Dysregulation of zinc and zinc transporters families has been associated with the genesis and progression of prostate cancer. The prostate epithelium utilizes two types of zinc transporters, the ZIP (Zrt-, Irt-related Protein) and the ZnTs (Zinc Transporter), to transport zinc from the blood plasma to the gland lumen. ZIP transporters uptake zinc from extracellular space and organelle lumen, while ZnT transporters release zinc outside the cells or to organelle lumen. In prostate cancer, a commonly observed low zinc concentration in prostate tissue has been correlated with downregulations of certain ZIPs (e.g., ZIP1, ZIP2, ZIP3, ZIP14) and upregulations of specific ZnTs (e.g., ZnT1, ZnT9, ZnT10). These alterations may enable cancer cells to adapt to toxic high zinc levels. While zinc supplementation has been suggested as a potential therapy for this type of cancer, studies have yielded inconsistent results because some trials have indicated that zinc supplementation could exacerbate cancer risk. The reason for this discrepancy remains unclear, but given the high molecular and genetic variability present in prostate tumors, it is plausible that some zinc transporters—comprising 14 ZIP and 10 ZnT members—could be dysregulated in others patterns that promote cancer. From this perspective, this review highlights novel dysregulation, such as ZIP-Up/ZnT-Down, observed in prostate cancer cell lines for ZIP4, ZIP8, ZnT2, ZnT4, ZnT5, etc. Additionally, an in silico analysis of an available microarray from mouse models of prostate cancer (Nkx3.1;Pten) predicts similar dysregulation pattern for ZIP4, ZIP8, and ZnT2, which appear in early stages of prostate cancer progression. Furthermore, similar dysregulation patterns are supported by an in silico analysis of RNA-seq data from human cancer tumors available in cBioPortal. The addition of carbonate minerals to seawater through an artificial ocean alkalinity enhancement (OAE) process increases the concentrations of hydroxide, bicarbonate, and carbonate ions. This leads to changes in the pH and the buffering capacity of the seawater. Consequently, OAE could have relevant effects on marine organisms and in the speciation and concentration of trace metals that are essential for their physiology. During September and October 2021, a mesocosm experiment was carried out in the coastal waters of Gran Canaria (Spain), consisting on the controlled variation of total alkalinity (TA). Different concentrations of carbonate salts (NaHCO 3 and Na 2 CO 3) previously homogenized were added to each mesocosm to achieve an alkalinity gradient between Δ 0 to Δ 2400 µ mol L -1. The lowest point of the gradient was 2400 µ mol kg -1 , being the natural alkalinity of the medium, and the highest point was 4800 µ mol kg -1. Iron (Fe) speciation was monitored during this experiment to analyse total dissolved iron (TdFe, unfiltered samples), dissolved iron (dFe, filtered through a 0.2 µ m pore size filter), soluble iron (sFe, filtered through a 0.02 µ m pore size filter), dissolved labile iron (dFe ′), iron-binding ligands (LFe), and their conditional stability constants (KFeL′) because of change due to OAE and the experimental conditions in each mesocosm. Observed iron concentrations were within the expected range for coastal waters, with no significant increases due to OAE. However, there were variations in Fe size fractionation during the experiment. This could potentially be due to chemical changes caused by OAE, but such an effect is masked by the stronger biological interactions. In terms of size fractionation, sFe was below 1.0 nmol L -1 , dFe concentrations were within 0.5–4.0 nmol L -1 , and TdFe was within 1.5–7.5 nmol L -1. Our results show that over 99 % of Fe was complexed, mainly by L1 and L2 ligands with kFe′L′ ranging between 10.92 ± 0.11 and 12.68 ± 0.32, with LFe ranging from 1.51 ± 0.18 to 12.3 ± 1.8 nmol L -1. Our data on iron size fractionation, concentration, and iron-binding ligands substantiate that the introduction of sodium salts in this mesocosm experiment did not modify iron dynamics. A natural plankton community from oligotrophic subtropical waters of the Atlantic near Gran Canaria, Spain, was subjected to varying degrees of ocean alkalinity enhancement (OAE) to assess the potential physiological effects, in the context of the application of ocean carbon dioxide removal (CDR) techniques. We employed 8.3 m3 mesocosms