1. Radiometric hazard assessment of soil and water samples adjacent to Bangladesh's first nuclear power plant before commissioning: Insights into human health and environmental radiological dynamics

Abstract

The current study provides a comprehensive examination of both natural and anthropogenic activity concentrations found in soil and surface water samples near the Rooppur nuclear power plant (RNPP), the first of its kind under construction in Bangladesh. The investigation covers a range of activity concentrations including 226Ra, 232Th, and 40K in fifty soil and fifty water samples, revealing values ranging from 18 to 38, 18–51, and 310–560 Bqkg−1 for soil, and 2.1–6.1, 2.1–5.5, and 67–115 Bqkg−1 for water, respectively. Although outdoor absorbed dose rate, outdoor effective dose, and gamma representative level index values in some soil samples exceed global averages, other radiological hazard parameters such as radium equivalent activity, annual effective dose, external and internal hazard indices, and lifetime carcinogenic risk for both soil and water samples consistently remain below safety thresholds established by international organizations. These findings indicate that the soil and water samples collected from the vicinity of the RNPP do not pose significant radiological hazards to the nearby populations. This study's comprehensive dataset is anticipated to play a crucial role in facilitating the identification and evaluation of any changes in environmental radioactivity, thereby assisting in the effective management and regulation of nuclear power plant activities in the years to come.

2. Hydroxymethylfurfural Content and Sugar Profile of Honey Available in Bangladesh Using Validated HPLC-PDA and HPLC-RID

Abstract

Background: Honey has a lot of reputation because of its supposed medicinal properties. In this study, Hydroxymethylfurfural (HMF), sugars, and Fructose/Glucose ratio of honey in Bangladesh were assessed for adulteration and authenticity evaluation. Methods: Seventy honey samples collected from different districts of Bangladesh were analyzed by High Performance Liquid Chromatography (HPLC) for HMF content and sugar profile. The samples were prepared by using Carrez I and Carrez II prior to injecting into HPLC. The samples were then filtered through syringe filter and taken in 1.5 ml vial for injecting into the HPLC system. Results: HMF values were ranging from 1.41 mg/kg to 2,063.90 mg/kg. The Limit of Detection (LOD) and Limit of Quantification (LOQ) was found 0.10 mg/kg and 0.33 mg/kg with R2=0.9994. The average values of fructose, glucose, and sucrose were in the range of 14.75-52.44%, 8.19-42.63%, and 0.10-21.12%, respectively. From validation parameters, LOD values for fructose, glucose, and sucrose were 0.003, 0.008, and 0.004%, respectively; and LOQ values were 0.01, 0.028, and 0.015%, respectively with an excellent linearity with R2 for fructose=1.0, glucose=0.9999, and sucrose=1.0. Conclusion: Some samples had higher HMF content which may be due to the storage time was increased and improper processing with high temperature or adulteration by High Fructose Corn Syrup (HFCS), sugar cane syrup, rice syrups or rice molasses. Abstract

With the advent of Industry 4.0, several cutting-edge technologies such as cyber-physical systems, digital twins, IoT, robots, big data, cloud computation have emerged. However, how these technologies are interconnected or fused for collaborative and increased functionality is what elevates 4.0 to a grand scale. Among these fusions, the digital twin (DT) in robotics is relatively new but has unrivaled possibilities. In order to move forward with DT-integrated robotics research, a complete evaluation of the literature and the creation of a framework are now required. Given the importance of this research, the paper seeks to explore the trends of DT incorporated robotics in both high and low research saturated robotic domains in order to discover the gap, rising and dying trends, potential scopes, challenges, and viable solutions. Finally, considering the findings, the study proposes a framework based on a hypothesis for the future paradigm of DT incorporated robotics.

