Abstract

Hydrogel‐based microchannels with biologically similar morphologies and properties can provide excellent platforms for advanced tissue/organ formation in vitro. However, there are still many restrictions on channel morphology, material selection, tubing connections, etc. Here, a novel and versatile method is proposed that couples cononsolvency photopolymerization, which enables the incorporation of porous structures into hydrogels, with on‐chip microchannels formed by buckling of a thin film. This method provides a hydrogel‐based microchannel with improved permeability while maintaining its mechanical properties by incorporating a continuous porous structure into a synthetic polymer network with excellent mechanical properties. Furthermore, by culturing vascular endothelial cells into the microchannel, it is demonstrated that the microchannel works as a vessel‐on‐a‐chip platform that can be used for the evaluation of barrier function, fabrication of various channel shapes, and development of co‐culture systems. This method, which can be adapted to various swellable hydrogels, can provide platforms with properties and functions tailored to the tissue/organ and, thus, it will contribute to the creation of physiologically relevant biological models. 20. TERT Promoter Mutations Increase Tumor Aggressiveness by Altering TERT mRNA Splicing in Papillary Thyroid Carcinoma.

Abstract

Context Telomerase reverse transcriptase promoter (TERT- p) mutations, which upregulate TERT expression, are strongly associated with tumor aggressiveness and worse prognosis in papillary thyroid carcinomas (PTCs). TERT expression is also observed in a proportion of PTCs without TERT -p mutations, but such tumors show less aggressiveness and better prognosis than TERT- p mutation–positive tumors. Objective TERT has multiple splicing variants whose relationships with the TERT- p status and clinicopathological characteristics remain poorly understood. We examined the relationship between the TERT -p mutational status, the TERT splicing pattern, and clinicopathological features. Methods We investigated the expression of 2 major variants, α deletion (dA) and β deletion (dB), in a series of 207 PTCs operated on between November 2001 and March 2020 in Nagasaki University Hospital and Kuma Hospital. Results The TERT -p mutations were found in 33 cases, and among 174 mutation-negative cases, 24 showed TERT expression. All cases were classified into 3 groups: the TERT -p mutation–negative/expression–negative group (mut−/exp−), the TERT -p mutation–negative/expression–positive group (mut−/exp+), and the TERT -p mutation–positive group (mut+/exp+). The +A+B/dB ratio in mut+/exp+ was significantly higher than that in mut−/exp+ PTCs. Analysis with clinicopathological data revealed that +A+B expression was associated with higher PTC aggressiveness, whereas dB expression counteracted this effect. Functional in vitro study demonstrated that dB strongly inhibited cell growth, migration, and clonogenicity, suggesting its tumor-suppressive role. Conclusion These results provide evidence that the TERT -p mutations alter the expression of different TERT splice variants, which, in turn, associates with different tumor aggressiveness. 21. Prevalence of panoramic radiographs findings of cementum‐osseous dysplasia with calcification in the department of oral surgery at hospital: A retrospective study.

Abstract

Aim: Cemento‐osseous dysplasia (COD) is a benign, non‐neoplastic, fibro‐osseous lesion that is often asymptomatic and discovered incidentally on dental radiographs. Radiological examination, rather than biopsy, is recommended for diagnosis because of the risk of infection. The dentist working at a hospital sometimes requests a family dentist to follow up on the progress of COD discovered accidentally during panoramic radiography at a hospital. However, there are few reports on the prevalence of COD on panoramic radiographs obtained at the oral surgery department of a hospital; therefore, we investigated this in this study. Materials and methods: We examined the prevalence of COD with calcification (classification, presence of infection, and complications with simple bone cysts) in 1194 patients for whom panoramic radiographs were obtained. Results: Twelve patients (1.0%) had findings of COD with calcification: four cases of periapical COD (three females and one male), five cases of focal COD (four females and one male), and three cases of florid COD (three females and zero males). Infection was seen in one case of Florid COD and one case of Focal COD. A simple bone cyst was observed in one patient with Florid COD. Conclusion: COD with calcification was observed in a certain percentage of patients, and many were asymptomatic. Dental healthcare workers need to understand that COD carries a risk of infection and that there are diseases that need to be differentiated. 32. Preconception Stress and Pregnancy Serum Glucose Levels Among Women Attending a Fertility Center.

Abstract

Context The association between women's stress and pregnancy glucose levels remain unclear, specifically when considering the preconception period as a sensitive window of exposure. Objective We investigated whether preconception perceived stress was associated with glucose levels during pregnancy among women attending a fertility center (2004-2019). Methods Before conception, women completed a psychological stress survey using the short version of the validated Perceived Stress Scale 4 (PSS-4), and blood glucose was measured using a 50-gram glucose load test during late pregnancy as a part of screening for gestational diabetes. Linear and log-binomial regression models were used to assess associations of total PSS-4 scores with mean glucose levels and abnormal glucose levels (≥ 140 mg/dL), adjusting for age, body mass index, race, smoking, education, physical activity, primary infertility diagnosis, number of babies, and mode of conception. Results Psychological stress was positively associated with mean abnormal glucose levels. The adjusted marginal means (95% CI) of mean glucose levels for women in the first, second, and third tertiles of psychological stress were 115 (110, 119), 119 (115, 123), and 124 (119, 128), and mg/dL, respectively (P for trend =.007). Also, women in the second and third tertiles of psychological stress had 4% and 13% higher probabilities of having abnormal glucose compared with women in the first tertile of psychological stress (P trend =.01). Conclusion These results highlight the importance of considering preconception when evaluating the relationship between women's stress and pregnancy glucose levels. [ABSTRACT FROM AUTHOR]

Abstract

Remotely controllable photoresponsive hydrogel actuators are promising for applications in multiple fields. However, simple deformation mechanisms, which rely on the general swelling/deswelling, limit their performance and application. Herein, we report a displacement amplification mechanism based on the buckling deformation of photoresponsive hydrogel film. The on‐chip buckled architecture of the hydrogel enables actuation between a flat 2D shape and tubular 3D buckled shape with remarkable performances, including high deformation ratio (height ratio: ≈360%), tunable cycle motion frequency (0.1—1 Hz) and high cyclic stability. Moreover, localized buckling deformation, such as tube opening and closing, can be controlled in response to photostimulation. Inspired by these biomorphic shapes and motions, an intestine‐mimetic device and demonstrate segmentation with substance crushing and peristalsis motion with substance propelling were further fabricated. This study will provide a useful design principle for hydrogel actuators and shed light on diverse applications in soft robotics, dynamic microfluidics and organs‐on‐chips. [ABSTRACT FROM AUTHOR]

Abstract

Retroperitoneal and mediastinal emphysema after colon resection is extremely rare, especially in the absence of anastomotic leakage. The feasibility and safety of conservative treatment for this complication are unknown. We report a patient who underwent open sigmoid colon resection for colon cancer and developed retroperitoneal and mediastinal emphysema that was not caused by anastomotic leakage. Retroperitoneal and mediastinal emphysema occurred as a result of diverticular perforation. We were able to treat this patient successfully with conservative management. [ABSTRACT FROM AUTHOR]