with a sediment trap attached to the bottom, creating a gradient in total alkalinity (TA). The lowest point on this gradient was 2400 μmol · L-1, which corresponded to the natural alkalinity of the environment, and the highest point was 4800 μmol · L-1. Over the course of the 33-day experiment, the plankton community exhibited two distinct phases. In phase-I (days 5–20), a notable decline in the photosynthetic efficiency (Fv/Fm) was observed. This change was accompanied by substantial reductions in the abundances of picoeukaryotes, small size nanoeukaryotes (nanoeukaryotes-1), and microplankton. The cell viability of picoeukaryotes, as indicated by fluorescein-di-acetate hydrolysis by cellular esterases (FDA- green fluorescence), slightly increased by the end of phase-I whilst the viability of nanoeukaryotes 1 and Synechococcus spp. did not change. Reactive oxygen species levels (ROS-green fluorescence) showed no significant changes for any of the functional groups. In contrast, in phase-II (days 21–33), a pronounced community response was observed. Increases in Fv/Fm in the intermediate OAE treatments of ∆900 to ∆1800 μmol · L-1 and in chlorophyll-a (Chl-a), chlorophyll-c2 (Chl-c2) , fucoxanthin and divinyl-Chl-a were attributed to the emergence of blooms of large size nanoeukaryotes (nanoeukaryotes-2) from the genera Chrysochromulina , as well as picoeukaryotes. Synechococcus spp. also flourished towards the end of this phase. In parallel, we observed a total 20 % significant change in the metaproteome of the phytoplankton community. This is considered a significant alteration in protein expression, having substantial impacts on cellular functions and the physiology of the organisms. Medium levels of ∆TA showed more upregulated and less downregulated proteins than higher ∆TA treatments. Under these conditions, cell viability significantly increased in pico and nanoeukaryotes-1 in intermediate alkalinity levels, while in Synechococcus spp., nanoeukaryotes-2 and microplankton remained stable. ROS levels did not significantly change in any functional group. The pigment ratios DD+DT : FUCO, and DD+DT : Chl-a increased in medium ∆TA treatments, supporting the idea of nutrient deficiency alleviation and the absence of physiological stress. Taken all data together, this study shows that there is minimal evidence indicating a harmful impact of high alkalinity on the plankton community. Objective. Dental caries remains a prevalent disease worldwide. Several epidemiological studies have shown that it affects the oral health of the pediatric population, and the Galapagos population in Ecuador is no exception. The aim of this study was to determine the prevalence of dental caries and its association, based on baseline information from the Galapagos Oral Health Study (ESSO-Gal), in children of the Galapagos Islands, Ecuador. Methods. A cross-sectional study was conducted involving 804 children aged 2–11 years. The prevalence of dental caries was assessed using the International Caries Detection and Assessment System (ICDAS II) criteria, while the presence of dental biofilm was assessed using the Silness–Löe index. Descriptive statistics, including frequency analysis and measures of central tendency and dispersion, were performed. Inferential statistical analyses were conducted to identify associations between variables. Statistical analyses were performed using the SPSS version 25.0 statistical program. Results. The caries prevalence rates based on ICDAS II codes 1–6, 1–2, and 3–6 were 98.01%, 96.9%, and 85%, respectively. A statistically significant difference was observed among the different islands regarding the cutoff point for ICDAS II codes 3–6 (p ≤ 0.001). Participants aged 6–11 years had the highest caries prevalence. Conclusions. The results show a high prevalence of dental caries among children in the Galapagos Islands, which increases with age. The coccolithophore Emiliania huxleyi shows a variety of responses to ocean acidification (OA) and to high-CO2 concentrations, but there is still controversy on differentiating between these two factors when using different strains and culture methods. A heavily calcified type A strain isolated from the Norwegian Sea was selected and batch cultured in order to understand whether acclimation to OA was mediated mainly by CO2 or H+, and how it impacted cell growth performance, calcification, and physiological stress