In this paper, a solution procedure is proposed to solve Fully Fuzzy Linear Fractional Programming (FFLFP) problem where all the variables and parameters are triangular fuzzy numbers. Here, FFLFP problem transformed into an equivalent Multi- Objective Linear Fractional Programming (MOLFP) problem. Then MOLFP converted into an equivalent multi objective linear programming problem by using mathematical programming approach. The proposed solution illustrated through numerical examples and compared with existing methods.

Abstract

Insulin-like growth factor binding protein-5 (IGFBP-5) is a key molecule in mammary gland development, which facilitates the removal of mammary epithelial cells (MECs) by apoptosis that takes place during remodeling of the mammary gland during involution. IGFBP-5 binds with IGFs for their bioavailability. IGFBP-5 has been reported to perform pleiotropic roles such as cellular apoptosis, proliferation and differentiation. To understand the role of IGFBP-5 during lactation and clinical mastitis, expression profiling of IGFBP-5 at the protein level was performed in both indigenous cows (Bos indicus) and buffaloes (Bubalus bubalis) belonging to two different breeds – Sahiwal cows and Murrah buffaloes. Reverse-transcriptase PCR (RT-PCR) of IGFBP-5 mRNA confirmed its expression in milk somatic cells and MECs of Sahiwal cows. ELISA was performed for quantitative measurement of IGFBP-5 concentrations in milk during different days (0, 50, 100, 150, 200, 250 and 300) of lactation, during the involution period and in animals exhibiting short lactation and clinical mastitis. The highest concentration of IGFBP-5 in milk was observed during the involution period followed by colostrum, late and early lactation, respectively, in both cattle and buffaloes. No significant difference in the concentration of IGFBP-5 was observed during the first 150 days of lactation between cows and buffaloes. However, higher concentration of IGFBP-5 was observed in cows during late lactation (200 to 300 days) in comparison with buffaloes. Abstract

The hadronization process of heavy hadrons with bottom and charm quarks, especially for baryons Λ c , in a dense QGP medium is largely not understood. We present within a coalescence plus fragmentation model the predictions for D 0 and Λ c spectra and the related baryon to meson ratios at RHIC and LHC. We will discuss how our model can predict values for Λ c / D 0 of the order of O(1), which is much larger than the expectations from fragmentation, and in agreement whit early data from STAR collaboration. Furthermore in the same scheme can be predicted a baryon to meson ratio Λ c / D 0 in pp collisions assuming that at the LHC top energies there can be the formation of QGP matter. The results show a considerable volume effects that significantly reduce the ratios, but still predict quite larger values with respect to fragmentation, in agreement with recent data from ALICE in pp collisions.

Abstract

Odonates were recorded from Kanha Tiger Reserve and its adjoining areas during January-December 2010. Thirty eight species were recorded belonging to seven families and 26 genera. Twelve species distribution is first time recorded from the reserve. With the addition of these newly recorded species with the previous records the species richness of the reserve increased up to 48 species, belonging to eight families. Among the collected Anisopterans Orthretum sabina sabina (Drury) was the most abundant species. A detailed annotated checklist of recorded odonates with the previous records is presented in the Table.

Abstract

Observations on the Indian Spiny-tailed Lizard Saara hardwickii (Gray, 1827) were undertaken in Tal Chhapar Wildlife Sanctuary, Rajasthan, India during the monsoons (July) following quadrat sampling that was time-constrained. The study revealed that the area is one of the preferable habitats for the species. A population analysis showed that the relative abundance of the subadults was higher, followed by juveniles and adults during the study period. The beginning of activity of the lizards was found to vary over the study period depending on prevailing weather conditions. The activity pattern was bimodal, except across rain events. The study revealed two important ecological findings about these lizards; complete sealing of burrow during rains which differed from partial sealing on normal days and complete diurnal cycle of body colour changes during the monsoon. Feeding was the predominant activity of this lizard followed by basking, resting and chasing each other. The adult lizards were found to be strictly herbivorous, in spite of an abundance of insects available in the area during the period. Subadults and juveniles were found to eat both plant parts, as well as insects. Microhabitat use such as inside grass clumps was found to be higher followed by barren ground, under shade and on stones.

