The safe and stable operation of the transformer is the key to ensure the reliable operation of the power grid. Loose windings will produce a vibration acceleration signal with twice the power supply frequency signal, i.e., 100 Hz, as the main spectral component. The realization of transformer winding vibration acceleration detection can effectively determine the operating status of the transformer winding. In this report, a dual-cantilever beam Fiber Bragg Grating (FBG) vibration acceleration sensor for transformer winding with a two-point package structure was designed based on theoretical analysis and finite element simulation of COMSOL Multiphysics software. Through the demodulation of the center wavelength change to achieve the detection of external vibration acceleration and to achieve the vibration acceleration sensitivity of 37.813 pm/g at 100 Hz, linearity up to 99.97%, cross-sensitivity is less than 17.4%. The resonant frequency of the sensor is 134 Hz, and the sensitivity of the vibration acceleration at the resonant frequency point is 425.52 pm/g. The sensor is installed under the transformer to measure the vibration acceleration of the transformer winding. The vibration component under 100 Hz frequency could be obviously captured, and the FFT of the center wavelength captured the amplitude signal to realize the measurement of the vibration acceleration of the transformer winding less than 0.02 g micro-vibration acceleration, which could satisfy the general vibration range of the transformer winding.

BackgroundThe European League of Rheumatology(EULAR)guidelines recommend Janus kinase (JAK) inhibitors for patients with moderate to severe rheumatoid arthritis (RA) who are insensitive or under-responsive to conventional synthetic disease-modifying anti-rheumatic drugs (csDMARDs). But there was no recommendation for which one was preferred in five currently approved JAK inhibitors. The objective of this network meta-analysis study was to evaluate the efficacy of five JAK inhibitors as monotherapy and combination therapy in patients with moderate-to-severe active rheumatoid arthritis.MethodsThe randomized controlled trials (RCTs) of tofacitinib, baricitinib, upadacitinib, filgotinib and peficitinib as monotherapy or combined with csDMARD in the treatment of active RA were searched in database of PubMed, Embase, Web of Science and Cochrane Library, up to December 2023. The control group included placebo or csDMARD. Outcome indicators included American College of Rheumatology 20% response (ACR20), ACR50, ACR70 and the percentage of patients achieving 28-joint disease activity score using C-reactive protein (DAS28(CRP))

Conventional plasma electrolytic oxidation treatments produce oxide coatings with micron-scale discharge pores, resulting in insulation and wear and corrosion resistance far below that expected of highly dense Al2O3 coatings. The introduction of cathodic polarization during the plasma electrolytic oxidation process, especially when the applied cathode-to-anode current ratio (Rpn) is greater than 1, triggers a unique plasma discharge phenomenon known as “soft sparking”. The soft spark discharge mode significantly improves the densification of the anode ceramic layer and facilitates the formation of the high-temperature α-Al2O3 phase within the coating. Although the soft spark discharge phenomenon has been known for a long time, the growth behavior of the coating under its discharge mode still needs to be studied and improved. In this paper, the growth behavior of the coating before and after soft spark discharge is investigated with the help of the micro-morphology, phase composition and element distribution of a homemade fixture. The results show that the ceramic layer grows mainly along the oxide–electrolyte direction before the soft spark discharge transformation; after the soft spark discharge, the ceramic layer grows along the oxide–substrate direction. It was also unexpectedly found that, under soft spark discharge, the silicon element only exists on the outside of the coating, which is caused by the large size and slow migration of SiO32−, which can only enter the ceramic layer and participate in the reaction through the discharge channel generated by the strong discharge. In addition, it was also found that the relative phase content of α-Al2O3 in the coating increased from 0.487 to 0.634 after 10 min of rotary spark discharge, which is an increase of 30.2% compared with that before the soft spark discharge transition. On the other hand, the relative phase content of α-Al2O3 in the coating decreased from 0.487 to 0.313 after 20 min of transfer spark discharge, which was a 55.6% decrease compared to that before the soft spark discharge transformation.

The correlation between negative pulse and the black electrolyte properties of magnesium alloy micro-arc oxidation and the treated area was investigated by introducing a negative pulse electric field. The physical phase composition, microstructure, elemental distribution, and content of the coating were analyzed using X-ray diffraction (XRD), scanning electron microscopy (SEM), and energy dispersive spectroscopy (EDS). The results showed that the introduction of negative pulses favored the generation of MgO and MgSiO3 contents in the coatings, and an increase in the MgO phase was found in the coatings formed in the failed electrolytes; the microporous size and microcracks of the coatings were gradually and significantly reduced; the average consumption of Cu ions was 0.0453 g/L·dm2, which is only 26% of that in the unipolar condition; the introduction of the negative pulses significantly improved the “anomalous consumption” of Cu ions. The introduction of negative pulse can significantly improve the “abnormal consumption” of copper ions, which is attributed to the change in the electric field by negative pulse, which makes the cathode-enriched Cu ions migrate to the anode and reduces the reduction and precipitation of Cu ions at the cathode.

