The outbreak of the Sars-Cov-2 pandemic has changed our perception of safety in shared and public living environments including healthcare facilities, shops, schools, and enterprises. The Internet of Things (IoT) represents a suitable solution for managing anti-pandemic smart devices (e.g., UV lights, smart cameras, etc.) and increasing citizens’ safety in public health crises. In this paper, we highlighted how IoT technologies can be exploited as non-pharmaceutical interventions presenting the SAFE PLACE project as an implementation of this concept. The project meant to design and develop an IoT system to ensure the safety and salubrity of shared environments. Advanced algorithms will be exploited to detect and classify humans’ presence, gathering, usage of personal protective equipment, and considering carefully the privacy protection of individuals.

Introduction: As a result of Industry 5.0’s technological advancements, collaborative robots (cobots) have emerged as pivotal enablers for refining manufacturing processes while re-focusing on humans. However, the successful integration of these cutting-edge tools hinges on a better understanding of human factors when interacting with such new technologies, eventually fostering workers’ trust and acceptance and promoting low-fatigue work. This study thus delves into the intricate dynamics of human-cobot interactions by adopting a human-centric view.Methods: With this intent, we targeted senior workers, who often contend with diminishing work capabilities, and we explored the nexus between various human factors and task outcomes during a joint assembly operation with a cobot on an ergonomic workstation. Exploiting a dual-task manipulation to increase the task demand, we measured performance, subjective perceptions, eye-tracking indices and cardiac activity during the task. Firstly, we provided an overview of the senior workers’ perceptions regarding their shared work with the cobot, by measuring technology acceptance, perceived wellbeing, work experience, and the estimated social impact of this technology in the industrial sector. Secondly, we asked whether the considered human factors varied significantly under dual-tasking, thus responding to a higher mental load while working alongside the cobot. Finally, we explored the predictive power of the collected measurements over the number of errors committed at the work task and the participants’ perceived workload.Results: The present findings demonstrated how senior workers exhibited strong acceptance and positive experiences with our advanced workstation and the cobot, even under higher mental strain. Besides, their task performance suffered increased errors and duration during dual-tasking, while the eye behavior partially reflected the increased mental demand. Some interesting outcomes were also gained about the predictive power of some of the collected indices over the number of errors committed at the assembly task, even though the same did not apply to predicting perceived workload levels.Discussion: Overall, the paper discusses possible applications of these results in the 5.0 manufacturing sector, emphasizing the importance of adopting a holistic human-centered approach to understand the human-cobot complex better.

Abstract This editorial summarises a longer note (95th Meeting of the EFSA Management Board, item 7. Available online: https://www.efsa.europa.eu/sites/default/files/event/mb95/item-07-doc1-partnership-with-mss-230622-d2.pdf) submitted to Management Board of the European Food Safety Authority (EFSA) for consideration prior to a discussion held in its 95th meeting, in June 2023. It presents the context and the framework of EFSA's current scientific cooperation activities and the vision of transitioning towards long‐term partnerships as the necessary evolution for the sustainability of food safety risk assessment in the EU. This new approach needs strategic alignment and political will to ensure the involvement of risk assessment organisations in the Member States. For that purpose, Management Board members can act as key advocates towards national risk managers and policy makers.

This paper introduces a design framework for mixed reality urban exploration (MRUE), based on a concrete implementation in a historical city. The framework integrates different modalities, such as virtual reality (VR), augmented reality (AR), and haptics-audio interfaces, as well as advanced features such as personalized recommendations, social exploration, and itinerary management. It permits to address a number of concerns regarding information overload, safety, and quality of the experience, which are not sufficiently tackled in traditional non-integrated approaches. This study presents an integrated mobile platform built on top of this framework and reflects on the lessons learned.