10. In silico Allergenicity Study of Insect resistant genetically Modified Rice (Oryza sativa L.) for assessment of biosafety

Abstract

India is one of the world's largest producers of rice (Oryza sativa), accounting for 20% of all world rice production. However, lepidopteran pests severely impact the harvest of rice, which leads to environmental pollution and increase production cost. Alternatively, genetic engineering methods may be used to prevent rice pests and increase production of rice in a safe and environmentally friendly manner. Bacillus thuringiensis (Bt) genes have been widely used to generate genetically modified (GM) crops because the expressed cry1Ab protein confers resistance to lepidopteron pests. The proteins expressed by these genes may lead to food safety problems. Thus, safety evaluations are necessary prior to commercialization. Bioinformatics analysis for allergenicity assessment of cry1Ab protein is performed using different allergen databases viz. FARRP SDAP, Allergome, and Algpred to identify any potential sequence matches to allergen proteins that might indicate allergenic cross-reactivity with the query sequence. A full FASTA search was performed to identify highly similar proteins. However; the full length search cannot identify discontinuous or conformational epitopes that depend upon the tertiary structure of the protein.So every possible contiguous 80-amino acid sequence of each query protein was searched for determining the similarity. The proteins sequence can be searched using FASTA/BLAST for broad homology to known allergens to identify any short sequence that might represent an allergenic epitope. The domains in the Cry protein sequences were searched using Interproscan for potential similarity at the domain level. The results showed neither significant alignment nor similarity of cry1Ab protein at full sequence, domain, and epitope level with any of the known allergen proteins in the full sequence matching. Matching the 80 amino acid and matching of 8 amino acids showed no similarity to determine the epitope potential. From literature survey, it is observed that no allergic problem is reported in animals.The results of allergenicity study suggests that Bt rice is found to be safe for commercial release.

Abstract

In this study, 5% Nd3+ doped Cerium oxide (Ce0.95Nd0.05O2) was prepared by a modified solid-state reaction method. Crystallite size and lattice strain were determined from the Williamson-Hall (W-H) plot. The four distinct peaks in the photoluminescence (PL) spectra in the visible region signify the structural defects within the crystal lattice. Room temperature ferromagnetism (RTFM) was observed from the hysteresis loop, and the mathematical fitting of the hysteresis loop suggests ferromagnetic dominance (∼83%) in the magnetic response. The virgin M-H loop was fitted by the ferromagnetic Brillouin function, which suggests possible domain formation in the sample. The in vitro antibacterial activity of 5% Nd3+doped CeO2 was evaluated against two clinically isolated multidrug-resistant pathogenic bacterial strains, namely MLD 2 (Escherichia coli) and MLD 4 (Staphylococcus aureus), utilizing the minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), disc agar diffusion (DAD) assays, etc. Notably, compared to the MLD 4 bacterial strain, the synthesized material revealed comparable better antibacterial results against the Gram-negative MLD 2 and in comparison, to the MLD 2 bacterial strain, a relatively higher concentration of the sample is required to completely eradicate the bacterial population of MLD 4. This implies that Ce0.95Nd0.05O2 can destroy the growth of the two tested bacterial strains. In the context of cytotoxicity, up to a certain concentration, the synthesized particle demonstrated no appreciable toxicity towards healthy PBMCs.