Abstract Objective Connexin 43 (Cx43) is a critical gene for maintaining myocardial homeostasis. Interestingly, Cx43 and stress‐induced phosphoprotein 1 (STIP1) were recorded to be lowly expressed in ischemia/reperfusion (I/R). However, their impacts on reperfusion arrhythmia (RA) remain to be explored. Our study aimed to find out the related underlying mechanisms. Methods After the establishment of an isolated heart model through Langendorff perfusion, the heart rate, conduction activation time, conduction velocity, and conduction direction of the left ventricle were evaluated, along with the apoptotic rate detection in the collected myocardial tissues. After the construction of a hypoxia/reoxygenation (H/R)‐induced cellular model, cell apoptosis, intercellular communication, cell viability, and the content of reactive oxygen species, superoxide dismutase, malondialdehyde, and lactic dehydrogenase were measured. The expression of Cx43 and STIP1 was determined in both rat heart and cell models. The bindings of STIP3 and Cx43 to heat shock protein 90 (HSP90) and heat shock protein 70 (HSP70) were verified. Results Relative to the corresponding controls, Cx43 and STIP1 were decreased in myocardial tissues of RA rats and H/R‐stimulated H9C2 cells, where Cx43‐binding HSP70 and HSP90 were respectively increased and decreased, and ubiquitination level of Cx43 was enhanced. STIP1 overexpression promoted protein expression of Cx43, intercellular communication, and cell viability, and reduced cell apoptosis and oxidative stress in H/R‐stimulated H9C2 cells. Conclusion STIP1 promoted Cx43 expression to improve intercellular communication and reduce oxidative stress in H/R‐stimulated H9C2 cells.

Abstract Magnetic resonance imaging (MRI) and magnetic resonance spectroscopy (MRS) have made great successes in clinical diagnosis, medical research, and neurological science. MRI provides high resolution anatomical images of tissues/organs, and MRS provides information of the functional molecules related to a specific tissue/organ. However, it is difficult for classic MRI/MRS to selectively image/probe a specific metabolite molecule other than the water or fat in tissues/organs. This greatly limits their applications on the study of the molecular mechanism(s) of metabolism and disease. Herein, we report a series of molecularly targeted MRI/MRS methods to target specific molecules. The optimal control method was used to efficiently prepare the singlet spin orders of varied multi-spin systems and in turn greatly expand the choice of the targeted molecules in the molecularly targeted MRI/MRS. Several molecules, such as N-acetyl-l-aspartic acid (NAA), dopamine (DA), and a tripeptide (alanine-glycine-glycine, AGG), have been used as targeted molecules for molecularly targeted MRI and MRS. We show in vivo NAA-targeted 1H MRS spectrum of a human brain. The high-resolution signal of NAA suggests a promising way to study important issues in molecular biology at the molecular level, e.g., measuring the local pH value of tissue in vivo, demonstrating the high potential of such methods in medicine.

As intelligent driving vehicles came out of concept into people’s life, the combination of safe driving and artificial intelligence becomes the new direction of future transportation development. Autonomous driving technology is developing based on control algorithms and model recognitions. In this paper, a cloud-based interconnected multi-sensor fusion autonomous vehicle system is proposed that uses deep learning (YOLOv4) and improved ORB algorithms to identify pedestrians, vehicles, and various traffic signs. A cloud-based interactive system is built to enable vehicle owners to master the situation of their vehicles at any time. In order to meet multiple application of automatic driving vehicles, the environment perception technology of multi-sensor fusion processing has broadened the uses of automatic driving vehicles by being equipped with automatic speech recognition (ASR), vehicle following mode and road patrol mode. These functions enable automatic driving to be used in applications such as agricultural irrigation, road firefighting and contactless delivery under new coronavirus outbreaks. Finally, using the embedded system equipment, an intelligent car was built for experimental verification, and the overall recognition accuracy of the system was over 96%.