management. Emiliania huxleyi responded differently to each acidification method. CO2-enriched aeration (1200 µatm, pH 7.62) induced a negative effect on the cells when compared to acidification caused by decreasing pH alone (pH 7.60). The growth rates of the coccolithophore were more negatively affected by high pCO2 than by low pH without CO2 enrichment with respect to the control (400 µatm, pH 8.1). High CO2 also affected cell viability and promoted the accumulation of reactive oxygen species (ROS), which was not observed under low pH. This suggests a possible metabolic imbalance induced by high CO2 alone. In contrast, the affinity for carbon uptake was negatively affected by both low pH and high CO2. Photochemistry was only marginally affected by either acidification method when analysed by PAM fluorometry. The POC and PIC cellular quotas and the PIC:POC ratio shifted along the different phases of the cultures; consequently, calcification did not follow the same pattern observed in cell stress and growth performance. Specifically, acidification by HCl addition caused a higher proportion of severely deformed coccoliths, than CO2 enrichment. The objective of this work is to analyse the density structure of the continental forearc in the northern segment of the 1960 Mw 9.6 Valdivia earthquake. Regional 2D and local 3D density models have been obtained from available gravity data in the area, complemented by new gravimetric stations. Models are constrained by independent geophysical and geological information and new TEM and MT soundings. The results show a segmentation of the continental wedge along and perpendicular to the margin, highlighting a high-density anomaly, below the onshore forearc basin, that limits the late Paleozoic–early Mesozoic metamorphic basement in the region where Chaitenia terrane has been proposed. A progressive landward shift of this anomaly correlates with the high slip patch of the giant 1960 Mw 9.6 Valdivia earthquake. Based on these results, we propose that the horizontal extension of the less rigid basement units conforming the marine wedge and Coastal Cordillera domain could modify the process of stress loading during the interseismic periods, and also that changes in position and extension of the late Paleozoic–early Mesozoic accretionary complex could be linked with the frictional properties of the interplate boundary. Summary: Ocean acidification (OA), a consequence of anthropogenic carbon dioxide (CO2) emissions, strongly impacts marine ecosystems. OA also influences iron (Fe) solubility, affecting biogeochemical and ecological processes. We investigated the interactive effects of CO2 and Fe availability on the metabolome response of a natural phytoplankton community. Using mesocosms we exposed phytoplankton to ambient (390 μatm) or future CO2 levels predicted for the year 2100 (900 μatm), combined with ambient (4.5 nM) or high (12 nM) dissolved iron (dFe). By integrating over the whole phytoplankton community, we assigned functional changes based on altered metabolite concentrations. Our study revealed the complexity of phytoplankton metabolism. Metabolic profiles showed three stages in response to treatments and phytoplankton dynamics. Metabolome changes were related to the plankton group contributing respective metabolites, explaining bloom decline and community succession. CO2 and Fe affected metabolic profiles. Most saccharides, fatty acids, amino acids and many sterols significantly correlated with the high dFe treatment at ambient pCO2. High CO2 lowered the abundance of many metabolites irrespective of Fe. However, sugar alcohols accumulated, indicating potential stress. We demonstrate that not only altered species composition but also changes in the metabolic landscape affecting the plankton community may change as a consequence of future high‐CO2 oceans. Rising concentrations of atmospheric carbon dioxide are causing ocean acidification and will influence marine processes and trace metal biogeochemistry. In June 2012, in the Raunefjord (Bergen, Norway), we performed a mesocosm experiment, comprised of a fully factorial design of ambient and elevated pCO2 and/or an addition of the siderophore desferrioxamine B (DFB). In addition, the macronutrient concentrations were manipulated to enhance a bloom of the coccolithophore Emiliania huxleyi. We report the changes in particulate trace metal concentrations during this