Abstract

Background: Assessments of age-specifc mortality and life expectancy have been done by the UN Population Division, Department of Economics and Social Afairs (UNPOP), the United States Census Bureau, WHO, and as part of previous iterations of the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD). Previous iterations of the GBD used population estimates from UNPOP, which were not derived in a way that was internally consistent with the estimates of the numbers of deaths in the GBD. The present iteration of the GBD, GBD 2017, improves on previous assessments and provides timely estimates of the mortality experience of populations globally. Methods: The GBD uses all available data to produce estimates of mortality rates between 1950 and 2017 for 23 age groups, both sexes, and 918 locations, including 195 countries and territories and subnational locations for 16 countries. Data used include vital registration systems, sample registration systems, household surveys (complete birth histories, summary birth histories, sibling histories), censuses (summary birth histories, household deaths), and Demographic Surveillance Sites. In total, this analysis used 8259 data sources. Estimates of the probability of death between birth and the age of 5 years and between ages 15 and 60 years are generated and then input into a model life table system to produce complete life tables for all locations and years. Fatal discontinuities and mortality due to HIV/AIDS are analysed separately and then incorporated into the estimation. We analyse the relationship between age-specifc mortality and development status using the Socio-demographic Index, a composite measure based on fertility under the age of 25 years, education, and income. There are four main methodological improvements in GBD 2017 compared with GBD 2016: 622 additional data sources have been incorporated; new estimates of population, generated by the GBD study, are used; statistical methods used in diferent components of the analysis have been further standardised and improved; and the analysis has been extended backwards in time by two decades to start in 1950. Findings: Globally, 18·7% (95% uncertainty interval 18·4-19·0) of deaths were registered in 1950 and that proportion has been steadily increasing since, with 58·8% (58·2-59·3) of all deaths being registered in 2015. At the global level, between 1950 and 2017, life expectancy increased from 48·1 years (46·5-49·6) to 70·5 years (70·1-70·8) for men and from 52·9 years (51·7-54·0) to 75·6 years (75·3-75·9) for women. Despite this overall progress, there remains substantial variation in life expectancy at birth in 2017, which ranges from 49·1 years (46·5-51·7) for men in the Central African Republic to 87·6 years (86·9-88·1) among women in Singapore. The greatest progress across age groups was for children younger than 5 years; under-5 mortality dropped from 216·0 deaths (196·3-238·1) per 1000 livebirths in 1950 to 38·9 deaths (35·6-42·83) per 1000 livebirths in 2017, with huge reductions across countries. Nevertheless, there were still 5·4 million (5·2-5·6) deaths among children younger than 5 years in the world in 2017. Progress has been less pronounced and more variable for adults, especially for adult males, who had stagnant or increasing mortality rates in several countries. The gap between male and female life expectancy between 1950 and 2017, while relatively stable at the global level, shows distinctive patterns across super-regions and has consistently been the largest in central Europe, eastern Europe, and central Asia, and smallest in south Asia. Performance was also variable across countries and time in observed mortality rates compared with those expected on the basis of development. Interpretation: This analysis of age-sex-specifc mortality shows that there are remarkably complex patterns in population mortality across countries. The fndings of this study highlight global successes, such as the large decline in under-5 mortality, which refects signifcant local, national, and global commitment and investment over several decades. However, they also bring attention to mortality patterns that are a cause for concern, particularly among adult men and, to a lesser extent, women, whose mortality rates have stagnated in many countries over the time period of this study, and in some cases are increasing. © 2018 The Author(s).