The improved SIFT algorithm effectively identifies a variety of commodities. At the same time, relying on the public cloud platform, discount information is recommended through WeChat programs, and the development of member users login interface. After comprehensive calculation, the total price is displayed by the EAIDK-610 user interface edited by Tkinter to complete the checkout. The LD-14 radar used can realize the functions of autonomous follow-up and pedestrian obstacles. When a sensor in the system fails or the detection deviation is too large, the multi-sensor fusion technology can ensure the normal operation of the system due to a certain degree of redundant. Improve the credibility and effectiveness of the data as a whole. This not only solves the problem of queuing and checkout in the supermarket, but also helps brand merchants to achieve precise marketing and scenario access. Finally, the use of computer vision technology and various embedded hardware devices to build a radar self-following the hardware prototype of the shopping cart for experimental verification. The average accuracy of APs in the system is as high as 98.31%and 97.28%, respectively.

Abstract Objectives To evaluate the therapeutic effect of neoadjuvant therapy (NAT) followed by radical hysterectomy and concurrent chemoradiotherapy (CCRT) in stage IB2 and IIA2 squamous cervical cancer (SCC) and investigate the value of apparent diffusion coefficient (ADC) in outcome evaluation of different treatment strategies in the patients. Methods A total of 149 patients with IB2 and IIA2 SCC who underwent pretreatment MRI and DWI scan were included. Patients were treated with NAT + RH or CCRT. Clinical indices and pathological factors were recorded. The imaging indices were measured including tumor size and tumor ADC values. Intraclass correlation coefficient was employed to evaluate the consistency of the indices measured by two observers. ROC curves were used to evaluate the cutoff values of clinical and imaging indices. Kaplan–Meier and Cox proportional hazard model were used to analyze the independent factors of disease-free survival (DFS). Results The median follow-up period was 42.3 months. SCC-Ag, ADCmax and ADCmin were independent factors for DFS in the entire cohort. SCC-Ag, ADCmin and vascular invasion were independent factors for DFS in NAT + RH group. ADCmax and ADCmin were independent factors for DFS in CCRT group. ADCmin was the strongest independent factor for DFS in NAT + RH group, while ADCmax was that in CCRT group. Conclusion The NAT + RH patients had similar DFS to that of CCRT in IB2 and IIA2 SCC, which could be a potential feasible alternative treatment. ADCmin and ADCmax were more valuable in evaluating the outcome of patients who underwent NAT + RH or CCRT, respectively.

An improved Gene Expression Programming (GEP) by Total Variation (TV) method is introduced in this paper for modeling mechanical properties of multi-components. To verify the scheme, the Glass Fiber Reinforced Pipe materials test under 10000h, and the Carbon Fiber Reinforced Self-lubrication materials for pantograph of high speed rail way are discussed by TV-GEP. The improved method obviously overcomes the Runge problem and has less error than Response Surface Method (RSM) in prediction. The algebra equations of results verify the wear/Hardness theory and discovers a relation between Resistivity/Compressive of Carbon Fiber Reinforced Self-lubrication materials.

Although many models are available to predict prognosis of heart failure patients, most tools combining survival analysis are based on proportional hazard model. Non-linear machine learning algorithms would overcome the limitation of the time-independent hazard ratio assumption and provide more information in readmission or mortality prediction among heart failure patients.The present study collected the clinical information of 1796 hospitalized heart failure patients surviving during hospitalization in a Chinese clinical center from December 2016 to June 2019. A traditional multivariate Cox regression model and three machine learning survival models were developed in derivation cohort. Uno's concordance index and integrated Brier score in validation cohort were calculated to evaluate the discrimination and calibration of different models. Time-dependent AUC and Brier score curves were plotted to assess the performance of models at different time phases.