Deformed templating is the process by which self-replicating protein conformations with a given cross-β folding pattern can seed formation of an alternative self-replicating state with different cross-β folding pattern. In particular, uninfectious but propagative PrP amyloid can transform into a bona fide infectious conformer, PrPSc through deformed templating. The process can take many rounds of replication (if taking place in vitro) or even several passages of the evolving PrP conformers through successive brains if in vivo, through experimental transmission. In all cases, deformed templating involves a forced conversion in which there is a mismatch between the template and the substrate and/or the templating environment, typically a recombinant PrP amyloid, adept at converting recombinant PrP under denaturing conditions (e.g., presence of chaotropic agents), encountering a glycosylated, GPI-anchored PrPC substrate under physiological conversion conditions. Deformed templating is characterized by emergence of intermediate conformers that exhibit biochemical characteristics that are intermediate between those of the initial PrP amyloid and the final PrPSc conformers. Here, we took advantage of the recent elucidation of the structure of a PrP amyloid by cryo-EM and the availability of a physically plausible atomistic model of PrPSc that we have recently proposed. Using modeling and Molecular Dynamics (MD) approaches, we built a complete molecular modelization of deformed templating, including an atomistic model of a glycosylated intermediate conformer and a modified model of PrPSc. Among other unanticipated outcomes, our results show that fully glycosylated PrP can be stacked in-register, and how 4-rung β-solenoid (4RβS) PrP architectures can share key structural motifs with parallel-in register intermolecular sheet (PIRIBS) PrP amyloids. Our results shed light on the mechanisms of prion replication.

BackgroundVoice assistants allow users to control appliances and functions of a smart home by simply uttering a few words. Such systems hold the potential to significantly help users with motor and cognitive disabilities who currently depend on their caregiver even for basic needs (eg, opening a door). The research on voice assistants is mainly dedicated to able-bodied users, and studies evaluating the accessibility of such systems are still sparse and fail to account for the participants’ actual motor, linguistic, and cognitive abilities. ObjectiveThe aim of this work is to investigate whether cognitive and/or linguistic functions could predict user performance in operating an off-the-shelf voice assistant (Google Home). MethodsA group of users with disabilities (n=16) was invited to a living laboratory and asked to interact with the system. Besides collecting data on their performance and experience with the system, their cognitive and linguistic skills were assessed using standardized inventories. The identification of predictors (cognitive and/or linguistic) capable of accounting for an efficient interaction with the voice assistant was investigated by performing multiple linear regression models. The best model was identified by adopting a selection strategy based on the Akaike information criterion (AIC). ResultsFor users with disabilities, the effectiveness of interacting with a voice assistant is predicted by the Mini-Mental State Examination (MMSE) and the Robertson Dysarthria Profile (specifically, the ability to repeat sentences), as the best model shows (AIC=130.11). ConclusionsUsers with motor, linguistic, and cognitive impairments can effectively interact with voice assistants, given specific levels of residual cognitive and linguistic skills. More specifically, our paper advances practical indicators to predict the level of accessibility of speech-based interactive systems. Finally, accessibility design guidelines are introduced based on the performance results observed in users with disabilities.

The cellular prion protein (PrPC), a cell surface glycoprotein originally identified for its central role in prion diseases (also called transmissible spongiform encephalopathies), has recently been implicated in the pathogenesis of other neurodegenerative disorders, such as Alzheimer’s and Parkinson’s diseases, by acting as a toxicity-transducing receptor for different misfolded protein isoforms, or in some case by exerting neuroprotective effects. Interestingly, PrPC has also been reported to play unexpected functions outside the nervous system, for example by contributing to myelin homeostasis, regulating specific processes of the immune system and participating in various aspects of cancer progression. Collectively, these observations point to a much broader role for PrPC in physiological and disease processes than originally assumed. In this manuscript, we provide an overview of what is known about the role of PrPC beyond prion disorders and discuss the potential implications of targeting this protein in different diseases.

Prions are unusual protein assemblies that propagate their conformationally-encoded information in absence of nucleic acids. The first prion identified, the scrapie isoform (PrPSc) of the cellular prion protein (PrPC), caused epidemic and epizootic episodes [1]. Most aggregates of other misfolding-prone proteins are amyloids, often arranged in a Parallel-In-Register-β-Sheet (PIRIBS) [2] or β-solenoid conformations [3]. Similar folding models have also been proposed for PrPSc, although none of these have been confirmed experimentally. Recent cryo-electron microscopy (cryo-EM) and X-ray fiber-diffraction studies provided evidence that PrPSc is structured as a 4-rung β-solenoid (4RβS) [4, 5]. Here, we combined different experimental data and computational techniques to build the first physically-plausible, atomic resolution model of mouse PrPSc, based on the 4RβS architecture. The stability of this new PrPSc model, as assessed by Molecular Dynamics (MD) simulations, was found to be comparable to that of the prion forming domain of Het-s, a naturally-occurring β-solenoid. Importantly, the 4RβS arrangement allowed the first simulation of the sequence of events underlying PrPC conversion into PrPSc. This study provides the most updated, experimentally-driven and physically-coherent model of PrPSc, together with an unprecedented reconstruction of the mechanism underlying the self-catalytic propagation of prions.