Abstract

Background: Efforts to establish the 2015 baseline and monitor early implementation of the UN Sustainable Development Goals (SDGs) highlight both great potential for and threats to improving health by 2030. To fully deliver on the SDG aim of “leaving no one behind”, it is increasingly important to examine the health-related SDGs beyond national-level estimates. As part of the Global Burden of Diseases, Injuries, and Risk Factors Study 2017 (GBD 2017), we measured progress on 41 of 52 health-related SDG indicators and estimated the health-related SDG index for 195 countries and territories for the period 1990–2017, projected indicators to 2030, and analysed global attainment. Methods: We measured progress on 41 health-related SDG indicators from 1990 to 2017, an increase of four indicators since GBD 2016 (new indicators were health worker density, sexual violence by non-intimate partners, population census status, and prevalence of physical and sexual violence [reported separately]). We also improved the measurement of several previously reported indicators. We constructed national-level estimates and, for a subset of health-related SDGs, examined indicator-level differences by sex and Socio-demographic Index (SDI) quintile. We also did subnational assessments of performance for selected countries. To construct the health-related SDG index, we transformed the value for each indicator on a scale of 0–100, with 0 as the 2·5th percentile and 100 as the 97·5th percentile of 1000 draws calculated from 1990 to 2030, and took the geometric mean of the scaled indicators by target. To generate projections through 2030, we used a forecasting framework that drew estimates from the broader GBD study and used weighted averages of indicator-specific and country-specific annualised rates of change from 1990 to 2017 to inform future estimates. We assessed attainment of indicators with defined targets in two ways: first, using mean values projected for 2030, and then using the probability of attainment in 2030 calculated from 1000 draws. We also did a global attainment analysis of the feasibility of attaining SDG targets on the basis of past trends. Using 2015 global averages of indicators with defined SDG targets, we calculated the global annualised rates of change required from 2015 to 2030 to meet these targets, and then identified in what percentiles the required global annualised rates of change fell in the distribution of country-level rates of change from 1990 to 2015. We took the mean of these global percentile values across indicators and applied the past rate of change at this mean global percentile to all health-related SDG indicators, irrespective of target definition, to estimate the equivalent 2030 global average value and percentage change from 2015 to 2030 for each indicator. Findings: The global median health-related SDG index in 2017 was 59·4 (IQR 35·4–67·3), ranging from a low of 11·6 (95% uncertainty interval 9·6–14·0) to a high of 84·9 (83·1–86·7). SDG index values in countries assessed at the subnational level varied substantially, particularly in China and India, although scores in Japan and the UK were more homogeneous. Indicators also varied by SDI quintile and sex, with males having worse outcomes than females for non-communicable disease (NCD) mortality, alcohol use, and smoking, among others. Most countries were projected to have a higher health-related SDG index in 2030 than in 2017, while country-level probabilities of attainment by 2030 varied widely by indicator. Under-5 mortality, neonatal mortality, maternal mortality ratio, and malaria indicators had the most countries with at least 95% probability of target attainment. Other indicators, including NCD mortality and suicide mortality, had no countries projected to meet corresponding SDG targets on the basis of projected mean values for 2030 but showed some probability of attainment by 2030. For some indicators, including child malnutrition, several infectious diseases, and most violence measures, the annualised rates of change required to meet SDG targets far exceeded the pace of progress achieved by any country in the recent past. We found that applying the mean global annualised rate of change to indicators without defined targets would equate to about 19% and 22% reductions in global smoking and alcohol consumption, respectively; a 47% decline in adolescent birth rates; and a more than 85% increase in health worker density per 1000 population by 2030. Interpretation: The GBD study offers a unique, robust platform for monitoring the health-related SDGs across demographic and geographic dimensions. Our findings underscore the importance of increased collection and analysis of disaggregated data and highlight where more deliberate design or targeting of interventions could accelerate progress in attaining the SDGs. Current projections show that many health-related SDG indicators, NCDs, NCD-related risks, and violence-related indicators will require a concerted shift away from what might have driven past gains—curative interventions in the case of NCDs—towards multisectoral, prevention-oriented policy action and investments to achieve SDG aims. Notably, several targets, if they are to be met by 2030, demand a pace of progress that no country has achieved in the recent past. The future is fundamentally uncertain, and no model can fully predict what breakthroughs or events might alter the course of the SDGs. What is clear is that our actions—or inaction—today will ultimately dictate how close the world, collectively, can get to leaving no one behind by 2030. Microwave popping characteristics of a particular variety of paddy were studied using a domestic microwave oven. The experiments were carried out at 4 levels of moisture content (around 13%, 14%, 15% and 16% wb), 3 levels of power (600 W, 850 W and 1000 W) and 5 levels of heating time (40 s, 60 s, 80 s, 100 s and 120 s). A general factorial experiment design was followed, and effect of different treatment combinations on popping percentage and expansion ratio of the paddy was evaluated. The maximum popping percentage of 63.47% was obtained at a moisture content of 14.15% and energy level of 80 kJ (1000 W and 80 s) while the maximum expansion ratio of 4.42 was obtained at 14.94% moisture content and energy level of 68 kJ (850 W and 80 s). Optimum values of microwave power, time of heating and moisture content of paddy were achieved at 1000 W, 80 s and 15%, respectively, corresponding to popping percentage and expansion ratio of 58.73 and 3.58.