Background: Microstructural changes in deep gray matter (DGM) nuclei are related to physiological behavior, cognition, and memory. Therefore, it is critical to study age-dependent trajectories of biomarkers in DGM nuclei for understanding brain development and aging, as well as predicting cognitive or neurodegenerative diseases. Objectives: We aimed to (1) characterize age-dependent trajectories of mean susceptibility, adjusted volume, and total iron content simultaneously in DGM nuclei using quantitative susceptibility mapping (QSM); (2) examine potential contributions of sex related effects to the different age-dependence trajectories of volume and iron deposition; and (3) evaluate the ability of brain age prediction by combining mean magnetic susceptibility and volume of DGM nuclei. Methods: Magnetic susceptibilities and volumetric values of DGM nuclei were obtained from 220 healthy participants (aged 10–70 years) scanned on a 3T MRI system. Regions of interest (ROIs) were drawn manually on the QSM images. Univariate regression analysis between age and each of the MRI measurements in a single ROI was performed. Pearson correlation coefficients were calculated between magnetic susceptibility and adjusted volume in a single ROI. The statistical significance of sex differences in age-dependent trajectories of magnetic susceptibilities and adjusted volumes were determined using one-way ANCOVA. Multiple regression analysis was used to evaluate the ability to estimate brain age using a combination of the mean susceptibilities and adjusted volumes in multiple DGM nuclei. Results: Mean susceptibility and total iron content increased linearly, quadratically, or exponentially with age in all six DGM nuclei. Negative linear correlation was observed between adjusted volume and age in the head of the caudate nucleus (CN; R2 = 0.196, p < 0.001). Quadratic relationships were found between adjusted volume and age in the putamen (PUT; R2 = 0.335, p < 0.001), globus pallidus (GP; R2 = 0.062, p = 0.001), and dentate nucleus (DN; R2 = 0.077, p < 0.001). Males had higher mean magnetic susceptibility than females in the PUT (p = 0.001), red nucleus (RN; p = 0.002), and substantia nigra (SN; p < 0.001). Adjusted volumes of the CN (p < 0.001), PUT (p = 0.030), GP (p = 0.007), SN (p = 0.021), and DN (p < 0.001) were higher in females than those in males throughout the entire age range (10–70 years old). The total iron content of females was higher than that of males in the CN (p < 0.001), but lower than that of males in the PUT (p = 0.014) and RN (p = 0.043) throughout the entire age range (10–70 years old). Multiple regression analyses revealed that the combination of the mean susceptibility value of the PUT, and the volumes of the CN and PUT had the strongest associations with brain age (R2 = 0.586). Conclusions: QSM can be used to simultaneously investigate age- and sex- dependent changes in magnetic susceptibility and volume of DGM nuclei, thus enabling a comprehensive understanding of the developmental trajectories of iron accumulation and volume in DGM nuclei during brain development and aging.

Abstract Background Diabetes mellitus (DM) is a complex metabolic disease that is caused by a complex interplay between genetic and environmental factors. This research aimed to investigate the association of genetic polymorphisms in PDX1 and MC4R with T2DM risk. Methods The genotypes of 10 selected SNPs in PDX1 and MC4R were identified using the Agena MassARRAY platform. We utilized odds ratio (OR) and 95% confidence intervals (CIs) to assess the correlation between genetic polymorphisms and T2DM risk. Results We found that PDX1-rs9581943 decreased susceptibility to T2DM among in a Chinese Han population (OR = 0.76, p = 0.045). We also found that selected genetic polymorphisms in PDX1 and MC4R could modify the risk of T2DM, which might also be influenced by age, sex, BMI, smoking status, and drinking status (p

Abstract Background Evidence from genetic epidemiology indicates that type 2 diabetes (T2D) has a strong genetic basis. Activated STAT4 has an inflammatory effect, and STAT4 is an important mediator of inflammation in diabetes. Our study aimed to study the association between STAT4 single nucleotide polymorphisms (SNPs) and T2D susceptibility in Chinese Han population. Methods We conducted a 'case–control' study among 500 T2D patients and 501 healthy individuals. 5 candidate STAT4 SNPs were successfully genotyped. The association between SNPs and T2D susceptibility under different genetic models was evaluated by logistic regression analysis. ‘SNP-SNP’ interaction was analyzed and completed by multi-factor dimensionality reduction (MDR). Finally, we evaluated the differences of clinical characteristics under different genotypes by one-factor analysis of variance. Results The overall results showed that STAT4 rs3821236 was associated with increasing T2D risk under allele (OR 1.23, p = 0.020), homozygous (OR 1.51, p = 0.025), dominant (OR 1.36, p = 0.029), and additive models (OR 1.23, p = 0.020). The results of stratified analysis showed that rs3821236, rs11893432, and rs11889341 were risk factors for T2D among participants ≤ 60 years old. Only rs11893432 was associated with increased T2D risk among female participants. There was also a potential association between rs3821236 and T2D with nephropathy risk. STAT4 rs11893432, rs7574865 and rs897200 were significantly associated with lysophosphatidic acid, cystatin C and thyroxine t4, respectively. Conclusion The genetic polymorphisms of STAT4 is potentially associated with T2D susceptibility of Chinese population. In particular, rs3821236 is significantly associated with T2D risk both in the overall and several subgroup analyses. Our study may provide new ideas for T2D individualized diagnosis/protection.