experiment. Our results show that particulate Ti and Fe were dominated by lithogenic material, while particulate Cu, Co, Mn, Zn, Mo and Cd had a strong biogenic component. Furthermore, significant correlations were found between particulate concentrations of Cu, Co, Zn, Cd, Mn, Mo and P in seawater and phytoplankton biomass (µ gC L -1), supporting a significant influence of the bloom in the distribution of these particulate elements. The concentrations of these biogenic metals in the E. huxleyi bloom were ranked as follows: Zn < Cu ≈ Mn < Mo < Co < Cd. Changes in CO2 affected total particulate concentrations and biogenic metal ratios (Me : P) for some metals, while the addition of DFB only significantly affected the concentrations of some particulate metals (mol L -1). Variations in CO2 had the most clear and significant effect on particulate Fe concentrations, decreasing its concentration under high CO2. Indeed, high CO2 and/or DFB promoted the dissolution of particulate Fe, and the presence of this siderophore helped in maintaining high dissolved Fe. This shift between particulate and dissolved Fe concentrations in the presence of DFB, promoted a massive bloom of E. huxleyi in the treatments with ambient CO2. Furthermore, high CO2 decreased the Me : P ratios of Co, Zn and Mn while increasing the Cu : P ratios. These findings support theoretical predictions that the molar ratios of metal to phosphorous (Me : P ratios) of metals whose seawater dissolved speciation is dominated by free ions (e.g., Co, Zn and Mn) will likely decrease or stay constant under ocean acidification. In contrast, high CO2 is predicted to shift the speciation of dissolved metals associated with carbonates such as Cu, increasing their bioavailability and resulting in higher Me : P ratios. We studied the effects of elevated CO2 concentrations on cell growth, calcification, and spectral variation in the sensitivity of photosynthesis to inhibition by solar radiation in the globally important coccolithophore Emiliania huxleyi. Growth rates and chlorophyll a content per cell showed no significant differences between elevated (800 ppmv) and ambient (400 ppmv) CO2 conditions. However, the production of organic carbon and the cell quotas for both carbon and nitrogen, increased under elevated CO2 conditions, whilst particulate inorganic carbon production rates decreased under the same conditions. Biometric analyses of cells showed that coccoliths only presented significant differences due to treatments in the central area width. Most importantly, the size of the coccosphere decreased under elevated CO2 conditions. The susceptibility of photosynthesis to inhibition by ultraviolet radiation (UVR) was estimated using biological weighting functions (BWFs) and a model that predicts photosynthesis under photosynthetically active radiation and UVR exposures. BWF results demonstrated that the sensitivity of photosynthesis to UVR was not significantly different between E. huxleyi cells grown under elevated and present CO2 concentrations. We propose that the acclimation to elevated CO2 conditions involves a physiological mechanism of regulation and allocation of energy and metabolites in the cell, which is also responsible for altering the sensitivity to UVR. Rising concentrations of atmospheric carbon dioxide are causing ocean acidification and will influence marine processes and trace metal biogeochemistry. The importance of the combined impacts of elevated CO2 and changes in trace metal availability on marine plankton remain largely unknown. A mesocosm experiment was performed to study changes in particulate trace metal concentrations during a bloom dominated by the coccolithophore Emiliania huxleyi. We employed a full-factorial experimental design, comprising all combinations of ambient and elevated pCO2 and dissolved iron (dFe). Particulate metal concentrations (Fe, Cu, Zn, Co, Mn, Cd, Mo, Ti and Pb) were determined by high-resolution inductively coupled plasma mass spectrometry (HR-ICPMS). We examined biogenic and lithogenic sources of particulate metals, and their evolution during the experiment. Biogenic metal concentrations were estimated from bulk particle measurements by comparing phosphorus (P)-normalised quotas with published ratios, as well as concentrations of particulate trace metals in the presence and absence of an oxalate-EDTA wash. Our results demonstrate that particulate Ti and Fe