Both heating and drying of high moisture paddy using vibrating platform coupled with infrared heating source dryer give uniform but quick drying by exposing each and every grain uniformly to the IR source. A laboratory apparatus capable of modulating both frequency and amplitude of vibration had been developed to study vibration aided drying characteristics of high moisture paddy. The effective moisture diffusivity of the grain during drying under this mode has been evaluated. The drying characteristics of high moisture paddy was studied under different vibrating conditions for five levels of radiation intensity (1509, 2520, 3510, 4520 and 5514 W/m(2)) and four levels of grain bed depths (single kernel thickness of 3, 6, 12 and 25 mm). The optimum frequency and amplitude of the vibrations were found to be in the range of 20-22 Hz and 8-9 mm for all the three grain bed depths of paddy (25, 12 and 6 mm) for obtaining nearly complete and uniform mixing (mixing index congruent to 1.0) [Das, I., Das, S.K., Bal, S., 2003a. Abstract. Class IV Emphysematous pyelonephritis (EPN) is a life-threatening infection of the kidney, and there is sparse data on immediate and long-term outcomes of medical management. The study aimed to review the clinical presentation, diagnosis, immediate and long-term outcomes of medical management, and outcome of patients presenting with class IV EPN Methods. This retrospective study was conducted in a tertiary care center in Dharwad, India. The study included class IV EPN patients admitted to the hospital between Jan 2012 to Dec 2019. The factors studied were demographics, comorbidities, radiological investigations, clinical presentation, urine, blood and pus culture, and sensitivity and treatment history. Immediate and long-term outcomes of medical management were determined. Descriptive analysis of the data was conducted. Continuous variables were presented as mean ± standard deviation and discrete variables in terms of number (%). Results. Twenty-one patients with class IV EPN, with a mean age of 55.7 ±13.5 years, were analyzed. The majority were males (M: F 15:6). The patients were followed up for 32.70±21.12 months. Fever, flank or abdominal pain, and acute kidney injury were observed in all 21 patients, while nausea and/or vomiting and dysuria were seen in 16 (76.2%) patients. E. Coli was the most common pathogen found. Double J stenting was done in 20 (95.2%) patients; Ultrasonography guided aspiration 8(38.1%) and percutaneous drainage in 5 (23.8%) patients. One (4.8%) patient died during the first episode and 3(19%) others due to recurrent infection and end-stage renal disease. Conclusions. Class IV Emphysematous pyelonephritis can be managed successfully by minimally invasive modalities. However, long-term morbidity and mortality risks are still negative.

AbstractThe present communication evaluates the antigen B (AgB) family of bubaline isolates of Echinococcus granulosuswith respect to their conformational propensity and also discusses the stretches of agretope. AgB, which is abundantly present in hydatid cyst fluid, is encoded by a gene family, AgB1?AgB5. Hydatidosis is of zoonotic and economic importance in India. Buffaloes serve as the intermediate host. However, to date the AgB family has not been fully analysed. During the present study two different primers used for amplification of AgB1revealed homology to Echinococcus canadensis(G8) as well as E. granulosus sensu stricto(G1/G2). The sequence of AgB3 is homologous to that of the well-defined species, Echinococcus ortleppi(G5), and the predicted amino acid sequence of AgB4 is homologous to bovine isolates identified earlier. ?- and ?-amphipathic structures were recorded in all the antigens designated as T-cell receptor sites. The antigenic index of different stretches correlated with hydrophilicity because the hydrophobic residues are not accessible to the cells. In this study, we investigated the binding propensity of AgB to MHC II in order to determine stretches of agretope. Agretopes began with four hydrophilic residues. Two to three additional hydrophilic residues were present in the internal motif. This comparison of AgB and its family of bubaline isolates, with respect to their sequence information, ?- and ?- amphipathic regions, antigenic index and stretches of agretope is the first such report from India. Abstract