Introduction Astaxanthin (ASX) is carotenoid with the highest antioxidant activity in various cell types and reverse atherosclerosis. However, the roles and detailed mechanisms of ASX in atherosclerosis associated endothelial injury remains unclear. Methods In vitro atherosclerosis model was established in HUVECs by incubation with oxidized low-density lipoprotein (ox-LDL). Cell viability and oxidative stress were measured. The mRNA and protein expressions of lectin-like ox-LDL receptor (LOX-1) and other related genes were determined. Results ox-LDL reduced cell viability of HUVECs, and induced oxidative stress, as evidenced by elevated cellular malondialdehyde (MDA) and decreased superoxide dismutase (SOD). Pretreatment with ASX (50, 100, 200, and 400 μM) markedly reversed the reduction in cell viability and an increase in migration of HUVECs induced by ox-LDL (50 μg/mL). ASX attenuated the increase in the endothelial-to-mesenchymal transition (EndMT) process, as evidenced by increased CD31 and decreased α-SMA and vimentin proteins by ASX treatment in HUVECs. Furthermore, ASX attenuated the increase in MDA and decrease in SOD induced by ox-LDL, increased supernatant NO production, attenuated the increase in iNOS and decrease in eNOS in HUVECs with ox-LDL. ASX enhanced mRNA and protein expressions of the lectin-like ox-LDL receptor (LOX-1), which was dependent on ASX’s antioxidant activity. The inhibitory effect of ASX on EndMT could be abolished by overexpression of LOX-1 in HUVECs induced by ox-LDL. Conclusions Our data speculate that ASX prevents ox-LDL-induced endothelial cell injury and EndMT by inducing antioxidant property (SOD and NO) and decreasing LOX-1 expression.

The phase structures, magnetic properties, electronic structures, and anisotropy of the full Heusler alloy Co2NbGa were studied using the first-principles method, and the results were compared with experiments. The L21-type cubic structure was the most stable, which was in good agreement with the experimental measurement. The Curie temperature obtained by the mean field theory was also very close to the experimental value. The electronic structures and magnetic properties showed that the Co2NbGa alloy was a half-metallic ferromagnet with 100% spin polarization at equilibrium, and the robustness of the half-metallic feature was preserved in lattice ranges of 5.79 to 6.42 Å. The Co2NbGa alloy was an anisotropic material with stable mechanical properties at L21-type cubic and XA-type tetragonal structures, and the influence of Hubbard U and vacancy defects on the physical nature was also studied and discussed. The 100% spin polarization, high Curie temperature, and stable equilibrium structure make Co2NbGa alloy a promising candidate in spintronics and magnetoelectronic.

Background: Patients with chronic obstructive pulmonary disease (COPD) and obstructive sleep apnea (OSAS) overlap syndrome (OS) are thought to be at increased risk for cardiovascular diseases.Objective: To evaluate the burden of cardiovascular diseases and long-term outcomes in patients with OS.Methods: This was a retrospective cohort study. The prevalence of cardiovascular diseases and 1-year mortality were compared among patients diagnosed with OS (OS group), COPD alone (COPD group) and OSAS alone (OSAS group), and Cox proportional hazards models were used to assess independent risk factors for all-cause mortality.Results: Overall, patients with OS were at higher risk for pulmonary hypertension (PH), heart failure and all-cause mortality than patients with COPD or OSAS (all p < 0.05). In multivariate Cox regression analysis, the Charlson comorbidity index (CCI) score [adjusted hazard ratio (aHR): 1.273 (1.050–1.543); p = 0.014], hypertension [aHR: 2.006 (1.005–4.004); p = 0.048], pulmonary thromboembolism (PTE) [aHR: 4.774 (1.335–17.079); p = 0.016] and heart failure [aHR: 3.067 (1.521–6.185); p = 0.002] were found to be independent risk factors for 1-year all-cause mortality.Conclusion: Patients with OS had an increased risk for cardiovascular diseases and 1-year mortality. More efforts are needed to identify the causal relationship between OS and cardiovascular diseases, promoting risk stratification and the management of these patients.

A novel spin gapless semiconductor material Ba2MnTeO6 is found and reported by first-principles calculations. In this paper, we study the influence of spin orbit coupling, strain and Coulomb interaction on the properties of ferromagnetic Ba2MnTeO6 in order to test the stability of physical properties. It turns out that the spin orbit coupling has no significant effect on the electronic structures of the Ba2MnTeO6 at equilibrium state. Under the action of uniform compressive strain and tensile strain, Ba2MnTeO6 transforms from spin gapless semiconductor to semiconductor and half-metal respectively. At the same time, as the strain increases, the semiconductor band gap gradually widens, and the half-metallic gap gradually narrows. After considering Coulomb interaction, Ba2MnTeO6 will also be transformed into a semiconductor, and the semiconductor band gap will increase with the increasing of U values. Furthermore, the mechanical performance studies show that Ba2MnTeO6 is an anisotropic material with stable structure. As the first pure double perovskite type spin gapless semiconductor, the high spin polarization and accurately adjustable physical properties of ferromagnetic Ba2MnTeO6 indicate that it has broad application prospects in the fields of spintronics and optoelectronic materials and devices.