In large acute medical wards treating heterogenous and complex patients intensity of patient care should be graduated according to clinical severity. We conducted a retrospective observational study on all unselected admissions (8838) to the internal medicine ward of the Santa Chiara Hospital of Trento from 2012 to 2017. During 2012 and 2013, a standard organizational model (SMC) was in place, while an organizational model for intensity of medical care (IMC) was introduced in 2014. In SMC, patient admission was performed according to bed availability only. In IMC, patients were allocated to three different ward settings (high, medium and post-acute care) based on the stratification of clinical instability. The National Early Warning Score (NEWS) was used for the stratification, together with the clinical judgment. The implementation of the IMC model led to a decrease of mortality and urgent transfers for clinical deterioration to Intensive Care Unit and to an increase of admissions from Intensive Care Unit and from regional spoke hospitals. Redesigning delivery processes based on IMC can play a pivotal role in improving patient outcomes and bed management.

Floods are an increasingly frequent phenomenon with catastrophic consequences for citizens and cities. Virtual reality (VR) has become a valuable training tool, simulating inaccessible or hazardous situations like emergencies and risk environments. This study evaluates a designed virtual training simulating a riverbank flood scenario. Beyond imparting factual knowledge, this environment aims to facilitate experiential learning by enabling users to repeatedly practice correct behaviors safely. We investigate the impact that different tutorials have on the ability of users to comprehend and retain the knowledge conveyed during the VR experience. To achieve this, three training experiences with different levels of contextualization and interaction tutorials were compared in a between-participant design. Data collection involved both self-reported measures (e.g., questionnaire) and objective measures (e.g., behavioral data). The results demonstrate the effectiveness of the virtual environment in acquiring knowledge of flood contents, particularly when the tutorial offers high levels of interaction, thereby reducing cognitive load. Furthermore, we observed that a lower level of contextualization in the tutorial phase enhances user efficiency in acquainting themselves with the virtual environment. This study emphasizes the potential of virtual training in augmenting preparedness, providing a valuable framework for the development of experiential learning tools in flood emergencies. Brain abscesses due to Listeria monocytogenes are rare and are a clinical challenge. We report the case of a 72-year old female patient under immunosuppressive treatment for Horton’s arteritis who was admitted with an initial diagnosis of ischemic stroke. Further workup revealed listerial abscesses as the cause of her neurological symptoms. She was treated with ampicillin plus gentamicin with an excellent outcome. In spite of the rarity of this entity, immunosuppresive drugs are the most important predisposing factors. These drugs are widely used and consequently there is an increased number of subjects at risk of listerial brain abscesses.

This study belongs to the ethnomethodological tradition of identifying the everyday practices accounting for the oiled machinery of social organization and applies this approach to understanding direction light usage. We observe a set of episodes videorecorded in North-East Italy in the urban traffic. We first unpack the meaning of direction light usage from a pragmatic perspective and then test our interpretation against the cases in our collection that seem to deviate from it. We argue that direction lights' usage works as an announcement to some road users and a request to a subset of them; in both cases, direction lights convey contextualized (indexical) coordinates about the vehicle's prospective trajectory. We then explain the cases in which signaling is omitted and draw some implications for traffic coordination and safety. [ABSTRACT FROM AUTHOR]

The present work considers the training in risk perception carried out with a motorcycle simulator, the Honda Rider Trainer (henceforth HRT), and investigates the effect of the social setting on the trainees’ performance. Through preliminary pilot studies a general procedure and three different social settings are defined: (a) a standard one (one trainee, one instructor); (b) standard with an audience of two schoolmates; and (c) two trainees and one instructor. The results show that in a school environment and with a careful procedure, the users’actions are aligned with the training purpose, and no gender difference emerges in the performance; also, the addition of a small audience, or a parallel training does not modify the trainees’ ability to cope with risks.