The present study was conducted on 412 crossbred (Jersey X Deshi) cows sired by 24 bulls, maintained over the period from 2005 to 2014. The economic traits studied were age at first calving (AFC; months), calving interval (CI; days), dry period (DP; days) and service period (SP; days). The averages for AFC, CI, DP and SP were 34.46±0.22 months, 505.95±3.24, 224.20±3.31 and 228.26±3.21 days, respectively. Effect of management system was significant for all the economic traits (AFC, CI and DP) except SP. It may be inferred that rearing crossbred Jersey cows under intensive system of management may be advocated to utilize the maximum genetic potential of crossbred cattle for all the traits. Semi-intensive system may help moderate exploration of the traits. But exclusively pasture dependent management should be avoided for rearing crossbred Jersey cows. Heritability estimates for AFC, CI and DP were found to be 0.462 ±0.179; 0.062 ±0.088; 0.270 ±0.138, respectively. Genetic correlations of AFC with CI and SP were high and positive. For maximum genetic improvement and their correlated response, it is necessary to include all the economic traits with differential weightage for achieving accuracy in selection programs.

Abstract

Recent advances in the area of genomics have led to the development of high throughput genotyping platforms that have immensely contributed to molecular breeding programs. Custom-designed single nucleotide polymorphism (SNP) arrays provide an efficient, cost effective, high throughput genotyping tool for QTL/gene mapping, variety identification, marker-assisted selection, etc. In the current study, two interspecific libraries of Chromosome Segment Substitution Lines (CSSLs) were evaluated under both drought and control conditions to identify lines with superior yield under drought. The CSSL libraries consisted of 48 BC4F3 lines derived from O. sativa cv. Curinga (tropical japonica) x O. rufipogon, and 32 BC4F3 lines derived from O. sativa cv. Curinga (tropical japonica) x O. meridionalis. The phenotypic screening of these 80 CSSLs led to the identification of three lines, MER-20, RUF-16, and RUF-44, that yielded well under drought stress. This line was backcrossed with popular rice variety of India, Swarna-Sub1 to introgress wild chromosome segments responsible for reproductive stage drought tolerance. During backcrossing, tracking of wild introgressions and monitoring of recurrent parent genome recovery was facilitated by the use of the Cornell 6K and 7K Infinium rice SNP arrays. The 6K and 7K SNP arrays assayed 5275 SNPs and 7099 SNPs, respectively, distributed across the 12 chromosomes. In our populations of (MER-20X Swarna sub1) BC2F1 lines, 1775 SNPs were polymorphic using the 6K array. The percentage of recurrent parent genome in these backcrossed lines ranged from 33-92% and the percentage of wild donor genome ranged from 8-67%. Using genotypic selection, 5% of plants were identified for further marker assisted backcrossing, based on the presence of the target donor (wild) segment and maximum recovery of recurrent parent background. In the next generation, BC3F1 lines were genotyped using the 7K SNP array, which identified 2521 polymorphic SNPs. In the BC3 generation, 60-95% of the recurrent parent genome was recovered and wild segments accounted for 5-40% of the genome. The best 5% progenies were again selected based on genotype, and selected BC3F2 individuals are now being evaluated for yield under drought stress. The use of the 6K and 7K SNP arrays improved the efficiency and accuracy of genotyping and greatly facilitated tracking of recurrent/donor genomes in backcross lines.