Background. Atherosclerosis is a chronic process that takes place in the vascular wall and causes various cardiovascular diseases (CVDs). Micro-RNA-149 (miR-149) mediates many physiological and pathological processes, including atherosclerosis. However, it is unclear about the roles of miR-149 in endothelial injury. Here, we explored the protective effect and related mechanism of miR-149 in endothelial cells induced with oxidized low-density lipoprotein (ox-LDL). Methods. Human endothelial cell lines (HUVECs) were exposed to ox-LDL to induce endothelial injury. Cell viability was determined by the CCK-8 assay. Autophagy was detected by immunofluorescence. RT-qPCR and western blot were carried out to determine the mRNA and protein expressions of Akt and mTOR. Results. The miR-149 level in HUVECs was reduced by ox-LDL (100 μg/mL) incubation in a time-dependent manner. miR-149-mimic transfection markedly protected HUVECs from ox-LDL-induced injury, with increased cell viability and reduced caspase-3 activity. miR-149 mimics enhanced HUVEC autophagy, which was induced initially by ox-LDL. miR-149 mimics also markedly downregulated the expression of Akt, p-Akt, mTOR, and p-mTOR in ox-LDL-treated HUVECs. The miR-149-induced protection against HUVECs injury could be reversed by cotreatment with 3-methyladenine (3-MA, an autophagy inhibitor) or insulin (an activator of Akt/mTOR pathway). Conclusions. miR-149 prevents ox-LDL-induced endothelial cell injury by enhancing autophagy via increasing Akt and mTOR expressions.

Studying the changes in extreme river runoff induced by climate change is of utmost importance, as the variability of floods directly affects life and human activities. This study examines the fluctuations and persistence of winter floods in 14 catchments in the Rika River Basin (Ukraine) and ten catchments in the Steyr River Basin (Austria). The catchments represent typical hydrological regimes in the Danube River region. The fluctuations and persistence of floods are analyzed by the hydro-genetic method and a seasonality analysis for the period 1951–2015. The results show a much more pronounced fluctuation pattern in the upper Rika catchments than in the upper Steyr catchments. This pattern indicates an increase in winter flood magnitudes between the mid-1960s and the 1990s, followed by a decrease until recently. The flood seasonality shows a large inter-annual variability in both regions. The most significant winter floods tend to occur in November and December. The winter flood fluctuations are compared with changes in associated climate characteristics, i.e., seven-day maximum precipitation, a melt index, and annual maximum snow depth. The seasonality of these characteristics has a strong inter-annual variability and only partly explains the winter flood fluctuations.

The role of soil moisture is widely accepted as a significant factor in the mass and energy balance of catchments as a controller in surface and subsurface runoff generation. The paper examines the potential of a new dataset based on advanced scatterometer satellite remote sensing of soil moisture (ASCAT) for multiple objective calibrations of a dual-layer, conceptual, semi-distributed hydrological model. The surface and root zone soil moisture indexes based on ASCAT data were implemented into calibration of the hydrological model. Improvements not only in the instrument specifications, i.e., better temporal and spatial sampling, but also in the higher radiometric accuracy and retrieval algorithm, were applied. The analysis was performed in 209 catchments situated in different physiographic and climate zones of Austria for the period 2007–2018. We validated the model for two validation periods. The results show that multiple objective calibrations have a substantial positive effect on constraining the model parameters. The combined use of soil moisture and discharges in the calibration improved the soil moisture simulation in more than 73% of the catchments, except for the catchments with higher forest cover percentages. Improvements also occurred in the runoff model efficiency, in more than 27% of the catchments, mostly in the watersheds with a lower mean elevation and a higher proportion of farming land use, as well as in the Alpine catchments where the runoff is not significantly influenced by snowmelt and glacier runoff.

Exploring travel time distribution and variability patterns is essential for reliable route choices and sophisticated traffic management and control. State-of-the-art studies tend to treat different types of roads equally, which fails to provide more detailed analysis of travel time characteristics for each specific road type. In this study, based on a vast amount of probe vehicle data, 200 links inside the Third Ring Road of Beijing, China, were investigated. Four types of roads were covered including urban expressways, auxiliary roads of urban expressways, major roads, and secondary roads. The day-of-week distributions of unit distance travel time were first analyzed. Kolmogorov-Smirnov test, Anderson-Darling test, and chi-squared test were employed to test the goodness-of-fit of different distributions and the results showed lognormal distribution was best-fitted for different time periods and road types compared with normal, gamma, and Weibull distribution. In addition, four reliability measures, that is, unit distance travel time, coefficient of variation, buffer time index, and punctuality rate, were used to explore the day-of-week travel time variability patterns. The results indicated that urban expressways, auxiliary roads of urban expressways, and major roads have regular and distinct morning and afternoon peaks on weekdays. It is noteworthy that in daytime the travel times on auxiliary roads of urban expressways and major roads share similar variability patterns and appear relatively stable and reliable, while urban expressways have most reliable travel times at night. The results of analysis help enable a better understanding of the volatile travel time characteristics of each road type in urban network.

