Electrochemical reactions' rate and their behavior at the electrode-electrolyte interface are better understood using electrochemical impedance spectroscopy (EIS). EIS is an analytical, electrochemical, and noninvasive technique, which provides both quantitative and qualitative analysis with higher resolution data over the traditional electrochemical techniques. It computes impedance of the concerned electrochemical system over a wide range of frequencies and measures the response (Current or voltage) with a phase shift. In this regard, this primer provides a guide for understanding the fundamentals of the EIS technique, and their theoretical background, followed by the discussion of electric circuit modeling, and deconvolution of impedance data using Nyquist and Bode plot. EIS data majorly describe the surface kinetics of the reaction, mass transport, and interpret the electronic properties of the electrode material. Thus, this review includes a detailed explanation of EIS in understanding the electrochemical behavior of electrode materials within supercapacitors and electrochemical sensing applications. ABSTRACT Vapour pressure deficit (VPD) is a primary determinant of stomatal behaviour and water balance in plants. With increasing global temperature, the accompanying rise in VPD is likely to have a significant impact on the performance of plant species in the future. However, the plasticity of stomatal response to VPD remains largely unexplored. This study examines the plasticity of whole plant stomatal conductance (gc) response to VPD in Pinus radiata plants grown under two temperatures and a water‐deficient treatment over a period of 3 months. The soil–stem water potential gradient (ΔΨ), gc and soil−stem hydraulic conductance (Ks‐s) were evaluated. The different treatment groups showed significant differences in maximum gc relating to differences in Ks‐s, however, gc dynamic response to VPD was very similar in all treatments such that ΔΨ was conserved once VPD increased above an average threshold of 0.64 kPa. The ability to robustly quantify water potential regulation in Pinus presents opportunities to explore variation in this globally important tree genus as well as providing a new approach to characterize the regulation of gas exchange in response to VPD. Background and Objective: Fermentation has a huge potential to improve the nutritional and functional properties by using the biological activity of the grain itself. The current study was performed to optimize the conditions for the fermentation of Quality Protein Maize (QPM) and maize further; the impact of fermentation treatment on the bio‐techno‐functional properties of QPM and maize flour was assessed. Findings: The technological properties, including oil absorption capacity, water solubility index, emulsion activity and stability, foaming capacity and stability, protein solubility, gel consistency, and least gelation concentration, were found to be increased with the fermentation treatment, while water absorption capacity, paste clarity, and swelling power observed a decline. The findings suggested elevated levels of all the bioactive constituents with the fermentation process. The modulations at the molecular level were confirmed by the scanning electron micrographs of fermented flour. Further, changes in the peaks of Fourier transform infrared spectra and the emergence of new peaks were also reported. Conclusions: The fermentation treatment altered the techno‐functional properties, bioactive constituents, macromolecular structure, and molecular interactions of QPM and maize flour. Significance and Novelty: Limited literature is available dedicated to the assessment of the nutritional, techno‐functional, and phytochemical components in QPM and maize as influenced by the fermentation process. Lung cancer is one of the most fatal malignancies, with the highest death rate (∼19%), and the NSCLC type accounts for ∼85% of lung cancers. In the search for new treatments, antimicrobial peptides have received much attention due to their propensity for selective destruction of cancer cells. In the current study, we evaluated the efficacy of the metastasis-specific tumour-homing-TMTP1 peptide against lung cancer using inhalable hybrid nano-assemblies of the PEG–PLGA copolymer as a carrier for pulmonary delivery which was assessed for aerodynamic and physicochemical properties, along with the peptide-release profile, physical stability, cellular uptake and biocompatibility, generation of reactive oxygen species, cell migration, autophagic flux, and apoptotic cell death in A549 lung cancer cells. Optimization of inhaled dose, lung retention, and efficacy studies was conducted to evaluate the formulation in an NNK (nicotine-derived nitrosamine ketone) induced tumour-bearing lung cancer murine model. After inhalation, the formulation with nano-scale physiognomies showed good lung deposition, retention, and metabolic stability. The inhalable nano-assemblies have shown enhanced generation of reactive oxygen species with increased autophagy flux and apoptotic cell death. Pre-clinical animal trials show substantial tumour regression by inhalable TMTP1-based nano-formulation with limited side effects. Our results on metastasis targeting and tumour-homing peptide TMTP1 demonstrate its effective tumour targeting and tumour-killing efficacy and provide a reference for the development of new therapeutics for NSCLC. Aims Endothelial cell (EC) dysfunction plays a key role in the initiation and progression of cardiovascular disease. However, studying these disorders in ECs from patients is challenging; hence, the use of human induced pluripotent stem cells (hiPSCs) and their in vitro differentiation into ECs represents a very promising approach. Still, the generation of hiPSC-derived ECs (hECs) remains demanding as a cocktail of growth factors and an intermediate purification step are required for hEC enrichment. Therefore, we probed the utility of a forward programming approach using transgenic hiPSC lines. Methods and results We have used the transgenic hiPSC line PGP1 ETV2 isoform 2 to explore the in vitro differentiation of hECs via doxycycline-dependent induction of the ETS variant transcription factor 2 (ETV2) and compared these with a standard differentiation protocol for hECs using non-transgenic control hiPSCs. The transgenic hECs were highly enriched without an intermediate purification step and expressed—as non-transgenic hECs and human umbilical vein endothelial cells—characteristic EC markers. The viability and yield of transgenic hECs were strongly improved by applying EC growth medium during differentiation. This protocol was successfully applied in two more transgenic hiPSC lines yielding reproducible results with low line-to-line variability. Transgenic hECs displayed typical functional properties, such as tube formation and LDL uptake, and a more mature phenotype than non-transgenic hECs. Transgenic hiPSCs preferentially differentiated into the arterial lineage; this was further enhanced by adding a high concentration of vascular endothelial growth factor to the medium. We also demonstrate that complexing lentivirus with magnetic nanoparticles and application of a magnetic field enables efficient transduction of transgenic hECs. Conclusion We have established a highly efficient, cost-effective, and reproducible differentiation protocol for the generation of functional hECs via forward programming.