concentrations were dominated by lithogenic material in the fjord. In contrast, particulate Cu, Co, Mn, Zn, Mo and Cd concentrations correlated with P concentrations and phytoplankton biomass, indicative of their strong biogenic character. Furthermore, ocean acidification changed the relative concentrations of particulate metals; a result mainly driven by the effects of ocean acidification on the growth of different phytoplankton phyla. This study demonstrates the utility and robustness of combining trace metal analyses of particles in a controlled mesocosm experiment with manipulations of CO2 and Fe concentrations using natural assemblages of marine phytoplankton. The assay of copper in zinc matrixes by reactor activation analysis presents significant difficulties, since the practicable capture reactions, Cu(n,γ)Cu and Cu(n,γ)Cu, are interfered by Zn(n,p)Cu and Zn(n,p)Cu reactions, as dominant activities. Taking into account this fact, the possibility of developing a method for copper determination in zinc compounds based on the measurement of Cu/Cu activity ratios is discussed. A method for manganese determination in iron concentrates by reactor activation analysis has been developed, on the basis of discounting the contribution of the Fe(n,p)Mn to the total Mn activity through the Mn/Mn activity ratio, employing the data on the nuclear constants involved. The possibility of using Fe as internal monitor, for the calculation of manganese concentration, is evaluated for different values of both the $$\phi_{\text{epi}} /\phi_{\text{th}}$$ ratios and the α parameter. Some of the most important effects of global change on coastal marine systems include increasing nutrient inputs and higher levels of ultraviolet radiation (UVR, 280–400 nm), which could affect primary producers, a key trophic link to the functioning of marine food webs. However, interactive effects of both factors on the phytoplankton community have not been assessed for the Mediterranean Sea. An in situ factorial experiment, with two levels of ultraviolet solar radiation (UVR+PAR vs. PAR) and nutrients (control vs. P-enriched), was performed to evaluate single and UVR×P effects on metabolic, enzymatic, stoichiometric and structural phytoplanktonic variables. While most phytoplankton variables were not affected by UVR, dissolved phosphatase (APAEX) and algal P content increased in the presence of UVR, which was interpreted as an acclimation mechanism of algae to oligotrophic marine waters. Synergistic UVR×P interactive effects were positive on photosynthetic variables (i.e., maximal electron transport rate, ETRmax), but negative on primary production and phytoplankton biomass because the pulse of P unmasked the inhibitory effect of UVR. This unmasking effect might be related to greater photodamage caused by an excess of electron flux after a P pulse (higher ETRmax) without an efficient release of carbon as the mechanism to dissipate the reducing power of photosynthetic electron transport. The effect of different ultraviolet radiation ( UVR) treatments combining PAR (P), UVA (A) and UVB (B) on the molecular physiology of Dunaliella tertiolecta was studied during 6 days to assess the response to chronic UVR exposure. UVR reduced cell growth but did not cause cell death, as shown by the absence of SYTOX Green labeling and cellular morphology. However, caspase-like enzymatic activities ( CLs), (regarded as cell death proteases), were active even though the cells were not dying. Maximal quantum yield of fluorescence ( Fv/ Fm) and photosynthetic electron transport rate ( ETR) dropped. Decreased nonphotochemical quenching ( NPQ) paralleled a drop in xanthophyll cycle de-epoxidation under UVB. Reactive oxygen species ( ROS) and D1 protein accumulation were inversely correlated. PAB exhibited elevated ROS production at earlier times. Once ROS decayed, D1 protein recovered two-fold compared with P and PA at later stages. Therefore, PsbA gene was still transcribed, suggesting ROS involvement in D1 recovery by its direct effect on mRNA-translation. We add evidence of an UVB-induced positive effect on the cells when P is present, providing photoprotection and resilience, by means of D1 repair. This allowed cells to survive. The effects of increased CO2 and irradiance on the physiological performance of the chlorophyte Dunaliella tertiolecta were studied at different PAR and UVR (UVA + UVB) irradiances, simulating