In this work, we synthesized monodispersed CdSe quantum dots (QDs) by a microfluidic method and via a bulk reaction. The structures of the CdSe QDs were characterized by X-ray powder diffraction (XRD) and high-resolution transmission electron microscopy (HR-TEM). The optical properties of the prepared CdSe QDs were determined using ultraviolet-visible absorption spectroscopy and photoluminescence spectroscopy. The CdSe QDs obtained by the microfluidic method have a faster crystal growth rate and a higher absolute photoluminescence quantum yield than those obtained via the bulk reaction. Additionally, we investigated the growth process of the CdSe QDs with increasing residence times.

The result of molecular diagnostic and detection greatly dependent on the quality and integrity of the isolated nucleic acid. In this work, we developed an automated miniaturized nucleic acid extraction device based on magnetic beads method, consisting of four components including a sample processing disc and its associated rotary power output mechanism, a pipetting module, a magnet module and an external central controller to enable a customizable and automated robust nucleic acid sample preparation. The extracted nucleic acid using 293T cells were verified using real-time polymerase chain reaction (PCR) and the data implies a comparable efficiency to a manual process, with the advantages of performing a flexible, time-saving (~10 min), and simple nucleic acid sample preparation.

Enhancing drug loading efficacy and stability of polymeric micelles remains a grand challenge. Here we develop adamantane-based star copolymers adamantane-[poly(ε-caprolactone)-b-poly(2-(diethylamino)ethyl methacrylate)-b-poly(poly(ethylene glycol) methyl ether methacrylate)]4 (Ad-(PCL-b-PDEAEMA-b-PPEGMA)4) and their self-assembled micelles for controlled drug delivery. Results show that the polymers have excellent stability in solution with low critical micelle concentration (CMC) (0.0025–0.0034 mg/mL) and the apparent base dissociation constant (pKb) of the polymers is from 5.31 to 6.05. Dynamic light scattering analysis exhibits the great environmental response capability of the pH-sensitive micelles according to particle sizes and zeta potentials. With the synergy effect of the adamantane and hydrophobic block, the micelles display the high Doxorubicin (DOX) loading efficacy (up to 22.4%). The DOX release study shows that the micelles are capable of controlled release for drug. This work indicates the Ad-(PCL-b-PDEAEMA-b-PPEGMA)4 micelles may provide new guidelines for drug control and release system in overcoming cancer treatment.

TAR DNA-binding protein 43 (TDP-43) is normally a nuclear RNA-binding protein that exhibits a range of functions including regulation of alternative splicing, RNA trafficking, and RNA stability. However, in amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration with TDP-43 inclusions (FTLD-TDP), TDP-43 is abnormally phosphorylated, ubiquitinated, and cleaved, and is mislocalized to the cytoplasm where it forms distinctive aggregates. We previously developed a mouse model expressing human TDP-43 with a mutation in its nuclear localization signal (ΔNLS-hTDP-43) so that the protein preferentially localizes to the cytoplasm. These mice did not exhibit a significant number of cytoplasmic aggregates, but did display dramatic changes in gene expression as measured by microarray, suggesting that cytoplasmic TDP-43 may be associated with a toxic gain-of-function. Here, we analyze new RNA-sequencing data from the ΔNLS-hTDP-43 mouse model, together with published RNA-sequencing data obtained previously from TDP-43 antisense oligonucleotide (ASO) knockdown mice to investigate further the dysregulation of gene expression in the ΔNLS model. This analysis reveals that the transcriptomic effects of the overexpression of the ΔNLS-hTDP-43 transgene are likely due to a gain of cytoplasmic function. Moreover, cytoplasmic TDP-43 expression alters transcripts that regulate chromatin assembly, the nucleolus, lysosomal function, and histone 3' untranslated region (UTR) processing. These transcriptomic alterations correlate with observed histologic abnormalities in heterochromatin structure and nuclear size in transgenic mouse and human brains.

