem to involve intestinal bacteria, especially the symbionts which are needed by the triatomines to complete nymphal development and to produce eggs. Development of the symbionts is regulated by the vector. T. cruzi and T. rangeli interfere with this system and induce the production of antibacterial substances. Whereas T. cruzi is only subpathogenic for the insect host, T. rangeli strongly affects species of the genus Rhodnius and this pathogenicity seems based on a reduction of the number of symbionts. Abstract Genetic relationships among 10 species of bugs belonging to the tribe Rhodniini (Hemiptera: Reduviidae), including some important vectors of Chagas disease, were inferred from allozyme analysis of 12 enzyme loci (out of 21 enzyme systems examined), using agarose gel electrophoresis. These species formed two clusters: one comprising Rhodnius brethesi , R. ecuadoriensis , R. pallescens and R. pictipes ; the other with Psammolestes tertius , Rhodnius domesticus and the Rhodnius prolixus group comprising R. nasutus , R. neglectus , R. prolixus and R. robustus . The resulting tree was [((R. ecuadoriensis , R. pallescens ) R. brethesi ) R. pictipes ], [R. domesticus (P. tertius {(R. nasutus , R. neglectus ) (R. prolixus , R. robustus )})]. Rhodnius nasutus and R. neglectus differed by only one locus, whereas no diagnostic loci were detected between R. prolixus and R. robustus (22 loci were analysed for these four species), despite considerable DNA sequence divergence between species in each of these pairs. Allozymes of the R. prolixus group showed greater similarity with Psammolestes tertius than with other Rhodnius spp., indicating that Rhodnius is paraphyletic and might include Psammolestes .