the solar radiation at different depths, at present (390 ppmv, LC) and predicted CO2 levels for the year 2100 (1000 ppmv, HC). Elevated CO2 resulted in higher optimum and effective quantum yields (Fv/Fm and ΦPSII, respectively), electron transport rates (ETR) and specific growth rates (μ). Cell stress was alleviated in HC with respect to LC as evidenced by a decrease in reactive oxygen species (ROS) accumulation. DNA damage showed a 42-fold increase in cyclobutane-pyrimidine dimer (CPD) formation under the highest irradiance (1100 μmol quanta m−2 s−1) in LC with respect to the lowest irradiance (200 μmol quanta m−2 s−1). Photolyase (CII-PCD-PL) gene expression was upregulated under HC resulting in a drastic decrease in CPD accumulation to only 25% with respect to LC. Proliferating cell nuclear antigen (PCNA) accumulation was always higher in HC and the accumulation pattern indicated its involvement in repair or growth depending on the irradiance dose. The repressor of silencing (ROS1) was only marginally involved in the response, suggesting that photoreactivation was the most relevant mechanism to overcome UVR damage. When the chlorophyte alga Dunaliella tertiolecta Butcher is placed in darkness, a form of programmed cell death with many similarities to apoptosis is induced, including the induction of caspase-like proteases. Many uncertainties about the regulation and mediators that participate in the process remain. To examine the relationship between caspase-like activities and different apoptotic events (i.e., phosphatidylserine [PS] translocation), increases in membrane permeability and numbers of dead cells revealed by SYTOX-green staining, and the generation of reactive oxygen species (ROS), we used the broad-range caspase inhibitor Boc-D-FMK to block the activity of the whole class of caspase-like proteins simultaneously. In the presence of the inhibitor, ROS were not produced, and cells did not die. Loss of membrane asymmetry, indicated by external labeling of PS by annexin V, was apparent at midstages of light deprivation, although it did not conform to the typical pattern for PS exposure observed in metazoans or vascular plants, which occurs at early stages of the apoptotic event. Thus, we have evidence for a link between ROS and cell death involving caspase-like enzymes in an alga. The fact that caspase-like inhibitors prevent not only cell death, but also ROS and loss of cell membrane integrity and asymmetry, suggests that caspase-like proteases might have regulatory roles early in cell death, in addition to dismantling functions. When the chlorophyte alga Dunaliella tertiolecta is placed in darkness, a form of programmed cell death with many similarities to apoptosis (including the induction of caspase-like proteases) is induced. Many uncertainties about this process remain, two of which are whether it requires protein synthesis and whether there is potential viral involvement. In order to examine the relationship between the induction and/or activation of proteolytic activities and the cell death event, we used inhibitors of cytoplasmic protein synthesis (cycloheximide, CHX), organellar protein synthesis (chloramphenicol, CAP) and DNA synthesis (mitomycin C, MMC). Use of MMC also allowed us to examine whether temperate viruses were present in the D. tertiolecta isolate, since MMC treatment has been shown to induce lytic cycles. Addition of protein synthesis inhibitors (100?µM CHX or 1500?µM CAP applied singly or both inhibitors added together) did not prevent cell death from occurring when cultures were placed in the dark. There were no differences in caseinolytic activities visualized using zymograms, or in caspase 1, 3, 8 and 9 -like activities. Surprisingly, 100?µM MMC prevented the cell death event. Caseinolytic activities that appeared in darkness in controls did not appear in MMC-treated cultures, and caspase-like activities remained the same as in controls maintained in the light. The lack of effect of CHX and CAP suggests that the cell death programme we observe does not depend on protein synthesis, but rather on post-translational modification of pre-existing proteins. Results for MMC discount the involvement of temperate virus, but are difficult to interpret. MMC affects DNA synthesis and presumably transcription, but since inhibitors of translation did not prevent cell death, it is not clear why inhibiting transcription would.