Er3+/Yb3+ codoped tellurite-zinc-niobium (TZNb) glass was prepared by the melt-quenching method and used for the construction of a point all-fiber temperature sensor. The glass thermal stability and network structural properties were studied by differential thermal analysis and Raman spectrum, respectively. High glass transition temperature is beneficial to widen the working temperature range. The dependence of fluorescence intensity ratio (FIR) of green upconversion emissions on the surrounding temperature from 276 to 363 K was experimentally investigated and the maximum temperature sensitivity is 95 × 10−4 K−1 at 363 K. Strong green upconversion emission, broad temperature measurement range and high sensitivity indicate this point temperature sensor is a promising optical device for application on optical temperature sensing.

Few comprehensive studies have investigated viraemia caused by human parvoviruses (HPAVs) in China. A total of 1626 of blood samples were collected from non-HBV and HBV infected Chinese subjects (adults, N = 1279; children, N = 347) from south-western and south-eastern China. DNA from three HPAVs was detected in blood samples using PCR-based assays. The epidemiological profiles and association with HBV co-infection were also analysed. Of the 1626 blood samples tested, 138 (8.49%) were found to exhibit HPAV viraemia, including 3.51% with B19, 3.75% with HBoV and 2.52% with PARV4. The presence of B19 DNA in both child and adult, as well as that of PARV4 DNA in adult,from the south-western region was significantly higher than that from the south-eastern region (P = 0.006 for B19 in children; P = 0.026 for B19 in adults; and P = 0.014 for PARV4 in adult).However, the frequency of HBoV DNA in adults from the south-western region was significantly lower than that observed in adults from the south-eastern region (P = 0.001). Furthermore, HBoV was more prevalence in male (4.9%) than in female (1.4%) individuals. In addition, no significant correlation between HBV and HPAV co-infection was found using serum samples from Chinese adults. In conclusions,the molecular prevalence of three HPAVs in blood samples exhibited variation among different populations depending on area, age and gender; No association between HPAV and HBV infection in adults was found. Our data provide a basis for improving blood safety and preventing HPAV infection in China.

OBJECTIVE: Few comprehensive studies have searched for viruses in infants and young children with community-acquired pneumonia (CAP) in China. The aim of this study was to investigate the roles of human herpes viruses (HHVs) and other respiratory viruses in CAP not caused by typical bacterial infection and to determine their prevalence and clinical significance. METHODS: Induced sputum (IS) samples were collected from 354 hospitalised patients (infants, n = 205; children, n = 149) with respiratory illness (CAP or non-CAP) admitted to Wenling Hospital of China. We tested for HHVs and respiratory viruses using PCR-based assays. The epidemiological profiles were also analysed. RESULTS: High rate of virus detection (more than 98%) and co-infection (more than 80%) were found among IS samples from 354 hospitalised infants and children with respiratory illness in this study. Of 273 CAP samples tested, CMV (91.6%), HHV-6 (50.9%), RSV (37.4%), EBV (35.5%), HBoV (28.2%), HHV-7 (18.3%) and rhinovirus (17.2%) were the most commonly detected viruses. Of 81 non- CAP samples tested, CMV (63%), RSV (49.4%), HHV-6 (42%), EBV (24.7%), HHV-7 (13.6%) and HBoV (8.6%) were the dominant viruses detected. The prevalence of several viral agents (rhinovirus, bocavirus, adenovirus and CMV) among IS samples of CAP were significantly higher than that of non-CAP control group. We also found the prevalence of RSV coinfection with HHVs was also higher among CAP group than that of non-CAP control. CONCLUSIONS: With sensitive molecular detection techniques and IS samples, high rates of viral identification were achieved in infants and young children with respiratory illness in a rural area of China. The clinical significance of rhinovirus, bocavirus, adenovirus and HHV (especially CMV) infections should receive greater attention in future treatment and prevention studies of CAP in infants and children.

Aims To investigate the capability, properties, and molecular mechanism of inulin fermentation by lactic acid bacteria (LAB) from Sichuan pickle. Methods and results A total of 79 LAB strains were purified from 30 aged Sichuan pickle brine samples, and only 21 Lactiplantibacillus plantarum strains (26.58%, 21/79) derived from 15 samples grew well through utilizing inulin as a carbon source. The fermentation tests through using long-chain inulin (lc-inulin) as a carbon source showed that only 6 L. plantarum strains grew well, while other 15 strains could only utilize short-chain oligofructose (FOS), and thin-layer chromatography analysis evidenced a strain specificity of inulin consumption patterns. Lactiplantibacillus plantarum YT041 is a vigorous inulin fermenter, and whole genome sequencing data revealed that sacPTS1 and fosRABCDXE operons might be associated with the fermentation of FOS and lc-inulin, respectively. Conclusions The phenotype of inulin consumption is commonly present in LAB from Sichuan pickle, which is strain-specific and largely depends on their specific ecological niche and degree of polymerization.
